2.Hospital Infection of Inpatients: Investigation and Analysis of 34 Cases

OBJECTIVE To investigate the status of hospital acquired infection correctly,and to clarify the difficulty of dealing with it as well as to improve the survey and management level.METHODS Thirty-four cases of hospital acquired infection were diagnosed by cross-sectional survey,followed and then analyzed.RESULTS The incidence of hospital acquired infection was 7.6% in our hospital,the average time of hospitalization was 38.68 days which 3.98 times of the average hospitalization time of whole patients.The average hospitalization fee was 24 065.9 Yuan,4.5 times of the average fee of whole patients.CONCLUSIONS Patients susceptible to hospital acquired infection include old patients(≥60-year-old),basic illness≥3,and operated patients,the main risk factors of hospital acquired infection in our hospital include invasive cathelization,operation,combination using of more than 2 antibiotics and using of steroid.

Oxidative damage theory is currently one of the predominant theories on the mechanisms of aging. Previous research has shown that antioxidants can extend the lifespan in the model organism by scavenging free radicals,inducing the expression of stress related genes and hormesis. However, recent studies have suggested that these pharmaceuticals may cause serious side effects,such as promoting oxidation,increasing the risk of cancer,and destroying the metabolic balance. The low absorption and targeting property also limit the efficiency of most antioxidants. As a result ,the correlation between antioxidants and lifespan extension remains to be demonstrated. We reviewed the research progress in the field of lifespan extension by antioxidants in recent years and provided references for future research in related areas.
4.Effects of PM2.5 on Expression of Pigment Epithelium-derived Factor in Bronchial Epithelial Cells

Objective To explore the effect of particulate matter (PM) 2.5 on the expression of pigment epithelium-derived factor (PEDF) protein in bronchial epithelial cells (BEAS-2B). Methods Human BEAS-2B were subcultivated, followed by low, medium and high concentrations of PM2.5 (25μg/ml, 50μg/ml, 100μg/ml) stimulation for 24 hours. The expression of PEDF protein in supernatant was ana-lyzed by enzyme-linked immunosorbent assay (ELISA), and the expression in BEAS-2B cells was detected by Western blotting. Results Compared with the control group, the expression of PEDF protein in supernatant and BEAS-2B cells induced by PM2.5 (25 μg/ml) in-creased, but no significance was found (t=-0.730, t=-1.840, P>0.05), and the expression induced by PM2.5 with the concentrations of 50μg/ml and 100μg/ml significantly increased (t>5.798, P<0.05). Conclusion PM2.5 with the concentrations of 50μg/ml and 100μg/ml could increase the expression of PEDF protein in a concentration-dependent manner both in supernatant and BEAS-2B cells.
5.Determination of cichoric acid in Enchinacea purpurea and its preparation by HPLC

Object To determine the amount of cichoric acid in Enchinacea purpurea and its preparation. Methods By the use of HPLC, mobile phase: 400 mmol/L ammonium acetate methanol (95∶ 5), flow rate: 1 mL/min, detection wavelength: 330 nm. Results The mean recovery of cichoric acid was 99 41%, RSD=3 23% (n=5). Conclusion The method can be applied in the determination of cichoric acid in E. purpurea extract and its preparation with satisfactory results.
6.Preparation and purification of theanine reference substance by preparative HPLC

Object To establish an isolation method of theanine reference substance by preparative HPLC. Methods After raw material extracted by water, the water phase was extracted by chloroform saturated with water, then concentrated on a thermostat. After centrifuged, supernatant was isolated and purified by preparative HPLC. Fraction was frozen and dried by Flexi Drier. Crude product was rinsed by methanol. The purity of product was determined by analytical HPLC. Results The purity of product is higher than 98% and theanine yield from raw material by this method exceeds 60%. Conclusion The developed method is simple, rapid, and at low production cost. The product owns the quality of reference substance.
7.Clinical application of Depulpin inactivation agent in emergency treatment of acute pulpitis

21 8 patients with acute pulpitis were randomly divided into 2 groups.1 09 cases were treated by Depulpin inactivation agent (group DI)for emergency management,another 1 09 cases were treated by pulp drainage with phenol camphor cotton ball(group CP).The effective analgesia rate of DI and CP group was 95.4% and 69.7% respectively(P<0.001 ).
8.Langerhans cell histiocytosis of the thyroid

Langerhans cell histiocytosis is an uncommon disease, involvement of the thyroid by langerhans cell histiocytosis is rare. Two cases of Langerhans cell histiocytosis of the thyroid confirmed by pathology were reported in this paper. The main clinical feature was enlargement of the thyroids. One patient had hypothyroidism. Meanwhile, the lungs and pituitaries of the two patients were affected. Langerhans cell histiocytosis of the thyroid had no typical symptoms and specific laboratory examination, and might be clinically diagnosed as chronic lymphocytic thyroiditis, benign or malignant thyroid tumors. Thyroid fine needle aspiration cytology might be useful in confirming the diagnosis. The effective treatments include systemic chemotherapy, local radiation therapy and surgical excision. The chemotherapy is still the major technological approach. Most patients can relieve partially after receiving the systemic chemotherapy.
9.High Throughput Screening of Drug Multi Residues in Fish by Quadrupole-Time-of-Flight Mass Spectrometry

A high-throughput method for screening drug multi residues was developed by quadrupole-time-of-flight( Q-TOF) mass spectrometry together with QuEChERS sample preparation technique. It has been used for the determination of 57 drugs, such as tetracyclines ( TCs ) , sulfonamides ( SAs ) , quinolones ( QNs ) , triphenylmethanes (TPMs), estrogens (ETs), androgens (AGs) and glucocorticoids (GCs) in fish. The optimized pretreatment conditions were examined. The target compounds were extracted with acetonitrile under the condition of Na2 EDTA-Mcllvaine buffer, and the usage of clean-up reagents was 25 mg anhydrous magnesium sulfate, 12. 5 mg octadecylsilane and 6. 25 mg N-Propylethylenediamine sorbent for extracted solvent of each milliliter. The positive results acquired by this high-throughput method were confirmed by liquid chromatography-tandem mass spectrometry ( LC-MS/MS) . The detection limit for these 59 drugs was in the range of 0. 5-5. 3 μg/kg. The method is time-saving, convenient, effective and wide coverage. Its sensitivity can meet the requirement of the detection of drug residues in aquatic products.
10.Experimental study on meiotic spindles and chromosomes of mouse mature (M II) stage oocytes under laser scanning confocal microscopy.

Taking the mouse as a model, the experimental method of observing the morphology of meiotic spindles and chromosomes in mature oocytes were investigated in order to evaluate the effects of various interventions on the quality of oocytes accurately and rapidly. Laser scanning confocal microscope (LSCM) was used to examine the meiotic spindles and chromosomes by the technologies of optical section and three-dimensional (3D) image reconstruction. The results showed that the configurations of meiotic spindles and chromosomes could be observed clearly by LSCM. The normal rate of meiotic spindles and chromosomes was 82% and 86% respectively. It was concluded that the LSCM was a valid instrument to observe the meiotic spindles and chromosomes of mature oocytes and could be used as a valid method to evaluate the quality of M II oocytes.
