1.Comparison of the safety of two treatment methods of cesarean scar pregnancy of type Ⅱ and Ⅲ in menopause within 7 weeks

Objective To comparison of the safety of two treatment methods of cesarean scar pregnancy (CSP) of typeⅡ and Ⅲ in menopause within 7 weeks. Methods Totally 70 cases of CSP of typeⅡ and Ⅲ within 7 weeks of the last menstrual period, hospitalized within Peking University First Hospital from January 2009 to May 2016, had been retrospectively studied in two groups of different treatments. The methotrexate (MTX) treatment group included 37 cases receiving ultrasound-guided complete curettage of uterine cavity combined with MTX therapy, while the uterine artery embolization (UAE) group had 33 cases treated with UAE combined with MTX therapy and subsequent ultrasound-guided complete curettage of uterine cavity. The bleeding measurements during operation had been documented and compared for the study. Results The comparative difference of bleeding measurements between the MTX treatment group and the UAE group was insignificant from the statistical perspective (median: 5.0 vs 5.0 ml, P=0.716).The comparative difference of the duration (median: 2.0 vs 6.0 days, P<0.01) and expenses (median: 2832.1 vs 10147.1 yuan, P<0.01) for hospitalization between the MTX treatment group and the UAE group were significant from the statistical perspective. Conclusions The bleeding risks may not increase during the treatment of ultrasound-guided complete curettage of uterine cavity combined with MTX therapy for CSP patients of type Ⅱ and Ⅲwithin 7 weeks of the last menstrual period. Meanwhile, the UAE adverse effects and complications will be avoided, and the duration and expenses for hospitalization will be reduced.

A themostable intracellular β-galactosidase from a thermophilic Bacillus stearothermophilus was purified by a combination of (NH4)2SO4 fractionation, ion-exchange (DEAE-22)and gel filtration (Sephades G-75). The optimum temperature and pH of the enzyme acivity were 60Cand pH6.4 respectively. The β-galatosidase activity exhibited thermosttability at 50 C. The enzyme was significaantly activated by alkali and alkali-earth metal ions. The activity was inhibited by Zn2+ 、 Fe3+ 、 Cu2+Reducing agents enhanced β- galactosidase activity. Thiol-binding agents drastically decreased the enzyme activity. The enzyme was specific for β-D glycosidic linkages,and the identity of the aglycone moiety also influenced enzyme activity. At 55Cthe Km for O-nitrophenyl-β-D-galactosidase (ONPG)and lactose were 2. 63mmol/L and 4.39mmol/L, respectively,and Vmax for both substrates were 1.93 × 10-5mmol. min-1 mg-1protein6.54 ×105 mmol. min-1. mg-1protein,respectively. Objective To study the effect of fusion protein FADDDEL-GFP in the treatment of type 1 diabetes by murine islet cell transplantion.Methods After transfecting the recombinate FADDDEL-GFP into mouse insulinoma cells NIT-1,insulin level secreted by the cells was examined.Type 1 diabetes was induced by STZ as animal model.NIT-fg cells,the NIT-1 cells modified by FADDDEL-GFP,were transplanted into the diabetic mice.Then the effects of islet cell transplantation on diabetic mice were checked.Results GFP was expressed in NIT-1 cells transfected with FADDDEL-GFP.Diabetic mice reached normoglycemia and prolonged the life span,which was obviously different from the control group.Conclusion FADDDEL-GFP may enhance the capability of resisting allogeneic transplantation rejection.

Objective To investigate the effect of aspirin on the incidence of thromboembolism in hypertensive patients associated with paroxysmal atrial fibrillation.MethodsFour hundred hypertensive patients with paroxysmal atrial fibrillation (mean age 59 years) free of thromboembolic events were randomized assigned to receive aspirin (0.3 g/d) or placebo. Objective Toinvestigatetheeffectofchronicrecombinantgrowthhormone(rhGH)stimulationonhepaticSTAT5in healthy young mouse and its molecular mechanism .Methods 32 normal young mice were divided into 4 groups :PBS ,PBS+single rhGH ,chronic rhGH ,chronic rhGH+ single rhGH groups .The mice in the treatment groups(chronic rhGH and chronic rhGH +single rhGH )received intraperitoneal injection of rhGH at 1 μg/g body weight once daily for three weeks ;the mice in the control groups(PBS and PBS+single rhGH) were injected with sterile phosphate buffered saline(PBS ,0 .1 mol/L) .At next day 8:00 after 16 h of injection of last rhGH and PBS ,at 30 min before killing mouse ,PBS 0 .1 mol/L was injected in the PBS group and the chro-nic rhGH group ,while the single dose of rhGH 1 μg/g was injected in the PBS+single rhGH and the chronic rhGH+single rhGH group .Results Compared with the control mice(PBS group and PBS+single rhGH group) ,the body mass in the rhGH stimulation mice (chronic rhGH group and the chronic rhGH + single rhGH group) was significantly increased ;compared with the PBS group ,the base p-STAT5 level within the hepatic GH signal pathway in the chronic rhGH was significantly decreased ;the combi-ning capacity of hepatic STAT5 and DNA detected by EMSA in the chronic rhGH group was significantly lower than that in the PBS group ;in the mice injected by single dose of rhGH 1 μg/g at 30 min before killing mouse ,the hepatic p-STAT5 level in the chronic rhGH+single rhGH was significantly decreased compared with the chronic rhGH + single rhGH ;compared with the con-trol group ,the CIS content in the chronic rhGH was obviously increased ,while the SOCS-2 content had no obvious difference .Con-clusion Chronic rhGH stimulation can inhibit the phosphorylation of hepatic STAT 5 in normal young mouse ,which mechanism could be related with the increase of CIS content .

Objective To investigate the adsorptive capacity of cross-linked agar entrapping attapulgite clay (CAA) capsule, a novel adsorbent for hemoperfusion. Methods We selected some drugs such as phenothiazines, methylene blue, and diazepam as the model components to test the adsorptive capacity of CAA in human blood in vitro by means of static stirred absorption experiment and repeated hemoperfusion. Results CAA was of strong adsorption for phenothiazines and methylene blue but of little effect on diazepam. The corresponding adsorptive mechanisms were also discussed here. Conclusion CAA could be used as a highly effective adsorbent for the cation drugs in human blood.

AIM: This study was conducted to evaluate the bone repairing ability of coral artificial bone(CAB) composite of human recombinant bone morphogenetic protein-2(rhBMP-2-CAB) implanted into extraction sockets. METHODS: 12 adult dogs served as the experimental animals. Immediately after extraction of the upper second and third incisors, rhBMP-2-CAB and CAB were respectively implanted into each site of extraction sockets. The bone repairing ability of the two grafts was analyzed with histology, image analysis system and radionuclide bone imagining at 4th, 8th and 12th week, respectively.
