1.Intracellular signal transduction pathway of integrin and hepatic stellate cell behavior

Integrins are the main mediators of the complicated interaction between hepatic stellate cells (HSCs) and extracellular matrix (ECM) components. Signal transduction by integrins is initiated by both occupancy and crosslinking of integrins by ECM components. Focal adhesion kinase (FAK) and mitogen- activated protein kinase (MAPK), which are the downstream molecules, involve in integrin-mediated cellular behaviors. In this review, we briefly summarize the integrin's structure, intracellular signaling cascades and its role in HSC behaviors,such as activation,proliferation and apoptosis.
2.Induction of apoptosis with Salvia miltiorrhiza monomer IH764-3 via downregulating focal adhesion kinase in H_2O_2-stimulated hepatic stellate cells

AIM: To investigate the effect of IH764-3 on the apoptosis of H 2O 2-stimulated hepatic stellate cells(HSCs) and the alteration of focal adhesion kinase (FAK). METHODS: The proliferation of HSCs was examined by direct cell count and the apoptosis was determined by Annexin-V/PI labeling, while the morphological change was observed by light microscopy and transmission electron microscopy. In addition, FAK mRNA was detected by RT-PCR. RESULTS: H 2O 2 promoted the proliferation of HSCs. IH764-3 induced the apoptosis of HSCs in a dose-dependent manner. The HSC apoptotic rates of different groups were 6.35%,9.28%,15.10%,19.69%,respectively, after treated with different concentrations of IH764-3 for 48 h while H 2O 2 group showed 2.30%. In 30 mg/L group, the apoptosis rates were 6.73%?10.34%?15.10% for the indicated time periods(12 h, 24 h, 48 h). In the presence of IH764-3, FAK mRNA decreased. The FAK mRNA reduction began at 2 h after adding IH764-3. CONCLUSION: IH764-3 induced the apoptosis of HSCs. Down-regulating the expression of FAK mRNA may be one of its mechanisms.
3.Investigation and management countermeasures on work situation of senior clinical nurses in a hospital

Objective To investigate the current work situation of senior clinical nurses to provide some advices for the nursing management .Methods The self-made questionnaires were adopted to investigate the work situation of 60 senior nurses for conduc-ting the descriptive analysis .Results The senior nurses′ satisfaction degrees to the professional self -identity ,the chief nurse′s scheduling ,the current work environment ,the approval by hospital and the emphasis degree by the department were not high .They thought that the suitable position for them was the office nurse and the second was the responsibility group leader .Whereas ,the un-dertaking key post at present was the responsibility group leader and the second was the office nurse .Conclusion Due to the partic-ularity of nursing work and the physiological and psychological change ,the senior nurses are unsuitable to the busy front -line work ;the senior clinical nurses have the rich clinical experience ,the stronger communicational and organizational coordination abili-ty ,and are more suitable for the posts of the office nurse and health education nurse ,and second is the post of responsibility group leader .
4.Role of extracellular signal-regulated kinase in apoptosis of hepatic stellate cells induced by Arg-Gly-Asp-Ser tetrapeptide

Objective To explore the role of extracellular signal-regulated kinase(ERK)in the apoptosis of hepatic stellate cells(HSC) induced by Arg-Gly-Asp-Ser (RGDS) tetrapeptide. Methods The cultrued HSC cells were divided into six groups including untreated control, fibronertin (FN), FN+RGDS (25 mg/L), FN+RGDS (50 mg/L), FN+RGDS(100 mg/L) and FN+Arg-Gly-Glu-Ser(RGES, 100 mg/L) groups. 3H-thymidine incorporation, transmission electron microscopy and TUNEL were employed to estimate the influence of RGDS on proliferation and apoptosis of HSC. 5.Protective effect of dexmedetomidine combined with flurbiprofen axetil on agitation during recovery peri-od

Objective To evaluate the efficiency of dexmedetomidine combined with flurbipro-fen axetil preventing agitation and reducing extubation reaction after general anesthesia. Methods Eighty patients,ASA Ⅰ or Ⅱ,scheduled for selective oral and maxillofacial surgery were randomly divided into four groups,20 patients in each group.30 mins before end of the operation, patients intravenously received flurbiprofen axetil 50 mg (group F),dexmedetomidine 0.5 μg/kg (group D),dexmedetomidine 0.25 μg/kg plus flurbiprofen axetil 50 mg (group DF),normal saline (group C),respectively.MAP,HR were recorded before extubation (T0 ),extubation (T1 ),5 mins after extubation (T2 ).The recovery time,extubation time,Riker sedation-agitation score(RSAS)be-fore extubation and Ramsay sedation score 5 min after extubation were observed.Results Compared with T0 ,MAP,HR at T1 ,T2 in group C and group F were significantly increased (P <0.05 or P <0.01),MAP,HR at T1 ,T2 in group D and group DF were significantly lower than those in group C (P <0.01 ).The recovery time,extubation time in group D were significantly longer than those in group C,group F and group DF(P <0.05).Ramsay scores in group D was significantly higher than other groups(P <0.05).The incidence of agitation in group D and group DF were significantly lower than those in group C(P <0.05 or P <0.01).Conclusion Dexmedetomidine 0.25 μg/kg plus flurbi-profen axetil 50 mg can effectively prevent agitation and reduce extubation cardiovascular reaction dur-ing recovery period,without the disadvantage of prolonging the recovery and extubation time.
6.Inhibitory effects of Arg-Gly-Asp-Ser(RGDS) tetrapeptide on proliferation and collagen synthesis via downregulating focal adhesion kinase in hepatic stellate cell

Objective To investigate the inhibitory effect of Arg-Gly-Asp-Ser (RGDS) on proliferation and collagen synthesis stimulated by fibronectin (FN) and the influence on the expression of focal adhesion kinase of hepatic stellate cell (HSC), in order to provide a theoretical ground for its use in prevention and treatment of hepatic fibrosis in clinic. Methods The adhesion rates were determined by toluidine blue colorimetric assay. The effects of RGDS on the proliferation and collagen synthesis of HSC were assessed by ~3H-TdR and 3H-proline incorporation assays, respectively. The FAK mRNA of HSCs was determined by RT-PCR. Results RGDS was found to inhibit the rates of adhesion, while RGES has no such ability; ②The proliferation and collagen synthesis of HSC were inhibited by RGDS in dose and time-dependent manners; ③The expression of FAK in the (25, 50, 100mg/L) RGDS groups was significantly lower than that in the control group at mRNA level. Conclusions RGDS can inhibit proliferation and collagen synthesis of HSC, and these effects correlate with its anti-adhesion ability. Meanwhile the expression of FAK is also down-regulated significantly. As a signaling molecule, FAK may be involved in the effect of RGDS. This may be one of the underlying mechanisms of anti-fibrosis effect of RGDS.
7.Analysis of the effect of combination therapy combined with psychological intervention on lumbar disc herniation

Objective To explore the drugs combined with psychological therapy for lumbar disc herniation, summarize the clinical experience.Methods For 60 cases of lumbar disc herniation were treated with drug therapy, and classified as the control group, the other 60 patients in the control group on the basis of psychological intervention therapy, and classified as the observation group, two groups of patients were in our hospital from January 2016 to January 2017 were treated.Results No significant difference between the two groups of patients before treatment, the pain score, after grouping after the intervention in the observation group were obvious pain relief;compared two groups of patients with the emotional state of the visible, before treatment had no significant difference after The observation group after the intervention group were improved more significantly, shows significant difference(P<0.05).Conclusion The analysis showed that the drugs combined with psychological therapy for lumbar disc herniation, which can help patients to improve the clinical situation, adjust the patient's psychological anxiety and depression, improve the clinical effect of intervention, so it is worthy of reference.
