1.Content Determination of Amygdalin in Runing Granules(Semen Persicae,etc.)by HPLC

The amygdalin content in Runing Granules has been determined on the column RP-C_(15), using acetonitrile:water(1:6)as mobile phase and detection wavelength at 240nm.This method showed a high sensitivity,simple operation,good reproducibility and reliable result.It provide a certain base for the quality control of Chinesemateria medica and traditional Chi- nese patent medicine with amygdalin.
2.The Application Research of Digitized Chromatographic Fingerprint Specturm to Quality Standard of Chinese Drug

An introduction of principle of chromatographic fingerprint spectrum and its application on guality standard of Chinese drug. The chromatogram is digitiged by its relative retention value. The digitized chromatogrzaphic fingerprint specturm of some chinese drug was obtained. This technic can be used to reseasch guality standard of chinese drug sucessfully.
3.Study on the alkaloids in Huanglianjiedu Decoction

Objective: To study the content of alkaloids in Huanglianjiedu Decoction and its disassemble prescription. Methods: The contents of four alkaloids such as berberine, jatrorrhizine, coptisine, palmatine in Huanglian jiedu Decoction and its disassemble prescription were determined by HPLC. Results: The content of four alkaloids extracted by tranditional decoction had a tendency to low. On the coutrary, when Rhizoma Coptis and Cortex Phellodendri were decocted together the quantity of alkaloids was additive, and when two herbs above decocted with Radix Scutella and Fructus Gardeniae the quantity of alkaloids decreased obviously. Conclusion:This study can provide a basis for establishing rational quanlitative standard for Huanglianjiedu Decoction.
4.Establishment of method for the analysis of Yun Zhi Polysaccharopeptide(PSP) by HPCE

Objective: To establish the analytical method for the characteristic electropherograms of PSP by High Performance Capillary Electrophoresis(Capillary Zone Electrophoresis). Method: Types, concentration and pH of electrolyte, voltage, temperature, length and size of column were investigated for the analysis of PSP and BSA which is as standard using capillary zone electrophoresis. Results: The optimum condition is that 10mmol?L -1 boric acid buffer, pH9.18, a column 50?m?57cm(HP, effective length of 48.5cm), 30KV, temperature 25 ?C ,DAD detection. The analysis is ended within 7min. Conclusion: The method is simple, rapid and reliable. The characteristic electropherograms of PSP can be established.
5.Quality study on evodiamine of Fructus Evodia by HPCE-FPS

Objective: A HPCE capillary zone eletrophoretic method for the determination of evodiamine in Fructus Evodia was established. Similarly, areliable HPCE-FPS (fingerprint spectrum) was applied to compare quality of Fructus Evodia to 14 samples of 3 species from different areas. Methods: The evodiamine was detected on 254nm and proteins was on 196nm、254nm by HPCE. The electrophoretic parameters were optimized by studying the efffect of the buffer composition. Results: Calibration graph was constructed in the range 1.944 ?g?mL -1~7.766 ?g?mL -1 for evodiamine (r=0.9974). The regression equation was y= 188.7x-32.1. The average recovery was 101.3 (n=3). In this paper, the proteins of Frutus Evodia HPCE-FPS was established at 196nm and 254nm. Conclusion: Content of evodiamine from Fructus Evodia was determined by HPCE-CZE and HPCE-FPS and distinguished three species from different growing areas related to intrinsic quality of them. The research provides new technique for evaluating quality of traditional Chinese medicine.

7.Detection of plasmid-mediated quionlone resistance gene in Enterobacteriaceae

Objective To explore the distribution and genotypes of plasmid-mediated quionlone resistance (PMQR)genes and intI1 integrase genes in Enterobacteriaceae isolates.Methods The PMQR genes and intI1 integrase genes were identified by polymerase chain reaction in the nonduplicate strains of E.coli (80),E.cloacae (18)and K.pneunoniae (27).The positive PCR products were subj ect to DNA sequencing analysis.The gene-positive strains were tested by conj ugation experiment.The minimum inhibitory concentrations (MICs)of donor,recipient strains and transconj ugants were tested by agar dilution method with quinolones and other antimicrobial agents.Results Sixteen (12.8%)of the 125 Enterobacteriaceae isolates were qnr gene positive,including 8 qnrS1 positive and 8 qnrB6 positive.Furthermore,the aac(6′)-Ib-cr gene was identified in 15 (12.0%) strains.Twenty PMQR-positive isolates harbored intI1 integrase gene.The conjugation experiments were successful in 12 of the 26 PMQR-positive isolates and 7 of the 20 intI1-positive isolates.The MICs of quinolones and other antimicrobial agents against the transconj ugants were higher than the MIC values against recipient strains.Conclusions The PMQR genes are prevalent in Enterobacteriaceae isolates.The PMQR-positive isolates can co-harbor integrase genes.These resistance genes have the feature of horizontal transfer,to which close attention should be paid.
8.Clinical Analysis of Death Causes in 100 Burn Patients

Objective To analyze the death causes of the burn patients so as to explore the effective procedures which can raise burn management level. Methods One hundred patients died of burn injury during the past 20 years were enrolled in the study. The died patients were grouped as A (1984~1993) and B (1994~2003) groups, each group containing 50 cases. The mortality, burn area and depth, etiology, pre-hospital treatment, admission time, survival time, tracheostomy, the application of respirator and fibrobronchoscope, operation times, continuous renal replace treatment (CRRT), the incidence of inhalation injury and the pathogenesis of burn death were analyzed and compared between A and B groups. Results There were no differences in burn severity (area and degree), etiology and causes of burn death between the two groups. 9.Determination of catechins and caffeine in tea polyphenols

Objective: To develop a HPLC method for simultaneous determination of catechins (EGC、DC、EGCG、EC、GCG、ECG) and caffeine in tea polyphenols and to quantify seven ingredients in twenty nine batches of tea polyphenols samples from thirteen producers. Methods: The study was achieved using a C 18 column with a methanol water 0.1% formic acid gradient elution system. Results: The peak resolutions of seven ingredients in mixed standards were all above 2; The resolutions in samples were all above 1.5 except EGCG; The average recoveries were 98.56%~100.14%. Conclusion: This method is accurate, stable, reproducible and suitable for quality control of tea polyphenols; the variations of contents are obvious among tea polyphenols samples from different even the same producers.
10.Application of overlapping ratio to fingerprints of green tea and tea polyphenols

AIM: To discuss the application of overlapping ratio to the fingerprints of plant and plant extract. METHODS: We took the HPLC fingerprints of green tea and tea polyphenols for decision procedure. RESULTS: Overlapping ratio could be applied to selecting representative samples for characteristic fingerprints of green tea and tea polyphenols,and could be used to deduce the raw material,process and producer of tea polyphenols.(CONCLUSION:) Overlapping ratio can be used to choose the suitable samples for establishing the characteristic fingerprint spectrum,and can supply references to relationship investigation among products,raw material and process.
