2.Research on Traditional Chinese Medicine Syndrome and Treatment Rules of Literature Information Data Based on Polycystic Ovary Syndrome

This study was aimed to mine correlation between the cause and traditional Chinese medicine ( TCM ) syndrome of polycystic ovary syndrome ( PCOS ) from different angles and at different levels . The study found
that the inherent laws of the cause and syndrome by the application of different statistical methods and data mining methods . The literature information database of PCOS was searched . And the Microsoft Office Excel and the Microsoft Office Access were used in the establishment o the database . And the SPSS17 . 0 was used as the statistical software . The Medisc-3S system was used as the mining tools to discover the rules between etiology and syndrome . The function of frequency statistics and complex networks were used to discover and summarize the TCM syndrome and treatment of PCOS . The TCM syndrome and treatment of PCOS were discov-ered and summarized according to the statistical results . And the results showed that three reasons of etiology and pathogenesis of PCOS contained kidney-deficiency , phlegm-dampness , and blood-stasis . The five types of TCM syndrome included spleen-kidney deficiency , kidney-deficiency and blood-stasis , spleen-deficiency and phlegm-dampness , kidney-deficiency and phlegm-dampness , kidney-deficiency and liver stagnation . It was concluded that the treatment should be mainly from kidney-tonification and assisted with blood-activation and phlegm-removing . The association rules and complex networks of data mining methods were applied in the lit-erature research of PCOS . The results provided literature references and statistical evidences for the phlegm-dampness accumulation in the uterus as the pathogenesis of PCOS , and the methods of kidney-tonification , blood-activation and phlegm-removing as the treatment principles .
3.A convenient and fast method to amplify DNA fragments from filamentous fungi

Objective:Using PCR to amplify filamentous fungi DNA fragments. Methods:Pour some liquid nitrogen to the hypha and grind them to powder. Then add some TE buffer and boil them for 15-20 minutes. Take a little supernatant as PCR template. After PCR do agarose gel electrophoresis. Collect the gel containing the objective fragment and add some TE. Take a little supernatant as PCR template (again.) After PCR, do agorose gel electrophoresis again. Results:We obtained a great deal of objective DNA. Conclusion:Amplifying filamentous fungi DNA fragment by this way, we can save time and money. We can also improve our work efficiently.

The polycystic ovray syndrome(PCOS) is a heterogeneous condition with genetic predisposition. It is characterized by a myriad of symptoms including oligomenorrhea or amenorrhea,anovulation or infertility,hirsutism or acne. Insulin resistance appears to be an important factor in PCOS though the lack of an etiology has led to symptom oriented therapy which includes lifestyle modification,the use of cyclical progestagens and antiandrogens. Therapeutic approaches to PCOS remain an ongoing source of debate. Insulin sensitizing agents may bring new hope in therapy. Future research is aimed at shedding light on the pathophysiology so as to optimize treatment of women with PCOS.
5.Effects of pseudolaric acid B on the metastatic activity and invasiveness of HeLa cells and the expression of matrix metalloproteinase

Objective To investigate the effects of pseudolaric acid B(PAB) on the invasiveness and metastatic activity of cervical cancer HeLa cells in vitro and the possible mechanism.Methods HeLa cells were divided into 5 groups: blank control group,DDP 3.125?g/ml group,20?mol/L PAB group,10?mol/L PAB group and 5?mol/L PAB group.The effect of PAB on invasive ability of HeLa cells was observed with a Transwell cell culture chamber.The mRNA expression of MMP22 and MMP29 in HeLa cells was detected by RT-PCR and the protein expression was detected by Western blotting.Results The average number of invaded HeLa cells was 15.60?2.45 in 20?mol/L PAB group and 20.14?2.48 in 10?mol/L PAB group,which was significantly lower than those in control group(46.50?4.70) and PAB 5?mol/L group(42.44?5.80,P

Polycystic ovary syndrome(PCOS),one of the most common causes of anovulation-induced infertility,is largely dependent on the influence of obesity and metabolic alterations,including an insulinresistant state and the metabolic syndrome,which consistently affect most women with PCOS.The clinical phenotype and the pathophysiological interact between obesity and PCOS have also been changed obviously in these paitients.Obesity has profound effects on the reproductive function of PCOS women and,particularly the abdominal phenotype,may add to the chances of androgen excess,PCOS,insulin resistance and hyperinsulinaemia.Obesity plays an important role in the pathogenesis of PCOS and obesity-associated metabolic alterations have a long-term impact on the patients' health.

8.Isolation of human serum exosome and the clinical value of exosomal miRNA detection

Objective To isolate and identify exosomes from human serum , explore the feasibility of detecting exosomal miRNA in human serum.Methods Retrospective study.Serum samples from 10 healthy individuals in January 2013 were randomly selected.Besides, from January 2013 to December 2014, serum samples from prostate cancer(PCa) patients (n=20), benign prostatic hyperplasia(BPH) patients ( n=20 ) and healthy controls ( n=20 ) were selected.Exosomes were isolated from these serum samples using ExoQuick , and then identified by using transmission electron microscopy , NanoSight nano particle analyzer and Western Blot for morphology and molecular phenotype.The quality of exosomal RNA was analyzed using Agilent 2100 Bioanalyser.Then quantificational real-time polymerase chain reaction ( qRT-PCR) was carried out to detect miRNAs in different components of human serum ,and nonparametric tests were used for difference analysis.Results Exosomes isolated from human serum showed round or oval vesicles, mainly in diameter 40-100 nm, and with maximum peak distribution of 58 nm.Moreover, they expressed HSP70 and four transmembrane protein CD 63.Agilent 2100 Bioanalyzer results showed that the major RNA component of exosome was about 25nt small RNA.qRT-PCR confirmed that 4 normal miRNAs were expressed in human serum exosome , and the expression of miRNAs in exosome pellets were higher than the whole serum (miR-21, U =16,P =0.007 2; miR-16, U =3,P<0.000 1; miR-20a, U =2,P <0.000 1;let-7a, U=13,P=0.003 2) and exosome-depleted supernatant ( miR-21, U=15,P=0.006 5;miR-16, U=2,P<0.000 1;miR-20a, U=1,P<0.000 1;let-7a, U=10,P=0.002 8).miR-141, the molecular marker of prostate cancer ,were analyzed by qRT-PCR in whole serum samples and serum exosome pellets isolated from the same serum in a cohort of 20 PCa patients , 20 BPH patients and 20 healthy control people.The results showed that , in three groups , exosomal miR-141 expression were all significantly higher than serum circulating miR-141 (Control group, U=66,P=0.000 3; BPH group, U=83,P=0.001 6;PCa group, U=54,P<0.000 1).In addition, the expreession of exosomal miR-141 in PCa patients was significantly higher than BPH patients or healthy controls (3.85 fold, U=74,P=0.000 7 and 4.06 fold, U=70,P=0.000 5).Conclusion Exosome can be efficiently isolated from human serum.Compared with the whole serum , isolation of serum exosome may helpful to improve the detection of circulating miRNA.
9.In vitro activity of an anti-prostate cancer polypeptide APP216

Objective:To investigate the in vitro the anti-prostate cancer effect of a novel polypeptide,APP216,so as to provide a basis for development of new drug for treatment of prostate cancer.Methods:The polypeptide drug included the amino sequences of BH3,K237 and DG2 domain and the peptide that could be digested by PSA.The anti-prostate cancer effects of the polypeptide prodrug on prostate cancer cell line LNCaP,22RV1(secreting PSA)and PC3,DU145(secreting no PSA)were determined by MTT test and Hoechst 33258 staining.Results:MTT test revealed that the surviving rates of LNCaP and 22PV1 cells were respectively 22% and 34% 48 h after APP216(270 ?g/ml)treatment,and 9.8% and 8.2% 72 h after APP216(270 ?g/ml)treatment.The surviving rates of PC3 and DU145 cells were respectively 90% and 95% 48 h after APP216(270 ?g/ml)treatment,and 87% and 92% 72 h after APP216(270 ?g/ml)treatment.Hoechst 33258 staining showed the typical features of cell apoptosis:cell shrinkage,chromatin condensation and hypodiploid genomic DNA content in LNCaP and PC3m cells.Flow cytometry showed an apotosis rate of 36.26% 48 h after APP216(270 ?g/ml)treatment in LNCaP cells,and of 1.63% after 48 h APP216(270 ?g/ml)treatment in PC3m cells.Conclusion:APP216 has a satisfactory in vitro cytotoxicity on human PSA-secreting prostate cancer cells and can induce tumor cell apoptosis,but not on non-PSA secreting prostate cancer cells,indicating APP216 polypeptide can be specifically digested by endonuclease enzyme.Meanwhile,BH3 domain can be transferred into cells and induce apoptosis through HIV-TAT.
10.Exosome-encapsulated miR-375 in urine as a non-invasive biomarker for prostate cancer diagnosis

Objective To analyze the expression of urine exosomal miR-375 in prostate tumors and investigate its clinical utility.Methods A total of 45 patients with PCa,24 with benign prostate hyperplasia (BPH) and 24 healthy individuals were enrolled into this study.Exosomes were isolated from the urine of PCa,BPH and healthy individuals and the total RNA was extracted from the exosomes.The exosomal miR-375 expression was assessed by quantitative real-time PCR and analyzed with the comparative quantification cycle method (2-△△CT).We performed comprehensive biostatistical analyses to explore the clinical value of miR-375 in prostate cancer.Results The urine exosomal miR-375 expression was significantly downregulated in the patients with PCa compared with BPH and the healthy controls (P < 0.01).No statistically significant difference of the urine exosomal miR-375 expression levels between the patients with BPH and healthy individuals was observed (P > 0.05).The urine exosomal miR-375 expression level was also found to be associated with clinical stage and bone metastasis status of the patients with PCa (P <0.05),and with the increase of Clinical stage.The expression level of miR-375 decreased.No significant relationship was detected between miR-375 level and the patient's age,gleason score and serum prostate-specific antigen level (P > 0.05).Receiver operator characteristic analyses demonstrated that the urine exosomal miR-375 expression could better differentiate PCa from BPH patients:AUC 0.715 (95% CI:0.589-0.842) vs PSA AUC 0.632 (95% CI:0.492-0.771) (P<0.01).Conclusion The urine exosomal miR-375 could serve as a non-invasive biomarker for the diagnosis of PCa.
