Enantioselective hydrolysis of 2-carboxyethyl-3-cyano-5-methylhexanoic acid (CNDE) is the key step in chemoenzymatic synthesis of pregabalin. We purified an intracellular carboxyl esterase from Morganella morganii ZJB-09203, which exhibited high enantioselectivity and activity towards CNDE. The carboxyl esterase was purified to electrophoretic homogeneity by ammonium sulfate fraction precipitation, Phenyl Sepharose 6 FF hydrophobic interaction chromatography, anion exchange with DEAE Sephadex A-50 and Bio-Scale CHT column. The purified enzyme was a monomer with molecular mass of 68 kDa determined by SDS-PAGE and gel chromatography. Substrate specificity of the enzyme towards p-nitrophenyl esters suggested that the purified enzyme was an esterase. The optimal reaction pH for CNDE hydrolysis was 9.0, and optimal temperature was 45 degrees C. The esterase was stable between pH 7.0 and 9.0, and at 40 degrees C. The enzyme activity was enhanced by Ca2+, Cu2+ and Mn2+, whereas strongly inhibited by Co2+, Fe3+, Ni2+ and EDTA. Meanwhile, we investigated the kinetic parameters of the esterase towards p-nitrophenyl esters and effect of CNDE concentration on conversion. The present study reported the esterase capable of stereospecific hydrolysis of CNDE for the first time. Our research will provide foundations for industrial production of Pregabalin using the new biocatalyst.

Clinical Medicine of China 2009;25(9):920-922
Objective To investigate the expression of MMP-2 and MMP-7 and their elinieopathologieal significance in eoloreetal carcinomas. Methods The expression of MMP-2 mRNA, MMP-7 mRNA and MMP-2, MMP-7 in 42 samples of eoloreetal carcinomas and its adjacent normal eoloreetal mueosa were examined using fluo-rescence quantitative RT-PCR (FQ-RT-PCR) and immunohistoehemistry. The relationship of their expression with some elinieopathologieal characteristics was analyzed. Results MMP-2 mRNA, MMP-7 mRNA and MMP-2, MMP-7 were significantly over-expressed in eoloreetal carcinomas compared with its adjacent normal eoloreetal mueosa( P < 0.05 ) , and they were positively associated with bowel wall invasion, the regional lymph node metastasis and Dukes stage. The expression of MMP-2 mRNA and MMP-7 mRNA was positively correlated with MMP-2 and MMP-7 in eoloreetal eareinornas. Conclusions MMP-2 and MMP-7 may play a key role in eoloreetal carcinogenesis, tumor in-vasion and metastasis. Examination of combined MMP-2 and MMP-7 expression may have an important significance to judge the malignant degree and biological behavior of human eoloreetal carcinoma and for evaluating the progno-sis.

Object To investigate the saccharide changes of Radix Rehmanniae when being processed. Methods The fresh roots were torrefied at 65 ℃ and sampled at 0,1, and 6 d. The samples and 4 h steamed slices of those dried roots were extracted with hot water respectively. The extracts were subjected to HPLC analysis, Sugar pak 1 column at 80 ℃,with water as the mobile phase at 0 7 mL/min and detected by RID. Results There are three principal components in the fresh roots, i.e. stachyose (11%-15%), sucrose (0 30%-0 92%), and catalpol (0 27%-0 88%). In the processing of the fresh roots, the HPLC chromatorams of the extracts differed to each other remarkbly. On the chromatogram of roots torrefied for 1 d, one distinct monosaccharide peak displayed at 10 2 min, which is likely to be galactose, and it become prominent in the dried roots together with raffinose. In steamed ones, the peaks of fructose and glucose became outstanding. Conclusion According to the HPLC chromatograms, the degalacto sylation of stachyose may take place during torrifying, while defructosylation during steaming. One of the principal purposes of the processing may be for stachyose degradation for its flatulence causing character.
