1.The study on the serum proteomic fingerprints in gastric carcinoma patients by SELDI technique

Objective To detect serum protein spectrum in gastric cancer, screen the serum proteins related to gastric carcinoma and build auxiliary diagnosis model with SELDI-TOF-MS, Methods The serum proteomic fingerprints were detected by SELDI-TOF-MS and CM10 proteinchip. The sera came from 50 first visit gastric cancer patients who were diagnosed by pathological method and 16 healthy peo- ple. The data were analyzed by Biomarker Wizard Software and the proteomic fingerprint of gastric carcinoma model was developed by BPS (biomarker pattern software). Results Compared with the healthy people group, there were 34 significant different protein peaks in the gas- tric carcinoma group. And the diagnosis model composed with 6 proteins (M/Z values were 6016, 6744, 2822, 4474, 7892 and 3242) could classify the 2 groups correctly. In the test group, the sensitivity and specificity were 96% and 93.75% respectively, the accuracy was 95.45%. Conclusion SELDI-TOF-MS method show features such as microcontent, fast and high- resolution etc. It could be utilized to screen significant proteins and develop an auxiliary diagnosis model in gastric carcinoma, which may be used to diagnose this disease.

3.Radiologic Findings and Clinical Significance of the Herniation Pits of the Femoral Neck

Objective To study the incidence and clinical significance of the herniation pits of the femoral neck.Methods 600 cases(299 men,301 women,18~82 years)were collected.The incidence,radiologic finding and clinical significance of the herniation pit of the femoral neck were analysed.Results Of 1200 hip joints in 600 cases,there was 58 cases(64 sides)(5.3%)with herniation pits of the femoral neck,including 39 men(68.7%)and 19 women(31.3%),the lesions localized in the left joint in 25 eases(39.1%),in the right joint in 27 cases(42.2%)and in bilateral joints in 6 cases(18.7%).There were a obvious significant differences on both sexes,and no significant differences on age groups.On X-ray film,the lesions appeared as a round radiolucency with thin clear sclerotic rim.Conclusion The incidence of the herniation pit of the femoral neck is 5.3%,which has a typical X-ray feature,and may indicate the femoroacetabular impingement.
4.A new technique for transmyocardial laser revascularization.

OBJECTIVETo investigate the effects of traditional transmyocardial laser revascularization (T-TMLR) and non-transmural myocardial laser revascularization (N-TMLR) on myocardial ischemic and necrotic areas and angiogenesis.
METHODSIn 30 rabbit models, Evans blue -TTC staining and HE staining were used for measuring ischemic and necrotic areas and observing angiogenesis.
RESULTSThe ischemic areas in the MI + T-TMLR (30.6 +/- 1.0)% and the MI + N-TMLR group (30.2 +/- 0.4)% were significantly decreased compared with the MI group (37.2 +/- 0.2)% (F = 21.04, P < 0.01). The necrotic areas in the MI + T-TMLR (17.0 +/- 0.7)% and the MI + N-TMLR group (16.9 +/- 0.6)% was not different from that in the MI group (17.9 +/- 0.5)% (F = 1.73, P > 0.05). The angiogenesis in the MI + T-TMLR (1.8) and the MI + N-TMLR (1.6) was significantly increased compared with that in the MI group (0.6) (F = 15.32, P < 0.01).
CONCLUSIONSMI + N-TMLR can achieve the same effects of MI + T-TMLR. Inducing angiogenesis via laser channels and decreasing ischemic area may be one of the predominant mechanisms of TMLR, whereas whether laser channels themselves are patent is not relation to the effects of TMLR.

5.Controlled release of fuctional proteins IGF-1, aFGF and VEGF through self-assembling peptide nanofiber hydrogel.

Peptide RADA can undergo spontaneous assembly into well ordered nanofibers and hydrogels in its water solution. In this work, a variety of proteins, including IGF-1, aFGF and VEGF with different molecular weight and isoelectric points, were chosen and encapsulated within the RADA peptide hydrogel. UV-vis spectroscopy was used to determine the concentration of the released proteins in the solution. The release kinetics suggested that protein diffusion through nanofiber hydrogels depended primarily on the size of the protein and the density of the peptide nanofiber. Circular dichroism (CD) spectroscopy indicated that the encapsulation and release by RADA hydrogel did not affect the secondary structure of the proteins studied.

6.Analysis of the genetic polymorphisms of 15 autosomal short tandem repeat in a Han population of Shandong.

OBJECTIVETo obtain population genetic data at 15 short tandem repeat(STRs) in Shandong Han population.
METHODSFifteen STRs were genotyped in 200 unrelated individuals from Shandong Han population using Identifiler(TM) Kit.
RESULTSIn this Shandong Han population, the observed heterozygosities of the 15 loci ranged from 0.605 to 0.882, and the expected heterozygosities were 0.625-0.862, the polymorphic information contents ranged from 0.57 to 0.85, the discrimination powers ranged from 0.795 to 0.958, and the exclusion probabilities ranged from 0.297 to 0.758. The non-differentiation exact P values at TPOX loci were more than 0.05 between Shandong and Han population of Henan, Jilin, and Jiangsu. The non-differentiation exact P values between Shandong Han population and others populations were more than 0.05 at others loci. The P value at D13S317 was more than 0.05 between Shandong Han population and Ewenki population.
CONCLUSIONThese loci are polymorphic in Shandong Han population, and the discrimination powers and exclusion probabilities were high in the 15 STRs except TPOX and TH01.

7.ALK gene fusion associated non-small cell lung cancer: automated immunostainer detection and clinicopathologic perspectives.

To explore the automated immunostainer screening anaplastic lymphoma kinase (ALK) gene fusion non-small cell lung cancer (NSCLC) and clinicopathological characteristics of the molecular subtype lung cancers. Methods Five hundred and sixty-six cases of NSCLC were collected over a 16 month period. The test for ALK was performed by Ventana automated immunostainer with anti-ALK D5F3. The histological features, treatment and outcome of patients were assessed. Results Thirty-eight cases (6.7%, 38/566) of NSCLC showed ALK gene fusion. The frequency of ALK gene fusion was higher in male (7.1%, 25/350) than that in female (6.0%, 13/216) patients, but not achieving statistical significance (chi2 = 0.270, P = 0.604). ALK + NSCLC was more significantly more frequent in patients < or = 60 years (9.9%, 28/282) than >60 years (3.5% , 10/284) of age. Histologically, the ALK + NSCLCs were mostly adenocarcinoma (81.6%, 31/38) , among which eighteen cases were solid predominant subtype with mucin production; nine cases were acinar predominant subtype; one case was papillary predominant subtype and three cases were invasive mucinous adenocarcinoma. The ALK + non-adenocarcinoma included three cases of squamous cell carcinoma, three cases of adenosquamous carcinoma and one case of pleomorphic carcinoma. Among the ALK + NSCLC patients, the number of non/light cigarette smokers (86. 8% , 33/38) was more than that of heavy smokers. Twenty-nine cases were stages III and IV; twenty-nine cases showed lymph node metastasis; twenty cases showed metastases mostly to brain and bone; and one case showed EGFR gene mutation coexisting with ALK gene fusion. Twelve of fifteen patients received crizotinib therapy and remained stable. Conclusions NSCLC with ALK gene rearrangement shows distinctive clinical and histological features. Ventana-IHC may he a feasible and valid technique for detection of ALK rearrangement in NSCLC.

8.Alveolar soft part sarcoma: a clinicopathologic analysis of 48 cases.

OBJECTIVETo study the clinicopathologic features and differential diagnosis of alveolar soft part sarcoma (ASPS).
METHODSThe clinical data and pathologic features of 48 cases of ASPS were evaluated. Immunohistochemical study, PAS staining and fluorescence in-situ hybridization (FISH) were carried out in selected examples. Relevant literature was reviewed.
RESULTSAmongst the 48 cases studied, there were 17 males and 31 females, with male-to-female ratio of 1.0∶1.8. The age of patients ranged from 2 to 60 years (median=26 years). The tumor was most commonly located in deep soft tissue, especially that of lower extremities. Histologically, the tumor cells were arranged in alveolar or solid patterns and separated by sinusoidal vessels. They were large and contained abundant eosinophilic granules or crystals in cytoplasm. The nuclei were round to polygonal and vesicular, often with prominent nucleoli. Intravascular tumor extension was common. Some cases showed necrosis, hemorrhage and cystic changes. Immunohistochemical study showed that the tumor cells were positive for TFE3 (100%, 33/33). FISH assay was carried out in 4 cases and all of them had TFE3-ASPL gene fusion.
CONCLUSIONSASPS is a rare malignant neoplasm, often occurs in young patients. TFE3 is a useful immunohistochemical marker for diagnosis. The diagnosis is further confirmed by other markers.

9.A clinicopathological and prognostic study of 22 cases central neurocytoma.

OBJECTIVETo investigate the clinicopathological features and prognosis of 22 cases of central neurocytoma (CNC), representing 0.48% of a series of 4 528 patients undergoing biopsy for central nervous system tumors.
METHODSThe histopathological, ultrastructral, immunohistochemical and clinical features of CNC were studied by electron microscopic examination and immunohistochemical stain for Synaptophysin (Syn), neuron special enolase (NSE), Leu-7, glial fibrillary acid protein (GFAP), MBP and proliferating cell nuclear antigen (PCNA).
RESULTSThe age of the cases ranged from 4 to 44 (average 27.9 years) with all tumors localized in the ventricles. In the 18 patients followed up, 14 were alive for 8 months to 14 years and 11 months after the operation, and 4 died. The average survival period was 70.7 months. Histologically, the tumor in all 22 cases had the oligodendroglioma-like pattern with honeycomb appearance and cell-free islands of eosinophilic matrix. Cellular anaplasia, mitosis and necrotic areas were rarely seen in the tumors. Immunohistochemical study demonstrated strong positivity for Syn, NSE and Leu-7, and negative for GFAP and MBP. Ultrastructural features showed presence of round tumor cells with abundant cell processes containing microtubules, neurosecretory granules, clear vesicles and lysosome-like structures.
CONCLUSIONSThe differential diagnosis between CNC and oligodendroglioma could not be established by routine light microscopy. The importance of immunohistochemical and electron microscopic studies for making a correct diagnosis is emphasized. The prognosis of patients is usually favorable, even if the tumor was resected subtotally. The relationship between the presence of anaplastic histological features in CNC and patient outcome remains unclear.

10.Study of the volatilization inhibitor for formalin.

OBJECTIVETo find an inhibitor to reduce the volatilization of formalin.
METHODThe saturated solution of sodium hydrosulphite (SHS) was sprayed on the surface of the anatomy specimens, then the concentration of formaldehyde in the air was tested.
RESULTSThe concentration of formaldehyde in the air of SHS sprayed group [(3.10 +/- 1.22) mg/m3] was significantly lower than that of the control group [(8.36 +/- 4.11) mg/m3, P < 0.01].
CONCLUSIONSHS may be a volatilization inhibitor for formalin, which could reduce the concentration of formaldehyde in the air.
