1.Relationship of adiponectin and brachial ankle pulse wave velocity in type 2 diabetes mellitus patients combined with hypertension

Objective To explore the relationship of adiponectin and brachial ankle pulse wave velocity (baPWV) in type 2 diabetes mellitus patients combined with hypertension.Methods Fifty-six patients with diabetes mellitus,69 diabetes mellitus patients combined with hypertension and 78 healthy people were recruited in our study who visited doctors in Fenyang Hospital from March 2010 to March 2012.The levels of adiponectin,brachial ankle pulse wave velocity and other biochemical indicator were determined.Analyzed the correlation between adiponectin and baPWV among the three groups.Results The levels of adiponectin in control group,diabetes mellitus group,and diabetes mellitus combined with hypertension group were (13.9 ±4.2) mg/L,(9.1 ± 3.8) mg/L and (7.6 ± 2.4) mg/L respectively,and the difference was statistically significant (F =62.53,P <0.01).The levels of baPWV in control group,diabetes mellitus group and diabetes mellitus combined with hypertension group were (1 617.4 ± 198.2) cm/s,(2 010.9 ±231.7) cm/s and (2 196.1 ±218.6) cm/s respectively,and the difference was also statistically significant (F =139.41,P < 0.01).The correlation between adiponectin level and baPWV among three groups were-0.629,-0.561,and-0.528,respectively (P =0.027,0.034,0.029).Conclusion The levels of adiponectin decreased in diabetes mellitus patients combined with hypertension,while the baPWV increased.There is a good correlation between adiponectin and baPWV.
2.MR virtual endoscopy: a study of clinical applications in cholangiopancreatography

Objective To determine the added clinical applications of MR virtual endoscopy (MRVE) plus MRCP in cholangiopancreatography. Methods In 14 month, 44 patients with obstructive jaundice and symptomatic bile trace were assessed by MRVE in biliary tract.The 2D data of heavily T 2 weighted images with FSE(non breath hold, respiratory trigger) and SSFSE(breath hold, multi slice single shot fast spin echo) sequence were transferred to the computer workstation. Navigator software (GEMS) was used to display intraluminal views of the pancreaticobiliary tract with a surface rendering technique. Results Patients suffered from neoplasm ( n =18), bile stone (17), postoperative complications of cholecystectomy( n =3), and other disease of biliary tract( n =6). MRVE could clearly demonstrate the anatomical structures in biliary tract, extended pancreas tract,biliary meatus narrowing and obstruction, bile duct stones, duct branches, and the inner wall of pathological obstruction.The main limitation of virtual endoscopy was inability to evaluate mucosa and lack of histological diagnosis. Sensitivity for detection of bile duct dilatation and site of obstruction was: 2D FSE 91 7%, and SSFSE 97 9%. Conclusion MRVE provided noninvasively endoscope like display of biliary tract.Its display ability was a supplement to image biliary trace. MRVE effect of multi slice SSFSE sequence was better than that of fast spin echo.
3.The phantom and clinical study of low-dose protocol of volumetric high resolution CT

Objective To explore the optimal low-dose protocol of volumetric high-resolution CT (VHRCT) of the lung and evaluate its diagnostic value.Methods Catphan phantom were scanned using GE Lightspeed VCT with different parmeters from 120 kV,10 mAs to 120 kV,250 mAs in 10 mAs increments.Other parameters included 0.969 pitch,20 mm scan coverage,0.625 mm collimation and 30 cm display field of view and bone recon kernel.The spatial and density resolution,noise and radiation dose of each scanning were measured to determine the low-dose VHRCT protocol.In clinical study,105 patients with diffuse lung diseases underwent standard-dose VHRCT with 120 kV,250 mA according to the clinical needs.Low-dose VHRCT was performed with 120 kV,120 mAs in follow-up.Two radiologists who were unaware of the CT technique reviewed randomized images for the detail of diffuse lung diseases,including linear or reticular opacities,micro-nodules or tree-in-bud patterns,bronchiolectasis,ground-glass opacities and emphysema using a 4-point scale.Results In phantom study,the spatial-resolution maintained at 9 LP/cm from 250 mAs to 120 mAs.Below 120 mAs,the spatial-resolution and density-resolution decreased and noise increased with the decrease of tube-current.In clinical study,there were no statistical differences between standard-dose and low-dose VHRCT in demonstrating the detail of diffuse lung diseases(P >0.05).The CTDIvol was 23.44 mGy at 250 mAs and 11.25 mGy at 120 mAs,with 52%dose reduction by low-dose VHRCT.Conclusions Low-dose VHRCT at 120 kV,120 mAs offers maximum dose reduction without compromising spatial resolution and diagnostic value.

Objective By finding out the Chinese official pharmacy talent training and education model in Sui and Tang dynasties,we can used it in the cultivation of contemporary talents in pharmacy.Methods pharmacy education in CNKI,Sui and Tang history books,Chinese herbal medicine and legal literature were collected and categorized in training organizations,education and evaluation three aspects.Results In the Sui and Tang dynasties,pharmacy institutions included imperial pharmacies,drug possession Bureau and the Department of imperial medicine disciplines.Meanwhile,there were relevant regulations in education,teaching and quality assessment.According to the traditional theory,practice and correlation characteristics,focusing on the connection of clinical pharmacy practice courses and lessons,the government trained a large number of professional talents in pharmacy.Conclusion In the Sui and Tang dynasties,Chinese material medical attaches great importance to the comprehensive quality of talents cultivation,which is worthy for modern reference.

Objective To undergo T 2 *-weighted first-pass perfusion imaging after dynamic contrast-enhanced T 1-weighted imaging at the same examination in patients with breast neoplasm, and to evaluate if T 2 *-weighted imaging can provide additional diagnostic information to that obtained with T 1-weighted imaging. Methods Twenty-nine patients with breast lesions verified by pathology(benign 12, malignant 17)underwent MR imaging with dynamic contrast-enhanced T 1-weighted imaging of the entire breasts, then followed immediately with six-sections, T 2 *-weighted first-pass perfusion imaging of the lesions.The diagnostic indices were got by the 3 D T 1-weighted enhancement rate criterion and T 2 * signal intensity loss rate criterion individually.The sensitivity and specificity were calculated and the two methods were compared.Results With the dynamic contrast-enhanced T 1-weighted imaging, there was a significant differences between the benign and malignant breast lesions (t=2.563,P=0.016), but a considerable overlap of the signal intensity increase between the carcinomas and benign lesions was found, with a sensitivity of 94% and a specificity of 25%.With T 2 *-weighted first-pass perfusion imaging, there was a very significant differences between the benign and malignant breast lesions (t=4.777,P
7.Clinical application of computer-aided detection in breast cancers on mammograms

Objective To evaluate the clinical value of computer-aided detection (CAD) system in the diagnosis of breast cancer on mammograms. Methods The mammograms of 136 pathology-proved breast cancer cases were analyzed by CAD system. The results of CAD were compared with that made by radiologists. Results The sensitivity, specificity, and accuracy of CAD system to microcalcification lesions were 93.2% (41/44), 22.8%(21/92), and 45.6%(62/136), respectively. The sensitivity, specificity, and accuracy of CAD system to mass lesions were 82.9%(107/129), 71.4%(5/7), and 82.4%(112/136), respectively. There was middle level agreement between the detection of CAD system and that of the radiologists (Kappa_(mass)=0.424, P_(mass)=0.000; Kappa_(cal)=0.365, P_(cal)=0.013). Conclusion CAD system has the characteristics of high sensitivity and low specificity. It could be the aid of radiologists in breast cancer detection on mammograms.
8.Mono-modality image registration based on modified mutual information

With modified mutual information(MI) measure,Powell search algorithm is utilized to realize mono-modality image registration.The MI's formulae are improved to reduce computation cost,and the replace of Powell's search direction is also rectified to keep the original search direction and eliminate local maxima.With rotation set in advance,local maxima can be avoided.The influence on registration velocity by Powell convergence threshold is also discussed.The results show that the modified MI measure can reach sub-pixel precision with obviously accelerated speed.It has good robustness,high speed and high precision.
9.Value of MRI in differential diagnosis of primary architectural distortion detected by mammography

Objective The aim was to evaluate the diagnostic value of MRI in lesions with architectural distortion manifested in mammography. Methods A retrospective analysis of MRI was performed in 60 patients with 61 lesions manifested as architectural distortion in full?field digital mammography (FFDM) and subsequently confirmed by pathology or two year's follow?up, 30 were malignant and 31 were benign. All the patients underwent MRI within 2 weeks of mammography. MRI protocol included conventional MR, DWI and dynamic contrast?enhanced MRI. The breast imaging reporting and data system (BI?RADS) was used as the reference standard. BI?RADS categories 1 to 3 were considered as negative for malignancy and BI?RADS categories 4 to 5 were considered as positive for malignancy. ADCs of suspicious lesion of interest and glandular tissue were calculated. nADC was then calculated using the following formula:nADC=ADC(lesion)/ADC(glandular tissue). ADC and nADC were compared by using t test. ROC analysis was carried out to define the most effective threshold ADC and nADC value to differentiate malignant from benign lesion in the breast. Diagnostic performance of the FFDM, DCE?MRI and DCE?MRI combined nADC were calculated. Results ADC value of malignant and benign lesions was (1.35±0.31)×10?3 mm2/s and (1.07±0.40)×10?3 mm2/s, respectively . nADC values were 0.83±0.17 and 0.59± 0.25, respectively (t values were 2.82 and 4.54, P<0.01). Area under the curve of ADC and nADC were 0.829 and 0.753 respectively. When threshold of ADC was set at 1.19×10?3mm2/s, sensitivity and specificity were 71.0%and 86.7%, respectively. For a nADC value threshold of 0.589, sensitivity and specificity were 93.5%and 76.7%, respectively. Sensitivity, specificity and accuracy with FFDM were 80.0%(24/30), 9.7%(3/31) and 44.3%(27/61), Sensitivity, specificity and accuracy with DCE?MRI were 90.0%(27/30), 41.9%(13/31) and 65.6%(40/61), Sensitivity, specificity and accuracy with DCE?MRI combined nADC were 93.3%(28/30), 77.4%(24/31) and 85.2%(52/61), respectively. Conclusion Sensitivity and specificity with DCE?MRI combined nADC is higher, and DCE?MRI combined nADC values is helpful to differentiate malignant from benign lesions with architectural distortion manifested in FFDM.
10.Expression of the thermostable carboxypeptidase Taq gene in Pichia pastoris GS115.

To express recombinant carboxypeptidase from Thermus aquaticus (Cpase Taq) in Pichia pastosis, the open reading frame coding thermostable Cpase Taq was optimized based on the preference of P. pastoris codon usage and synthesized in vitro. The novel gene was cloned into P. pastoris expression vector pHBM905A and the sequence coding 6xHis tag was fused with the ORF of Cpase Taq gene. The recombinant plasmid was named pHBM905A-Cpase Taq and transformed into P. pastoris GS 115. Transformants were induced with 1% methanol for 72 h until the enzyme yield reached 0.1 mg/ml. The enzyme was purified and its enzymatic properties were analyzed. The results showed that the specific enzyme activity reached maximum at 75 °C and pH 7.5, which was about 80 U/mg. It was the first report about the secretory expression of Cpase Taq in P. pastoris GS115. Because of its large-scale preparation, this enzyme may be applied in industrial hydrolysis of peptides into amino acids in the future.
