1.Effect of Eucommia on Expression of Matrix Metal Proteinase-2 in Rats with Unilateral Ureteral Obstruction

[Objective] To investigate the effects of Eucommia on renal interstitial fibrosis and its mechanism.[Methods]Using renal interstitial fibrosis in rats with Unilateral Ureteral Obstruction(UUO).Adult female SD rats(n=32) were randomly divided into 4 groups: Sham operation group (Sham); UUO group,Irbesartan group; Eucommia group.Undergo light microscopy by Masson and HE staining to observe the renal interstitial fibrosis index.To detect the expression of MMP-2 by immunohistochemistry 2w after surgery.[Results] 1.The levels of BUN and Scr in UUO group were significantly higher than in Sham group.Compared with UUO group,the levels of BUN and Scr in irbesartan and eucommia groups were significantly lower.There was no significant difference between irbesartan group and eucommia group.2.The renal tissue was stained with HE.There was no change in Sham group.But in UUO group,there was tubular epithelial cells denaturalization,renal tubular expand or atrophy,interstitial macrophage and monocyt infiltration,interstitial expand,and there was no glomeruli change.While the change in irbesartan and eucommia group was slighter than that in UUO group.There was no significant difference between irbesartan group and eucommia group.3.The renal tissue Masson staining showed that there was interstitial expands fibrous tissues proliferation and there was no glomeruli change in UUO group.Compared with UUO group,the level of pathological change in irbesartan and eucommia groups was significantly lower.There was no significant difference between irbesartan group and eucommia group.4.The expression and distribution of MMP-2 were strong in Sham group,while in other groups lower in renal interstitial.Compared with UUO group,the level in irbesartan and eucommia groups was significantly higher.There was no significant difference between irbesartan and eucommia groups.[Conclusion] Eucommia inhibits the renal interstitial fibrosis lesion up-regulating the MMP-2 expression and blocking renal fibrosis.
2.Effect ofFeng-Shi-NingCapsule on Balance of Peripheral Blood Th17/Treg Cells and Local Inflammatory Media of Rats with Rheumatoid Arthritis

This study was aimed to observe the effects ofFeng-Shi-Ning (FSN) capsule on the balance of IL-17+CD4+T (Th17) / Foxp3+CD4+CD25+regulatory T (Treg) cells in peripheral blood and the inflammatory media in synovial fluid of rats with rheumatoid arthritis (RA), in order to further study the mechanism of FSN capsule in RA treatment. The collagen-induced arthritisⅡ (CIA) was used in the RA rat model establishment. The balance of Th17 and Treg cells in peripheral blood was analyzed by the flow cytometry method for effects of FSN capsule treatment. The levels of TNF-α, IL-6, IL-17A, IL-4 and IL-10 in synovial fluid were measured by cytometric bead array (CBA) method. The effects of the normal rats, CIA model rats and the rats treated by tripterygium wilfordii polyglycoside tablet (TPT) were compared. The results showed that compared with the model group, the percentage of Th17 cells in both the FSN capsule group and high-dose CIA group were obviously decreased (P < 0.05). The percentage of Treg cells had the tendency of increasing in all treatment groups with no significant difference (P > 0.05). There was no significant difference on Th17 and Treg cells among each treatment group and TPT group (P > 0.05). Compared with the model group, the high-dose and middle dose FSN can obviously reduce the levels of TNF-α, IL-6 and IL-17A in synovial fluid of rats in each FSN group (P < 0.05). There was no obvious upregulating effect on IL-4 and IL-10 in synovial fluid of each FSN group (P > 0.05). Compared with the TPT group, there were no significant differences on IL-6, TNF-α, IL-17A, IL-4 and IL-10 in each treatment group (P > 0.05). It was concluded that FSN can obviously reduce the percentage of Th17, decrease the secretion of IL-6, TNF-α and IL-17A in synovial fluid. However, it had no obvious effects on the percentage of Treg cells, or the secretion of IL-4 and IL-10 in synovial fluid. The mechanism of FSN capsule in RA treatment may be through regulating Th17 cells, adjusting body immune imbalance, and inhibiting the excessive secretion of inflammatory media in synovial fluid.
3.Effects of Shaoqiduogan on cellular immunity in mice with delayed-type hypersensitivity

Aim To investigate the effects of Shaoqiduogan(SQDG)on cellular immunity in mice with delayed-type hypersensitivity(DTH). Methods DTH model induced by 2, 4-dinitrochlorobenzene(DNCB)and cyclophosphamide(Cy)suppressed and potentiated DTH model were used. SQDG(30, 60 and 120 mg ? kg-1)were given intragastrically(ig)once daily for 7 consecutive days. Concanavalin A(Con A)-induced T cells proliferation was measured by MTT assay. The activity of IL-2 in culture supernatants of Con A-induced T cells proliferation was measured by testing its ability to support the proliferation of Con A stimulated thymocytes which was detected by MTT assay. Results In mice with DTH, SQDG(120 mg?kg-1)reduced ear swelling, thymus index, Con A-induced T cells proliferation and the activity of IL-2 in culture supernatants of Con A-induced T cells proliferation. The suppression and potentiation of DTH in mice were induced by given an intraperitoneal(ip)injection of Cy at the dose of 150mg?kg-1 on primary sensitization day and 250 mg?kg-1 three days before primary sensitization. SQDG(60 and 120 mg?kg-1)upregulated ear swelling, Con A-induced T cells proliferation and the activity of IL-2 in culture supernatants of T cells from mice with Cy-suppressed DTH; and downregulated levels of the aforementioned indexes in mice with Cy-potentiated DTH. Conclusion SQDG possesses a dual modulatory effect on cellular immunity in mice.
4.Feasibility and accuracy of quantification of fat content using iterative decomposition of water and fat with asymmetry and least squares estimation-quantitative fat imaging:a phantom study

Objective To validate the feasibility and accuracy of iterative decomposition of water and fat with asymmetry and least squares estimation-quantitative fat imaging (IDEAL-IQ) in fat quantification using fat-water model. Methods A homogeneous fat-water mixture model consisting of various known fat-fractions were described, and the fat fraction was 0.00, 0.01, 0.02, 0.04, 0.06, 0.08, 0.10, 0.14, 0.18, 0.22, 0.26, 0.30 g/ml respectively. A water-vaseline separated model was also described. IDEAL-IQ was performed. Thin slices were acquired for fat-water mixture model and repeated 3 days later. Nineteen slices of 14 mm-thick parallel to the water-vaseline boundary in 1 mm steps from vaseline to water
were acquired. The fat-fractions in 11 slices of fat-water mixture model were measured on FatFrac images. Accuracy was assessed through single sample t test or Kolmogorov-Sirmov test. Measured fat-fractions of the same known fat-fraction were assessed through independent samples t test between two scan times. Linear regression was used to assess the relationship between known fat-fractions and measured fat-fractions. Slices containing the water-vaseline boundary were measured with ROI in the middle of the FatFrac images. The relationship between measured fat-fractions and locations of scanning was exploded using curve fitting. Results (1) Fat-water mixture model: no significant difference(P>0.05) was found between measured fat-fractions and known fat-fractions when it was 0.00, 0.02, 0.06 and 0.08 g/ml with the measured fat-fractions 0.60%, (2.30 ± 0.60)%, (5.76 ± 1.40)%, (7.62 ± 1.40)% respectively for the first time. No significant difference(P>0.05) was found between measured fat-fractions and known fat-fractions when it was 0.00, 0.02, 0.10 g/ml with the measured fat-fractions 0.04%, (2.32 ± 0.60)%, (9.41 ± 1.00)%respectively for the second time. Measured fat-fraction was inlinear relation with known fat-fraction:Y=0.898X+0.224, r2=0.993, P<0.01, F=36 129.548.(2) Water-vaseline separated model: measured fat-fraction increased as scanning location changed, Y=0.045X2-0.499X-4.474, r2=0.978, P<0.05, F=350.623.Conclusions IDEAL-IQ can be used to quantify fat content with good repeatability and can accurately assess the actual fat content from the linearrelationship.
5.The correlation between serum estrogen level and the expression of mismatch repair genes in colonic mucosa

Objective To investigate the effects of estrogen on mismatch repiar gene expression in colonic mucosa in vivo. Methods A total of 42 healthy individuals underwent colonoscopy were enrolled in the study. Half an hour before colonoscopy examination, blood sample was taken for determining the serum estradiol (E2) level. N ormal colonic mucosal tissues determined by naked eye under colonoscopy examination were taken in the right hemi colon to detect HMLH1 and hMSH2 gene expression by semi-quantitative RT-PCR and immunohistochemistry staining. Then the correlation of serum E2 levels with hMLH1 and hMSH2 expression in colonic mucosa was analyzed. Results A bimodal curve was presented for the correlation between serum E2 level in healthy individuals and hMLH1 expression in colonic mucosa. A strong positive correlation of E2 level with hMLH1 expression in normal colonic mucosa was observed when serum E2 level was more than 45 pg/ml (For mRNA, P=0. 003, r=0. 701; for immunohistochemistry positivity index, P=0. 000, r=0. 874).However there was no correlation between E2 level and hMSH2 expression. Conclusion High serum E2 level might increase the hMLH1 gene expression in colonic mucosa in vivo.
6.Bladder symptoms and its impact on quality of life in type 2 diabetes mellitus

Objective To estimate the bladder symptoms, and explore its impact on patients′ quality of life in type 2 diabetes mellitus. Methods Overactive bladder symptom score (OABSS) was used to evaluate patients′ bladder symptoms, and Medical Outcome Study 12-Item Short Form Health Survey Scale (SF-12), Overactive Bladder Questionnaire Short Form(OABq-SF)were conducted to test the effect of bladder symptoms on general and disease-specific quality of life. Results Among the four bladder symptoms, nocturia was the most prevalent symptom (46.7%,479/1025 ), followed by urinary frequency (14.9%,153/1025 ),urinary urgency (14.2%,146/1025 ), urge urinary incontinence (7.9%, 81/1025). Nocturia and urge urinary incontinence significantly increased symptom bother (nocturia B=0.337,P<0.01;urge urinary incontinence B=0.437,P<0.01) and decreased disease-specific quality of life (nocturia B=-0.364,P<0.01;urge urinary incontinence B=-0.346, P<0.01) in these patients. Meanwhile, urge urinary incontinence was negatively associated with general mental health (B=-0.255, P=0.011). Conclusions The prevalence of bladder symptoms in type 2 diabetes patients was high, and was found to be associated with patients′general and disease-specific quality of life. Health care providers should be aware of the high prevalence of bladder symptoms and its impact on QOL to facilitate early diagnosis and treatment of bladder symptoms during diabetic care.
7.Application of online tests through two-dimensional code in the course of Fundamental Nursing

Objective To explore the application effects of online tests through two-dimensional code in the course of Fundamental Nursing. Methods A total of 79 nursing students in 2016 were included in the study. Compared with traditional method in the first term, online tests through two-dimensional code were implemented in the second term. The students′ appraisal on online tests through two-dimensional code was investigated with a self-designed questionnaire. Results The students showed a positive attitude to online tests through two-dimensional code. 97.3%(71/73) of the students thought online tests through two-dimensional code was easy to implement, and 93.2%(68/73) of them agreed that online tests help them to know how they learned and improved the efficiency and quality of class. Conclusions Online tests through two-dimensional code, which are novel, energy-saving and environmental friendly,can improve students′learning interest and enhance learning effect.Therefore,it is worthy to be applied and recommended.
8.Clinical Application Value of Lactobacillus Plantarum PS128 in Patients with Anxiety Disorders

Objective:
PS128 is a novel psycho biotic strain, it has been reported to play an important role in neuropsychiatric disorders. This study investigated the clinical effect of PS128 supplementation on patients with anxiety.
Methods:
A total of 200 patients with anxiety were recruited, and divided into two groups (n = 100/group). The control group received oral treatment with citalopram, and the PS128 group received PS128 capsules based on citalopram treatment. Hamilton Anxiety Scale (HAMA) and Self-Rating Anxiety Scale (SAS) were used to evaluate the anxiety levels.After 2 months of continuous administration, clinical efficacy was evaluated according to HAMA score.
Results:
There was no significant difference in HAMA and SAS scores between the two groups before treatment. With the treatment prolonged, the HAMA and SAS score decreased gradually in both control and PS128 groups, and the decrease rate of PS128 group was significantly greater than that of the control group. The clinical effective rates of PS128 group were higher than those in the control group, high levels of clinical cure rate were also detected in the PS128 group. Compared with the control group (22%), the incidence of adverse reactions was significantly reduced for patients in the PS128 group (4%).
Conclusion
The treatment effect of citalopram combined with PS128 against anxiety is satisfactory clinically. It can greatly improve the anxiety symptoms of patients, increase the cure rate, reduce adverse reactions.
9.Effects of different dialysates on apoptosis and expression of PKCδ of U937 cell line

Objective To investigate the effects of different dialysates on expression of protein kinase C-δ (PKCδ) and apoptosis of U937 cell line. Methods Different dialysates were added into culture fluid with U937 cell line at exponential phase of growth, and groups were divided: fluid A+fluid B group (dialysate A+dialysate B), fluid A+fluid B+rottlerin (PKCδ specific inhibitor)group, fluid A+powder B group (dialysate A+powder B) and fluid A+powder B + rottlerin group. Besides, blank control group and normal control group were established. Cells were harvested 24 h and 48 h after treatment, morphological changes were observed by Hoechst33258 fluorescence staining, cell apoptosis was measured by Annexin-V-FITC/PI double staining, and expression of PKCδ mRNA and protein was detected by RT-PCR and Western blotting, respectively. Results Cell apoptosis significantly increased in fluid A+powder B group, with typical morphology of apoptosis. After treatment for 24 h and 48 h, cell apoptosis rates in fluid A+powder B group were significantly higher than those at corresponding time points in blank control group , normal control group and fluid A+powder B+rottlerin group (P<0.05). Compared with normal control group, blank control group and fluid A+powder B+rottlerin group, the expression of PKCδ mRNA and protein of U937 cells in fluid A+powder B group were significantly increased (P<0.05). There was no significant difference in cell apoptosis rates and expression of PKCδ mRNA and protein between fluid A+fluid B group and blank control group, normal control group and fluid A+fluid B+rottlerin group (P>0.05). Conclusion Fluid A+powder B can significantly increase apoptosis of U937 cell line, the mechanism of which may be associated with the up-regulation of expression of PKCδ. Compared with fluid A+powder B, fluid A+fluid B is superior in reducing apoptosis of peripheral blood monouclear cells.
10.Overview of Gp96 mediated immunity.

As a member of the HSP90 family, heat shock protein (HSP) Gp96 is one of the most abundant proteins in the endoplasmic reticulum (ER), which displayed important molecular chaperones function in cells. Gp96 can stimulate the production of cytokines by activating the antigen presentation cells (such as dendritic cell, et al) in innate immunity. It is capable of eliciting an antigen-specific cytotoxic T lymphocyte (CTL) immune response to eliminate pathogens and tumors by facilitating antigen cross-presentation in adaptive immunity. Gp96 is also an ideal adjuvant in many recent researches. Here, we review the progress that addresses the role of biological characteristics, immunogenic mechanism that may be involved in the induction of anti-infection immune response and antitumor immunity, which may guide the new vaccine strategies with the knowledge of Gp96-antigen complexes.
