1.Effects of HIF - 1α on mitochondrial function of cardiomyocytes stimulated by lipopolysaccharide

ObjectiveTo investigate the effects of HIF-1α,on mitochondrial function of cardiomyocytes stimulated by lipopolysaccharide (LPS).MethodsCultured neonatal cardiomyocytes were randomly (random number) distributed to four groups (n =5).Control group:the neonatal cardiomyocytes were cultured in normal condition without any treatmeat; LPS group:cardiomyocytes were exposed to 1 μg/ml LPS for 2 h; YC-1 + LPS group:cardiomyocytes were exposed to 4 μmol/L YC-1 for 8 h before LPS treatment; YC-1 group:cardiomyocytes were exposed to 4μmol/L YC-1 for 8 h.As indexes of mitochondrial function,MMP,the content of ATP and the activity of cytochrome oxidase were measured,COX1/2 protein expression was determined by Western blotting.Results Compared with control group,LPS decreased MMP,the content of ATP and the activity of COX obviously ( P <0.05 ),depressed the expression of COX- 1 protein ( P < 0.05 ),induced the expression of COX-2 protein ( P < 0.05 ).Compared with the LPS group,MMP,the content of ATP and the activity of COX were markedly attenuated ( P < 0.01 ),the expression of COX-1 was significantly increased and the expression of COX-2 was decreased in theYC-1 +LPS group (P < 0.01 ).ConclusionsYC-1 attributes to the dysfunction of mitochondrial of myocyte by alterating the expression of COX-1 and 2, HIF-1α may attenuate the mitochondrial dysfunction of cadiomyocytes stimulated by LPS.
2.Clinical Study of Acupuncture Intervention in Gastroscopy Side Reaction by Superficial Needling at Point Fenglong

Objective To investigate the clinical intervention effect of superficial needling at point Fenglong on gastroscopy side reaction.Methods Eighty-two gastroscopy patients were randomly allocated to treatment and control groups, 40 cases each. The treatment group received gastroscopy and superficial needling at point Fenglong, and the control group, gastroscopy and sham acupuncture at point Fenglong. The pain score and the incidence of nausea and vomiting were observed in the two groups during gastroscopy.Results There was a statistically significant difference in the pain score between the two groups (P<0.05). The incidence of nausea and vomiting was 31.7% in the treatment group and 65.9% in the control group; there was a statistically significant difference between the two groups (P<0.05).Conclusion Superficial needling at point Fenglong can effectively reduce the incidence of gastroscopy side reaction.
3.Clinical effect of laparoscopy combined with leuprorelin treatment on mild endometriosis and infertility and fertility function of patients

Objective To investigate the clinical effect of laparoscopy combined with leuprolide on patients with mild endometriosis and infertility and effects on fertility function of patients. Methods A total of 60 patients with mild endometriosis and infertility in our hospital were randomly divided into the control group (n=30) and the observation group (n=30). The control group was treated with laparoscopic surgery, the observation group was treated with leuprorelin treatment, and the levels of FSH, E2, luteinizing hormone and PROG were tested by automatic biochemical analyzer between two groups before and after treatment, the pregnancy rate as assessment of reproductive function in patients. Clinical curative effect and effect on fertility of 2 groups were compared. Results Levels of FSH, E2, LH and PROG in the observation group were lower than those in the control group (P<0.05) in 3 months after treatment, and the pregnancy rates in the observation group after 1 months, 2 months and 3 months were higher than that in the control group (P<0.05). Conclusion The treatment of Laparoscopy combined with Leuprolide for mild endometriosis and infertility in patients has ideal effect, and is worthy of popularization and application.
4.Clinical Application of Multi-Planar Reconstruction with 64-slice Spiral CT on Protrusion of Lumbar Disc

Objective To investigate the value of multi planar reconstruction (MPR) with 64-slice spiral CT for diseases of lumbar disc, vertebra and accessory etc. Methods 50 cases of MPR were compared with 64-slice spiral CT on protrusion of lumbar disc at radom. Results MPR with 64-slice spiral CT showed not only the protrusion of lumbar disc but also the diseases of the vertebra,the accessory, and the adjacent tissue. Conclusion MPR with 64-slice spiral CT has a good value in diagnosis of disease of lumbar disc, vertebra, accessory and the adjacent tissue.
5.Effect of CHOP/GADD153 on cardiomyocyte apoptosis induced by angiotensin Ⅱ in vitro

AIM: To investigate the relationship between alteration of CHOP/GADD153 protein expression and cardiomyocyte apoptosis induced by angiotensin Ⅱ (AngⅡ), and inhibitory effects of CHOP/GADD153 antisense oligodeoxynucleotide (anti-ODN) on apoptosis in vitro.METHODS: Cultured neonatal cardiomyocytes were exposed to AngⅡ with or without preincubation of CHOP/GADD153 anti-ODN. Variability of myocytes was measured by MTT assay, the lactate dehydrogenase (LDH) release was also detected, the percentage of annexin V positive myocytes was monitored by flow cytometry as apoptosis rate, CHOP/GADD153, Bcl-2 and Bax expressions were determined by Western blotting. The changes of erythrocytic membrane band 3 protein and intraery-throcytic and extrar rythrccytic gases and electrolytes were studied in 69 cases of cor pulmonale and 50 normal subjects.It was found that in the patients of cor pulmonale accompanied with type Ⅱ respiratory failure,the relative low level of erythrocytic membrane band 3 protein and the restriction of HCO-3/Cl-exchange were the factors to aggravate CO2 retention and respiratory acidosis,relative intraerythrocytic alkalosis resulted from the relative increase of intra-erythrocytic HCO-3([HCO-3]),and prompt adminstration of oxygen to cor pulmonale patients with hypoxemia could not only improve extraerythrocytic acid-base imbalance but also increase intraerythcocytic P5O2 and the tissue capacity to store oxygen.
8.Language analogy teaching method by French and English comparison

Our college has applied French-English analogy-educating method ( trilingual education )to the medicinal French bilingual-educating course by means of the interactions of different languages. As the kin relations between English and French in the axea of medicine, as well as their interactions from the origin to the modem medical applications, it can be an effective method to use analogy-educating method in the French study course in medicine, which means that the multi-lingual interactions can be used in the bilingual, even the multi-lingual educating activities of bilinguai educating process of other subjects.
9.Role of NHE-1 in the proliferation and apoptosis of pulmonary artery smooth muscle cell in rats

AIM: To evaluate the role of Na+/H+ exchanger-1(NHE-1)in the proliferation and apoptosis of pulmonary artery smooth muscle cells in rats. METHODS: Twenty Wistar rats were equally randomized into the control group and 3-week hypoxic group. Intracellular pH (pHi) of the smooth muscle was determined with fluorescence measurement of the pH-sensitive dye BCECF-AM and the expression of NHE-1 mRNA was detected with RT-PCR. The primary culture of pulmonary artery smooth muscle cell in vitro was performed. In situ cell death detection kit (TUNEL) was used to study the effect of specific NHE-1 inhibitor, dimethyl amiloride (DMA), on the apoptosis of muscle cells which had intracellular acidification. RESULTS: pHi value and expression of NHE-1 mRNA of pulmonary artery smooth muscle cell were significantly higher respectively in the hypoxic group than those in the control group (P<0.01). DMA elevated the apoptotic ratio significantly. The effect was enhanced when DMA concentration was augmented and the time was prolonged. CONCLUSION: With the function of adjusting pHi, NHE-1 may play an important role in the proliferation and apoptosis of pulmonary artery smooth muscle cells.
10.The role of reactive oxygen species and GADD153 protein in angiotension Ⅱ-induced eardiomyocyte apoptosis in vitro

Objective To examine the role of the reactive oxygen species (ROS) and GADD 153 protein in angiotensin Ⅱ (Ang Ⅱ)-induced cardiomyocyta apeptosis in vitro.Methods Cardiomyocytes were isolated from ventricles of healthy 1-2 day old Wistar mrs of both sexes weighing 5-10 g and cultured in high glucose culture medium in an incubator filled with 5% CO2 at 37℃.The cardiomyocytes were randamiy allocated to one of 9 groups (n=5 each): group Ⅰ control (C); group Ⅱ ,Ⅲ,Ⅳ: cardiomyocytes were exposed to Ang Ⅱ 100 nmol/L for 6 h,12 h or 24 h respectively ( Ang Ⅱ 6,Ang Ⅱ12,Ang Ⅱ24) ; group Ⅴ:cardiomyocytes were preincubated with N-acetyl-L-cysteine (NAC) 5 mmol/L for 2 h before being exposed to Aag Ⅱ 100 nmol/L for 24 h (NAC + Ang Ⅱ); group Ⅵ:cardiomyocytes were exposed to NAC 5 mmol/L for 24 h (NAG); group Ⅶ:cardiomyocytes were tranfected with GADD153 antisense oligo-denxynueleotida (onti-ODN) 10 μmol/L for 24 h before being exposed to Ang Ⅱ 100 nmol/L for 24 h; group Ⅷ: cardiomyocytes were transfected with missense oligo-deoxynucleotide (mis-ODN) 10 μmol/L for 24 h before being exposed to Ang Ⅱ 100 nmol/L for 24 h and group Ⅸ: cardiomyocytes were exposed to transfection agent of same concentration for 24 h.Viability of myocytes was measured by MTT assay; ROS production was determined by spectropbotometry; apoptosis in cardiomyocytes was detected by staining with Hoechst33342; the percentage of Annexin V positive and PI negative myocytes was measured by flow cytometry as apoptosis rote and GADD153 protein expression was determined by Western blotting.Remits The viability of myocytes was significantly lower,while the ROS production,apoptosis rate and GADD153 protein expression in myocytes were significantly higher in group Ⅱ,Ⅲ,and Ⅳ(Ang Ⅱ6,Ang Ⅱ12,AngⅡ24) than in control group (P<0.05).The cell viability was significantly higher and the ROS production,apoptosis rate and GADD153 protein expression in myocytes were significantly lower in groupⅤ(NAC+AngⅡ)and Ⅶ(anti-ODN + Ang Ⅱ) than in group Ang Ⅱ24(Ⅳ).Conclusion Angiotensin Ⅱ induces cardiomyocyte apoptosis by increasing ROS production and up-regulating GADD153 protein expression.
