Objective To investigate the effect of electric stimulation applied during absolute refractory period on contraction and relaxation and Ca 2+ transient of ventricular myocytes from normal guinea-pigs. Methods The ventricular myocytes of guinea-pigs were obtained by enzyme digesting, and the extent of the contraction and relaxation and Ca 2+ transient were recorded through the motion edge detection system. The Ca 2+ transient was reflected by fluorescence ratio (360/380) ( F 360/F 380). Results The ventricular myocytes from the normal guinea-pigs: ①The contraction extent of guinea-pig ventricular myocytes increased by (16.55?5.49)%. The peak velocity of shorting (contraction) and the peak velocity of relengthening (relaxation) increased by (17.43?7.08)% and (19.74?9.08)%, respectively(n=10); ②The extent of F 360/F 380 increased (25.79?6.88)%. The peak velocity of F 360/F 380 increased by (29.47?9.25)% and (22.52?7.81)% during shortening and relengthening, respectively(n=10). The ventricular myocytes from failure guinea-pigs: ①The contraction extent of guinea-pig ventricular myocytes increased by (15.53?5.31)%. The peak velocity of shortening (contraction) and the peak velocity of relengthening (relaxation) increased by (10.60?3.02)% and (23.32?8.26)%, respectively(n=6); ②The extent of F 360/F 380 increased (16.82?7.03)%. The peak velocity of F 360/F 380 increased by (16.27?5.91)% and (10.32?2.46)% during shorenting and relengthening, respectively(n=6). Conclusion Appropriate electric stimulation applied during absolute refractory period might strengthen the contracting and relaxing function of normal and failing ventricular myocytes in guinea-pigs.

Objective To investigate the role of ECRG2,a novel tumor suppressor gene,in spindle assembly checkpoint. Methods Using siRNA approach to deplete the expression of ECRG2, using immunofluorescence to test the distribution of ECRG2,using Western blotting to examine the expression cell cycle proteins. Results ECRG2 localized to centrosomes during interphase and kinetochores during mitosis.Further analysis revealed that ECRG2 participates in the spindle assembly checkpoint.Depletion of ECRG2 abolished the spindle assembly checkpoint. Conclusion Our results indicated that ECRG2 is important for ensuring spindle assembly checkpoint, accurate chromosome segregation, and its depletion may contribute to chmmosome instability and aneuploidy in human cancers.

Objective:To investigate the animal model of guinea pig chronic heart failure and compensatory hypertrophy. Methods:Thirteen chronic heart failure models were made by placing a constricting band around the ascending aorta of guinea pigs, raised in the same house of 10 normal guinea pigs. The contrasting indexes in 6 weeks include: clinical features, hemodynamics, the mass ratio of ventricular over body, the width of left ventriculat hypertrophy. Results:One model died, 4 models without dyspnea showed increasing in left ventricular pressure and the mass ratio of ventricular over body, whereas the left ventricular end diastolic pressure showecl no significant different. Eight models with dyspnea showed striking increasing in left ventricular pressure and left ventricular end diastolic pressure, as well as the mass ratio of ventricular over body and the width of left ventricular hypertrophy. Conclusion:The animal model of guinea pig chronic heart failure and compensatory hypertrophy may be formed by constricting the ascending aortas of guinea pigs for 6 weeks.

AIM: To investigate the feasibility of transplanting endothelial progenitor cells(EPCs) obtained from spleen in vascular endothelium repairmen after vascular injury.METHODS: EPCs were isolated by using a Ficoll density gradient centrifugation, and were cultured in plate. The endothelial characteristics of EPCs were identified by immunochemical staining and fluorescent labeling.Dil-Ac-LDL labeled spleen-derived EPCs were transplanted into the rats by intravenous injection directly after induction of arterial injury and again 24 hours later.Rats received FITC-labeled lectin intravenously before euthanasia. The distribution of fluorescent labeled EPCs was traced. The morphology of arterial intima and media was studied by optical microscopy and image analysing system.RESULTS: The adherent cells were considered EPCs that showed spindle shape and form blood-siland-like structures during development. The adherent cells had many endothelial characteristics.Fluorescent labeling showed that the intravenously injected EPCs specifically restricted to the vascular injury site, and lectin binding confirmed the endothelial phenotype. The ratio of neointimal/media area in EPCs transplantation group was obviously reduced than that in injury group and M199 group(0.82?0.09 vs 1.52?0.21,1.48?0.19,P

OBJECTIVETo study the anatomy basis for the clinical application of the adjacent horn shaped perforator fasciocutaneous flap for the reconstruction of small and medium-sized defects in the trunk area.
METHODS(1) Ten adult antiseptic cadavers (20 sides) were perfused with red latex. The skin blood supply, line of the blood vessels, branches in accordance with the distribution and crossing were observed. (2) Fifteen cases with defects in the trunk were treated with the adjacent horn shaped perforator fasciocutaneous flaps. The defects size ranged from 5 cm x 5 cm to 13 cm x 13 cm with the size of the flaps ranging from 10 cm x 6 cm to 35 cm x 15 cm.
RESULTSThe trunk skin is supplied by mainly 17 groups arteries such as thyrocervical trunk, internal thoracic artery, posterior intercostal arteries, superior epigastric artery, arteria epigastrica inferior, lumbar arteries, and so on. The perforators (diameter > 0.5 mm) numbers are about 20, 40, 24, 6, on the chest, abdomen and perineum, upper back, waist, respectively. All the flaps survived completely with primary healing both on donor and recipient sites. The flaps color, texture, function and appearance were satisfactory during the follow-up period of 1-24 months.
CONCLUSIONSThe adjacent horn shaped perforator fasciocutaneous flap should be designed flexibly. The defects in the donor sites could be closed directly without skin graft. It is an effective, easy and ideal method for the reconstruction of large defects in the trunk.
