1.The effects of different opiate peptides and ACTH on the phagocytosis of mouse macrophages
Chinese Journal of Immunology 1986;0(04):-
Peritoneal macrophages were obtained from normal LACA mice and the phagocy-tosis of neutral red was measured after 24 hr.incubation with different opiate peptidesand ACTH.?-endorphin(10~-(10)-10~_6M)and dynorphin A(10~_(10)-1(10~_6M)were found tostimulate the phagocytosis and these increases in phagocytosis could be reversed byopiate receptor blocker,naltrexone(10~_5M).[D-Ala~2,D-Leu~5]-enkephalin(10~_(12)-10~_6M),however,had no effect on the phagocytosis.ACTH(10~_(10)-10~_6M)was fo-und to have suppressive effect on the phagocytosis. These results suggest that diffe-rent opiate peptides may cause different effects on phagocytosis of macrophages andthe increase in phagocytosis induced by ?-endorphin and dynorphin appears to be me-diated by opiate receptors on peritoneal macrophages.
2.EFFECTS OF CILIARY NEUROTROPHIC FACTOR ON CELL CYCLE AND EXPRESSION OF Fos PROTEIN IN ASTROCYTES IN VITRO
Acta Anatomica Sinica 1957;0(04):-
Objective To explore the effects of ciliary neurotrophic factor on cell cycle and expression of Fos protein in cultured astrocytes from cerebral cortex of rat in vitro Methods Astrocytes from cerebral cortex of rat were cultured in vitro. The cultured astrocytes were treated with ciliary neurotrophic factor(CNTF),and then cell cycle was measured by flow cytometer and Fos protein was examined by means of immunocytochemistry at different intervals. Results At 6h following CNTF treatment, astrocytes cell cycle process was promoted obviously, the percentage of cell in G-1/G-0 cycle phase was reduced, while that of G-2/M and proliferation index(PI) increased. The proliferous effect peaked at 12h,and then gradually declined at 24h,48h,but PI was higher than that of control group. The expression of Fos protein was enhanced from 2h to 24h after CNTF treatment, the pretreatment of dexamethasone inhibited the expression of Fos protein. Conclusion CNTF may promote proliferation and the expression of Fos protein in astrocytes.

6.Impact of the evaluated plasma homocysteine on T2DM patients with diabetic nephropathy and diabetic retinopathy
Journal of Chinese Physician 2014;(4):491-494,497
Objective To investigate the impact of plasma homocysteine ( Hcy) on type 2 diabetics ( T2DM) patients with diabetic nephropathy ( DN) and diabetic retinopathy ( DR) , and to explore the correlation among the levels of plasma Hcy , vascular endothelial growth factor (VEGF), homeostasis model assessment of insulin resistance (HOMA-IR), and insulin resistance (ISI). Methods A total of 241 cases of diabetes was recruited as subjects according to the 2010 China guideline for type 2 diabetes. The history of smoking, hypertension, duration of diabetes, height, and weight were measured.Fasting blood were collected to analyze the levels of fasting insulin (FINS), fasting plasma glucose (FPG), glycated hemoglobin a1c (HbA1c), plasma Hcy (Rate method), and VEGF [enzyme-linked immunosorbent assay, ELISA)].Results The age, body mass index (BMI), the history of hyperten-sion, duration of diabetes, HbA1c, homeostasis model assessment of insulin resistance (HOMA-IR), microalbuminuria (MAU), and VEGF were positively correlated with the levels of plasma Hcy or VEGF .The levels of ISI were reduced with the increased levels of Hcy or VEGF.The history of smoking was positively correlated with the VEGF .Spearman correlation analysis demonstrated that the levels of plasma Hcy were positively correlated with the levels of VEGF ( r =0.498 , P <0.01 ) .In the groups of diabetic retinopathy and early diabetic nephropathy , it could find that increased duration of diabetes , increased IR, higher levels of HbA1c and VEGF, and the levels of plasma Hcy≥10μmol/L might be the risk factors of DR ( P <0.05 ) .Only the history of hypertension and high levels of VEGF and Hcy in early DN showed a strong correlation ( P <0.05 ) .Conclusions Spearman correlation analysis demonstrated that the levels of plasma Hcy were positively correlated with the levels of VEGF .Higher levels of Hcy and VEGF might be the risk factors of the early DN and DR.At the same time, lower level of ISI might be the risk factor of DR , but the reduction of ISI might not be the main risk factor of the early DN .
7.Blood pressure regulation of statins
Chinese Journal of Clinical Pharmacology and Therapeutics 1999;0(04):-
Statins is first choice of hypercholesteroiemia. Recently, clinical investigations have shown that statins can correct blood pressure in hypertension patients accompanied with lipid abnormalities. This result can be observed not only in untreated patients but also in patients treated with antihypertensive drugs. But some researches did not support this result completely. To some extend, the mechanisms responsible for the hypotensive effect seem to be largely independent on the effect of statins on lipid profile, and may be related to the improvement of endothelial function and the regulation of angiotensin II receptors.

Objective To observe the effects of mouse indoleamine 2,3-dioxygenase (IDO) gene on CD4+ T cells by transfecting an eukaryotic expression vector containing mouse IDO gene fused with enhanced green fluorescent protein (pEGFP-N1). Methods The immature dendritic cells (imDCs) derived from mouse bone marrow were cultured in vitro and morphologically identified by transmission electron microscope, scanning electron microscope and flow cytometry. The immature dendritic cells were transfected by DOTAP liposome, and the expression of green fluorescence protein was confirmed with inverted fluorescence microscope. The spleen-derived CD4+ T lymphocytes from mice were isolated and purified in vitro, and then carried to mixed lymphocyte reaction (MLR) with 3H-TdR incorporation. The effect of IDO gene on the proliferation of spleen-derived CD4+ T lymphocytes of the allergized mouse challenged by ovalbumin was assayed by 3H-TdR MLR incorporation, and the apoptosis of spleen-derived CD4+ T lymphocytes was assayed by TUNEL. Results The proliferation of spleen-derived CD4+ T lymphocytes of the allergized mouse challenged by ovalbumin were inhibited by the imDCs transfected with IDO gene, the number of CD4+ T lymphocytes treated with the imDCs transfected pEGFP-N1-IDO eukaryotic expression vector decreased significantly compared with that in control group (P

In the present study, A fluorescent imaging-based high-throughput screening method was developed for identifying anti-migratory compounds with 96-well Transwell plates. The correlation, precision and stability of this method were examined and the incubation time of dye Hoechst 33342 in addition to migration time was optimized. In addition, The inhibitory activity of anti-cancer drug paclitaxel on tumor cell migration was assayed and an IC50 value of 0.717 micromol x L(-1) was obtained. Using this method, 24 components from Rhizoma Alismatis were screened and one component with anti-migration activity was found. These results show that the new proposed method with good precision, stability and linear range has the potential to assay the inhibitory activity of anticancer compounds.

Chinese Journal of Experimental Ophthalmology 2015;33(2):142-148
Background Small incision lenticule extraction (SMILE) is increasingly used for myopia. However, the size of the incision varied in different operators. The influence of 2 mm micro-incision SMILE on postoperative diopter and aberrations is not clear yet. Objective The aim of the current study was to perform a clinical analysis of the efficacy, safety, stability and predictability of 2 mm micro-incision SMILE and investigate the influence of preoperative sphere and cylinder power on postoperative aberrations. Methods A serial cases observational study was carried out under the approval of Ethic Committee of Tianjin Eye Hospital and informed consent of each patient.Fifty-five eyes of 31 subjects with myopia and myopic astigmatism were enrolled in this study in Tianjin Eye Hospital from January to April 2013 and a 2 mm micro-incision SMILE was performed on the eyes.Follow-up visits were performed at 1 week,1 month and 6 months after operation.Uncorrected visual acuity (UCVA),best corrected visual acuity (BCVA),manifest and cycloplegic refraction were examined at each postoperative time points to evaluate the efficacy, safety, stability and predictability of 2 mm micro-incision SMILE.Wave-front aberrations were measured by WaveScan aberrometer.Repeated measurement analysis of variance was used for the comparison of the differences of aberrations before and after the surgery. And the differences between various follow-up visits were compared with LSD-t test. The relationship between preoperative sphere or cylinder power and aberrations postoperatively were analyzed by Spearman rank correlation. Results At 1 week,1 month and 6 months postoperatively, the effective index was 1.2,and the safety indexes were 1.2,1.2 and 1.5 respectively. The spherical equivalent remained 0.00 D at each postoperative time point. And 96.4% and 97.8% of eyes were within ±0.5 D of intend correction and 100% were within ± 1.0 D of the intend correction at 1 month and 6 months postoperatively.Vertical coma (Z7) and spherical aberration (Z12) were significantly raised at 1 week,1 month and 6 months after surgery compared with the values preoperatively (Z7:t =9.668,10.607,9.772,all at P < 0.001 ;Z 12:t =-6.227,-6.923,-7.441,all at P<0.001).The Zernike coefficients were relatively symmetrical between various time points postoperatively (all at P>0.05).Third-order to sixth-order aberrations and higher order abberration (HOA) showed significant increase at various time points after the surgery compared with the values before surgery (all at P<0.05),and no significant differences were seen between various follow-up visits (all at P>0.05).Positive correlations were found between preoperative sphere and S3,S4 and HOA at 1 week,1 month and 6 months postoperatively (P<0.05).Negative correlations were found between preoperative cylinder and S3,S4 and HOA at postoperatively various time points (all at P<0.05).In addition, significant positive correlations were seen between Z7 and preoperative cylinder as well as between Z12 and sphere preoperatively, and negative correlations also were observed between Z7 and preoperative sphere as well as between Z12 and cylinder preoperatively at each follow-up visits(all at P<0.05).Conclusions SMILE with 2 mm micro-incision is a safe, effective, stable and predictable procedure for myopic and myopic astigmatism eyes. The aberrations and Zernike terms remain stable after surgery until 6 months although these parameters present elevation to a certain extent.Preoperative SE and astigmatism power affect postoperative aberrations.
