During digital impression, to scan the traditional dental casts is still the mainstream technology for data acquisition technology. Future investigations on scanning the patient alveolar ridge and its surrounding soft tissue directly and digitally recording the jaw relation directly are warranted. To develop 3D-printed artificial teeth and base materials with proper strength, aesthetics, comfort levels as well as 3D printing of the artificial tooth and the base as the final denture will make digital denture technology more mature.

Objective To set up the method of estimation of age for Chinese girls at the age of 11-16 years according to their characteristic of radiographic changes in articulations of bone. Methods The samples of the X-ray photograph were come out of 6 articulations of humeri, cubiti, manus, coaxe, genus and ankle from 150 girls, ranging in age from 11-16 years old, in Henan province, North of China. The radiographic characteristics of osteoepiphysis at these joints were observed carefully and classified. The data were analyzed with the SPSS package. Result Regression equation was made for radiographic determine of age for Chinese girls at the age of 11-16 years. Conclusion The equation can be used for forensic practice about the determine of age.

Objective To investigate the expression of TMS1/ASC gene induced by gemcitabine(GEM) in pancreatic carcinoma cell line PANC-1.To study the relationship between cysteine aspartase(caspase-1),nuclear factor-κB(NF-κB) and the expression of TMS1/ASC.Methods The pancreatic carcinoma cell line PANC-1 was cultured in Dnlbecco's modification of Eagle's medium(DMEM).Methyl thiazolyl tetrazolium (MTT)method was used to measure the effect of GEM at different time points(24,48 h)at different concentrations(1,2,4,8,16 μg/ml) on growth of PANC-l.RT-PCR was used to detect the expression of TMS1/ASC mRNA stimulated by medium alone and by GEM(4.27μg/ml)for 24 h and 48 h.Western blot analysis was performed with inhibitory protein of NF-κB multiclonal antibody,caspase-1 multiclonal antibody and β-actin monoclonal antibody to observe the expression of β-actin,caspase-1 and IκBα in GEM group and in the control group.The activation state of caspase-1 and NF-κB was examined.Results GEM inhibited the growth of PANC-1 cells in a concentrationand-time-dependent manners and its half maximal inhibitory concentration(IC50)was 4.27 μg/ml on 24 h.The expression of TMS1/ASC was 0.3 ±0.004 and 0.63 ±0.007 respectively in GEM group while it was 0.1 ±0.001 and 0.21 ± 0.006 in the control group on 24h and 48h.The difference between the 2 groups at the same time point had statistical significance (P ＜ 0.01).Western blot showed that GEM caused the activation of caspase-1.The expression of IκBα had no obvious differencebetween the 2 groups.GEM couldn't induce the activation of NF-κB.Conclusions GEM can inhibit proliferation of PANC-1 cells and induce their apoptosis.The drug sensitivity decreased with prolongation of exposure time.GEM might induce and increase the expression of TMS1/ASC,which might influence the apoptosis of the cells later.The apoptosis of PANC-1 cells induced by GEM is dependent of caspase-1 signaling pathway and independent of NF-κB signaling pathway.

Objective:To investigate the structural alterations of mitochondria and its role in neutrophil death induced by ONO-AE-248.Methods:Human neutrophils were cultured in vitro with ONO-AE-248(5?10-5 mol/L)and medium alone. Transmission electron microscopy(TEM) was used to detect the structural alterations of mitochondria and the level of mitochondria membrane potential by flow cytometry using mitocapture dying.Results:ONO-AE-248 resulted in extremely swollen mitochondria within 6 hours. Meanwhile, a rapid loss of mitochondrial membrane potential of neutrophils occurred, especially in 3 hours and 6 hours. There were obviously differences between spontaneous apoptosis and non-apoptosis programmed cell death induced by ONO-AE-248.Conclusion:The experiment results suggest that changes of mitochondrial structure and function be typically morphological, physiological and biochemical features in this unique form of neutrophil death, and that the mitochondrial pathway might play a more important role in ONO-AE-248-induced death of neutrophils.

Objective:To investigate the roles of apoptosis-associated speck-like protein containing CARD(ASC),one of differentially expressed proteins in neutrophil death induced by ONO-AE-248.Methods:Human neutrophils were cultured in vitro with or without ONO-AE-248(5?10-5 mol/L).The expression of ASC mRNA was detected by RT-PCR.PMN proteins were extracted at 12 h and then separated by 2D electrophoresis.20 differentially expressed proteins were selected and determined by PDQest 2-DE software.Results:ONO-AE-248 decreased the expression of ASC mRNA strikingly in 2 hours;ASC was one of the important differentially expressed proteins between spontaneous apoptosis and non-apoptosis programmed cell death induced by ONO-AE-248 in 12 hour and ASC protein point was weaker in ONO-AE-248 group.Conclusion:ASC might act as double switch that leads to apoptosis of neutrophils in the high expression and removes the forbiddance of pathway of ONO-AE-248 signals in the low expression,resulting in non-apoptosis programmed cell death of neutrophils.

Objective To observe the clinical effects of insulin glargine combined with novonorm in treatment of patients with newly diagnosed type 2 diabetes. Methods 112 cases of newly diagnosed type 2 diabetes patients were randomly separated into observation group(56 cases)and control group(56 cases). Observation group was treated with insulin glargine combined with novonorm. Control group was treated with novolin30R. The levels of FPG,2hPG and HbA1c before and after treatment,the control time of blood sugar,amounts of insulin and incidence of low blood sugar were observed in both groups. Results Compared with pre-treatment, FPG,2hPG and HbA1 c were significantly decreased (all P ＜0.05) after treatment, but there were no significant differences in both two groups (P ＞0. 05). The control time of blood sugar, amounts of insulin and incidence of low blood sugar in observation group were significantly lower than those in control group (all P ＜ 0.05). Conclusion Insulin glargine combined with novonorm in treatment of patients with newly diagnosed type 2 diabetes could effectively control blood sugar,shorten the control time of blood sugar,decrease amounts of insulin,and low incidence of low blood sugar.

Objective To construct four types of glucagon-like peptide-1 (GLP-1) and human serum albumin (HSA) fusion proteins that can be realeased at different rate in vivo by introducing protease cleavage sites between these two moieties.The therapeutic effect and release rate are studied to achieve balanced pharmacokinetics ( PK) and pharmacody-namics ( PD) of GLP-1 and HSA fusion proteins.Methods The gene with different polypeptide joint of GLP-1 and HSA fusion proteins were synthesized by overlap extension PCR amplification, cloned into expression vector pPIC9 and transformed into Pichia pastoris GS115.Then, fusion proteins were obtained by protein purification after being induced by methanol.The preliminary PK and PD of the fusion proteins were studied after purification.Results The fusion protein Gly2-GLP-1-GGGGG-HSA showed no release while Gly2-GLP-1-VTR-HSA, Gly2-GLP-1-SARSVRA-HSA, and Gly2-GLP-1-GRSRVTRSV-HSA showed a slow, medium and fast release rate, respectively, after incubation with furin.In vitro biological activity test results dispalyed that each type of fusion protein promoted insulin secretion of MIN6 cells.In vivo PK test indicated the half-life size of fusion proteins was the largest in Gly2-GLP-1-GGGGG-HSA, followed by Gly2-GLP-1-VTR-HSA, Gly2-GLP-1-SARSVRA-HSA, and Gly2-GLP-1-GRSRVTRSV-HSA.In vivo PD test exhibited hypoglycemic activity that was the highest in Gly2-GLP-1-VTR-HSA, followed by Gly2-GLP-1-SARSVRA-HSA, Gly2-GLP-1-GRSRVTRSV-HSA, and Gly2-GLP-1-GGGGG-HSA.Conclusion GLP-1 can be released from fusion proteins with full activity after the introduction of protease cleavage sites.Releasable fusion proteins at an appropriate release rate have the most balanced PK and PD.

Objective To study the PFGE type of Salmonella (S.) strains isolated from poultry production chains (hatching,breeding,slaughter,distribution and retail) of four cities in Heilongjiang province.Methods DNA collected from S.strains in 2012 was digested by Xba Ⅰ according to the standard PFGE protocol of US CDC.The PFGE patterns were then analyzed by BioNumerics software.Results The contamination of S.appeared most serious during the process of slaughtering (13.84％).PFGE was used to determine the genetic relationships between these isolates from poultry production chains,89 pulsotypes from 150 S.enteritidis isolates and 55 pulsotypes from 65 S.indiana isolates showed considerable diversity.The same pulsotypes ofS.enteritidis can be found between different food chains and cities.In contrast,no identical pulsotypes of S.indiana were found between different food chain and cities.In these four cities,the above said two kinds of S.were from different sources.The source of S.contamination in HLJ2 city had been traced back to the chain of poultry hatching.Conclusions The distribution of pulsetypes of the S.enteritidis and S.indiana isolates was from different regions and the dominant bands were also different between the chains of poultry production.Cross contamination existed in slaughterhouses and contamination can be traced back to the poultry hatching.

Objective To summarize the observation and nursing strategy of side effects for children wearing Ortho-K contact lens in order to improve the effects of treatment and reduce side effects. Methods A total of 1 066 myopic children wearing Ortho-K CL, who could recheck on time in Beijing Children′s Hospital from May 2013 to June 2014 were included. The reason of dyed cornea, dislocation, and discomfortable were analyzed, and corresponding nursing and healthy propaganda were performed. Results Among 1 066 children, 350 children (32. 8% ) wearing Ortho-K CL had epithelial dyed cornea after 6 months, and most of them appeared 1 month after wearing the lens. It would disappear after the treatment by levofloxacin eye drops, sodium hyaluronate eye drops and recombinant bovine basic fibroblast growth factor gel, stopping using the Ortho-K contact lens and providing healthy progaganda after 1 week. A total of 170 children (16% ) wearing Ortho-K CL had dislocation, in which 2 children slipped into conjunctival sac and it was taken out under guidance of nurse by parents. A total of 128 children (12% ) suffered from diplopia after wearing the lens, and 234 children(22% ) wearing Ortho-K CL had discomfortable feeling. No children had irreversible impairment and ocular infection. Conclusions According to the reason of side effects, appropriate nursing strategy and healthy propaganda can improve the success rate of Ortho-K contact lens, prolong their service time, contribute treatment effectiveness, and reduce its side effects.
