BACKGROUND:The long-term contraction of skeletal muscle can lead to the decrease of resting potential, affect its contraction, and result in the occurrence of fatigue. Whether acupuncture and injury-free magnetic lifting needle are effective on the recovery of resting potential? OBJECTIVE: To observe and compare the influence on the recovery of resting potential of skeletal muscle by taking acupuncture and magnetic needle as the recovery means.DESIGN: A controlled study.SETTINGS: College of Physical Education, Shanxi Normal University;Binzhou College of Shandong Province.MATERIALS: Thirty experimental toads, which were provided by Beijing University of Physical Education, were randomly divided into acupuncture group (n=10), magnetic needle group (n=10), magnetic needle+pressing group (n=10). Left and right semitendinosus muscles were taken as controls.METHODS: The in vitro toad's semitendinosus muscles (experimental side and control side) were treated with electrostimulation for 40 minutes,and then the resting potential of skeletal muscular cells were measured with the intracellular glass microelectrode technique. After the electrostimulation, filiform needle acupuncture, magnetic needle, magnetic needle plus pressing were also given.MAIN OUTCOME MEASURES: The resting potentials were observed before exercise, after electrostimulation and other treatments in the groups.RESULTS: ① The resting potentials of skeletal muscular cells after electrostimulation were decreased as compared with those before electrostimulation [experimental side: (59.48±7.37), (84.54±5.72) mV; control side:(59.43±7.14), (83.62±5.86) mV]. ② After the treatments of acupuncture,magnetic lifting needle, magnetic lifting needle plus pressing, the decreased resting potentials after electrostimulation were obviously recovered (P ＜ 0.01). ③ The recovered percentage of resting potential in the magnetic needle group was obviously smaller than those in the acupuncture group and magnetic needle+pressing group [(42.50±12.41)%, (53.71±10.93)%,(49.10±8.39)%, P ＜ 0. 01].CONCLUSION: Acupuncture, magnetic lifting needle, magnetic lifting needle plus pressing plays a role in promoting the recovery of resting potential of skeletal muscular cells after long-term electrostimulation, and the effect of simple magnetic needle is smaller than that of acupuncture and magnetic needle plus pressing.

AIM: To investigate the effects of estrogen interference on ab dominal adipose accumulation in C7BL/6J mice. METHODS: Estrogen was administrated to the ovariectomized mice (estrogen-deprived model) and the changes of abdominal adipose weight of the mice in the normal estrous cycle and pro-menopausal were observed. Mice of 3 months old were randomly divided into c ontrol, ovariectomized (OVX), OVX+estrogen 15-days (E d 15 ) and OVX+ estrog en 60-days (E d 60 ) groups. Each group had 10 mice. Mice were raised and sa crificed until 7 months old after ovariectomized. 5 months and 10 months old mic e were divided into control, estrous cycle normal + estrogen 60 days, pro-menop ausal and pro-menopausal+60 days groups. 17?-estradiol time-release pellets ( 0.18 mg/pellet) were given ic in 15 days and 60 days before mice w ere sacrificed. RESULTS: The abdominal adipose weight in OVX gro up increased significantly (P 0.05 ). There was no difference in the abdominal adipo se weight between control and estrous cycle normal + estrogen 60-days groups, a nd between pro-menopausal and pro-menopausal+ estrogen 60-days groups. CONCLUSION: Ovariectomy significantly influences the metabolism of abd ominal adipose that causes adipose accumulation. Estrogen interference, especial ly long-term administration, can attenuate the abdominal adipose accumulation. Exogenous estrogen interference has no effect on abdominal adipose accumulation on cycle normal and pro-menopausal mice.

Objective To establish an in vitro oxygen-glucose deprivation/reoxygenation axon injury model in rat hippocampal neurons in order to found a basis for research concerning the injury of oxygen glucose deprivation and axons regeneration. MethodsThe hippocampal neurons isolated from rats 24 h after born and cultured for 7 d were exposed to D-hanks solution with nitrogen gas instead of the original culture medium for 0.5, 1.0 and 1.5 h respectively, and then continue to be cultured in the original culture medium with oxygen. LDH content in the culture media was measured at 1, 5, 24, 48 and 72 h after reoxygenation. The morphological changes of neuron and axon were observed with inverted phase contrast microscopy. ResultsAfter oxygen-glucose deprivation/reoxygenation injury, hippocampal neurons became darker, swollen, and were with shorten axons. With the elapse of time, the LDH content was increased. The survival rate of hippocampal neurons was higher and the change of axon length was more obvious in the group of oxygen-glucose deprivation for 0.5 h than in the other groups. ConclusionAn in vitro oxygen-glucose deprivation/reoxygenation axon injury model in rat hippocampal neurons is successfully established.

Objective:To compare the proliferative and periodontal specific differentiation abilities of induced pluripotent stem cells (iPSCs) at different passages,and to investigate whether long term culturing would have a negative influence on their proliferation and specific differentiation capacity,thus providing a theoretical basis for further in-depth research on periodontal regeneration and the possible clinical applications of iPSCs.Methods:IPSCs derived from human gingival fibroblasts at passages 5,10,15 and 20 were recovered and cultured in vitro.Their morphology and proliferation rates were observed respectively.We further induced the iPSCs at different passages toward periodontal tissue under the treatment of growth/differentiation factor-5 (GDF-5) for 14 days through the EB routine,then compared the periodontal differentiation propensities between the different passages of iPSCs by detecting their calcified nodules formation by Alizarin red staining and assaying their relative periodontal tissue related marker expressions by qRT-PCR and immunofluorescence staining,including bone related markers:osteocalcin (OCN),bone sialoprotein (BSP);periodontal ligament related markers:periostin,vimentin;and cementum related markers:cementum attachment protein (CAP),cementum protein 1 (CEMP1).The untreated spontaneous differentiation groups were set as negative controls respectively.Results:iPSCs at different passages all showed a high proliferative capacity when cultured in vitro and turned into a spindlelike shape similar to fibroblasts upon periodontal specific differentiation.All iPSCs formed typical calcified nodules upon GDF-5 induction by Alizarin red staining in comparison to their untreated controls.The relative calcium deposition at all passages had been significantly upgraded under the treatment of GDF-5 (P5:t =2.125,P =0.003;P10:t =2.246,P =0.021;P15:t =3.754,P =0.004;P20:t =3.933,P =0.002),but no significant difference in their calcium deposition were detected within passages 5,10,15 and 20 (periodontal differentiation:F =2.365,P =0.109;spontaneously differentiation:F =2.901,P =0.067).Periodontal tissue related marker expressions of iPSCs at all passages had also been significandy upgraded under the treatment of GDF-5 (P ＜ 0.05),but still,no significant difference in their expression levels of periodontal tissue related proteins were detected within passages (BSP:F =0.926 7,P=0.450;vimentin:F=0.917 1,P=0.455;CEMP1:F=2.129,P=0.1367).Conclusion:Our results preliminarily confirmed that long term culturing won't influence the proliferation capacity and periodontal specific differentiation propensity of iPSCs,as they can still proliferate and differentiate toward periodontal cells with high efficiency upon growth factor induction after continuous passaging.Therefore,iPSCs could be recognized as a promising cell source for future possible application in periodontal tissue regeneration.

Objective:By exploring the epidemic level and related factors of depressive symptoms,to provide the references for improving mental health of the physical disabled.Methods:The data were from the 2013 China Health and Retirement Longitudinal Study database.Persons with self-reported physical disabled were selected as subjects,and a total of 974 valid ones were obtained,with the average age of (62 ± 10) yeats.The depressive symptoms were measured with the Center for Epidemiologic Studies Depression (CES-D),with the cut-off score of equal or higher than 10 as having depressive symptom.The independent variables included age,gender,education level,marital status,urban and rural areas,self-rated general health,self-rated heating,self-rated memorizing,life satisfaction,smoking,drinking,sleep duration,chronic diseases,activity of daily life,social activity,family support,and disability periods.Chi-square test was used to analyze the effect of each factor on prevalence of depressive symptoms.Binary logistic regression was used to analyze the effect of multi-factors.Results:The prevalence of depressive symptom among the physical disabled was 46.1％.The subjects who were female,single,suffered from chronic disease,without family support,short-time sleeper were more likely to have depressive symptoms than the controls (OR =1.35,1.62,1.60,1.67,2.58;P ＜0.05).The subjects who had better self-rated general health,better self-rated hearing,better self-rated memorizing,better life satisfaction were less likely to have depressive symptoms than the controls (OR =0.38,0.53,0.47,0.09;P ＜ 0.05).Conclusion:It suggests that the prevalence of depressive symptoms is higher among the physical disabled aged 45 years of age or older.It should take appropriate measures to reduce emotinal problems for them.

Lymphocytic hypophysitis(LH) is a rare but increasingly recognized autoimmune endocrine condition that causes partial or total hypopituitarism and is often associated with peripartum young women.We here report a 28-year-old patient who had a spontaneous and uneventful pregnancy following LH that had been treated with transspenoidal surgery and followed by anti-inflammatory agent.The woman failed to lactate and developed frontal headaches 3 months after normal delivery of her first child 3 years ago.Lab test showed the reduced concentrations of thyroxine,estradial and cortisol,suggesting hypopituitarism.Magnetic resonance imaging of the brain with contrast was performed and showed a uniformly enhancing pituitary mass with elevated optic chiasm.She underwent transsphenoidal surgery and histological examination of the resected specimen was consistent with lymphocytic hypophysitis.Anti-inflamation was started with prednisolone 40 mg per day because of a recurrence of headache that had completely recovered after surgery and regularly withdrawn to a long term maintenance dose of 10 mg per day.Physiological thyroxine replacement therapy was maintained.Her menstruation was restored without sex hormone replacement after 3 months.Three years after surgery,she got pregnant spontaneously and had normal breastfeeding after delivery.LH did not recur during this peripartum.

Aim To investigate the alteration of Wnt/β-catenin signaling pathway in early diabetic rat myo-cardium and clarify its role in development of diabetic cardiomyopathy. Methods The diabetes mellitus ( DM) model was prepared by intraperitoneal injection of streptozotocin ( STZ, 60 μg · g-1 ) . The alteration of Wnt/β-catenin signaling pathway was determined by Western blot and immunohistochemistry. HE staining was used to analyze the change of myocardial pathologi-cal structure. Results Cardiac histological analyses revealed that DM induced cardiomyocyte degeneration and necrosis. Myocardial Wnt2, β-catenin and c-Myc were enhanced in 2 wk DM compared with control group while DKK1 showed no significant alteration. Conclusion Wnt/β-catenin signaling pathway is acti-vated in early diabetic myocardial injury. Further re-searches on its role in DM myocardium may find a new therapeutic target for diabetic cardiomyopathy.

Objective:To evaluate the levels of interleukin 18(IL-18) mRNA in peripheral blood mononuclear cells(PBMCs),livers and spleens in monkey endotoxic shock models.Methods:Cynomolgus monkeys were injected i.v. with lipopolysaccharide(LPS) (2.8 mg/kg) to prepare the endotoxic shock models. The mRNA levels of IL-18 in PBMCs,livers and spleens were tested by fluorescence semi-quantitative realtime reverse transcription(RT)-PCR and compared with that of TNF-? and IL-1?.Results:IL-18 mRNA expression in PBMCs remarkably increased at 120 min after LPS administration, so did in livers and in spleens at the same time. mRNA levels of TNF-? in PBMCs peaked at 60 min after LPS injection, and also increased markedly at 120 min in livers and spleens. IL-1? mRNA levels peaked at 60 min in PBMCs, and did not change so much in livers and in spleens.Conclusion:IL-18 mRNA expression in PBMCs can be up-regulated by LPS, but changed latterly than TNF-? and IL-1? in endotoxic shock cynomolgus monkeys. It is presumed that IL-18 can be produced by PBMCs and liver Kupffer cells(maybe splenic macrophages),and TNF-? is produced by a variety of cells, but IL-1? in bloodstream mainly come from PBMCs after LPS challenge.
