Tumor dormancy is a well-known clinical phenomenon and the underlying mechanisms are poorly understood.It has been speculated that one of the major factors accounting for the dormant behavior is impaired angiogenesis.As stimuli result in the imbalanced concentration between the pro-angiogenic factors and anti-angiogenic factors,the tumors would escape from dormancy.However,why a dormant(silent)tumor be-comes angiogenic has not been explained and the molecular events that participate in it are under investigation.Despite all that,anti-angiogenic therapy is a very promising strategy to induce tumor dormancy.

Objective To observe the analgesic effect of progressive muscle relaxation training (PMR) and compound lidocaine cream in hemodialysis patients undergoing puncturing on arteriovenous fistula.Methods 96 hemodialysis patients undergoing puncturing on arteriovenous fistula were randomly divided into the progressive relaxation training group (group A),compound lidocaine cream group (group B),progressive relaxation training and compound lidocaine cream group (group C),and the blank control group (group D),with 24 patients in each group.Compound lidocaine cream were applied on the central puncture point 60 minutes before operation in group B and group C,and the patients of group A and group C were conducted with progressive relaxation training before puncture and sleep,while no intervention were assigned to group D.The pain of each patients were measured in 10 minutes with Visual Analog Scale(VAS).Results The pain scores were respectively (3.042 ± 0.999) in group A,(2.500 ±0.978)in group B,(1.333 ± 0.817) in group C and (5.042 ± 1.122) in group D.The AVOVA under the significant level of 0.05 for factorial date indicated that the main effect of factor A (progressive relaxation training) was statistically significant and the main effect of factor B (compound lidocaine cream) was also statistically significant.And that statistically significant interaction between the two factors was found,too.Conclusions Compound lidocaine cream with progressive relaxation training could alleviate the pain during puncturing arteriovenous fistula from hemodialysis significantly and was better than single use of each method.

Objective To compare the effects of accelerated solvent extraction (ASE), ultrasonic extraction (USE) and soxhlet extraction (SE), by extraction and determination of pesticide residues in Chinese herbal medicines. Methods Pesticide residues of Chinese herbal medicines were extracted by accelerated solvent extraction, ultrasonic extraction and soxhlet extraction, then the extract were cleaned up by sulfonation treatment, alumina neutral-florisil column and gel permeation chromatography (GPC). The extract was separated by HP-5 capillary column and detected by electron-capture detector. Results Extraction efficiency of USE was significantly lower than that of ASE and SE, there was no significant difference between ASE and SE. Pairwise comparison of the recoveries of three purification methods showed no significant difference. Conclusion The extraction efficiency of three methods was ASE>SE>USE. The extraction method should be selected according to the requirement.

Objective To establish a gas chromatography method for the determination of 50 organochlorine and pyrethroid pesticides residues in Zigyphussp. Methods The organochlorine and pyrethroid pesticides were extracted from Zigyphussp with solvents of acetonitrile (1%acetic acid) by accelerated solvent extraction (ASE) for 5 min at 100℃. Then the extracts were cleaned up by alumina neutral-florisil column and eluted by mixed solvents of ethyl acetate and hexane (15∶85, V/V). The extract was separated by HP-5 and DB1701 capillary dual-column and detected by electron-capture detector. Results The average recoveries and RSD ranged from 71.4% to 110.9%and 1.2% to 14.1% respectively, at three spiked mixed organochlorine and pyrethroid levels. Conclusion The method has good separation and repeatability, and can be used in determination of organochlorine and pyrethroid pesticide residues in Zigyphussp.

Objective To study the biological effects of nucleic transcript factor NF-?B/RelB gene silencing on bone marrow derived dendritic cells(BMDCs).Methods For constructing a new type of DCs,RNAi RelB DC,mouse BMDCs were cultured through IL-4 and GM-CSF cytokine inducing system and were purified with CD11c positive immunomicrobeads by flow cytometer,and RelB gene was silenced with PCR expression cassettes.The RelB RNA interference effect in DCs was proved at both mRNA and protein levels.The biological characteristics of RNAi RelB DCs were investigated by means of cell morphology,cell surface markers and mixed lymphocyte reaction ability.The cell microstructure was observed using scanning electron microscopy and transmission electron microscopy.The cell surface markers,such as MHC-Ⅱ,CD86 and CD40,were detected by flow cytometer.The mixed lymphocyte reaction ability,as well as the ability of DCs in stimulating allogeneic T cells proliferation,was detected by 3H-TdR incorporation method.Results RNAi RelB DCs seemed to have fewer and shorter spikes and many folds under scanning electron microscopy.Many phagocytic vacuoles were observed in the cytoplasm under transmission electron microscopy.DCs surface molecules,MHC-Ⅱ,CD86 and CD40,were expressed at a relatively lower level compared with the control DCs.This RNAi RelB DCs also showed weak allogeneic T cells stimulating ability in vitro in MLR.Conclusion The dendritic cells derived from bone marrow are characterized the biological features and immunological function of immature DC with silencing of RelB gene.This kind of RNAi RelB DCs may be used to induce immunotolerance as a novel tolerogenic DCs.

AIM:To investigate the inhibition of breviscapine on apoptosis of cultured myocardial cell of neonatal rat induced by hypoxia/reoxygenation. METHODS:Myocardial cell hypoxia/reoxygenation model was established by culturing primary myocardial cells of neonatal rats in vitro. Cultured myocardial cells were divided into 5 groups:control group,hypoxia/reoxygenation group and 3 groups pretreated with breviscapine of final concentration 25,50 and 100 mg/L,respectively. The cell viability was measured with MTT; apoptotic rates were determined by AnnexinV-FITC/PI; the expression of Bcl-2 was detected by immunohistochemical method. Expressions of Cytochrome C (CytC) and Caspase-3 were detected by Western blot. RESULTS:Compared with the control group,the viability of myocardial cell decreased and apoptosis rate elevated after hypoxia/reoxygenation. However after pretreatment with 25,50 and 100 mg/L breviscapine,respectively. Cell viabilities increased and apoptotic rates lowered,and the protective effect on myocardial cell had concentration-dependent. In addition,Expression of Bcl-2 decreased but Caspase-3 activity and CytC release increased in myocardial cells induced hypoxia/reoxygenation. Pretreated with breviscapine,expression of Bcl-2 elevated but Caspase-3 activity and CytC release reduced obviously. CONCLUSION:It is associated with the increase in Bcl-2 expression,inhibition of CytC release and Casepase-3 activity that breviscapine could significantly protect myocardial cell against apoptosis induced by hypoxia/reoxygenation.

Objective To research the nutritive condition and establish prediction model of chlorophyll-a(Chla) in a surface water source of Zhengzhou, China. Methods The water temperature(WT), Secchi-depth(SD), chemical oxygen demand(CODMn), total nitrogen(TN), total phosphorus(TP), Chla were determined in two sampling sites of the water source by standardized methods from Mar, 2004 to Oct, 2004. Nutritive condition of the water source was analyzed by trophic level index(?)[TLI(?)]. The influence of some environmental factors on Chla was analyzed by correlation analysis and stepwise multiple regression. Chla was predicted by establishing a stepwise multiple regression equation. Results In the spring, summer and autumn, nutritive condition of the water source was from the maximum of mesotropher, the maximum of light eutropher to the minimum of middle eutropher. A positive correlations were found between the standardized Chla and WT, CODMn, TP. A negative correlation was found between the standardized Chla and SD. Objective:To investigate the effect of cPLA 2(cytosolic PLA 2, 85-kD PLA 2) on the signaling pathways of IL-1?、IL-6 release in LPS induced Hela cells.Methods:①LPS stimulated Hela cells were exposed to the complex of cPLA 2 initiation site-directed antisense oligonucleotide and liposome in the indicated doses and time, and then,analyzed the changes of cPLA 2 both in mRNA and in protein by RT-PCR and Western blot.②At the same time, the supernatants were collected to detect the level of IL-1? and IL-6 under radioimmunoassay(RIA).Results:①The transfection led to a dose-dependent decrease in cPLA 2 protein, while the change of mRNA is not notable.②The levels of IL-1?、IL-6 in the transfected cells were reduced following the repression of cPLA 2 compared with that of LPS added only.Conclusion:cPLA 2 probably plays an important role in the signaling mechanisms on IL-1?、IL-6 release in LPS-induced Hela cell. Objective To observe whether leptin protects rat islet ?-cells from lipotoxicity induced by free Taity acid ( FFA). Methods Pancreatic islets were isolated from Wistar rats.
