1.Effects of mesenchymal stem cells conditioned media on the transforming growth factor-beta signaling pathway in normal and hypertrophic scar fibroblasts

BACKGROUND:Numerous studies have shown that mesenchymal stem cel s (MSCs) can effectively attenuate the fibrosis of damaged heart, lung and kidney by secreting various bioactive factors. OBJECTIVE:To evaluate the anti-fibrotic therapeutic potential of bone marrow MSCs conditioned media in vitro. METHODS:Normal fibroblasts and hypertrophic scar fibroblasts were treated with bone marrow MSCs conditioned media, then transforming growth factor-βand col agen production were analyzed by ELISA, and mRNA expression level of Smad7 and hydroxyproline content were detected by RT-PCR and colorimetry, respectively. RESULTS AND CONCLUSION:Bone marrow MSCs conditioned media significantly inhibited the production of both transforming growth factor-βand col agen in hypertrophic scar fibroblasts (P<0. 01), and up-regulated the mRNA expression level of Smad7 (P<0. 01), a major inhibitory regulator in the SMAD family. However, the normal fibroblasts were scarcely influenced by bone marrow MSCs conditioned media. These findings indicate that bone marrow MSCs conditioned media is considered a promising candidate for the treatment of hypertrophic scars, which may provide new theoretical supports to reduce cutaneous scarring.
2. Development and application of real-time qPCR assay for detecting Sendai virus gene copy number

Objective:
To develop a real-time quantitative PCR (qPCR) assay for detecting Sendai virus gene copy number.
Methods:
The recombinant plasmid was constructed and a reference used to establish the real-time qPCR method with the TaqMan probe and verified for reproducibility and sensitivity.
Results:
The linear range of the developed real-time qPCR assay was 1.024 × 103 ~5 × 1010 copies/μl, with an R2 value of more than 0. 99. The standard recovery of the tested samples was 84.56%~119.48%.
Conclusions
Compared with traditional hernagglutination assay for particle number detection, this method has higher sensitivity, better repeatability and high quantitative accuracy. It can be used for the detection of particle number of vaccine with sendai virus as the carrier, so as to detect the specific activity of vaccine products.

4.Research progress of pyroptosis in pediatric surgical diseases

Pyroptosis is a kind of programmed cell death accompanied by inflammatory responses, which is mainly induced by the Caspase-1/4/5/11 activated mediated via inflammasomes or lipopolysaccharide.Pyroptosis causes the cleavage of various Gasdermin family members, including Gasdermin D, in which a large number of pro-inflammatory factors are released to cause inflammatory responses in various diseases.At present, pyroptosis has not been extensively analyzed in pediatric surgical diseases.This review summarizes the research progress of pyroptosis in pediatric surgical diseases like Hirschsprung′s disease, biliary atresia, neonatal necrotizing enterocolitis, and tumors.It is found that pyroptosis is involved in the occurrence and development of pediatric surgical diseases.The research on the specific mechanism of pyroptosis in pediatric surgical disease and involved signaling pathways contributes to the early diagnosis and treatment of some pediatric surgical diseases.
5.Influence of tablet computer games on the treatment compliance and maladaptive behaviors of patients undergoing painless gastroscopy

Objective? To observe the improve effect of tablet computer games on treatment compliance and maladaptive behaviors of patients undergoing painless gastroscopy. Methods? The 133 patients undergoing painless gastroscopy for the first time in Xinxiang Central Hospital from June 2018 to June 2019 were selected and divided into routine care group (n=65) and tablet computer game group (n=68) by picking method. Routine care group was given routine nursing intervention, while tablet computer game group was given tablet computer games intervention 30 minutes before painless gastroscopy based on routine nursing intervention. The treatment compliance was compared by cooperation questionnaire between two groups, and after painless gastroscopy for 1 week, the incidence of postoperative behavior change were evaluated by postoperative behavior scale (PHBQ). Results? The differences of coordination scores during examination between the two groups were not statistically significant (P> 0.05). The scores of coordination awareness, preparation before examination, posture and other coordination, as well as total scores were higher in tablet computer game group than those in routine care group. The result was statistically significant (P<0.05). The incidence of sorting anxiety, sleep anxiety, eating disorder and indifference and withdrawal was lower in tablet computer game group than those in routine care group. The result was statistically significant (P<0.05). There were no statistical differences in the incidence of separation anxiety and aggression between the two groups (P>0.05). Conclusions? The tablet computer game intervention before painless gastroscopy can improve the examination cooperation and reduce the incidence of inappropriate behaviors after examination. It is worthy of clinical promotion.
6.Efficiency analysis of early cardiac rehabilitation procedures in treating patients after percutaneous coronary intervention

Objective To investigate the efficicy of early cardiac rehabilitation procedures in treating patients after percutaneous coronary intervention. Methods From August 2016 to August2017, 51 patients underwent routine intervention forpercutaneous coronary intervention were selected as control group. From September 2017 to September 2018, 51 patients with early cardiac rehabilitation procedures for percutaneous coronary intervention were selected as observation group. The cardiac function before and after treatment, quality of life and the incidence rate of adverse cardiovascular events after 6 months of operation were compared between two groups. Results After 2 weeks of intervention, the left ventricular ejection fraction (LVEF) in the observation group was significantly higher than that in the control group, and the levels of left ventricular end-systolic volume (LVESV), left ventricular end-diastolic volume (LVEDV) and wall motion integral index (WMSI) were significantly lower than those in the control group (P < 0. 05). After 2 weeks of intervention, the scores of physiological function, physical pain, physiological function, emotional function, social function, energy, general health status and mental health in the observation group were significantly higher than those in the control group (P < 0. 05). The incidence rate of malignant arrhythmia in the observation group was significantly lower than that in the control group (P < 0. 05). Conclusion Early cardiac rehabilitation procedures can improve cardiac function of patients with percutaneous coronary intervention, improve quality of life, and reduce cardiovascular adverse events.

8.Research progress on influencing factors of quality of life of breast cancer long-term survivors

A large number of investigations and studies abroad have focused on the quality of life of breast cancer long-term survivors (BCLS) and its influencing factors. Although their long-term quality of life has been improved, the improvement of their quality of life is affected by many factors. At present, there are few researches on BCLS in China. This research systematically searches CNKI, Wanfang, PubMed and Web of Science, includes relevant literature on quality of life of BCLS in recent years and describes the current domestic and foreign research status of quality of life of BCLS. It reviews from four aspects, including general demographic factors, disease-related factors, psychosocial factors and exercise, in order to provide reference for the follow-up research and practice.
9.Systematic review and Meta-synthesis of the return to work real experience of breast cancer patients

Objective:To systematically review the return to work (RTW) experience of breast cancer patients.Methods:Qualitative research on the RTW real experience of breast cancer patients were retrieved by computer in China National Knowledge Internet, WanFang Data, China Biology Medicine Literature Database, Web of Science, ScienceDirect, EBSCO and PubMed from database building to July 2020. "Joanna Briggs Institute Critical Appraisal Tool for qualitative studies in Australia" was used to evaluation, and the collective synthesis method was used to integrate the result.Results:A total of 18 articles were included, and 56 clear research results were refined. The similar results were grouped together to form 8 new categories and integrated into 3 synthesis results. Synthesis result 1 was the transformation of own working status and external environment. Synthesis result 2 was improving subjective initiative, and actively seeking adaptation and development. Synthesis result 3 was cherishing themselves, being grateful for the status quo, and realizing personal value.Conclusions:Health care providers should pay attention to the physical and mental experience, behavioral performance and needs of breast cancer patients during the RTW process, and increase the evaluation and management of this population, build a dynamic, multidisciplinary and targeted guidance program, and actively encourage patients to RTW and return to normal life.
10.Improving Effect of Curcumin Analogue H8 on Glucose and Lipid Metabolism Disorder in db/db Mice

Objective To investigate the influence of curcumin and its analogue H8 on glucose and lipid metabolism disorder in db/db mice. Methods The type 2 diabetes mouse model (db/db mice) was intragastrically administrated with curcumin and analogue H8 for 8 weeks.The blood biochemical indexes were measured.The expression of PEPCK and G6Pase mRNA was detected by real-time PCR in liver tissues.The expression of PEPCK and G6Pase protein was detected by Western blotting. Results Curcumin analogue H8 reduced blood glucose and lipids in db/db mice (P<0.01) and improved liver function related enzymes significantly.The levels of PEPCK and G6Pase mRNA in db/db mice were significantly decreased (P<0.01) and the expression levels of PEPCK and G6Pase protein were significantly decreased(P<0.01). Conclusion Curcumin analogue H8 improves the glucose and lipid metabolism disorder in db/db mice,and it is related to inhibiting the expression of PEPCK and G6Pase gene and protein.
