3.Comparison of methods for detection of antibody level in serum immunized with SARS-CoV-2 mRNA vaccine
Chinese Journal of Biologicals 2023;36(12):1414-1418
Objective To compare three methods for detection of antibody level in serum immunized with SARS-CoV-2mRNA vaccine. Methods Enzyme-linked immunosorbent assay(ELISA),pseudo virus-based neutralization assay(PBNA)and micro-cytopathic effect neutralization test(MCPENT)were used to detect the antibody levels of a total of 120 serum samples(40 before immunization and 80 after immunization)before and after 2 doses of mRNA vaccine immunization, and the consistency and correlation of the three methods were analyzed. Results The consistency rates of the three methods detecting 120 serum samples were all over 90%,the Kappa coefficients were all more than 0. 7,and each P was less than0. 01. The correlation coefficient(r)between the antibody potency results of positive serum samples detected by the three methods was 0. 825~0. 902,and each P was less than 0. 01. Conclusion The three methods have good consistency and correlation in detecting antibody level of serum immunized with SARS-CoV-2 mRNA vaccine.
4.Establishment and validation of pseudovirus-based neutralization assay for in vivo potency of SARS-CoV-2 mRNA vaccine
Chinese Journal of Biologicals 2024;37(12):1470-1475+1483
Objective To establish and verify a stable pseudovirus-based neutralization assay(PBNA) for the detection of the potency in vivo of SARS-CoV-2 mRNA vaccine so as to use the method for the quality control of SARS-CoV-2 mRNA vaccines. Methods Three different immunization schedules of BALB/c mice(half male and half female) were set up, and each immunization schedule had two doses of vaccine. The titer of neutralizing antibody(Nab) in serum was detected by PBNA, and the appropriate immunization schedule was screened. After the immunization schedule was determined, the mice were immunized at the dose in the range of 0. 5-6 μg/mouse to further determine the linear range between serum Nab titer and immunization dose. The PBNA method for detecting the potency of SARS-CoV-2 mRNA vaccine in vivo was established, and the relative accuracy, intermediate precision, linearity and range of the method were verified. In addition, the potency in vivo of three batches of Omicron XBB.1.5 variant SARS-CoV-2 mRNA vaccine was detected by using the established method, and the results were compared with those of the original detection method of the enterprise. Results The immunization schedule of SARS-CoV-2 mRNA vaccine was determined as follows: the blood samples were taken 7 d after the booster immunization which was conducted 14 d after the primary immunization, the immune dose was 2 μg/mouse, with the linear dose range of 0. 5-3 μg/mouse. A PBNA method for detecting the potency in vivo of SARS-CoV-2 mRNA vaccine in mice was established. The results of methodological verification showed that the relative bias(RB) of each titer level detected by the method was within ± 20%, meeting the requirements of relative accuracy. The geometric coefficient of variation(GCV) of each titer level measured six times was less than 50%, meeting the intermediate precision requirements. The linear regression equation was fitted as y = 1. 003 4 x-0. 011 7, r = 0. 887 0, and the linear regression was significant(P = 9. 67 × 10~(-15)),in accord with the linear acceptability criteria. The titer level ranged from 25% to 150%, which met the quality standard range of in vivo potency of common vaccines. The method was used to detect three batches of Omicron XBB.1.5 variant SARS-CoV-2 mRNA vaccine, and the results were consistent with those of the original method, indicating that the method could be used for in vivo potency detection. Conclusion A stable PBNA method was established to detect the potency of SARS-CoV-2 mRNA vaccine in vivo, and can be applied to the quality control of SARS-CoV-2 mRNA vaccines.
5.The application of N-terminal pro-B-type natriuretic peptide, C-reactive protein and D-dimer in evaluating prognosis of elderly patients confined to bed
Chinese Journal of Postgraduates of Medicine 2013;36(28):16-19
Objective To evaluate the value of plasma N-terminal pro-B-type natriuretic peptide (NT-proBNP),C-reactive protein(CRP) and D-dimer(D-D) in prognosis of elderly patients confined to bed. Methods The clinical data of 150 elderly patients confined to bed were reviewed retrospectively, and 35 patients were in dead group and 115 patients were in survival group. The level o f plasma NT-proBNP,CRP and D-D were measured before treatment, during treatment and after treatment.Their characteristics and dynamic change were analyzed, and the value of NT-proBNP,CRP and D-D was evaluated in prognosis of elderly patients confined to bed. Results The level of NT-proBNP,D-D and CRP in dead group before treatment, during treatment and after treatment were significantly higher than those in survival group (P < 0.05 or < 0.01).The peak level of NT-proBNP,D-D and CRP in dead group emerged after treatment.By the way,the peak level of NT-proBNP,D-D and CRP in survival group emerged before treatment.Incidence of two or three markers positive in dead group [37.1% (13/35),54.3% (19/35)] was significantly greater than that in survival group [20.9% (24/115),8.7% (10/115)] (P =0.032,0.008).Multivariate Logistic regression analysis showed that the level of NT-proBNP,D-D and CRP were independent predictors of 28-day mortality (P <0.05).Conclusions The level of NT-proBNP,CRP and D-D are increased in severe elderly hospitalized patients. The level of above indexes have a dynamic change and reach peak before death. They are the independent prognostic markers of hospital mortality.Combined detection of NT-proBNP,D-D and CRP can improve the predictive value of hospital mortality in elderly patients confined to bed.
6.Molecular regulation mechanism of pancreatic islet β-cell remodeling in neonatal stage
Chinese Journal of Endocrinology and Metabolism 2013;(4):351-353
The neonatal period is important for the dramatic increment as well as function improvement of islet β-cell.β-cell mass augmentation is primarily regulated by neogenesis, replication, and apoptosis. The mechanisms involve cell cycle related proteins and mitosis-promoting signaling pathways. This remodeling period has far-reaching effects on the body in adapting to the metabolic needs and regulating glucose homeostasis later in life.
7.Analysis of heart rate variability in the patients with essential hypertension and coronary heart disease
Chinese Journal of Primary Medicine and Pharmacy 2008;15(4):639-641
Objective To study autonomic nerve regulator character and arrhythmias in the patients with essential hypertension(EH) and coronary heart disease(CHD). Methods The heart rate variability(HRV) and arrhythmias in 50 EH, 44 CHD, 52 EH + CHD patients and 35 healeh elderly subjects were investigated. Results There were significant differences of HRV in the patients with EH, CHD and healthy elderly subjects(P<0.05).There was much significant difference of HRV between the patients with EH + CHD and healthy elderly subjects(P<0.01), but no significant difference of HRV between the patients with EH and CHD(P>0.05). The incidence rate of the premature atrial contraction, premature ventriculax contraction and atrial tachycardia in EH, CHD, EH+CHD and healthy elderly subjects, there were much significant differences( P<0.01 ). Except the incidence rate of premature atrial contraction there was significant difference between the patients in CHD and EH + CHD (P<0.05) ,and no significant differences of the incidence rate of other kinds of arrhythmias in the patients with EH,CHD and EH + CHD(P>0.05 ). Conclusion The autonomic never function of EH and CHD patients was damaged, and the incidence rate of arrhythmias was increased(the premature atrial contraction often happened, then the premature ventricular contraction), and arrhymias happened in EH + CHD patients is most oftenly. The vagal hypoacfivity and sympathetic hyperactivity were major causes of reduction of HRV and probable causes in the elderly patients.
