[Objective] To optimize the extraction process of Weiyanping granules. [Method]The orthogonal test method was adopted to explore the influences of 4 factors(quantity of water,soaking time,extraction time and times) on the yielding rate of berberine and dry paste rate. [Results]Analysis by synthesis,the optimum condition for extraction was A2B3C1D3.The medicinal substances should be soaked in 10 times water for 2 hours and distilled for 3 times,with 1 hour each time.[Conclusion]The experiment result showed that the optimum extraction process condition was stable,reasonable and economical,which could be used to guide industrialized production.

Objective:To study the optimal process of reflux extraction for lignans from Acanthopanax sessiliflorus roots. Methods:Sesamin as the reference substance, a colorimetric method was used to detect the absorption value of the samples at 558 nm to calculate the content directly. The extraction time, the extraction times and the volume of extraction solvent were applied to optimize the extrac-tion conditions for total lignans by orthogonal experiment. Results:The absorbance had a linear relationship with the amourt of sesamin within the range of 10. 6-53. 0 μg·ml-1(r=0. 999 7). The average recovery of sesamin was 102. 2%(RSD=1. 6%, n=6). The content of total lignans (measurement by sesamin) in the extracts was 10. 0 mg·g-1. The optimum reflux extraction conditions were as follows:adding 40-fold 50 % methanol, extracting once for 30 min. Conclusion:The UV method for determining the total lignans is feasible, stable and reliable as well as precise, and the optimal extraction process is reasonable.

Objective] To observe the diagnosis value of noninvasive diagnosis model S index used to evaluate the fibrosis degree of slight chronic hepatitis B(CHB). [Method] Trace back and investigate the routine serological indicators GGT, PLT, ALB and liver aspiration biopsy results of 63 cases of CHB;use noninvasive diagnosis formula to calculate:S index=1000×GGT/(PLT×ALB2),use ROC curve to analyze and evaluate the clinical diagnosis value of S index. [Result] S index is in positive relation with liver fibrosis degree. S index can forecast the fibrosis having or not with best truncation point 0.04h, the sensitivity 87.0%, specificity 47.5%and area under the curve(AUC) 0.690,close to 0. 7; in forecasting obvious fibrosis, with best truncation point 0.04h, sensitivity 100%,specificity 36.8%,AUC 0.664. [Conclusion] S index used for forecasting slight CHB having or not fibrosis has high value of diagnosis, but it has low correctness for diagnosing having or not obvious liver fibrosis.

OBJECTIVE To study the correlation between mutations of gyrA and parC genes and ciprofloxacin-(resistant) clinical isolates of Acinetobacter baumannii from Chengdu.METHODS The genes of gyrA and parC DNA in 28 ciprofloxacin-resistant and 2 ciprofloxacin-susceptible isolates of A.baumannii were amplified by PCR and then analyzed by restriction fragment length polymorphism(PCR-RFLP) and DNA sequencing.(RESULTS) Hinf Ⅰ digestion of the gyrA gene products of susceptible isolates(generated) two fragments,but resistant isolates(generated) one fragment.The parC gene products generated 2 fragments.DNA sequencing of 5 resistant isolates revealed mutations in gyrA gene that resulted in amino acid substitutions: Ser83→Leu and Ala88→Thr,especially,Ser83 point mutation accounting for the disappearance of sequence of Hinf Ⅰ.There was not any mutation in(gyrA) of 1 susceptible isolate;the substitution of Ser108→Ile in parC(gene) of 1 susceptible and 1 resistant isolates were identified,the remaining four isolates had more nosense mutations.CONCLUSIONS Compared with parC gene mutations,(gyrA) gene mutations of A.baumannii appears to be the main molecular mechanism responsible for ciprofloxacin resistance.The high-level ciprofloxacin resistance in A.baumannii probably needs a variety of mutations.

Objective:To establish a method for the determination of related substances in enoxacin gluconate injection by HPLC with gradient elution. Methods:HPLC was performed on a DiamonsilTM C18 column (250 mm × 4. 6 mm, 5 μm), the mobile phase A was 0. 025 mol·L-1 phosphonic acid solution (adjusting pH to 3. 0 with triethylamine)-methanol- acetonitrile (80∶10∶10) and the phase B was 0. 025 mol·L-1 phosphonic acid solution (adjusting pH to 3. 0 with triethylamine)-methanol-acetonitrile (350∶325∶325) with gradient elution at a flow rate of 1. 1 ml·min-1 , the detection wavelength was 269 nm and 284 nm, the injection volume was 20μl, and the column temperature was 40℃. Results:Under the HPLC conditions, the samples had good stability and separation. The calibration curve was linear within the concentration range of 19. 80 ng·ml-1-19. 80 μg·ml-1(r=0. 999 9) for 5-hydroxymethylfur-fural with the detection limit of 0. 18ng, and the average recovery was 99. 68% with RSD of 0. 12% (n=9). Conclusion:The method is accurate, sensitive, specific and reproducible, and can be used in the determination of related substances in enoxacin gluconate in-jection.

Aim To study gyrA and parC mutations of clinical Pseudomonas aeruginosa strains. Methods MIC values of 55 clinical P.aeruginosa isolates were determined by agar dilution test and 1 sensitive strain and 8 resistant strains were selected with standard sensitive strain ATCC27853 as control, the quinolone determining region (QRDR) of the gyrA and parC genes were amplified by PCR, the lengths of PCR products were 351 bp and 397 bp. The gyrA PCR products(351 bp) were digested with enzyme sacⅡ. The gyrA and parC gene were sequenced. Results In this study, gyrA genes of all resistant strains had an ACC to ATC mutation in codon 83, leading to the amino acid substitution of an isoleucine for a threonine, and three high level resistant strains also showed a GAC to GGC mutation in codon 87, leading to the substitution of a glycine for an aspartic acid. In addition, four resistant strains also had an TCG to TTG mutation in codon 87 of parC gene, leading to the amino acid substitution of a serine for a leucine. The strains with both gyrA and parC mutations were two to sixteen times more resistant than the strains which had only gyrA mutations. At the same time, a silent mutation (CAC to CAT) in codon 132 of gyrA gene and a silent mutation(GCT to GCG) in codon 115 of parC gene occured, which did not lead to amino acid change. Conclusion The mutations of 83 and 87 codons of gyrA and the mutatations of 87 codon of parC gene were related to fluroquinolone resistance, and the mutations of the 83 codon of gyrA gene were more important.

【Objective】To observe therapeutic effect of Guishen Pill(GP) combined with acupuncture(AP) for male immune infertility(MII) and to investigate influence on acid phosphatase(ACP) in seminal plasma.【Methods】One hundred and five MII patients were equally randomized into three groups.Group A received GP combined AP,group B received GP and group C received prednisone.The therapeutic effect was evaluated in the three groups after treatment,and the changes of ACP were observed before and after treatment.【Results】The total effective rate was 91.43%,88.57% and 68.57%,and the cure rate was 45.71%,22.86% and 11.43% in groups A,B and C respectively.The difference of total effective rate was significant in groups A and B as compared with that in group C(P

Objective To investigate the correlation between carotid artery intima media thickness (IMT) and cerebrovascular hemodynamic analysis (CVA) and the diagnostic value in ischemic stroke.Methods Using colour Doppler IMT,IMT/D ratio and CVA were performed in 120 patients with hyperacute and acute ischemic stroke as compared with the health controls.Results IMT and IMT/D ratio were higher in patients with ischemic stroke than the health aduts. Vmean and Qmean in ischemic stroke hyperacute group were lower, and R,DR were higher obvious in hyperacute group than that of acute group. IMT and IMT/D ratio were negatively correlation with Vmean and Qmean and positively correlation with R and Zc.Conclusion The measurement of carotid artery IMT and CVA have clinical significance in early diagnosis of ischemic stroke.

Objective To determine if the shortened exposure of oocytes to sperm can influence the insemination rate and the results of IVF-ET. Methods A total of 217 women were included.They were divided into study group (105 cases) and control group (112 cases). In study group the shortened insemination was used.The oocytes were washed 2 hours after the insemination and transferred into the fresh medium.In control group the classical procedure was used.The oocytes were inseminated by ICSI.The number of retrieved oocytes,fertilized oocytes,the number of embryos were counted and the pregnancy rate was evaluated. Results There was no statistically significant difference between the study and control group in the number of retrieved oocytes (1163 vs 1752),number of embryos (1084 vs 1097),and pregnancy rate ( 42.9% vs 42.9%). Conclusions The shortened exposure of oocytes to sperm is a simple and effective way in IVF-ET.
