1.Proteinase-activated receptors regulated the expression of type I/Ⅲ collagen protein of hepatic stellate cell isolated and cultured from SD rats liver

Objective:To investigate the role of proteinase-activated receptor 1 and 2(PAR1,PARs)in regulation ofthe expression oftype I/Ⅲ collagen protein of hepatic stellate cell isolated and cultured from SD rats liver.Methods:Hepatic stellate cell isolated and cultured from SD rats livers with a modification technique of Friedman and centrifugation of Nycodenz Gradient.Expression of mRNA for PAR1, PAR2,Ⅰcollagen and Ⅲ collagen were examined by reverse transcription polymerase chain reaction(RT-PCR)and protein examined by immunocytochemistry,observated characteristic cell shape,and examined expression of hexadecanoic acid(HA)and laminin(LN)of supernatant from culturesd HSC.Results:In the course of culturing rat HSC,vitamin A autofluorescence and lipid droplet were decreased gradually or lost following HSC activation,the cells morphology changed to stellate cells and cells proliferation activated.Ⅰ/Ⅲ collagen protein expressions were also increased,the expression of HA,LN,PAR1 and PARs paralleled with the expression ofⅠ/Ⅲ collagen protein and correlated with activation and proliferation of HSC.Conclusion:The expression of PARs correlated with activation and proliferation of HSC,could promote the expression ofⅠ/Ⅲ collagen protein,HA and LN.PARs might play an important role in the mechanisms of hepatic fibrosis.
2.Clinical efficacy of terlipressin in treatment of type II hepatorenal syndrome

Objective To investigate the clinical efficacy of domestic terlipressin in the treatment of type II hepatorenal syndrome (HRS -II).Methods A total of 25 HRS -II patients admitted to our hospital from November 2011 to June 2014 were recruited into the treatment group,and 28 HRS -II patients treated with dopamine before 2011 were recruited into the control group.Patients in the treatment group were randomly divided into two subgroups:one subgroup (n =12)was given terlipressin once every 8 h,and the other subgroup (n =13) was given terlipressin once every 12 h.Both groups received albumin (Alb)infusion to expand the blood volume before treatment with terlip-ressin or dopamine,and the course of treatment was 7 days.The improvement in clinical symptoms,levels of blood urea nitrogen (BUN), serum creatinine and electrolytes,urine volume,changes in liver function,and ascites disappearance in the two groups before and after treat-ment were compared.Comparison of categorical data between the two groups was made by χ2 test,and comparison of continuous data was made by t test.Results Patients in the control group showed no obvious symptom relief,but those in the treatment group had varying de-grees of improvement in clinical symptoms.Neither group had significant changes in liver function and serum sodium level after treatment. The treatment group had significantly more patients whose ascites volume had decreased from large to medium than the control group (χ2 =5.705,P <0.05).There was a slight but not significant decrease in the levels of BUN and serum creatinine in the control group after treat-ment with dopamine (all P >0.05),whereas the urine volume showed significant difference after the treatment (t =15.534,P <0.01). The treatment group showed significant differences in the levels of BUN and serum creatinine and urine volume after terlipressin treatment (t=11.535,9.941,and 19.685,respectively;all P <0.01),and significant differences in those levels were also observed compared with the control group after dopamine treatment (t =7.317,9.284,and 9.839;all P <0.01).Larger changes in BUN,serum creatinine,and urine volume were observed in the subgroup receiving terlipressin every 8 h than in that receiving terlipressin every 12 h,but the differences were not significant (all P >0.05).Patients treated with terlipressin had mild and tolerable adverse reactions.Conclusion The domestic terlipressin combined with albumin infusion has good clinical efficacy and mild adverse effects in treating HRS -II.Its clinical application is highly recommended.
3.Expression and relationship between HDAg and bcl-2/bax in liver tissue of the patients with hepatitis D

Objective To study the role of bcl-2 and bax in the pathogenesis of hepatitis D. Methods Expression of HDAg, bcl-2 and bax in liver specimens of 79 patients with hepatitis D were studied by immunohistochemistry technique. Meanwhile, the relationship between expression of HDAg and that of bcl-2/bax in liver specimens of patients were studied by double labelling technique and serial sections.Results bcl-2 was mainly expressed in the cytoplasm of hepatocytes, bax mainly in the cytoplasm of hepatocytes and partly in the nucleus of hepatocytes, and HDAg mainly in the nucleus of hepatocytes. Most of HDAg and bax positive cells were distributed among infiltrating lymphocytes at the periportal region especially at the advancing edges of areas of “piecemeal necrosis”. Most of hepatocytes of bax positive was found to locate near to positive cells of HDAg, and there were positive correlations between degrees of bax expression and HDAg expression(P<0.05). Conclusions The distribution and the expression of bax and HDAg are significantly correlated with the activity of inflammation and the severity of the liver damage, and HDV infection may induce the expression of bax in the hepatocytes,and hepatocyte apoptosis mediated by bcl-2/bax may play an important role in the liver cell injury by HDV infection.
4.Case Series Reports of 40 Pediatric Cold Children with Lung-stomach Heat Retention Syndrome Treated by Yinlai Decoction Addition and Subtraction

Objective]To observe the clinical effect of Yinlai Decoction Addition and Subtraction on treating Pediatric Cold about lung-stomach heat retention syndrome.[Method]This study adopted the research design method of case series reports, continuously included 40 Pediatric Cold children with lung-stomach heat retention syndrome treated by Yinlai Decoction Addition and Subtraction from November 2011 to December 2012. Through clinical visiting, we could assess the treatment outcome of this time on the basis of evaluation standard. [Results]33 patients were cured, 5 patients turned better and 5 patients had no effect, the total effective rate was 95.00%. [Conclusion]Yinlai Decoction had clearing and dispersing function, it could treat lung and stomach simultaneously, therefore, it had obvious efficacy in Pediatric Cold about lung-stomach heat retention syndrome.
5.Inductive effect of high fat diet on uncoupling protein 2 in rats with nonalcoholic fatty liver

BACKGROUND: Up to now the pathogenesy of nonalcoholic fatty liver(NAFL) is unclear. Uncoupling protein (UCP) of mitochondrial membrane can make oxidative phosphorylation uncoupling in mitochondrium, and then result in decreasing the efficiency of ATP synthesis by mitochondrium and participate the progression of fatty liver by changing energy metabolism.OBJECTIVE: To observe the expressive rule of UCP-2 in NAFL rat models at different time phases.DESIGN: Randomized control animal trial.SETTING: Department of Gastroenterology, Research Institute of Surgery,Daping Hospital, Third Military Medical University of Chinese PLA.MATERIALS: The experiment was performed at National Key Laboratory,Research Institute of Surgery, Daping Hospital, Third Military Medical University of Chinese PLA from November 2004 to December 2005. A total of 64 clean-grade Wistar adult male rats were selected. After being fed with common feed for 1 week, the rats were randomly assigned into model group and normal control group with 32 rats in each group. In each group there were 4 time points: feeding for 2, 4, 8 and 12 weeks, 8 ones at each time point.METHODS: The rats in model group received high fat diet (88% basic feed, 10% pork fat and 2% cholesterol) for 12 weeks. Those in normal control group received common feed in different cages (6 rats in each cage).MAIN OUTCOME MEASURES: Blood was collected from anesthetic rats of the two groups at corresponding time point. Animals were killed,and then liver was extracted rapidly. ①Change of expression of UCP-2in liver was determined with immunohistochemical method and Western blot technique. ②Activity of triacylglycerol, free fatty acid (FFA) and alanine aminotransferase (ALT) in serum was measured with biochemical method. ③Content of malondialdehyde (MDA) in liver tissues was measured.RESULTS: Totally 64 rats were involved in the result analysis. ①Biochemical indicators in serum: Levels of triacylglycerol, FFA and ALT in serum were significantly higher in the model group than in the normal control group at weeks 4, 8 and 12, especially at weeks 8 and 12 (P < 0.01). ②The expression of UCP-2 enhanced gradually with the exacerbation of fatt yliver in the model group, while it was negative in the normal control group.③Content of MDA in liver tissues: It was significantly higher in the model group than in the normal control group at weeks 4, 8 and 12, especially at weeks 8 and 12 [(4.07±0.15), (4.93±0.14), (5.20±0.20) μmol/g; (3.14±0.20), (3.12±0.18), (3.13±0.16) μmol/g,P < 0.01].CONCLUSION: With the formation and aggravation of nonalcoholic fatty liver, levels of triacylglycerol, FFA and ALT in serum and content of MDA in liver tissues increase, and the expression of UCP-2 also increase. It is indicated that high fat diet can induce expression of hepatocyte UCP-2,decrease of ATP progression in cells and accelerate the formation and development of fatty liver.
6.Analysis of SCL-90 test results of 809 physical examinees

Objective To explore the mental health status of physical examination population,and provide the basis for mental health guidance.Methods The SCL-90 scale in the cloud service system of PEM was used to evaluate the gender, marriage, age, education, income and other factors in the physical examination cases of our hospital.Results The positive rate of SCL-90 in 809 patients was 27.94%.Female scores,and the scores of somatization, obsessive-compulsive symptoms, depression, anxiety, terror of women were higher than those of men except paranoid.Unmarried population scores and the scores of all factors were higher than those of married and divorced people.The scores of all factors in the 18-<30 year-old cases was the highest among all cases;the scores of depression,hostility and paranoid was decreased with the increase of age;the score of somatization, obsessive-compulsive symptoms, interpersonal sensitivity, anxiety, terror,spirit of 50-60 year-old cases were the lowest among all cases.The higher the degree of education,and the more economic income, the lower the psychological evaluation of each factors scored.Conclusion The mental health problems of female,unmarried,low age,low education,low-income groups are grim.
7.Level of Alpha-Synuclein Oligomers in Speripheral Blood Plasma and Red Blood Cells in Parkinson's Disease

Objective To investigate the level of alpha-synuclein (α-syn) oligomers in peripheral blood plasma and red blood cells (RBC) in patients with Parkinson's disease (PD), and the value of it for diagnosis and evaluation of PD. Methods From March, 2013 to De-cember, 2014, peripheral blood samples were collected from 30 PD patients and 30 healthy coutrols, and the level ofα-syn oligomers in plas-ma and RBC was detected with enzyme linked immunosorbent assay. Results The level of α-syn oligomers was less in both plasma and RBC in the controls than in the patients (t>2.346, P<0.05), and was more in RBC than in plasma in both the controls and the patients (t>2.242, P<0.05). The level ofα-syn oligomers of patients did not correlate with their course, Hoehn-Yahr Stage, the scores of Unified Parkin-son Disease Rating Scale Part 3, Non-Motor Symptom Quest and Mini-Mental State Examinatlon, both in plasma and RBC (P>0.05). Con-clusion The level ofα-syn oligomers in peripheral RBC and plasma may be helpful for diagnosis of PD, but less for the evaluation of the ill-ness.
8.Study on the health related quality of life and influencing factors in emigrants with rheumatoid arthritis in Three-Gorge Reservoir area

Objective To analysis the health related quality of life (HRQOL)in emigrants with rheumatoid arthritis(RA) in Three‐Gorge Reservoir area ,and to study the influencing factors on HRQOL .Methods Self‐rating depression scale(SDS) was used to assess degree of depression in emigrants with rheumatoid arthritis in Three‐Gorge Reservoir area ,and SF‐36 scalec was used to assess the HRQOL of these patients .The one‐way ANOVA and stepwise regression analysis were used to study the influence fac‐tors of HRQOL .Results SDS scores showed that there were 110 peoples with depressive symptoms (67 .5% ) ,including mild de‐pression 58 cases(35 .6% ) ,moderate depression 39 cases(23 .9% ) ,severe depression 13 cases(8 .0% ) .Stepwise regression analysis showed that :SDS score ,DAS‐28 classification ,age ,sex ,treatment ,education ,occupation ,physical exercis ,economic income ,health insurance and alcohol consumption influenced the HRQOL scores of patients with IBS .DAS‐28 classification ,economic income , health insurance ,sex ,treatment were the main influencing factors ,with the unstandardized partial coefficients of -39 .211 ,31 .512 , 25 .866 ,-42 .348 ,-51 .235 .Conclusion Most of emigrants with RA in Three‐Gorge Reservoir area suffering from depressive symptoms ,and the HRQOL scores of them is not optimistic .The results suggested that more attention must be paid to the elderly , low‐income ,women ,low education ,DAS‐28 classification and no occupation .Quiting drinking ,receiving treatment ,psychological treatment ,strengthen exercise and expanding the scope of medical insurance all can improve patients′health related quality of life .
9.Clinical application of GlucoWatch in Chinese diabetic patients

The clinical application of GlucoWatch used in 32 Chinese diabetic patients was evaluated to determine its value in following blood glucose trends and track patterns and to know the compliance of patient. The results confirmed the accuracy of GlucoWatch and showed that it could follow blood glucose trends and track patterns. The skin irritation was mild and most subjects thought that blood glucose monitoring by GlucoWatch could reduce the pain caused by traditional methods.
10.Investigation of HBV-DNA in peripheral blood mononuclear cells and hepatocytes with molecular hybridization

The existence of HBV-DNA in the mononuclear cells of peripheral blood and in the serum of 61 patients with HBV infection was determined with southern blot and dot blot hybridization,and that in the liver tissue of 31 patients out of the 61 with southern blot and in situ hybridization.The positive rate of HBV-DNA in the serum,mononuclear cells and hepatocytrs was 26.2% (16/61),24.6% (15/61) and 44.8% (13/31) respectively.There was no concordance of the existence of HBV-DNA in the serum,peripheral mono-nuclear cells and hepatocytes in an individual.For example,HBV-DNA was absent in the serum but present in mononuclear cells and hepatocytes in 11 cases.In fact,peripheral mononuclsar cells can serve as the reservoir for HBV to replicate.It cannot be denied that HBV can replicate in an individual even though HBV-DNA is negative in the serum.
