1.Investigation on the protective mechanism of anisodamine on ischemic intestine

It is reported that anisodamine exhibited cell protection action during anoxia and ischemia, the mechanism of which is still obscure. Present study found that: (1) 654-2 significantly reduced the reperfusion mucosal damage of ischemic intestines and the accumulation of intracellular calcium; (2) anisodamine could blunt the contraction of the high potasium depolarized guinea pig ileum with calcium; (3) intermittent injections of 5mg/kg 654-2 significantly inhibited the cardiac function.The above results suggest that 654-2 possesses calcium-antagonist action, which may be one of the chief mechanism of its cell protection action.
2.One case report: removal of foreign body in ethmoidal sinus-medial orbital wall through nose assisted by endoscope.

The patient was male, 20 years old, and complained of pain, bleeding and decreased vision after the right eye was injured by nail for an hour.
PHYSICAL EXAMINATION
right exophthalmos, conjunctival edema, skin laceration at the lower eyelid of right eye, limitation of eye movement, asymmetric eyes and weak light-reflecting. X-ray showed: metallic foreign body shadow in the right orbit, 0.5 cm x 0.4 cm approximately. Orbit and paranasal sinus CT showed: 1 hematoma of the right eye and inside. 2 high density foreign body embedded in the bone wall of the inside of right orbit and ethmoid. 3 medial wall fracture of right eye orbit. 4 bilateral maxillary sinus and right ethmoidal sinus effusion. 5 slightly left side of nasal septum. The patient was initially diagnosed as foreign body in right ethmoidal sinus, skin laceration of right lower eyelid, retrobulbar hematoma.

3.Analysis of apelin level and related factors in patients with obesity and different glucose tolerances

Objective To evaluate apelin level in patients with obesity and different glucose tolerances, investigate relevance of apelin with body mass index(BMI), blood lipid and blood glucose. Methods According to different glucose results of oral glucose tolerance test, 120 subjects were divided into three groups: normal glucose tolerance(NGT),impaired glucose tolerance(IGT) and type 2 diabetes(T2DM). All of the subjects were also divided into obese and non-obese subgroups based on their BMI(≥25kg/m~2 or <25kg/m~2 ). Plasma apelin levels were compared among groups of NGT, IGT and T2DM. Fasting plasma glucose(FPG)and 2h postprandial plasma glucose (2hPG) after OGTT, lipid profile and BMI were measured. Results Both fasting and 2h postprandial plasma apelin had significant differences in these groups (P<0.05,or P<0.01). The overweight and obese subgroups in the IGT group and diabetes group had higher apelin levels than their counterpart subgroups in the NGT group. BMI, FPO, systolic blood pressure(SBP) were independent influencing factors for apelin (P<0.05 ,or P<0.01). Conclusions Plasma apelin level is related to obesity and type 2 diabetes.
4.Reversal effect of isotetrandrine, an isoquinoline alkaloid extracted from Caulis Mahoniae, on P-glycoprotein-mediated doxorubicin-resistance in human breast cancer (MCF-7/DOX) cells

This study investigated the reversal effect of isotetrandrine, an isoquinoline alkaloid extracted from Caulis mahoniae, on P-glyeoprotein-mediated multidrug resistance in human breast cancer doxorubicin-resistant (MCF-7/DOX) cells. RT-PCR assay and immunity histochemistry assay were used to determine the expression level of mdrl gene and P-gp in MCF-7/DOX cells to elucidate resistant character of MCF-7/DOX cells. The activity of isotetrandine to enhance doxorubicin cytotoxicity was tested using MTT (3-(4,5-dimethyhhiazol)-2,5-diphenyltetrazolium bromide) assay and was evaluated by the reversal fold (RF) values. Intracellular accumulation of doxorubicin was assessed by the determination of doxorubicin-assoeiated fluorescence intensity. Effect of isotetrandrine on the expression level of P-gp in MCF-7/DOX cells was then determined by immunity histochemistry assay. The ability of isotetrandrine to inhibit P-gp function was evaluated by detecting the accumulation and efflux of rhodamine123 ( Rh123 )with flow cytometry (FCM). Verapamil was employed as a comparative agent in whole experiment. The results indicated that MCF-7/DOX cells had phenotype of MDR and that the positive expression of P-gp MCF-7/DOX cells and reversal fold (RF) was significantly higher than that of verapamil (P < 0.05 ), butit hardly affected cytotoxicity of DOX in MCF-7 cells and the expression level of P-gp in MCF-7/DOXcells. The ability of isotetrandrine to inhibit P-gp function was reversible, because incubation of MCF-7/DOX cells with isotetrandrine caused a marked increase in uptake and a notable decrease in efflux of Rh123 and a marked increase of intracellular DOX concentrations. In conclusion, isotetrandrine exhibited potent effect on the reversal of P-gp-mediated MDR in vitro, suggesting that it might become a candidate of effective MDR reversing agent in cancer chemotherapy.

6.Optimization of Reflux Extraction for Total Lignans from Acanthopanax Sessiliflorus by Orthogonal Experi-ment

Objective:To study the optimal process of reflux extraction for lignans from Acanthopanax sessiliflorus roots. Methods:Sesamin as the reference substance, a colorimetric method was used to detect the absorption value of the samples at 558 nm to calculate the content directly. The extraction time, the extraction times and the volume of extraction solvent were applied to optimize the extrac-tion conditions for total lignans by orthogonal experiment. Results:The absorbance had a linear relationship with the amourt of sesamin within the range of 10. 6-53. 0 μg·ml-1(r=0. 999 7). The average recovery of sesamin was 102. 2%(RSD=1. 6%, n=6). The content of total lignans (measurement by sesamin) in the extracts was 10. 0 mg·g-1. The optimum reflux extraction conditions were as follows:adding 40-fold 50 % methanol, extracting once for 30 min. Conclusion:The UV method for determining the total lignans is feasible, stable and reliable as well as precise, and the optimal extraction process is reasonable.
7.Study on mastery situation of knowledge about infection diseases among primary and middle school students in Chongqing

Objective To understand the mastery situation of the knowledge about common infectious diseases among the prima-ry and middle school students,in order to provide the evidence for the prevention and treatment of infectious diseases.Methods The stratified random cluster sampling method was adopted.The survey was performed by the questionnaire form.The data were analyzed by the Chi square test,logistic regression model in the SPSS17.00 statistical software.Results Among the primary and middle school students in Chongqing,the students mastering 75% of the basic knowledge about infectious diseases accounted for 24.6% of the total respondents (P<0.01).From the view of the distribution of disease entities,the top six diseases with the awareness rate of less than 60% were hepatitis B,bacterial dysentery,cholera,measles,pinworm and rabies respectively(P<0.01), whose awareness rates are under 60%(P<0.01).As for the level of mastery among different grades,the senior middle school students had a better knowledge of tuberculosis,flu,SARS,AIDS,hepatitis B,poliomyelitis,etc(P<0.01).Comparatively,the pri-mary school students grasped the knowledge of hand-foot-and-mouth disease and pinworm better(P<0.01).Conclusion The pri-mary and middle school students have a low awareness rate of infectious diseases.Pertinently strengthening the popularization of the knowledge about infectious diseases is the powerful measure for reducing the occurrence of infectious diseases and promote the healthy development of students.
8.The value of prenatal ultrasound examination in diagnosing twin reversed arterial perfusion sequence

Objective To explore the clinical significance of prenatal ultrasound examination in diagnosing twin reversed arterial perfusion sequence (TRAP). Methods This was a retrospective study on 5 cases of TRAP sequence conifrmed by prenatal ultrasound and clinical method. The clinical data and ultrasound images were analyzed. Cases were collected at Hubei Maternal and Child Health Hospital from 2001 January to 2013 February. Results Four-iffths of TRAP were not corrected diagnosed and another one was misdiagnosised as asymmetrical conjoined twins in early pregnancy. Five cases of TRAP were correctly diagnosed during the second trimester. Sonographic ifndings of ifve acardia without blood lfow were as follows:(1) All acardias had no blood perfusion in body and umbilical cord, were absence of the fetal head (acephalous) and heart (acaidia), without skin edema, and a linear object resemble as umbilical cord extend from umbilical region of acardia to plcenta could be seen. Four (case 2-5) had lower limbs or dysplasia of lower limbs (two had spine), and one (case 1) had an irregular mass with bone. (2) Four pump twins had no obvious abnormity, while one had two clubfeet (case 2). Three died in uterus, one underwent induced labor due to oligohydramnios and heart dysfunction, and another one survived. Prenatal ultrasound images were consistent with MRI, X-ray examination, autopsy and postnatal neonatal examination results. Conclusions Acardia without blood lfow which was generally misdiagnosed as single gestation in utero can be diagnosed according to the special disformity of acardia and the linear umbilical cord extend from umbilical region to placenta, besides, it still need to strengthen monitoring the pump twin even if there was no blood perfusion to acardia.
9.Experimental Study of Anti-Sports Fatigue Effect Mechanisms of Cistanche Deserticola

Objective To study the effect of cistanche deserticola on lactate dehydrogenase(LDH) isoenzyme,glycogen and nitric oxide synthase 3(NOS3) of liver in the mice burden swimming and explore relevant molecular mechanisms of anti-sports fatigue.Methods The mice were devided into the normal control group,the sport control group and the cistanche deserticola experimental group.Each mice of the normal control group and the sport control group was given saline 0.2 mL per day.Each mice of the cistanche deserticola experimental group was given water decoction of cistanche deserticola 0.2 mL(3 g/kg) per day.The administrations were for 15 days.The burden swimming for 90 minutes was carried out on the mice of the sport control group and the cistanche deserticola experimental group at 1 hour after the last administration.Livers of the mice were removed after 10 hours of swimming.One part of liver was fixed in the neutral formalin liquid to prepare the paraffin sections and the others was used for measurement of LDH activity.The structure of the liver was observed by staining of HE and the liver glycogen were measured by staining of glycogen.NOS3 was examined by S-P immunohistochemical method.Results The liver structure of the sport control group was injured seriously,the isoenzyme of LDH4 and LDH5 were higher,the liver glycogen were poor,NOS3 was decreased compared with the normal control group(P
