1.THE ANTI-FATIGUE EFFECT OF BLACK CURRANT EXTRACTS IN MICE

Objective To study the anti-fatigue function of black currant extracts in mice. Method Kunming mice (SPF) were randomly divided into black currant extracts high, middle, low dose group, black currant natural juice dose group and control group according to their body weights. Mice were administered ig daily for 30 d. The loaded-swimming time ,hepatic glycogen ,blood urea nitrogen were measured. Results Black currant extracts high, middle, low dose groups could increase the time of loading swimming,and the hepatic glycogen content,and decrease blood urea nitrogen in mice. All indices above were more than those in black currant natural juice except the blood urea nitrogen content was lower. Conclusion Black currant extracts could alleviate physical fatigue in mice, and be better than black currant natural juice.
2.Effect of dexmedetomidine on cell apoptosis during acute lung injury induced by blunt chest trauma in rats

Objective To evaluate the effect of dexmedetomidine on the cell apoptosis during the acute lung injury induced by blunt chest trauma in rats.Methods Thirty pathogen-free male SpragueDawley rats,weighing 240-270 g,aged 8 weeks,were divided into 3 groups (n=10 each) using a random number table:shan operation group (group Sham),blunt chest trauma group (group T) and blunt chest trauma plus dexmedetomidine group (group TD).The model of acute lung injury was induced by dropping a 300 g weight onto a precordium in anesthetized rats.After the model was established,dexmedetomidine was intravenously infused at a rate of 5 μg · kg 1 · h-1 for 6 h in group TD.At 6 h after the model was established,blood samples were collected from the femoral artery for determination of concentrations of serum tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6).The rats were sacrificed after blood sampling,and lungs were removed for examination of the pathological changes (with light microscope) and for determination of Bax and Bcl-2 expression in lung tissues (by immuno-histochemistry) and cell apoptosis (by TUNEL).Bcl-2/Bax ratio and apoptosis index (AI) were calculated.Results Compared with group C,the serum TNF-α and IL-6 concentrations and AI were significantly increased,the expression of Bax and Bcl-2 was up-regulated,and Bcl-2/Bax ratio was decreased in T and TD groups (P< 0.01).Compared with group T,the serum TNF-α and IL-6 concentrations and AI were significantly decreased,the expression of Bax in lung tissues was down-regulated,the expression of Bcl-2 in lung tissues was up-regulated,Bcl-2/Bax ratio was increased (P<0.05),and the pathological changes of lung tissues were significantly attenuated in group TD.Conclusion The mechanism by which dexmedetomidine mitigates the acute lung injury induced by blunt chest trauma is related to inhibition of cell apoptosis in rats.
3.Early Pelvis Control Training for Patients with Hemiplegia

Objective To explore the effect of early pelvis control training on the balance function in patients with hemiplegia. Methods 60 early stroke patients were divided into the treatment group and the control group. The patients in the treatment group accepted the early pelvis control training based on the normal early rehabilitation which was given to the control. They were evaluated with Berg Balance Scale and the Modified Bathel Index before and 2 months after treatment. Results The scores of Berg Balance Scale and the Modified Bathel Index of the treatment group improved more than that of the control group (P<0.05). Conclusion The early pelvis control training can improve the balance dysfunction after hemiplegia effectively.
4.Dose-effect relationship between epidermal growth factor and its protection of the intestinal tract from the radioactive damage

BACKGROUND: Epidermal growth factor can inhibit gastric acid secretion, promote the healing from the ulcer, protect mucous membrane, and enhance the growth and proliferation of mucous membrane cells of the gastrointestinal tract.OBJECTIVE: To investigate the effect of epidermal growth factor at different doses on the barrier function of the intestinal mucous membrane in rats after radioactive damage.DESIGN: A randomized controlled trial.SETTING: Department of Clinical Trophology, First Affiliated Hospital and College of Public Health.MATERIALS: The experiment was completed at the Public Health Department of Xinjiang Medical University and the First Affiliated Hospital of Xinjiang Medical University from August 2001 to May 2003. Totally 80 newborn Wistar rats, half male and half female, were involved in the study.METHODS: Wistar rats were randomly divided into the control groups(positive and negative) and intervention groups(30 μg/kg, 20 μg/kg, 10 μg/kg) with 16 rats in each group and half male and half female. The models were made by a single radiation of 9.5Gy on the abdomen except for the negative control group. Epidermal growth factor was injected subcutaneously once per day for intervention. Eight days later, the body mass, food intake, efficiency of food utilization were observed and the morphology of the intestinal mucous membrane was checked.dermal growth factor on the increased body mass, food intake and food intake observation of the intestinal mucous membrane under the optic microscope and the electric-microscope.group of rats with epidermal growth factor of 30, 20, 10 μg/kg and the intake of the female and male rats in the group with epidermal growth factor of 30, 20, 10 μ g/kg was significantly different compared with that of the rats in the negative control group( P < 0.05), while there was no significant under the optic microscope and electric microscope showed that when epidermal growth factor was administrated, epidermal growth factor could apparently improve the intestinal mucosal barrier function with the increase of the dose, especially in the group with epidermal growth factor of 30 μ g/kg,which indicates that epithelial cells were increased obviously and cup shaped cells were also increased.CONCLUSION: Epidermal growth factor might not affect the body mass,food intake and efficiency of food intake of the rats, but large doses of epidermal growth factor can improve the morphology of the intestinal mucous membrane and promote the proliferation and repairs.
5.Association between dietary patterns during pregnancy and gestational diabetes mellitus

Objective:
To explore the association between dietary patterns during pregnancy and the development of gestational diabetes mellitus (GDM), so as to provide the guidance of diet balance and GDM prevention for pregnant women.
Methods:
Pregnant women who underwent prenatal examination at the First Affiliated Hospital of Xinjiang Medical University from December 2021 to July 2022 were selected as the subjects. The oral glucose tolerance test (OGTT) was employed at 24 to 28 weeks of gestation to diagnose GDM. The questionnaire of "The Survey on Nutrition and Health of Pregnant Women in China" was used to collect general information, food intake frequency and amount during a month before OGTT. Dietary patterns were determined by factor analysis, and the association between dietary patterns and the development of GDM was identified using a multivariable logistic regression model.
Results:
Totally 449 pregnant women were included, with a median (interquartile range) age of 31.00 (5.00) years and a median (interquartile range) gestational age of 35.00 (3.00) weeks. There were 7 dietary patterns named potato-cereal pattern, poultry-meat-dessert pattern, milk-egg-aquatic product pattern, mushroom-vegetable-offal pattern, fruit-bean pattern, pickle-wheaten food pattern, and beverage-nut pattern according to the food types with high factor loads. There were 89 cases diagnosed as GDM, with a prevalence rate of 19.82%. Multivariable logistic regression analysis showed that the development of GDM was associated with the poultry-meat-dessert pattern (OR=0.242, 95%CI: 0.086-0.678) and the fruit-bean pattern (OR=0.093, 95%CI: 0.025-0.342), compared with the potato-cereal pattern.
Conclusion
Compared to the potato-cereal pattern, the poultry-meat-dessert pattern and the fruit-bean pattern may reduce the risk of GDM.

6.Preparation and testing of hepatitis C core antigen ELISA reagent

Objective To establish a sandwich enzyme-linked immunosorbent (ELISA) method for early diagnosis of hepatitis C. Methods Immunization of Balb/c mice with hepatitis C core antigen were prepared by genetic engineering. Mouse monoclonal antibodies (McAb) to anti-HCV-core Ag were obtained. A sandwich ELISA kit for detecting HCV-core Ag was developed by using four strains of anti-HCV-core Ag McAb. One hundred and twenty five serum specimens with increased ALT but negative for anti-HCV, anti-HIV, and anti-Tp tests were tested. Results Nine of the 125 specimens were positive for HCV-core Ag. Conclusion The double sandwich ELISA kits for detecting HCV-core Ag may be useful for the early diagnosis of hepatitis C .
7.Research advances in association between diseases outside the gallbladder and gallstones

Gallstones is a common digestive system disease in China and has a complex pathogenesis. It is often considered that it is mediated by many factors in the genetic background, including biliary cholesterol supersaturation, intestinal cholesterol hyperabsorption, abnormal cholesterol nucleation in bile, dysfunction in gallbladder dynamics, and cholestasis. This article reviews the role of the above mechanisms in the association between diseases outside the gallbladder and gallstones. Based on current evidence, the above diseases may promote the formation of gallstones, and active intervention of the above diseases outside the gallbladder may help to expand new strategies for the prevention and treatment of gallstones.
8.Study on the complex hepatitis C virus antigen with multi-genotype E1 epitopes

Objective To design a complex hepatitis C vires(HCV)=E1 antigen,and to search its application in HCV vaccine and diagnosis test.Methods Through consulting the database and widely comparison of sequences from HCV E1 of different genotype,the representative immunodominant epitope sequences were selected from all the six genotypes.Their genes were deduced according to the preference codon in E.coli and three fragments were designed to contain the six epitopes.They were chemically synthesized and cloned into pBVIL1 vector separately.The cloned fragments were conjugated together profiting from pBVIL1's property,and then a doubled pan-DR helper T cell epitopes(PADRE)gene was inserted to form a complex expression plasmid.The transformed E.coli cells with this plasmid were cultured and induced to express recombinant protein and the antigenic activity of the product was tested.Results An expressing plasmid containing 8 epitopes from HCV genotype 1a,1b,2a,3a,4a,6a and a doubled PADRE sequences was constructed successfully and the engineering E.coli transformed with this plasmid highly expressed after inducing culture at 42℃ in an inclusion manner.The immunological activity of the purified recombinsnt multi-epitope antigen shows that:(1)It can react with a great part of sera from HCV positive patients by indirect ELISA.(2)It can induce notable specific humoral immunity in injected mice.Conclusion The novel constructed expressing plasmid and its product may be useful in study of a newly HCV vaccine as well as an antigen for HCV immunoassays.

Objective To research CpG and Al(OH)3 adjuvants enhancing immunogenicity of hepatitis C virus(HCV) recombinant ptotein combined vaccine(TIE).Methods BALB/c mice were immunized with candidate vaccine TFE using CpG,Al(OH)3,Al(OH) 3 + CpG,or freund's adjuvant(FA) as the adjuvant.Five mice were sacrificed after 10 d of the last immunization.Specific antibodies in sera were tested by enzyme-linked immunosorbent assay(ELISA).Splenic cells were isolated and levels of IFN-γ,IL-4 and cytotoxic T lymphocyte(CTL) cytotoxicity assay were messuredin vitro.The remaining mice were subcutaneouly injected with 1 × 106 SP2/0-NS3 cells on the back to investigate the protective effects.The differences of means between groups were compared by LSD-t test.Results The specific CTL activity of TFE + A1(OH) 3 + CpG group was higher than TFE + FA group and TFE + CpG group(P < 0.05).The level of IFN-γsecreting cells in TFE + Al(OH)3 + CpG group was higher than that in TFE + M(OH)3 group or TFE + CpG group(P < 0.05).Conclusion Combining Al(OH) 3 and CpG could enhance specific cellular immunogenicity of candidate HCV vaccine TFE.TFE + M(OH) 3 + CpG could effetively prevent the attack of tumor cell SP2/0-NS3 expressing nonstructural protein NS3 of HCV.
