1.Diagnostic value of four serum tumor markers in lung cancer

Objective To investigate the diagnostic value of serum tumor markers carcinoembryonic antigen (CEA) ,neuron spe-cific enolase(NSE) ,cytokeratin-19 fragments(CYFRA21-1) ,squamous cell carcinoma antigen(SCCA)in the lung cancer .Methods By using electro chemiluminescence immunoassay ,the concentrations of CEA ,NSE ,CYFRA21-1 and SCCA were determined in 98 cases of lung cancer and 30 cases of healthy control group ,and the positive rate ,sensitivity ,specificity ,accuracy were calculated .Re-sults In different lung cancer pathological groups ,the concentrations of the 4 serum tumor markers were significantly higher than the control group(P<0 .05) .CEA had the highest diagnostic sensitivity for detecting adeno carcinoma ,NSE had the highest diag-nostic sensitivity for detecting small cell lung cancers ,CYFRA21-1 had the highest diagnostic sensitivity for detecting squamous carcinoma .The combined tests of four serum tumor markers had higher diagnostic sensitivity and accuracy than single test for lung cancers .Conclusion The four serum tumor markers are valuable in the pathological classification ,diagnosis and differential diagno-sis of lung cancer .The diagnostic sensitivity and the accuracy are obviously higher in combined tests .
2.The protective effects of TGP on local cerebral ischemia in rat

AIM To observe the protective effects of TGP on local cerebral ischemia. METHODS The local cerebral ischemia in rat was made by middle cerebral artery occlusion(MCAO). The infraction weight was determined by NBT stain. SOD, MDA and apoptosis were determined with different method respectively. RESULTS TGP 20 mg*kg-1 ip markedly improved the abnormal nervous symptoms, increased the SOD activity and reduced contentes of MDA in MCAO rat. TGP 20 mg*kg-1 ip significantly decreased the numbers of apoptotic cells in ischemic cortex. CONCLUSION TGP has protective effects against cerebral infarction, and its mechanism may be related to anti-apoptosis and free radical.
3.THE EFFECT OF TOTAL FLAVONES OF HIPPOPHAE RHAMNOIDES L. ON BEATING AND ELECTRICAL ACTIVITY OF CULTURED CARDIAC CELL

In this paper, we studied the effect of total flavones of Hippophae rhamnoides L (TFH) on beating and transmembrane potential of cultured sucking rat cardiac cell. The results indicated that cellular beating rate and beating amplitude were significantly reduced, the action potential duration of 50% and 90% repolarization was sbortened, the slope of phase 4 depolarization was decreased, and other parameters of the action potential showed no significant change. In addition, we also observed that TFH could make the spontaneous abnormal beating of cell change into regular beating. These findings suggested that the mechanism of TFH on cultured cardiac cell might have some relation to antagonizing inward flow of Ca~(2+) and aocelerating the move of k' outward.
4.Development and Identification of Monoclonal Antibodies against the Recombinant P38 Antigen of Schistosoma japonicum

Objective To express P38 of Schistosoma japonicum in Escherichia coli BL21 and develop the monoclonal antibodies (McAb) against rSjP38. Methods The recombinant plasmid pET32(a)-P38 was transformed into E.coli BL21(DE3). 1 mmol/L IPTG (isopropyl-beta D-thiogalactopyranoside) was used to induce the expression of the recombinant rSjP38. The rSjP38 was soluble in supernatant after sonication and further purified by His-Ni chromatography. BALB/c mice were immunized with the purified rSjP38 and hybridomas were generated with traditional technique. McAbs were screened by ELISA with limited dilution. The subtype and specificity of McAb were identified by kit and Western blot respectively. Results Eight hybridoma cell lines secreting monoclonal antibodies to rSjP38 were obtained. The subtype of all the 8 McAbs are IgG1. Western blotting showed that the 8 McAbs reacted strongly and specifically with native antigen (P38) of Schistosoma japonicum. Conclusions Eight hybridoma cell lines secreting highly specific McAbs against P38 have been established.
5.Prevalence of the Mild Cognitive Impairment among Elderly in Beijing

Objective:To study the prevalence of mild cognitive impairment (MCI) among elderly people and associated factors to it.Method:A sample of 1865 elderly people aged 60 or above living in Beijing metropolitan area was investigated by a two-stage survey.In the first stage, a door-to-door visit to the subjects was performed by a trained interviewer to conduct general questionnaire including activity daily living assessment, memory and cognitive test.On the second stage, neurologists examined all screened subjects to collect family history and to finish general and neurological examination as well as neuropsychological testing.MCI was diagnosed by the consensus of 2 neurologists.Result:Among the 1865 subjects visited, 217 (11.6%) were defined to have MCI.After adjusted for age and gender the standardized prevalence reaches 8%.Within the people with MCI 22.6% was classified as vascular MCI.The prevalence of MCI was higher in people living in rural area, in people with lower educational attainment, and in people who were labor workers.The prevalence increased with age advancing.Conclusion:Similar to dementia, people with advance aged, with low education and living in rural area are those at high risk of developing MCI.
6.Study of the Alendronate inhibiting growth and inducing apoptosis of giant cell tumor of bone in vitro

[Objective]Giant cell tumor of bone is notorious for its local aggressive behavior and its tendency to recur after operative treatment.Bisphosphonates is the drug of anti-osteoporosis.It is found also have anti-cancer effect recently.We conducted experiment examing the effect of bisphosphonates alendronate on the growth and survival of the cells.To study if bisphosphonates are capable of inducing cells death and significantly inhibiting their growth in vitro.[Method]Cells viability was detected by MTT Assay after the tumor cells were cured with different concentration and different time.Tumor cells apoptosis with in situ TUNEL assay and flow cytometry was detected.The active Caspase-3 was also detected.[Result]After exposure to alendronate,the cells exhibited the characteristic features of cell shrinkage,rounding and partial detachment,and demonstrated the lobulated appearance of apoptotic cells.It was much more prominent while the treating time prolonged or the concentration increased.Alendronate((5 200) M) treatment for 24 h,resulted in 2.79%~31.17% decrease in cell viability,and 11.13%~49.94% for 72 h,respectively.A significant dose-dependent and time-dependent decrease in the number of viable cells was observed in the GCT cells.After Alendronate treat for 24 h,the mean cell population in apoptosis was 14.32% at concentration 5 mmol/l,and 40.24% at 200 mmol/l.It was up to 18.41% and 42.22% respectively after 48 h.In Alendronate-treated GCT cells,Caspase-3 activation was observed.The cell response varied with doses of Alendronate showing the levels of Caspase-3 expression with a dose dependent response.[Conclusion]In conclusion,we demonstrated that bisphosphonate alendronate could inhibit GCT cells in the present study.This response was time-dependent and dose-dependent.Alendronate inducing apoptosis in GCT cells is mediated by the activation of Caspase-3.
7.Effects of chidamide combined with curcumin on human cutaneous T-cell lymphoma cell line Hut78 and their molecular mechanisms

Objective To evaluate the inhibitory effect of chidamide combined with curcumin on the proliferation of cutaneous T-cell lymphoma (CTCL) cell line Hut78,as well as their promotive effect on its apoptosis,and to explore their therapeutic mechanisms in CTCL.Methods Some Hut78 cells were treated with different concentrations of chidamide (0.3,0.6,1.2,2.4 μmol/L) and 10 μ mol/L curcumin alone or the combination of 1.2 μ mol/L chidamide and 10 μ mol/L curcumin for 24,48 and 72 hours separately.MTS assay was conducted to estimate cell viability at each time point.After selection of chidamide concentrations,some Hut78 cells were treated with chidamide (0.6 and 1.2 μmol/L)and curcumin (10 μmol/L) alone or in combination (1.2 μmol/L chidamide and 10 μmol/L curcumin) for 24 hours,then,flow cytometry was performed to detect cell apoptosis and analyze cell cycle,real-time (RT)-PCR and Western-blot analysis were conducted to quantify the mRNA and protein expressions of apoptosis-associated genes Fas,caspase 8,nuclear factor (NF)-κB p65 as well as cell cycle-associated genes P21,CDK2 and cyclin E respectively.Statistical analysis was carried out by repeated-measures analysis of variance,one-way analysis of variance and the least significant difference (LSD)-t test.Results Chidamide could significantly inhibit the proliferation of Hut78 cells in a dosedependent and time-dependent manner (F =266.558,564.966,respectively,both P < 0.001).After 48-and 72-hour culture,the combination of 1.2 μmol/L chidamide and 10 μmol/L curcumin showed significantly stronger inhibitory effect on cell proliferation compared with 1.2 μmol/L chidamide or 10 μmol/L curcumin alone (all P < 0.001).As flow cytometry showed,the percentage of apoptotic cells was significantly higher in the combined treatment group than in the 0.6-,1.2-μmol/L chidamide groups and 10-μmol/L curcumin group (all P < 0.001).Compared with the 0.6-μmol/L chidamide group and 10-μmol/L curcumin group,the combined treatment group and 1.2-μmol/L ehidamide group both showed significantly increased proportion of cells at G0/G1 phase and mRNA expressions of Fas,caspase 8 and P21,but decreased proportion of cells at S phase or G2/M phase and mRNA expressions of NF-κB p65,CDK2 and cyclin E (P <0.05 for proportion of cells at different phases,P < 0.001 for mRNA expressions of different genes).Furthermore,the mRNA expression of Fas was significantly higher in the combined treatment group than in the 1.2-μmol/L chidamide group (P < 0.001),while no significant differences were observed in the mRNA expressions of caspase 8,P21,NF-κB p65,CDK2 and cyclin E or the proportion of cells at any phase between the combined treatment group and 1.2-μmol/L chidamide group (all P > 0.05).Western-blot analysis showed that protein expressions of Fas,caspase 8 and P21 significantly increased,but those of NF-κB p65,CDK2 and cyclin E significantly decreased in the combined treatment group compared with the 0.6-,1.2-μmol/L chidamide groups and blank control group receiving no treatment,which were in accordance with the above changes in mRNA expressions of these genes.Conclusion Chidamide can inhibit the growth of the CTCL cell line Hut78 by directly decelerating cell proliferation and inducing cell apoptosis,and the combibation with curcumin can markedly enhance the inhibitory effect of chidamide on the growth of Hut78 cells.
8.Clinical Observation of Aerosol Inhalation of Magnesium Sulfate Combined with Salbutamol Sulfate in the Treatment of Acute Attack of Severe Bronchial Asthma in Children

OBJECTIVE:To investigate the clinical efficacy and safety of aerosol inhalation of magnesium sulfate and salbuta-mol sulfate in the treatment of acute attack of severe bronchial asthma in children. METHODS:85 children with acute attack of se-vere bronchial asthma were divided into control group(45 cases)and intervention group(40 cases)in accordance to even and odd number of hospitalization time. Based on routine treatment of glucocorticoid intravenous injection,oxygen uptake,sedation and nu-tritional support,control group was given aerosol inhalation of salbutamol sulfate,0.6 ml each time,once every 20 min,for 1 h;intervention group was additionally given aerosol inhalation of magnesium sulfate,0.6 ml each time,once every 20 min,for 1 h, on the basis of control group. Therapeutic efficacy,improvement time of shortness of physical examination indexes (breath, wheeze,coughing),lung function indicators and ADR were observed in 2 groups,and discharge time was recorded in 2 groups. RESULTS:The total effective rate of clinical efficacy of intervention group was 95.00%after 1 h treatment,which was significant-ly higher than 77.78% in control group,with statistical significance (P<0.05);the time of shortness of breath improvement, wheeze improvement,coughing improvement and discharge in intervention group were significantly shorter than in control group, with statistical significance (P<0.05);PEF and FEV1 of intervention group were improved significantly compared to control group,with statistical significance(P<0.05). Both groups suffered from slight ADR,there was no statistical significance in the in-cidence of ADR between 2 groups (P>0.05). CONCLUSIONS:On the basis of conventional treatment,the aerosol inhalation of magnesium sulfate and salbutamol sulfate shows good clinical efficacy and safety in the treatment of acute attack of severe bronchi-al asthma in children.
9.Protection of the external branch of the superior laryngeal nerve in carotid endarterectomy

Objectives To investigate the methods of protecting external branch of the superior laryngeal nerve (EBSLN)in carotid endarterectomy and to observe the effect of using these methods in clinical surgery. Methods EBSLN (20 sides)of 10 heads of corpse were studied by using microanatomy from January 2013 to December 2013. The occurrence probability of EBSLN on the lower edge of posterior belly of digastric muscle,medial edge of external carotid artery and upper edge of superior thyroid artery in anatomy triangle was analyzed. The distances from the midpoint of the EBSLN to carotid bifurcation, mandibular angle and mastoid tip were measured. Sixty-five patients with carotid endarterectomy in Tianjin Huanhu Hospital from December 2013 to November 2014 were treated with the protective methods of the relevant EBSLN by using anatomy triangle as a mark. Whether the patients had injury symptoms of EBSLN were followed up after procedure. Results (1)The occurrence probability of 20-side EBSLN in anatomy triangle was 95%(19 sides). The midpoint of EBSLN in the anatomy triangle at the posterior mandibular angle was median 0. 34 (-1. 62 to 2. 43)cm,at the inferior mandibular angle was 1. 28 (-1. 33 to 3. 42) cm,at anterior mastoid tip was 2. 84 (0. 51 to 5. 14)cm,at inferior was 4. 51 (2. 82 to 6. 39)cm,and at anterior superior of the carotid bifurcation was 1. 64 (0. 57 to 3. 78)cm. (2)65 patients who underwent carotid endarterectomy used the protective methods of intraoperative EBSLN. There was no manifestation of EBSLN injury at 3 weeks to 9 months after procedure. Conclusion In carotid endarterectomy,taking an anatomic triangle as a symbol,it is no more than 2 cm of the anterior superior of carotid bifurcation during the separation process. As for the patients with higher or lower position of carotid bifurcation,in the range of crossing rear mandibular angle 0. 50 cm or below the mastoid tip 4. 50 cm for arterial separation should be avoided,and this can effectively protect EBSLN.
10.Systematic Evaluation on Danshen Chuanxiongqin Injection in the Treatment of Angina Pectoris

Objective To systematically evaluate the clinical efficacy and safety of Danshen Chuanxiongqin Injection (DCI) in the treatment of angina pectoris. Methods Randomized controlled trials (RCTs) regarding DCI in the treatment of angina pectoris were searched in CNKI, VIP, Wanfang Database, CBM, PubMed, Embase and Cochrane Library by Feb. 2014. Two researchers independently retrieved the RCTs and extracted the information. The Cochrane risk of bias method was used to assess the quality of the included studies, and a meta-analysis was conducted with Review Manager 5.2 software. Results A total of 12 RCTs with 1145 participants were included. The meta-analysis indicated that the combined use of DCI and conventional treatment with western medicine was more effective in the outcomes of the total clinical effective rate [RR=1.27, 95%CI (1.19,1.35), P<0.000 01], the total effective rate of ECG [RR=1.34, 95%CI (1.23,1.46), P<0.000 01], and decrease of plasma viscosity [MD=-0.15, 95%CI (-0.25,-0.05), P=0.004] and fibrinogen [MD=-0.96, 95%CI (-1.14,-0.78), P<0.000 01]. And there were no adverse drug reaction reports. Conclusion Based on this systematic evaluation, DCI combined with conventional therapy is effective and relatively safe in treating angina pectoris, but it still needs larger samples, multi-center, and high quality RCT to verify.
