1.THE KARYOTYPE ANALYSIS OF THE TRADITONAL CHINESE MEDICINAL HERB COIX LACHRYMA-JOBI L

The counts of the chromosome and karyotypeanalysis of coix lachryma-jobi L. (a traditional Chi-nese medicinal herb) are represented in this pa-per. The results show that the number of the chro-mosome of coix lachryma-jobi L. is 2n=20. All ofthe homologous chromosomes have median kine-tochrores. The absolute length of the chromo-somes tanges from 2. 26~10. 5?m. The relativelength is 7. 19~13. 02%. The karyotype formulais K (2n)=20=20m. These results provide morecritical cytological criteria for identification of Coixlachryma--jobi L. on the chromosomal level. Inaddition, It can be provided the data for studyingof the fraditional Chinese medicinal herbs againstcancer.

3.Nodal marginal zone B-cell lymphoma with monocional IgM:2 cases report and review of literatures

Objective To elucidate the clinical and pathological features, diagnosis, treatment and prognosis of nodal marginal zone B-cell lymphoma (NMZL). Methods 2 patients, with NMZL who has extremely elevated monoclonal IgM were analyzed and review were included. Results The common pathological diagnosis caused by the association of monoclonal gammopathy with indolent B-cell non-Hodgkin lymphomas was marginal zone lymphoma or B small cell lymphocytic lymphoma/B-cell chronic lymphocytic leukemia. According to our cases and literatures, the main distribution of involved sites in NMZL were lymph nodes, spleen and bone marrow. Sixty percent of the NMZL patients presented with advanced stage with, high risk of IPI. The diagnosis of NMZL was based on pathology of the lymph node. The proliferation and distribution of the differentiation monocytoid cell had a specified significance. Prognosis of early stage ( I or Ⅱ) NMZL were much better than that of advanced stage NMZL even the patients had received chemotherapy.Conclusion NMZL mainly involved lymph nodes, bone marrow and spleen, sporadically associated with the elevated monoclonal IgM. Most of the NMZL patients presented with advanced stage.The prognoses were poor.
4.Histological changes of pure titanium micro-implant under inflammatory stimulation and orthodontic force as well as bone remodeling

BACKGROUND:Implant stability is mainly influenced by peri-implant inflammatory stimulation.
OBJECTIVE:To build a beagle model of peri-implantitis under orthodontic force and to observe the bone remodeling of the Beagle dog model.
METHODS: Micro-implants were randomly implanted into the maxilary interradicular region at the center of the mesial and distal roots of bilateral P2, P3, P4 and M1 of Beagle dogs. One side served as a loaded micro-implant with peri-implantitis under 100 g of orthodontic force at 1, 2, 3, 4 weeks of peri-implantitis, and the force lasted for 1 month. After that, the animals were kiled to prepare specimens with micro-implants. Bone-to-implant contacts were calculated and histological changes of the bone interface under continuous orthodontic force at different stages of peri-implantitis were observed under light microscope.
RESULTS AND CONCLUSION:There were a large number of inflammatory cels after micro-implants were implanted with silk thread ligation to the cervical part. Over time, inflammatory cels were gradualy diffused to the tip of micro-implant, and there were a great quantity of colagenous fibers, osteocytes and active bone remodeling. When the inflammation was diffused to the tip of micro-implant after 2 weeks of peri-implantitis, woven bones composed of newly formed trabeculae appeared, and imflammatory cels dispersed. The medulary cavity was irregular after colagen fibrils absorption, and there were 3-4 layers of osteoblasts in the bone lacunae, with active bone formation. These findings indicate that the Beagle model of pure titanium peri-implantitis under orthodontic force was successfuly built in the experiment, and bone formation became active at 2 weeks after modeling.
5.Effects of Different Processing Methods on the Contents of Oleanolic Acid and Ursolic Acid in Chaenomeles Medicinal Materials

Objective To investigate effects of different processing methods on the contents of oleanolic acid and ursolic acid in chaenomeles medicinal materials. Methods The contents of oleanolic acid and ursolic acid in samples processed by different processing methods in producing area were determined by HPLC with Agilent HC-C18 (2) chromatographic column (4.6 mm×250 mm, 5 μm), mobile phase of methanol-water-acetic acid-triethylamine (89∶11∶0.1∶0.05), detection wavelength of 210 nm, column temperature at 25 ℃, the flow rate of 1.0 mL/min. Results The total contents of oleanolic acid and ursolic acid in the samples prepared through 4 different processing methods were 0.86%, 0.93%, 1.29%, and 1.47%, respectively, showing that different processing methods in producing area had a significant effect on contents of oleanolic acid and ursolic acid in chaenomeles medicinal materials. Conclusion This study provides a basis for processing methods of chaenomeles medicinal materials in producing area.
6.Heat shock protein 70 in liver fibrosis in infants with biliary atresia and its impact on prognosis

Objective To investigate the role of heat shock protein 70 (HSP70) in liver flbrosis in infants with biliary atresia (BA) and its impact on prognosis. Methods Fourty-six (46) cases of infants with BA undergoing elective Kasai surgery were selected. In the same period, 30 cases of children with choledochal cyst and 17 cases of children with portal vein cavernous transformation were selected. The expressions of HSP70 in liver tissues were detected using immunohistochemical staining. The liver flbrosis in children with BA was detected using Sirius red-saturated picric acid staining. The expressions of HSP70 proteins in different flbrotic liver tissues were detected by using double staining. All postoperative BA infants were followed up and ended at June 30, 2016.Results The proportion of high expression of HSP 70 proteins in BA infants were signiflcantly higher than that in children with choledochal cyst and vein cavernous transformation (P<0.05). Rank correlation analysis showed that the expressions of HSP70 in liver tissues were positively correlated with the degree of liver flbrosis (r=0.861,P<0.05). 15 patients died among 46 cases of BA infants. The survival rate in BA children with mild liver flbrosis was 82.4%, which was signiflcantly higher than 58.6% in the severe group (P<0.05). The survival rate in HSP70 protein low expression group was 85.0%, while in HSP70 protein high expression group was 53.8%. Kaplan-meier survival analysis showed that the median survival time in the HSP70 protein low expression group was (34.0±2.6) months, while in the HSP70 protein high expression group was (18.3±2.2) months, the difference was statistically signiflcant (χx2=4.765,P=0.029).Conclusions The expressions of HSP70 proteins in liver tissues in infants with BA were high and were positively correlated with the degree of hepatic flbrosis. It suggested the possible involvement of HSP70 in the process of liver fibrosis. The upregulated expressions of HSP70 often indicated poor prognosis in children. It could be used as determining biomarker for prognosis.

8.Community-acquired infections and risk factors in patients with diabetes:A Meta-analysis

Objective To analyze the types and risk factors of community-acquired infections (CAI)in diabetic patients by system analysis method of evidence-based medicine.Methods China National Knowledge Infrastructure (CNKI),Wanfang database,VIP database were searched by computer,domestic published researches on CAI and related risk factors in dia-betic patients were aggregated,Meta-analysis was conducted by stata 1 1 .0 software.Results A total of 1 2 literatures were included in the study .The average rate of CAI in diabetic patients was 39.55% (22.12%-55.86%).The major infec-tions were respiratory system infection(40.74%),urinary tract infection(27.35%),tuberculosis(10.80%),skin and soft tissue infection(9.19%),and hepatobiliary system infection (5.57%).Stratified analysis on risk factors revealed that OR and OR95%CI of chronic complication,age,disease course,glycemic control,gender,type of diabetes,subtype of ketoac-idosis was 1.63(1.45,1.82),1.30(1.19,1.42),1.47(1.35,1.61),0.68(0.61,0.76),0.69(0.64,0.75),1.37 (1.13,1.66 )and 0.87(0.62,1.23),respectively.There was no publication bias and combined results were stable. Conclusion The main CAI in diabetic patients are respiratory system infection,urinary tract infection,tuberculo-sis,skin and soft tissue infection,and so on ;several factors,such as female,older age,long-term disease course, poor glycemic control,and complication,can contribute to the increase of CAI in diabetic patients.
9.Preparation and Tumor Inhibition Effect of Transferrin Modified Paclitaxel-loaded Liposome

OBJECTIVE:To prepare transferrin modified paclitaxel-loaded liposome(TF-PTX-LP),and to study the tumor in-hibition effect. METHODS:TF-PTX-LP was prepared by thin-film method,and morphology of TF-PTX-LP was observed. Qualita-tive and quantitative investigation were used to value the uptake efficiency of TF-LP and LP by HepG2 cells. The proliferation inhi-bition rate of HepG2 cells was investigated after treated with PTX,PTX-LP and TF-PTX-LP for 24,48 and 72 h. Tumor spheres were prepared by using HepG2 cells. Effects of normal saline,PTX,PTX-LP and TF-PTX-LP on the volume of tumor spheres were investigated after 0,1,2,4,5,6 and 7 d treatment. HepG2 tumor-bearing nude mice model was induced. Inhibitory effects of normal saline,PTX,PTX-LP and TF-PTX-LP(8.5 mg/kg by PTX)on transplantable tumor of tumor-bearing nude mice were in-vestigated. RESULTS:TF-PTX-LP showed uniform spherical shape,with particle size of 100-120 nm. The fluorescence intensity of HepG2 cells treated with TF-LP was stronger than that treated with LP(P<0.01). Compared with PTX and PTX-LP,TF-PTX-LP showed higher proliferation inhibition rate(P<0.01). Compared with normal saline,PTX and PTX-LP,tumor spheres were small-er in volume after treated with TF-PTX-LP,and inhibition rate of tumor was higher in tumor-bearing nude mice;there were statisti-cal significance after treated for 6,7 d(P<0.01). The proliferation inhibition rate and tumor spheres volume changed in time-de-pendent manner. CONCLUSIONS:TF-PTX-LP which owns good tumor inhibition effect is prepared successfully.
10.The integrin linked kinase promotes migration and invasion of lung cancer cell by upregulating matrix metalloproteinase-9 through nuclear factor-κB pathway

Objective To investigate the role of integrin-linked kinase (ILK) on migration and invasion of lung cancer cell by upregulating matrix metalloproteinase-9 through nuclear factor-κB (NF-κB) pathway.Methods A549 cell line were overexpressed ILK and matrix metalloproteinase-9 (MMP-9) confirmed by cell transfection,siRNA interference,cell scratch test,real-time quantitative PCR and Western Blot.Results Over-expression of ILK stimulated MMP-9 expression in lung cancer cells(P < 0.01).The addition of MMP-9 inhibitor doxycycline and anti-MMP-9 neutralizing antibody significantly impaired the wound healing capacity of ILK-transfected cells(P < 0.01),as well as by in vitro matrigel invasion assay (P < 0.01).In addition,overexpression ILK induced phosphorylation and nuclear translocation of nuclear factor-κB subunit p65.Upregulation of MMP-9 was severely abolished by either BAY 11-7028,a specific NF-κB inhibitor,or siRNA targeted to NF-κB p65 in ILK over-expression cells.Conclusion The finding indicate that over-expression of ILK can promote the migration and invasion of lung cancer cell,and upregulate MMP-9 through the NF-κB pathway.
