Objective To observe the effect of Bufalin on controlling ECA109 cells , and to explore its potential anti-tumor mechanism. Method The effect of Bufalin on the proliferation of human esophageal cancer ECA109 cells was evaluated by CCK-8 assay and it effects on apoptosis of esophageal cancer ECA109 cells were determined by flow cytometry (FCM). Protein expressions of NF-κBp65, ERK, PERK1/2 and Caspase-3, Cleaved Caspase-3, PARP, Cleaved PARP in esophageal cancer ECA109 cell were observed through Western blot. Results The proliferation of human esophageal cancer ECA109 cells was significantly inhibited in bufalin group in a time- and concentration-dependent manner. Bufalin can induce the apoptosis in human esophageal ECA109 cells. Results of Western blot showed the protein expressions of apoptosis-related protein Cleaved Caspas-3 and Cleaved PARP in esophageal cancer ECA109 cells could be markedly up-regulated by Bufalin , but p-ERK1/2, NF-κBp65 in esophageal cancer ECA109 cells could be markedly down-regulated by Bufalin. Conclusion Bufalin can potently inhibit the growth of esophageal cancer ECA109 cells and the potential anti-tumor mechanism might be involved in inhibiting ERK/ NF-κB signal pathway.

Objective To elucidate the clinical and pathological features, diagnosis, treatment and prognosis of nodal marginal zone B-cell lymphoma (NMZL). Methods 2 patients, with NMZL who has extremely elevated monoclonal IgM were analyzed and review were included. Results The common pathological diagnosis caused by the association of monoclonal gammopathy with indolent B-cell non-Hodgkin lymphomas was marginal zone lymphoma or B small cell lymphocytic lymphoma/B-cell chronic lymphocytic leukemia. According to our cases and literatures, the main distribution of involved sites in NMZL were lymph nodes, spleen and bone marrow. Sixty percent of the NMZL patients presented with advanced stage with, high risk of IPI. The diagnosis of NMZL was based on pathology of the lymph node. The proliferation and distribution of the differentiation monocytoid cell had a specified significance. Prognosis of early stage ( I or Ⅱ) NMZL were much better than that of advanced stage NMZL even the patients had received chemotherapy. Conclusion NMZL mainly involved lymph nodes, bone marrow and spleen, sporadically associated with the elevated monoclonal IgM. Most of the NMZL patients presented with advanced stage. The prognoses were poor.

Chinese Journal of Clinical Oncology 2015;46(4):226-230
Objective: To investigate clinical and hematological features of malignant lymphoma (ML) combined with venous thromboembolism (VTE). Methods:The clinical data of 65 patients with ML combined with VTE were collected between October 2010 and April 2014 and then retrospectively reviewed. Coagulation function and hemorheology were analyzed. Results:The proportion of males and females among patients with ML combined with VTE was 2.61:1. ML combined with VTE was mainly observed in patients with end-stage cancer. The highest incidence (81.54%) of ML combined with VTE was found in StageⅢB-Ⅳtumor. Among 65 patients with VTE, 43 (66.15%) were found after ML was diagnosed, 55 (84.62%) were deep vein thrombosis (DVT), 7 (10.77%) were pulmonary embolism (PE), and 3 (4.62%) were complicated with both DVT and PE. Common DVT locations were the upper limb and the neck vein of 37 cases (67.27%). Clinical manifestations of ML combined with DVT are limb pain, swelling, and increased skin temperature. Unexplained difficulty in breathing, chest pain, and syncope are also found in ML combined with PE. The total effective rate of 55 cases with DVT was 49.09%, and the rate of those cases with PE was 14.29%. The levels of platelet aggregation, D-dimer, high cut blood viscosity, low cut blood viscosity, plasma viscosity, hematocrit, assembly index of erythrocyte, and rigidity index of erythrocyte were significantly higher in patients with ML combined with VTE than in patients with ML alone;by contrast, APTT, erythrocyte sedimentation rate, deformability index of erythrocyte, and average velocity of blood flow were reduced in patients with ML combined with VTE than in patients with ML alone. Conclusion:DVT, which exhibits high incidence in males, is the most common type observed in ML combined with VTE. DVT is commonly manifested in the upper limb and the neck vein;furthermore, DVT is mainly observed in patients with end-stage cancer. Hematological indexes indicated changes in the sensitivity state of thromboembolism.

Objective To construct and expr es s a recombinant plasmid of nonstructural protein NS3-NS4 of hepatitis C virus ( HCV), and to identify the antigenicity of the expressed protein. Methods A gene region encompassing the nonstructural protei n NS3-NS4 of HCV was amplified by polymerase chain reaction (PCR) from the pUC1 9/HCV template. The recombinant expression plasmid containing the pBV220/NS3-NS 4 sequence was constructed, and the nonfused NS3-NS4 recombinant protein was ex pressed in E.coli DH5? efficiently. The recombinant protein was det ected by SDS-PAGE and ELISA. Results We successfully constructed and expressed the recom binant plasmid in prokaryote. Its antigenicity was detected with 50 standard ser a. Compared with the second-generation diagnostic Kit, the total detection rate was 96%. Conclusion The whole NS3-NS4 protein, a region of dominant immunogenicity, should be the effective component of the HCV diagnostic Kit and provide the clue for developing HCV DNA vaccine.
