1.Studies of the Effects of Hydrogen Peroxide on the Ultrastructure and Permeability of Candida Albicans

The Effects of hydrogen peroxide(H2o2) on the ultrastructure, cellular pea-meability barrier and 3H-TdR-DNA leakage of C. albicans were studied. Damage of the cell, especially leakage of cytoplasm were found under transmission electron microscope. Morphological changes and small hole like structures were also found under scanning electron microscope. Radioactivity assay of the leakage of 3H-TdR-DNA leakage of C. albicans further suggested that the 3H-TdR-DNA leakage of C. albicans was directly due to the damage of cellular permeability barrier. Negative correlation (r=+0.8392--0.9915)between the extent of 3H-TdR-DNA leakage and the number of surviving colony-forming units was found. This indicated that the damage of cellular permeability barrier and leakage of DNA of C. albicans caused by H2o2 was one of the most importent causes of cell killing.
2.Effects of treatment with different fluids on systemic inflammatory response in newborn rabbits with septic shock

Objective To evaluate the effects of treatment with different fluids on systemic inflammatory response in newborn rabbits with septic shock. Methods One hundred New Zealand white rabbits (3-5 days, 45-70 kg) were randomly divided into 5 groups (n = 20 each):group Ⅰ sham operation (group S); group Ⅱ sepsis (group CLP); group Ⅲ lactated Ringer's solution (group R); group Ⅳ 6% HES (group HES) and group V (group R+ HES). The animals were anesthetized with intraperitoneal ketamine and midazolam. Sepsis was induced by cecal ligation and puncture (CLP). The animals in group R and HES received lactated Ringer's solution and 6% hydroxyethyl starch (HES) 30 ml/kg infused iv over 1 h respectively immediately after CLP,while group R+ HES received Ringer's solution 20 ml/kg and 6% HES 10 ml/kg. Nio fluid was infused in groups S and CLP. Carotid artery and jugular vein were cannulated for MAP monitoring, blood sampling and drug and fluid administration. MAP was recorded immediately (T1) and at 0.5, 1, 2, 4, 8 h (T2-6) after CLP. Arterial blood samples were collected at T6 for determination of plasma IL-6 and IL-10 concentrations. Results Compared with those at T1, MAP decreased significantly after CLP at T4-6 in group CLP and at T5.6 in group R but hypotension did not develop after CLP in group HES and R + HES. Plasma IL-6 concentration was significantly higher in group CLP and R than in group S. There was no significant difference in plasma IL-6 concentration between group S and group HES and R + HES. Plasma IL-10 concentration was significantly lower in group CLP and higher in group HES than in group S. There was no significant difference in plasma IL-10 concentration between group S and group R and R + HES. Plasma IL-6 concentration was significantly lower, while plasma IL-10 concentration was significantly higher in group R + HES and HES,and plasma IL-10 concentration was significantly higher in group R or6% HES infused iv alone in maintaining the balance between pro-and anti-inflammatory cytolines in newborn rabbits with septic shock.
3.Correcting Astigmatism with Corneal Incision during Cataract Surgery Guided by Corneal Topography Preoperatively

Objective To explore the effect of correcting astigmatism with corneal incision guided by the corneal topography preoperatively.Methods 97 patients(102 eyes) having undergone phacoemulsification and foldable introcular lens implanation,were divided into two groups according to the incision.Superior temple sclera tunnel incision was made on 51 eyes of the group A,and the clear cornea incision was made on the steepest meridian for the group B(51 eyes).The preexisting cornea astigmatism of all patients reneged from 0.25 D to 5.75 D.Corneal topography examination was made before the operation and one day,1 week,1 month and 3 months after surgery except for visual acuity,slit lamp and fundus examination.The value of surgical induced cornea astigmatism was observed.Results In the group A,the differences were not statistically significant between pre-operation and 1 day,1 week,1 and 3 months after surgery for the value of corneal astigmatism(P>0.05).In the group B,the value of corneal astigmatism in 1 day and 1 week after surgery were a little greater than that of pre-operation,but 1 month later,that was significant better than that pre-operation.The value of corneal astigmatism was reduced from 1.5 D before surgery to 0.52±0.14 D after surgery,from 1.5~2.5 D to 0.88±0.53 D and from over 2.5 D to 1.5±0.72 D respectively.Conclusion The incision guided by corneal topography can correct cornea astigmatism existed pre-operation and get the best visual outcome.
4.Research progress on obesity, adipokines, and esophageal adeno-carcinoma

Obesity has rapidly become a pandemic and is associated with increased carcinogenesis, especially esophageal adeno-carcinoma (EA). Adipose tissue is considered an endocrine organ because of its capacity to secrete a variety of adipokines, such as leptin, adiponectin, and resistin. Adipokines have recently been found to be associated with different types of chronic esophageal diseas-es, including gastro-esophageal reflux diseases, Barrett esophagus, and carcinogenesis. First-hand evidence supporting the inhibitory and/or active function of adipokines has been collected on human EA. This review aims to provide important insights into the potential mechanisms of adipokines in EA progression.
5.Clinical Aspects of Epilepsy in Children with Cerebral Palsy:a Review

The occurrence of epilepstic seizures in patients with cerebral palsy (CP) remains a serious event and is very disruptive for children who already suffered from orther disabilties. Data from population-based studies or neurodevelopmental clinic studies showed that 8%~62% of patients with CP suffer from epilepsy. The incidence is highest in spastic tetraplegia and hemiplegia. Half of the patients were onset within the first year of age. Children with tetraplegia CP tended to have an earlier onset of epilepsy than chlidren with other CP types. Partial seizures were the most common seizure types, followed by infantile spasms and Lennox-Gastaut syndrome. Low birth weight, neonatal seizures, family history of epilepsy, lower intelligence and grey matter damage were found to be related to significantly increased risk of epilepsy. Epilepsy in children wih CP usually had poor outcome, half of them were intractable and needed polytherapy. Only 37%~65.2% of patients became seizure-free.
6.Clinical characteristics of the newly diagnosed Crohn's disease in the elderly

Objective:To investigate a classification on age of diagnosis and its clinical characteristics in hospitalized elderly patients with first diagnosed Crohn's disease.Methods:This was retrospective case-control study.A total of 181 newly diagnosed CD patients were admitted to Huadong Hospital from January 2005 to December 2019.According to Montreal CD classification criteria based on the age at the time of diagnosis(sub-grouping A3 into A3 and A4 in this paper), 181 CD patients were classified into four groups: the A1 group(≤16 years, n=4), A2 group(17-40 years, n=60)and A3 group(41-59 years, n=63)(above three as control groups), and the A4 group[≥60 years, with elder-onset Crhon's disease(EOCD), n=54]as a study group.Results:A total of 181 first diagnosed CD patients who met the diagnostic criteria were included in this study.During the investigation period, the total incidence rate of CD was 37.90/100, 000, and the total prevalence rate of CD was 142.79/100, 000.Among the 181 first diagnosed CD patients, there were 4 patients(2.2%)with age of diagnosis of ≤16 years(group A1), 60 patients(33.2%)with age of diagnosis of 16-40 years(A2), 63 patients(34.8%)with age of diagnosis of 41-59 years(A3)and 54 patients(29.8%)with age of diagnosis of ≥=60 years(A4). Of these first diagnosed CD patients, male were dominant in A2 group(51 males / 9 females), while the ratio of males and females tended to be the same(32 males /31 females)in A3 group, and the proportion of female patients was greater than that of male patients in A4 group(25 males /29 females)( P<0.0001). The main CD lesion was ileocolic type in the EOCD group, accounting for 57.4%( P=0.0077). The incidence rate of ileus type CD was 42.6%( P=0.1942). Among the 163 CD patients who underwent colonoscopy, under Simple Endoscopic Score for CD(SES-CD)evaluation, the proportion of SES-CD severity type in EOCD group was as high as 82.2%, which was higher than that in the A3 group(51.7%, P=0.0187). All CD patients underwent pathological examination.There were 173 cases(95.6%)with focal lymphocytic infiltration, 120 cases(66.3%)with inflammatory infiltration on the crypt epithelial and 82 cases(45.3%)with non-caseous granuloma tissue.In the EOCD group, non-caseous granuloma accounted for 61.1%, which was higher than that in the A3 and A2 groups( P=0.0318). In the EOCD group, the non-complication rate was 1.89%, and the incidence rates of 3, 4 and ≥5 complications were 14.8%, 22.2% and 24.1%, respectively( P<0.0001, P=0.0280, 0.0141 and 0.0013). The sulphasalazine(SASP)alone was the main treatment method, accounting for 42.6%( P=0.0038), and the surgery accounted for 24.1%( P=0.9598). Conclusions:The incidence and trend of EOCD are basically consistent with those of adult-onset CD and showed an rising volatility.Incidence rate of EOCD is higher in females than in males.EOCD lesions of visual observation under endoscope are serious.The detection rate of non-caseous granulomas and related scars is high under microscope.EOCD patients have many complications, and SASP alone is the main treatment method.
7.Effects of PI3-K/AKT signal transduction pathway in rat neural stem cells

Objective To incubate rat neural stem cells (NSCs) with selection-specific inhibitor ( LY294002) of PI3-K, and explore the effects of PI3-K/AKT signal transduction pathway in NSCs. Methods NSCs derived from E15-16 rats were isolated and cultured. After incubation of NSCs with different concentrations of LY294002 (0 to 40 μmol/L), cell survival ( WTS-8 assay), proliferation ( BrdU immunohistochemical identification), differentiation (β Tubulin-III immunohistochemical identification) and AKP phosphorylated protein (Western blotting) were detected. Results The effects of LY294002 on the survival, proliferation and differentiation of NSCs was concentration-dependent. The survival of the NSCs decreased significantly with a higher concentrations of LY294002 (25, 30, 35, 40 μmol/L) (P < 0. 05). The numbers of BrdU positive cells with LY294002 of 30, 35 and 40 μmol/L were significantly smaller than that of the control (LY294002 0 μmol/L) (P < 0.05). The numbers of β Tubulin-III positive cells with LY294002 of 35 and 40 μmol/L were significantly smaller than that of the control (P < 0. 05). The expression of AKT was blocked by LY294002 in a concentration-dependent manner. Conclusion PI3-K/AKT signal transduction pathway plays a vital role in the survival, proliferation and differentiation of NSCs.
8.Advances in the research of extravascular lung water after severe burn.

Pulmonary edema frequently occurs after severe burn. It not only impairs pulmonary function directly, but also can induce or exacerbate other pulmonary complications such as lung infection, acute lung injury ( ALI), and ARDS. Extravascular lung water (EVLW) is closely related to the pulmonary edema. Dynamical monitor of EVLW has been used to predict and quantify the degree of pulmonary edema clinically. This review focuses on the recent progresses at home and abroad on the formation mechanism, monitoring approach, and prevention and treatment of EVLW after severe burn injury.

9.Study onMedication Rules of Jiaotong XinshenTreating Principle Based on Data Mining

Objective To analyze the application rules of Jiaotong Xinshentreating principle in modern clinic based on TCM inheritance auxiliary platform.MethodsJiaotong Xinshen treating principle related prescriptions in CNKI and Wanfang databases were analyzed.“Jiaotong Xinshen”and “Xinshen Bujiao” were set as key words to retrieve relevant prescriptions in the databases. TCM inheritance auxiliary platform (V2.5) software was used to establish database. Data mining methods, such as frequency statistics and association rules were used to analyze new prescriptions aboutJiaotong Xinshen treating principle guided medication rules.ResultsTotally186 prescriptions were collected and analyzed. Medicine frequency and association rules among medicines in the prescriptions were confirmed. 12 medical combinations and 6 new prescriptions were mined.ConclusionThe data management platform and data mining methods can be applied in discovering the prescription patterns and medications rules ofJiaotong Xinshen treating principle in clinical application.

Objective] To summarize the clinical key points of acupoint sticking therapy in the application of treating disease before its onset. [Method] From the treatment timing, treatment principle, acupoint selection to expound the academic perspective and clinical experience of our department in the application of acupoint sticking therapy by consulting the ancient literature and summarizing clinical practice. [Result] Through the correct selection of treatment timing for acupoint sticking therapy, which is mainly used in summer, secondly used in winter, and focusing on tonifying Yang-qi, and point selection by pattern identification, paying attention to the accumulation of curative effect, this therapy has good curative effect in the treating disease before its onset.[Conclusion] Nowadays, there are more and more sub-health people. Since acupoint sticking therapy has advantages of“easy, convenient, cheap, uesful”and safe, it is a good therapy to apply for treating disease before its onset.
