2.Epithelial-mesenchymal transition and the airway remodeling of chronic obstructive pulmonary disease
Journal of Medical Postgraduates 2015;(9):1004-1008
Chronic obstructive pulmonary disease ( COPD) is a group of chronic airway inflammatory disease characterized by sustained airflow limitation and pulmonary function damage.Inflammation and tissue repair repeatedly result in airway remodeling. Now the mechanisms of airway remodeling remain unclear.More and more researches indicated that epithelial mesenchymal transition ( EMT) is crucial in the airway remodeling of COPD.EMT could be induced by various cell factors and signal channels in the airway epithelium, thus the airway remodeling happened. This review systematically introduces EMT and focuses on the relationship of EMT and airway remodeling of COPD and the probable mechanisms.

Chinese Journal of Tissue Engineering Research 2013;(40):7132-7137
BACKGROUND:Scientists began to use stem cells to treat Alzheimer’s disease since human beings achieved embryonic stem cells successful y in 1998.
OBJECTIVE:To review the research progress in stem cells treatment for Alzheimer’s disease.
METHODS:An online search of CNKI and PubMed databases was performed for articles published from 1998 to 2012 (CNKI) using key words of“Alzheimer’s disease, stem cells, neural stem cells, mesenchymal stem cells, induced pluripotent stem cells”in Chinese and English, respectively. The articles were included which related to Alzheimer’s disease, neural stem cells, mesenchymal stem cells and induced pluripotent stem cells, and for articles in the same field, those published recently or in authorized journals were selected. A total of 85 articles were searched. According to inclusion and exclusion criteria, 29 were included.
RESULTS AND CONCLUSION:The possible mechanisms of Alzheimer’s disease include differentiation of neural-like cells, secretion of nutrition factor, improvement of endogenous repair, stimulation of vascular regeneration and action on inflammatory cells. This review highlighted research status of neural stem cells, mesenchymal stem cells and induced pluripotent stem cells in the treatment of Alzheimer’s disease. There were advantages and disadvantages for al of them. The low immunogenicity and oncogenicity of neural stem cells are advantages, but limited source reduces their application. Mesenchymal stem cells also have low immunogenicity and oncogenicity, but their differentiation potential is not good as embryonic stem cells. Induced pluripotent stem cells also have many unsolved problems, but they have been regarded as promising stem cells.

Objective To investigate the long-term programming effects on pubertal reproductive function by immunological challenge in early life. Methods Female Sprague-Dawley rats were administered by endotoxin (lipopolysaccharide, LPS) at a dosage of 50 μg/kg and saline intraperitoneally on postnatal day 3 and 5. Body weight was measured weekly. Puberty onset ( vaginal opening) and oestrous cyclicity were monitored from postnatal day 30. At the age of 6 weeks, bilateral ovariectomy was performed. The histological and morphological change of the ovaries (the thickness of the theca interna and the number of different kinds of follicles) were observed and the immunoreactivity of the ovarian sympathetic nerve markers (low affinity receptor of nerve growth factor, p75NGFR) was evaluated by immune staining. Results Immunological challenge (exposed to LPS) in early life delayed vaginal opening significantly [LPS-treated (40.6 ±0.7) days versus controls ( 38. 6 ± 0. 5 ) days, P < 0. 05], decreased the percentage of normal oestrous cyclicity ( LPS-treated 26. 1% versus controls 66. 8% , P< 0. 05 ) , decreased the total number of different types of follicles (primordial follicles: LPS-treated 610 ±47 versus controls 1181 ±57, P < 0. 05; primary follicles: LPS-treated 624 ±41 versus controls 960 ± 30, P < 0. 05 ; preantral follicles: LPS-treated 183 ± 16 versus controls 260 ± 14, P < 0. 05; antral follicles: LPS-treated 32 ± 4 versus controls 79 ± 7, P < 0. 05) and increased the thickness of the theca interna [LPS-treated ( 15. 8 ±0. 4) μm versus controls (11.4 ±0. 3) μm, P < 0. 05]. The immunostaining of p75NGFR was obviously enhanced in the LPS-treated ovaries when compared with that of controls ( P < 0. 05 ) . Conclusions Immunological stress during early critical developmental windows could have long dysfunctional effects on the pubertal reproductive function. It delayed puberty onset, reduced the percentage of the normal oestrous cycles, decreased follicles reserve and increased the thickness of the theca interna which might involve the up-regulation of the local ovarian sympathetic nerve activity.

Journal of Biomedical Engineering 2015;32(2):455-459
This paper presents a surgical optical navigation system with non-invasive, real-time, and positioning characteristics for open surgical procedure. The design was based on the principle of near-infrared fluorescence molecular imaging. The in vivo fluorescence excitation technology, multi-channel spectral camera technology and image fusion software technology were used. Visible and near-infrared light ring LED excitation source, multi-channel band pass filters, spectral camera 2 CCD optical sensor technology and computer systems were integrated, and, as a result, a new surgical optical navigation system was successfully developed. When the near-infrared fluorescence was injected, the system could display anatomical images of the tissue surface and near-infrared fluorescent functional images of surgical field simultaneously. The system can identify the lymphatic vessels, lymph node, tumor edge which doctor cannot find out with naked eye intra-operatively. Our research will guide effectively the surgeon to remove the tumor tissue to improve significantly the success rate of surgery. The technologies have obtained a national patent, with patent No. ZI. 2011 1 0292374. 1.

Journal of Medical Postgraduates 2015;(2):221-224
Bronchial asthma is the chronic airway inflammatory disease that characterized by reversible airflow limitation .The presence of airway inflammation could lead to airway remodeling .In recent years , many studies have found that arginase plays an im-portant role in the airway inflammation and airway remodeling of asthma .It not only reduces airway release nitric oxide and increases airway hyperresponsiveness , but also its metabolites such as putrescine , proline, etc can induce cell proliferation and collagen synthe-sis, leading to airway remodeling .In this paper , the arginase and its role in the pathogenesis of asthma are discussed .

Chinese Journal of Tissue Engineering Research 2015;(25):4002-4006
BACKGROUND:Biliary stent is widely used in different biliary tract surgeries, and there are some defects in the use of biliary stent. OBJECTIVE:To explore the biodegradability and compatibility of polylactide/glycolide biliary stent. METHODS:The polylactide/glycolide biliary stent was prepared, and immersed in the bile. At 1, 2, 3, 4 and 5 weeks after soaking, eight experimental stents were dried and placed under the scanning electron microscope. In the sterile condition, the stent was implanted under the rat skin. At 1, 2, 3, 4, and 5 weeks after implantation, two rats were sacrificed. The stent was taken out. The appearance of the stent and muscle structure of experimental animals was observed. The gross appearance of stent was observed at different time by scanning electron microscopy. The rate of degradation was calculated. The muscle structure of experimental animal and the stent were understood. RESULTS AND CONCLUSION:At 1 week, stent shape remained almost intact, but the texture became soft;surface roughness and cracks gradual y appeared;stent wal col apse was visible. The stent was completely degraded at 5 weeks. Before degradation, the stent presented clear three-dimensional reticular structure observed by scanning electron microscopy. With the time extended, the stent surface and section corrosion became worsened. By gel permeation chromatography detection, at 1 week of implantation, the molecular mass of the stent declined rapidly, and then gradual y declined. At 2 weeks, the relative molecular mass decreased to 15 000. At 4 weeks, stent mass loss was approximately 40%. Al the experimental animals were survived, no poisoning, al ergy or heat source response. The operative wound was healed wel , and no infection occurred. By histological observation, at 5 weeks, the stent was completely degraded, with a degradation rate of 100%. Surrounding muscles recovered to a normal state. These results indicate that the polylactide/glycolide biliary stent has good degradation in vitro and biocompatibility.
