Tachyarrhythmia is a common type of arrhythmia in children.Continuous of frequent tachy cardia can bring serious harm to children's physical and mental health.With the help of electrophysiological study,electrophysiological mechanisms of various arrhythmias have been stated constantly,while the medical technology develops rapidly.The treatment strategy in pediatric arrhythinia has undergone major reforms,and radiofrequency catheter ablation has come into being.Through the thermal efficiency of the radiofrequency current,radiofrequency catheter ablation can damage the arrhythmia foci or block the atrioventricular reentrant pathway,so achieve the purpose of cure.The clinical application of radiofrequency catheter ablation can avoid the inconvenience of antiarrhymic drugs.Comprehensive assessments of the impacts and possible prognosis should be achieved by clinicians.Strict indications are very important.Currently the ablation has matured in atrioventricular re entrant tachycardia,atrioventricular nodal reentrant tachycardia,ventricular tachycardia,atrial tachycardia and atrial flutter,with high successful rate and low recurrence rate.Complications are closely related to the experience of the surgeon.

Objective To observe the histopathological features of early skin lesions and serum inflammatory mediators changes in the diabetes mellitus(DM) rats.Methods A total of 50 Wistar rats at 8 to 10 weeks of age were divided into normal control group(C group,n=25) and diabetic group(DC group,n=25).The rats of DC group were given a high-glucose and high-fat diet for 2 weeks to induce insulin resistance and then an intraperitoneal injection of 35 mg/kg streptozotocin.All 25 rats were identified to be diabetic models with their fasting blood glucose over ≥7.0 mmol/L.Four rats of C group or DC group were killed to take the skin tissues in the 4th,8th and 12th weekend.In the 16th week,the heart blood samples of the left 13 rats of C group and 10 rats of DC group were collected to measure TNF-?,IL-6 and C-RP levels,and the skin samples were also taken for pathological observation and full-thickness and dermal thickness measurement.Results In the 16th week,the TNF-?,IL-6 and C-RP levels of DC group were significantly higher than the normal group(P

Objective To study the protective effects of rosiglitazone against early skin changes in experimental diabetic rats. Methods Diabetic models were established in male Wistar rats aged 8 to 10 weeks by using streptozotocin (STZ). Then, 39 experimental diabetic rats were equally divided into insulin-treated diabetic group (DI group), rosiglitazone-treated diabetic group (DR group), diabetic control group (DC group),and 13 normal rats served as the control (C group). The rats were given subcutaneous insulin (1 - 2 U) twice daily in DI group, intragastric rosiglitazone (5 mg/kg) once daily in DR group, and intragastric sterile water in DC and C groups. Sixteen weeks later, heart blood samples were collected from all the rats for the measurement of tumor necrosis factor (TNF)-α, interleukin (IL)-6, C reactive protein (CRP), superoxide dismutase (SOD) and P substance (SP) levels, then the rats were killed and tissue samples were obtained from the medial area of the dorsal skin and subjected to pathological observation, measurements of skin as well as dermal thickness, and immunohistochemical examinations for the expression of advanced glycation end products(AGEs) and peroxisome proliferator activated receptor-γ (PPAR-γ). Results The levels of IL-6, TNF-α and CRP in the DC group were significantly higher than those in the C group (135.05 ± 43.39 ng/L vs. 99.92 ±32.36 ng/L, 1.45 ± 0.67 μg/L vs. 0.86 ± 0.60 pg/L, 3.51 ± 0.62 mg/L vs. 2.54 ± 1.31 mg/L, all P < 0.05),while no significant difference was found between C group and DI group or DR group (all P> 0.05). Decreased levels of SOD and SP were noted in the DC group (70.71 ± 37.52 U/ml, 22.22 ± 7.93 ng/L), compared withthe C group (137.76 ± 27.6 U/mL, 29.57 ± 3.74 ng/L, both P< 0.01), DI group (149.96 ± 13.25 U/mL, P<0.01; 29.79 ± 5.21 ng/L, P< 0.05) and DR group (128.50 ± 38.27 U/mL, P< 0.01; 33.35 ± 15.0 ng/L, P<0.05 ). Micrometer measurements indicated that the skin thickness was significantly lower in the DC group than in the C group, DI group and DR group (0.77 ± 0.18 mm vs. 1.59 ± 0.26 mm, 1.47 ± 0.50 mm and 1.22 ±0.47 mm, P < 0.01, 0.01, 0.05 respectively). Histological observation found a mild disarrangement of epidermal cells, shrinkage, swelling and degeneration of dermal collagen as well as progressive atrophy or disappearance of subcutaneous fat in the DC group. No obvious histological changes appeared in the DI or DR group. Immunohistochemistry indicated that AGEs and PPAR-γ proteins, which were stained into brown granules, were located in the vascular basement membrane, stromal cells, etc, of skin. The DC group showed the highest expression of AGEs but lowest expression of PPAR-γ. Conclusions There is an accumulation of AGEs and elevated expressions of inflammatory factors in the skin of experimental diabetic rats, and rosigLItazone shows a protective effect against the early skin changes.

[ ABSTRACT] Diabetes is characterized by an absolute or relative deficiency inβ-cell mass, which cannot be re-versed with existing therapeutic strategies.The restoration of the endogenous islet β-cells can stabilize the level of blood glucose.The isletβ-cells can be obtained from the directional differentiation of stem cells, but the process is complex and has the risk of teratomas generation.Cell direct reprogramming, one terminal differentiated cell can transdifferentiate into another kind of terminal differentiated cell, which is other than directional differentiation from stem cells.Direct reprogram-ming gives rise to the generation of isletβ-cells from one terminal differentiated cell, may be preferable for diabetes therapy because of its unique advantage.

Human herpesvirus-8 (HHV-8) also referred to as Kaposi’s sarcoma-associated herpes-virus (KSHV),is a novel tumor causing virus,which is thought to be the etiologic agent of KS,and also is associated with primary effusion lymphoma(PEL) and multicentric castleman’s disease(MCD) HHV-8 encodes a number of viral proteins, including the proteins associated with HHV-8 latent infection and lytic infection,and the proteins unique to HHV-8,which are critical players in the development of KS and HHV-8-associated diseases. The proteins encoded by HHV-8 and association between HHV-8 and KS were reviewed.

Objective To investigate the efficacy of oxaliplatin plus S-1 versus the XELOX(oxaliplatin + capecitabine) program in the adjuvant chemotherapy in the patients with postoperative gastric cancer .Methods A total of 81 cases with postoperative gas-tric cancer were randomly divided into the treatment group and the control group .The treatment group (41 cases) received the chemotherapy of oxaliplatin +S-1 ,while control group was given the chemotherapy of XEOLX program .Results The 1-year and 2-year recurrence-free survival (RFS) of the treatment group and the control group were 51 .3% vs .61 .5% ,25 .6% vs .20 .5% ,there were no statistically significant differences between the two groups (P=0 .361 ,P=0 .591) .The 1-year and 2-year overall survival period of the treatment group and the control group were 64 .1% vs .69 .2% ,30 .8% vs .25 .6% ,respectively ,there were also no sta-tistically significant differences between two groups(P=0 .631 ,P=0 .615) .The mainly adverse reactions of the two groups were myelosuppression ,gastrointestinal reactions ,hand-foot syndrome ,mucositis ,peripheral nerve toxicity ,hepatic and renal function damage .The occurrence rate of hand-foot syndrome in the control group was significantly higher than that in the treatment group (P=0 .001) ,the adverse reactions were improved after symptomatic treatment .Conclusion Oxaliplatin plus S-1 and oxaliplatin plus capecitabine are therapeutic equivalence and well tolerated in the patients with postoperative gastric cancer .

The role of insulin pump in the treatment of diabetes is becoming more and more significant. Based on the systematic literature review, collation and analysis, this article introduces the management of insulin pump outside the hospital from the development trend of insulin pump, application and common problems. In order to enhance the therapeutic satisfaction of patients and improve their quality of life, our aims are to put forward the strategy of standardized management and provide a reference for establishing and improving the standardized management system of insulin pump therapy outside hospital.

AIM: To compare the therapeutic effects of transplantation of human umbilical cord mesenchymal stem cells (hUCMSC) through different ways on diabetic mice.METHODS: hUCMSCs were labeled with enhanced green fluorescent protein (EGFP) and luciferase (Luc) reporter gene, and then the cells were transplanted into the diabetic mice through pancreas or tail vein to monitor the migration of the hUCMSCs in vivo.The pathological changes of pancreas tissue sections were determined by HE staining.Weight and blood glucose of the mice were measured dynamically.To compare the therapeutic effects, serum insulin levels were analyzed and glucose tolerance test were also performed.RESULTS: In vivo bioluminescence imaging results showed that the hUCMSCs transplanted into pancreatic capsule was mainly located in the pancreas while the hUCMSCs transplanted through vein tail injection was mainly located in the lung.HE staining illustrated that islet cells presented distinctive boundary and no infiltration of inflammatory cells in pancreatic capsule transplantation group was observed, but a little inflammatory cell infiltration and fibrosis formation in tail vein injection group were seen.A significant decrease in blood glucose level and a significant increase in serum insulin level in pancreas transplantation group were showed as compared with vein tail injection group.CONCLUSION: Transplantation of hUCMSCs through different approaches demonstrates different effects.The transplantation of hUCMSCs into pancreatic capsule is more effective on hyperglycemia reversion, insulin secretion and improvement of beta-cell function than that through tail vein.

BACKGROUND:To construct tissue engineering cartilage would open up a novel way for the repair of cartilage damage in avoidance of the disadvantages of traditional therapeutic method.OBJECTIVE: To probe the techniques for the isolation of mesenchymal stem cells (MSCs) from bone marrow, as well as the in vitro differentiation into chondrocytic phenotype in a specific culture fluid.DESIGN:A complete randomized experimentSETTING:The Department of Traumatic Orthopedics and Hand Surgery of the First Affiliated Hospital of Guangxi Medical University, and Teaching and Research Faculty of Histology and Embryology of Guangxi Medical University.METHODS: The experiment was carried out at Guangxi Medical University between August 2002 and April 2003. Twenty SD neonatal weaning rats were selected. Bone marrow was aspirated from the bones of rat limbs and was isolated by gradient centrifugation in Percoll, and MSCs could be obtained in combination with adherent screening method, which were then cultured in DMEM-LG with 15% fatal bovine serum (FBS) in the incubator of 37℃ with 5% CO2 for 10-14 days. The passage cells were induced in DMEM-HG with 15% FBS (containing TGF-β1 10 μg/L, 10-7 mol/L dexamethasone, 50 mg/L VitC).MAIN OUTCOME MEASURES :The morphology, growth, as well as proliferation and specific expression of chondrogenic matrix of in vitro cultured MSCs due to specific induction.RESULTS: Totally 20 SD rats were observed and analyzed with no loss SCs grew in visible symmetric colonies, displaying a long-spindle shape,and the morphological characteristics of marrow-derived MSCs had no obvious changes during passage-culture, but its proliferation time was found from a shuttle fibroblastic appearance to polygonal shape, displaying posiHC staining of type Ⅱ collagen of cartilage specific matrix.bronectin adherent screening technique is a convenient, effective and practical method to separate and collect MSCs from rat bone marrows in chondrogenic phenotype when induced by a specific medium and can secrete cartilage specific matrix, and they can be the optimal seed cells for cartilage tissue engineering.
