1.Clinical effect ofHaizhu-Chuanxdingi tablet with MORA-super Bio-Resonance in bronchial asthma and its influence on the levels of serum MMP-9, TIMP-1, TLR2 and ECP in patients

Objective To explore the clinical effect of Haizhu-Chuanxidingtablet with MORA-super bio-resonance in bronchial asthma.Methods A total of 94 patients met the inclusion criteria in our hospital were divided control group and observe group by a random way, 47 in each group. The control group was treated with MORA-super bio-resonance while the treatment group withHaizhu-Chuanxiding tablet on the base of control group. The treatment time was 50 days in the 2 groups. After the treatment, the effects in the 2 groups were compared. Before and after the treatment, the levels of lung function (FVC, PEF, FEV1% and FEV1), inflammation index (hs-CRP, IL-8,TNF-α and IL-4), matrix metalloproteinase-9 (MMP-9), tissue inhibitors of metalloproteinase 1 (TIMP-1), toll like receptor 2 (TLR2) and eosinophil cationic protein (ECP) were detected in the 2 groups.Results After the treatment, the effect rate was 91.5% (43/47) in the observe group, significantly higher than 72.3% (34/47) in the control group (Z=4.201,P=0.037). After the treatment, the levels of FVC (2.45 ± 0.37 Lvs.1.93 ± 0.32 L,t=3.561), PEF(6.16 ± 1.25 L/svs. 5.56 ± 1.10 L/s,t=3.413), FEV1/FVC (68.39% ± 6.44%vs. 62.31% ± 5.97%,t=3.315) and FEV1 (2.61 ± 0.56 Lvs. 2.21 ± 0.47 L,t=3.672) in the observe group significantly higher than the control group (P<0.05). After the treatment, the levels of hs-CRP (8.14 ± 0.42 mg/Lvs. 12.41 ± 0.65 mg/L,t=3.424), IL-8 (113.47 ± 11.13 ng/Lvs. 153.81 ± 19.73 ng/L,t=3.692), TNF-α (0.31 ± 0.04 ng/Lvs. 0.70 ± 0.13 ng/L,t=3.923)], IL-4 (39.43 ± 6.23 ng/Lvs. 47.75 ± 8.23 ng/L, t=3.783) in the observe group significantly lower than the control group (P<0.05). After the treatment, the levels of MMP-9 (84.89 ± 9.19 ng/mlvs. 113.35 ± 11.35 ng/ml,t=3.671), TIMP-1 (91.53 ± 8.78 ng/mlvs. 131.15 ± 12.45 ng/ml,t=3.782), ECP (19.43 ± 1.33 ng/Lvs. 25.75 ± 2.13 ng/L,t=3.561) in the observe group significantly lower than the control group (P<0.05). The level of TLR2 (270.31 ± 7.01 ng/Lvs. 198.70 ± 10.13 ng/L;t=3.871,P=0.038) in the observe group significantly higher than the control group (P<0.05).ConclusionsHaizhu-Chuanxidingtablet with MORA-super bio-resonance in bronchial asthma showed a good effect on patients with bronchial asthma and could promote the inflammation and lung function in patients.
2.Effect of different chemotherapy combined with radiotherapy for the treatment of non-small cell lung cancer complicated with type 2 diabetes

Objective To evaluate the effect and the incidence of adverse reaction of different chemotherapy combined with radiotherapy for the treatment of non-small cell lung cancer (NSCLC) complicated with type 2 diabetes.Methods A total of 123 midterm advanced NSCLC patients complicated with type 2 diabetes were randomly divided into A,B,C three groups through the random number generated by computer,and each group was 41 cases.Three groups of patients were treated with three-dimensional conformal radiotherapy and synchronous chemotherapy.Group A received gemcitabine and cisplatin,group B received paclitaxel and cisplatin,and group C received pemetrexed and cisplatin.The treatment efficiency,time to tumor progression,median survival time,survival rate and incidence of adverse reaction were observed and compared.Results The total effective rates of group A,B and C were 82.93%,78.05% and 80.49%,with no statistically significant difference (x2 =0.365,P =0.357).The times to tumor progression of the three groups were (6.54 ± 1.23) months,(5.96 ± 1.27) months,(6.27 ± 1.08) months,with no statistically significant difference (F=1.235,P =0.314);the median survival times of the three groups were (16.54 ± 1.87) months,(15.36 ± 1.25) months,(16.29 ± 2.08) months respectively,with no statistically significant difference (F =2.226,P =0.245);1-year survival rates of the three groups were 80.49%,82.93%,85.37% respectively,with no statistically significant difference (x2 =1.234,P =0.665);2-year survival rates were 48.78%,51.22%,43.90%,with no statistically significant difference (x2 =2.354,P =0.451).The incidence rates of radioactive pneumonia and bone marrow suppression in group C were 24.39% and 24.39%,those were significantlylower than31.71%,53.66% of group B (x2 =18.687 P=0.017;x2 =25.336,P=0.011) and 41.46%,31.71% of group A (x2 =17.543,P=0.019;x2 =24.358,P =0.013).Conclusion The clinical efficacy of the three groups are similar,but the incidence rate of adverse reaction in pemetrexed and cisplatin plus radiotherapy group is the lowest.Therefore,pemetrexed plus cisplatin is the safe and effective treatment for the previously untreated patients with NSCLC.

5.Methods in Analyzing Abdominal Fat of Obese Children and Adolescents

Objectives To assess the clinical value of ultrasonography (US) and bioelectrical impedance analysis (BIA) in analyzing abdominal fat contents of obese children and adolescents through comparison with MRI. A correlation with other obese related metabolic parameters was conducted. Methods Ninety 7-17-y-old obese children and adolescents (60 boys and 30 girls with mean age of 9.6 ± 2.9 y and mean BMI of 24.5 ± 4.5 kg/m2) were recruited. Metabolic parameters were measured, and insulin resistance was estimated according to homeostasis model assess-ment (HOMA-IR). On the same day abdomen subcutaneous fat thickness (SFTUS) was measured by US. Body fat mass (FMBIA) and abdominal visceral fat area (VFABIA) were analyzed by bioelectrical impedance analysis (BIA). After obtaining informed consent, abdominal MRI was performed in 20 subjects. Each section of umbilicus level was analyzed by image threshold value segmentation using SigmaScan Pro 5 and abdominal subcutaneous fat area (SFAMRI) and visceral fat area (VFAMRI) were calculated. Results (1) A strong positive association was found between SFTUS and SFAMRI (P< 0.05), VFABIA and VFAMRI (P < 0.01) respectively. (2) FMBIA and SFAMRI, VFAMRI, SFTUS also showed significant correlations (P < 0.05). (3) VFAMRI showed extremely significant positive correlations with TG, Insulin,C-peptide and HOMA-IR (P < 0.01 ) ; SFAMRI was also correlated positively with them (P < 0.05). (4) SFTUS was correlated positively with UA (uric acid), Insulin, 2HIns (insulin measured at 2 hours after meal), C-peptide,2HC-peptide (C-peptide measured at 2 hours after meal) and HOMA-IR (P < 0.01). (5) VFABIA was correlated significant positively with UA, insulin, TG, 2HIns and HOMA-IR. FMBIA showed positive correlation with UA, Insulin,2HIns, C-peptide, 2HC-peptide and HOMA-IR. Conclusions abdominal subcutaneous and visceral fat of obese children and adolescents evaluated by US and BIA are correlated well with those assessed by MRI, and also correlated well with TG, insulin, C-peptide and other metabolic biochemical parameters. Our data support the value of using cost effective, simple and convenient methods such as BIA and US to evaluate the obese and related metabolic risk of children and adolescents in clinical practice.
6.Experimental study of the oriented differentiation of bone marrow derived mesenchymal stem cells into chondrogenic phenotype in a specific culture fluid

BACKGROUND:To construct tissue engineering cartilage would open up a novel way for the repair of cartilage damage in avoidance of the disadvantages of traditional therapeutic method.OBJECTIVE: To probe the techniques for the isolation of mesenchymal stem cells (MSCs) from bone marrow, as well as the in vitro differentiation into chondrocytic phenotype in a specific culture fluid.DESIGN:A complete randomized experimentSETTING:The Department of Traumatic Orthopedics and Hand Surgery of the First Affiliated Hospital of Guangxi Medical University, and Teaching and Research Faculty of Histology and Embryology of Guangxi Medical University.METHODS: The experiment was carried out at Guangxi Medical University between August 2002 and April 2003. Twenty SD neonatal weaning rats were selected. Bone marrow was aspirated from the bones of rat limbs and was isolated by gradient centrifugation in Percoll, and MSCs could be obtained in combination with adherent screening method, which were then cultured in DMEM-LG with 15% fatal bovine serum (FBS) in the incubator of 37℃ with 5% CO2 for 10-14 days. The passage cells were induced in DMEM-HG with 15% FBS (containing TGF-β1 10 μg/L, 10-7 mol/L dexamethasone, 50 mg/L VitC).MAIN OUTCOME MEASURES :The morphology, growth, as well as proliferation and specific expression of chondrogenic matrix of in vitro cultured MSCs due to specific induction.RESULTS: Totally 20 SD rats were observed and analyzed with no loss SCs grew in visible symmetric colonies, displaying a long-spindle shape,and the morphological characteristics of marrow-derived MSCs had no obvious changes during passage-culture, but its proliferation time was found from a shuttle fibroblastic appearance to polygonal shape, displaying posiHC staining of type Ⅱ collagen of cartilage specific matrix.bronectin adherent screening technique is a convenient, effective and practical method to separate and collect MSCs from rat bone marrows in chondrogenic phenotype when induced by a specific medium and can secrete cartilage specific matrix, and they can be the optimal seed cells for cartilage tissue engineering.
7.In vitro bactericidal assay of linezolid on multidrug-resistant tuberculosis and nontuberculous mycobacteria

Objective To investigate the mycobactericidal efficacy of linezolid on multidrugresistant tuberculosis (MDR-TB) and nontuberculous mycobacteria (NTM) in vitro.Methods Seven hundred and sixty-two Mycobacterium tuberculosis and 20 NTM isolates from Hangzhou were identified by using hsp65 gene sequencing from March to June,2013.Seventy MDR-TB isolates,which were screened by using proportion method,and twenty NTM isolates were tested the MIC to linezolid,and the differences of MICs to linezolid between MDR-TB and NTM werw analyzed in vitro.Results All 70 MDR-TB isolates were inhibited at concentrations of ≤ 1 mg/L by linezolid,MIC50 was 0.5 mg/L and MIC90 1 mg/L.Of all 8 isolates inhibited at concentration of 1 mg/L,there were 6 isolates resistant to isoniazid,rifampin,streptomycin and ethambutol,suggesting this resistance pattern may reduce sensitivity to linezolid.Among the NTM isolates,18 Mycobacterium intracellulare isolates,whose MICs to linezolid at 8-16 mg/L,were non-resistant isolates,while 2 Mycobacterium abscessus isolates displaying MICs > 32 mg/L,were resistant to linezolid.Conclusions Linezolid showed great activity to MDR-TB and there were no linezolid-resistant MDR-TB isolates identified in this study.However,MICs of MDR-TB against linezolid increased with their resistant numbers to first-line agents.This study also showed that different NTM species have varied sensitivity to linezolid.
8.Detection of integration frequency catalyzed by integrase using phenotypic screening method

Objective To establish a system for detecting integration frequency of antibiotic resist-ante integron.Methods We cloned integron and aadA2 gene cassette into different sites of plasmid pACYC 184,and the plasmid was transformed into E.coli BL21(DE3)containing plasmid overexpressing integrase.The positive clone was cultured overnight and then was spread on LB agar plate with or without streptomycin respectively,and with appropriate amount of bacteria.Clones after cultured overnight were counted to detect the integration frequency.Meanwhile we used positive clones in LB agar plate containing streptomycin as templates to carry out PCR.The purified PCR products were sequenced to identify the integration sites.Re-suits The integration frequency of integron capturing aadA2 gene cassette in BL21(DE3) host was 1.1 x 10-3 mainly at attI site.Conclusion This system can be used to detect integration frequency.
9.Inhibitory effect of hydroxysafflor yellow A against PMN activation induced by LPS.

Carthamus tinctorius L. is a traditional Chinese medicine with the effect of promoting blood circulation and removing blood stasis. HSYA (hydroxysafflor yellow A) is the main effective component of Carthamus tinctorius L. In order to study the inhibitory effects of HSYA against PMN (polymorphonuclear) activation induced by LPS (lipopolysaccharide), rabbit PMN adhesion potency which was activated by LPS through colorimetry method was observed. Cellular free calcium concentration was determined by fluorescence spectrophotometry. RT-PCR was applied to study the effect of HSYA on PMN TNF-alpha and IL-6 mRNA expression; The inhibition of HSYA on NF-kappaB activation was monitored with immunofluorescence. The results showed that after treated with HSYA, the increase of adhesion potency (HSYA dose 1.01 x 10(-4) mol x L(-1)), free calcium concentration (HSYA dose 3.1 x 10(-5) mol x L(-1)), TNF-alpha and IL-6 mRNA expression elevation (HSYA dose 5.2 x 10(-1) mol x L(-1)) induced by LPS were inhibited. HSYA can inhibit NF-kappaB p65 subgroup nuclear translocation (HSYA dose 5.2 x 10(-5) mol x L(-1)). It is suggested that HSYA is effective in PMN activation induced by LPS.
10.Application of in situ Y chromosomal hybridization in the detection of mesenchymal stem cells

BACKGROUND:Transplantation of mesenchymal stem cells (MSCs) to multiple sclerosis (MS) is a new developed treatment to be given more and more attention.However,whether the MSCs function by cell replacement after going cross the blood-brain barricr or immune suppression needs further confirmation.OBJECTIVE:To establish a steady and effective method of Y chromosome in situ hybridization (ISH) and to detect the distribution of MSCs in a mouse model with MS.DESIGN,TIME AND SETTING:The observational study was performed at the Laboratory of Molecular Immunology,Institute of Healthy Science from September 2007 to February 2009.MATERIALS:A total of 30 C57BL/6 mice,aged 6-8 weeks and weighing 15-20 g,were selected.Female mice were used to establish experimental autoimmune encephalomyelitis (EAE).METHODS:Y chromosme specific DNA probe labled with digoxine (DIG) was designed and ISH was performed to confirm that the designed probe was Y chromosome specific and biologically sensitive.We transplanted the MSCs from male mice into the female EAE mice,and tracked the MSCs by Y chromosome ISH.MAIN OUTCOME MEASRUES:Distribution of MSCs in mice with EAE was observed under the optical and fluorescence microscope.RESULTS:The probe was confirmed to be Y chromosome specific and biologically sensitive.What's more,a steady and effective method of ISH was established.Some MSCs were detected in the periphery organs-spleen and lymph node of mice with EAE;seldom in spinal cord which indicated that MSCs might play its roles by immune suppression.CONCLUSION:Some MSCs were detected in the periphery organs-spleen and lymph node of mice with EAE,but seldom in spinal cord in the central nervous system.
