1.Analysis of relationship between severe radiation pneumonitis and dose-volume histogram parameters for non-small cell lung cancer treated with three-dimensional conformal radiation therapy

Objective To explore the relationship between severe(≥grade 3 ) radiation pneumonitis (RP) and dose-volume histogram (DVH) parameters for non-small cell lung cancer (NSCLC) treated with three-dimensional conformal radiation therapy (3-DCRT). Methods Ninety-four patients with NSCLC treated with 3-DCRT were retrospectively analyzed. Clinical parameters were analyzed. DVH parameters analyzed were V20, V30, V40,mean lung dose (MLD),normal tissue complication probability(NTCP) ,and total dose. Results Age,sex, Karnofsky scored, performance status, forced expiratory volume in 1 second,presence of weight loss, preexisting lung disease, history of thoracic surgery, and history of chemotherapy were not associated with the risk of severe RP(P>0.05). However,in univariate analyses, V20, V30, V40, MID and NTCP were associated with severe RP(P<0.01). In multivariate analysis, MID and V30 were variable associated with severe RP(P<0.01). The severe RP was 0 when MLD < 10 Gy and 21%(8/39) when MLD between 10 Gy and 20 Gy but 35%(7/20) when MLD > 20 Gy,it was 0 when V30 < 25% and 12%(4/33) when V30 between 25% and 35% but 38%(11/29) when V30 >35%. Conclusion MLD and V30 are significant predictive factors for severe RP and they should be limited to ≤20 Gy and ≤ 35% in order to reduce severe RP.
2.The radiological manifestation of hypophosphatasia

ObjectiveTo explore the radiological features of hypophosphatasia.MethodsFive cases of hypophosphatasia were definitely diagnosed,which included 3 males and 2 females aged from 5 months to 23 years.The laboratory assays were analyzed,radiological appearances of bone were determined and differential diagnoses were made.ResultsThe alkaline phosphatase in blood serum of five patients decreased,which were 8,20,13,21,and 18 U/L respectively.Phosphoethanolamine increased in blood serum of the five patients,which were 16.5,13.5,21.6,18.7,and 28.9 μmol/L respectively.Phosphoethanolamine also increased in urine,which were 2350,9120,3520,5280,and 1820 μmol/L respectively.Calcium in blood serum increased,which were 4.2,5.6,4.9,6.1,and 3.5 mmol/L respectively.X-ray images displayed that the density of bone decreased in 5 cases,the metaphyses exhibited widening and cupping in 4 cases,the provisional calcification zone of the metaphysis became thinning or disappearing in 4 cases,long bone bended in 4 cases and pathologically fractured in 1 case.Conclusion Hypophosphatasia can be indicated by its relatively special radiological appearance,and it can be diagnosed and differentiated from rickets,osteomalacia and osteogenesis imperfect by the laboratory examination combined with its clinical presentation.
3.Application of in situ Y chromosomal hybridization in the detection of mesenchymal stem cells

BACKGROUND:Transplantation of mesenchymal stem cells (MSCs) to multiple sclerosis (MS) is a new developed treatment to be given more and more attention.However,whether the MSCs function by cell replacement after going cross the blood-brain barricr or immune suppression needs further confirmation.OBJECTIVE:To establish a steady and effective method of Y chromosome in situ hybridization (ISH) and to detect the distribution of MSCs in a mouse model with MS.DESIGN,TIME AND SETTING:The observational study was performed at the Laboratory of Molecular Immunology,Institute of Healthy Science from September 2007 to February 2009.MATERIALS:A total of 30 C57BL/6 mice,aged 6-8 weeks and weighing 15-20 g,were selected.Female mice were used to establish experimental autoimmune encephalomyelitis (EAE).METHODS:Y chromosme specific DNA probe labled with digoxine (DIG) was designed and ISH was performed to confirm that the designed probe was Y chromosome specific and biologically sensitive.We transplanted the MSCs from male mice into the female EAE mice,and tracked the MSCs by Y chromosome ISH.MAIN OUTCOME MEASRUES:Distribution of MSCs in mice with EAE was observed under the optical and fluorescence microscope.RESULTS:The probe was confirmed to be Y chromosome specific and biologically sensitive.What's more,a steady and effective method of ISH was established.Some MSCs were detected in the periphery organs-spleen and lymph node of mice with EAE;seldom in spinal cord which indicated that MSCs might play its roles by immune suppression.CONCLUSION:Some MSCs were detected in the periphery organs-spleen and lymph node of mice with EAE,but seldom in spinal cord in the central nervous system.

5.Influence of glycine supplement on partial biochemical indicator in exhausive exercise mice

Objective To investigate the influence of glycine supplement on partial biochemical indicator in exhaustive exer‐cise mice .Methods Divided Kunming′s male‐mice into 3 group(10/group):exhaustive swimming group(E) ,exhaustive swimming group with glycine supplement(G) and silent control group (S) .Mice in the G group was treated with 4‐weeked intragastric admin‐istration of glycine ,while mice in other 2 groups received physiological saline .After 4 weeks ,mice in E and G group did the exhaus‐tive swimming exercise .After sampling ,the activity of complement hemolytic activity was measured ,and the serum levels of some biochemical indexes were determined .Results Time of exhaustive swimming in G group was significantly longer than E group(P<0 .01) .After exhaustive swimming ,the ALT ,AST ,ALP ,LDH ,CK ,N ,Fe3+ and CHA in E group were significantly changed com‐pared with S group(P<0 .01);while those indicators in G group were significantly improved than E group (P<0 .01) ,the ALP and LDH level can regain to the level of S group (P>0 .05) .Conclusion Exhaustive swimming mice can have part indicator abnormali‐ty ,but Glycine supplement can effectively improve that condition and enhance the exercise ability to some extent .
6.Effects of dexmedetomidine on expression of aquaporins 1 and 5 during lung ischemia/reperfusion in rats: an in vitro experiment

Objective To evaluate the effects of dexmedetomidine on the expression of aquaporins 1 (AQP1) and AQP5 during lung ischemia/reperfusion (I/R) in rats in an in vitro experiment.Methods SPF healthy male Sprague-Dawley rats,aged 3-4 months,weighing 250-320 g,were used in this study.Forty-five isolated rat lungs in which the model of isolated lung perfusion was successfully established were divided into 3 groups (n=15 each) using a random number table:sham operation group (S group),I/R group and dexmedetomidine group (D group).The isolated rat lungs were continuously perfused for 150 min in group S.After 15 min of perfusion,the isolated rat lungs were subjected to 60 min of ischemia and apnea followed by 75 min of ventilation and reperfusion to establish the model of isolated lung I/R injury in group I/R.In group D,the isolated rat lungs were perfused for 15 min with K-H perfusion fluid containing 10 nmol/L dexmedetomidine,and then subjected to 60 min of ischemia and apnea followed by 75 min of ventilation and reperfusion with K-H perfusion fluid containing 10 nmol/L dexmedetotnidine.The lung compliance,airway resistance,perfusion flow and partial pressure of arterial oxygen (PaO2) were recorded at 10 min of perfusion and 15,45 and 75 min after restoration of perfusion.Lung tissues were obtained at 75 min after restoration of perfusion for determination of wet/dry weight ratio (W/D ratio) and for examination of the pathological changes and changes in the uhrastructure.The expression of AQP1 and AQP5 protein and mRNA in lung tissues was detected by Western blot and real-time polymerase chain reaction,respectively.Results Compared with S group,the airway resistance was significantly increased and lung compliance,perfusion flow and PaO2 were decreased during reperfusion,and W/D ratio was increased in I/R and D groups (P<0.05),the expression of AQP1 and AQP5 protein and mRNA in lung tissues was significantly down-regulated in group I/R,and the expression of AQP 1 and AQP5 protein and mRNA in lung tissues was significantly up-regulated in group D (P<0.05).Compared with I/R group,the airway resistance was significantly decreased and lung compliance,perfusion flow and PaO2 were increased during reperfusion,W/D ratio was decreased,the expression of AQP1 and AQP5 protein and mRNA in lung tissues was up-regulated (P<0.05),and the pathological changes of lung tissues was significantly attenuated in group D.Conclusion The mechanism by which dexmedetomidine reduces I/R injury may be related to up-regulation of the expression of AQP1 and AQP5 in rats in an in vitro experiment.
7.The factors affecting pulse transit time during anesthesia

Objective To investigate the factors affecting pulse transit time during anesthesia. Methods Sixty cases of gastrointestinal surgery under general anesthesia were investigated.The pulse transit time (PTT),BP and HR were measured at the time before and after intravenous anes-thesia induction,and at the time before and after the injection of vasoactive drugs,respectively. Results Compared to at the time before injection,HR,SBP and DBP increased significantly,and PTT decreased significantly (P <0.01 )at the time after injection of atropine and ephedrine.Com-pared to at the time before injection,HR and PTT increased significantly,while SBP and DBP de-creased significantly (P < 0.01 )at the time after injection of nicardipine.While HR and PTT de-creased significantly,and SBP,DBP increased significantly (P <0.01 )with the injection of pheny-lephrine.Obvious negative correlation between SBP and PTT was observed before and after induc-tion;meanwhile,only weak correlation between DBP and PTT was observed,and no obvious correla-tion between HR and PTT was detected.SBP and PTT were well negatively correlated with the injec-tion of atropine,ephedrine,nicardipine or phenylephrine.Conclusion All the PTT changes during anesthesia were found to be negatively correlated with the systolic blood pressure.
8.Experimental study of correlation between the focus-to-flat panel detector distance and entrance surface dosed in digital radiography of lumbar spine

Objective To investigated the correlation between the focus-to-flat panel detector distance and entrance surface dosed in digital radiography of lumbar spine.Methods The spine phantom used for bone mineral density(BMD) calibration was radiographed at different value of FFD (100 ~ 240 cm with the interval 10 cm,totally 15 times) using 70 kV and 90 kV with the automatic exposure control (AEC) mode without anti-scatter grid.The parameters measured by QUART dido 2100K type X-ray dose meter.So record the value parameters of the tube current (mAs) and entrance surface dose (ESD).The relation curve were counted between FFD,ESD,mAs and Exposure index (EI) use excle.Pearson correlation analysis and related coefficient difference analysis were performed using the SPSS 19.0 software (P <0.05 for the statistically significant level).The FFD and ESD values of regression equation were obtained with regression analysis.Results (1) When the tube voltage was 70 kV:FFD and ESD were negative correlation (r =-0.922,P < 0.001),FFD and mAs were positive correlation (r =0.991,P <0.001),FFD and EI negative correlation (r =-0.938,P < 0.001).(2) When the tube voltage was 90 kV:FFD and ESDwere negative correlation (r=-0.955,P <0.001),FFD and mAs were positive correlation (r=0.994,P <0.001),FFD and EI showed no significant correlation (r=-0.523,P>0.05).(3) Different position correlation coefficient analysis of the difference was not statistically significant difference.(4) The regression analysis results:The regression equation of the lumbar anteriorposterior position FFD and the ESD at 70 kV:y=0.805 x + 319.731 (F=74.175,P <0.001).The regression equation of the lumbar lateral position FFD and the ESD at 90 kV:y =3.152 x + 457.613 (F =133.524,P < 0.001).Conclusions The FFD and ESD in the lumbar spine X-ray radiography were relevant and highly negative correlation.It can effectively reduce the radiographic ESD of lumbar spine with the increasing of the FFD.
9.Using cell membrane chromatography and HPLC-TOF/MS method for in vivo study of active components from roots of Aconitum carmichaeli

An offline two-dimensional system combining a rat cardiac muscle cell membrane chromatography time-of-flight mass spectrometry (CMC-TOF/MS) with a high performance liquid chromatography time-of-flight mass spectrometry (HPLC-TOF/MS) was established for investigating the parent components and metabolites in rat urine samples after administration of the roots of Aconitum carmichaeli.On the basis of the analysis of the first dimension,retention components of the mine sample were collected into 30 fractions (one fraction per minute).Then offline analysis of the second dimension was carried out.34 compounds including 24 parent alkaloids and 10 potential metabolites were identified from the dosed rat urine,and then binding affinities of different compounds on cell membranes were compared and influences of some functional groups on activity were estimated with the semi-quantification and curve fitting method.As a result,binding affinities decreased along with the process of deacylation,debenzoylation and demethylation,which may be related to the alleviation of toxicity in the procedure of herb processing or metabolism.Moreover,some minor components in rat urine (Songorine,14-benzoylneoline,Deoxyaconitine,etc.) exerted relatively strong affinity on cell membranes are worth exploring.The results delivered by the system suggest that the CMC can be applied to in vivo study.
10.Studies of the mechanism and inhibitory effect of aescine on SGC-7901 cells in vitro

Aim To study the mechanism and inhibi-tory effect of Aescine on SGC-7901 cells in vitro.Methods MTT method was used to detect SGC-7901 proliferation.Flow cytometer method was used to determine cell cycles.AnnexinⅤ-FITC apoptosis derection kit was used to detect apoptosis of cells.Western blot was employed to analyze the expression of Bcl-2.Results MTT result showed that Aescine could significantly inhibit the growth of SGC-7901.AnnexinⅤ-FITC apoptosis derection kit and Western blot results suggested that Aescine could induce the apoptosis of SGC-7901.Conclusions Aescine can inhibit the growth of SGC-7901 and induce the apoptosis of cells in vitro.
