1.Effect of Roux-en-Y gastric bypass on type 2 diabetes mellitus and its initial mechanisms

Objective: to observe the effect of Roux-en-Y Gastric Bypass (RYGB)on STZ induced type 2 diabetic rat model, and to clarify its initial mechanism. Methods: 42 type 2 diabeticrats were randomly allocated into operation group(group O 12 rats); sham group(group S 10 rats); diet control group(group F 10 rats); control group(group C 10 rats). Fasting glucose, Insulin Growth Factor-1 ( IGF1), fasting plasma insulin levels, weight and food intake were measured before and 1st , 2nd, 3 rd ,4th, 8th and 16 th week after operation. Results: 16 weeks after operation in the Group O, fasting glucose fell to (8.8?4.9)mmol/L, IGF1 increase to (148.6?7.3)ng/L ,fasting plasma increase to (74.7?9.2)mIU/L and weight decline to (212.6?15.1)g ,there was statistical significance compared with the levels of preoperation (P
2.An Experimental Study of the Effect of Skin Temperature Detecting on Blocking Flap

Objective To further study the blood supply mechanism of blocking flap and establish the standard of transferring the flap.Methods 7 groups of flap were made on 5 mini-Banna pigs.For each group,we had research flap and control flap on the right side and left side of each pig.After flap blocking operation,we detected the skin temperature of the flap on the 3rd,7th and 10th day.Results There was no statistic difference between each group before operation,but there was statistic difference after operation.Conclusion Temperature detecting may help choose the right time(32.5~33.5℃) for transferring the blocking flap.
3.Mechanism of Roux-en-Y gastric bypass and gastric banding on type 2 diabetes

Objective To clarify mechanism of Roux-en-Y gastric bypass (RYGB) and gastric banding on diabetes induced by STZ injection. Methods 40 rats with STZ induced diabetes were randomly allocated into Roux-en-Y gastric bypass (RYGB) group (group RYGB, n=10), gastric banding group (group GB, n=10 ), diet control group (group F, n=10), control group (group C,n= 10). The fasting blood glucose, the fasting insulin IGF-1, the fasting Plasma leptin, the fasting plasma insulin level, the weight and the food-intake, the operation time, the death rate were measured and recored before and after operation on 1st , 2nd, 3 rd ,4th, 8th and 16 th week postoperatively. Results The fasting blood glucose of the group of gastric banding(GB) descended to (12.6±3.7) mmol/L, the fasting plasma insulin rose to (58.7±9.2) mIU/L, the fasting plasma leptin descended to (14.6±3.3) pg/ml, the weight was (212.6±15.1) g.There were significant differences between before and after operation on 16 th week(P<0.01). The fasting blood glucose of the group of Roux-en-Y (RYBG) descended to 8.8±4.9 mmol/L in the sixteenth week, the fasting insulin IGF-1 rose to (148.6±7.3) ng/L, the fasting plasma insulin rose to (14.1±3.5) pg/ml, the fasting plasma leptin descended to 14.1±3.5 pg/ml, the weight was (200±15.1) g. There were significant differences between before and after operation 16 th week (P<0.01). There were significant differences of the fasting plasma insulin and the the fasting plasma leptin between group F and group C during the 3 rd to 4th week after operation (P<0.05). Compared the weight of the group F and the group C on the third week of operation, there were significant differences (P<0.05), and there were no significant differences in other time. The fasting blood glucose of the group F and the group C had no sig-nificant differences between before and after operation.(P<0.05). Conclusions The fasting blood glucose and the fasting insulin level of the group F improve more than of the group GB at the same time. The plasma insulin and the plasma leptin of the two groups all work in glucose control. The diet control and the modification of the plasma insulin and the plasma leptin all play a major role in the gastric banding mechanism, and the IGF-1 may work in the descending the blood glucose after the operation of Roux-en-Y. In the operation time and die rate, the group of F surpass the group of GB.

5.A preliminary study of anti-aging and wound healing of recombination cytoglobin.

In this paper, the preliminary study on antioxidant, enhancement of antioxidant enzymes activity, reducing the content of oxygen free radicals, delaying skin aging of the recombination cytoglobin (rCygb) purified by our lab were investigated through human keratinocyte cell line (HaCAT) H2O2 oxidative stress model, mouse skin aging model caused by continuous subcutaneous injection D-gal, rat acute liver injury model induced by CCl4 and rat skin wound healing model. The results showed that rCygb improved the activities of total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px) and catalase (CAT), reduced the activities of lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) as well as decreased the content of malondialdehyde (MDA). Skin biopsy showed that rCygb promoted angiogenesis, increased expression of collagen and improved the anti-inflammatory ability. All results displayed that rCygb improved the oxygen free radical scavenging ability, delayed skin aging and promoted wound healing.
6.Investigation on the Learning Results and the Medical Students' Attitude towards Study

Objective:In order to understand the learning results and the medical students' attitude to study better,we took effective measures to better improve the quality of education.Methods Questionnaire were taken to inquire students' professional thinking and the attitude of study and the SPSS software was used to analyze the learning results.Results:Students had the problems of"four insufficiency and a pressure"during the learning procedure,and learning results of medical students showed big differences in different majors,different semesters and different genders.Conclusion:Our college should take necessary measures in enhancing education to regulate the professional thinking and the medical students' attitude of study.Our college teachers should get the total message and handle the students' learning results and give every student individual education.Meanwhile,we should pay more attention to letting students find the ways of obtaining necessary information and effective learning methods.
7.Clinical comparative investigation using intensity-modulated radiotherapy combined with concurrent chemotherapy for the local advanced nasopharyngeal carcinoma

Objective To research the early effects and side-effects of the local advanced nasopharyngeal carcinoma patients using intensity-modulated radiotherapy(IMRT)combined with concurrent chemotherapy.Methods From January 2005 to January 2007,60 patients with nasopharyngeal carcinoma of stage m-IV b were received IMRT combined with concurrent chemotherapy in our center.Sixty patients were divided into paclitaxel concurrent group(32 patients)and cisplatin concurrent group(28 patients).The prescribing doses of the primary tumor were 68-72 Gy for each group.The patients of paclitaxel concurrent group patients of the cisplatin concurrent group got earlier radiodennatitis and radiation-induced mucositis but also got significantly higher rate of radiodermatitis,radiation-induced mucositis,radiation-induced leucopenia and gastrointestinal toxicity,as well as the loss of weight.No significant difference was found on liver and renal funcfons between two groups.Four patients(12.5%)of the paclitaxel concurrent group were broken-off,which was much better than the cisplatin concurrent group.There was no significant difference on the specific length of break-off time,the 2-year overall survival rate and the 2-year diseaee-free survival rate between two groups.Conclusions IMRT combined with concurrent chemotherapy of paclitaxel liposome for local advanced nasopharyngeal carcinoma results in less side-effects and better tolerance than IMRT combined with concurrent cisplatin chemotherapy.
8.The method of continuous measuring extracellular acetylcholine and choline levels in striatum of freely moving awake rats

AIM To establish the method of continuous measuring extracellular Acetylcholine and Choline levels in[FQ(10*2。46,X-WZ]-striatum of freely moving awake rats. METHODS The microdialysis and high performance liquid chromatography(HPLC)-post column Immobilized enzyme reactor(IMER)-electrochemical detection(ED) were used to continuously measure extracellular acetylcholine and choline levels in striatum of freely moving awake rats. RESULTS Extracellular acetylcholine and choline levels were higher at the beginning of perfusion, then decreased gradually. The ACh concentration reached a stable level after 240 min while the Ch continuously decreased in the whole perfusion period of 450 min. The detection limit for ACh and Ch, at a S/N ratio of three, was found to be 150 fmol per injection. In the range of 0.125~1.0 ?mol?L -1 , the relation between the amounts and the peak amplitude was linear. CONCLUSION HPLC-IMER-ED is a sensitive and stable method to measure the ACh and Ch. Coupled with microdialysis, it can been successfully used for monitoring the extracellular ACh and Ch levels of freely moving awake rats. -
9.Effect of atorvastatin therapy before percutaneous coronary intervention on periprocedural myocardial injury

ObjectiveTo explore the effect of statin in reduction of my ocardial injury in patients undergoing percutaneous coronary intervention(PCI).MethodsTotal of 185 patients undergoing PCI were included,140 patients were received atorvastatin before PCI,while 145 were not given statins at the time of PCI.Creatine kinase-MB and troponin Ⅰ were measured at baseline and at 8 and 24 hours after the procedure.ResultsPost procedurul peak levels of troponin Ⅰ and creatine kinase-MB were(0.12 ±0.26) μg/L and(2.61 ± 3.07) μg/L in the observation group,which were significantly lower than(0.51 ± 1.14) μg/L and( 6.85 ± 14.38 ) μg/L in the control group( t =3.951,3.414,P < 0.05).ConclusionPretreatment with atorvastatin for 7 days could significantly reduces procedural myocardial injury in elective PCI.
10.Comparative study on immunomodulatory effects of mesenchymal stem cells derived from different sources

Objective To study the difference in immunomodulatory effects among non-Hodgkin lymphoma (NHL)-, Hodgkin lymphoma (HL)- and normal adult bone marrow-derived mesenthymal stem cells (MSCs). Methods MSCs were obtained from HL, NHL and normal adult bone marrow and cultured in expanded medium. Immunophenotype was investigated by FACS. The levels of cytokines were evaluated by enzyme linked immunosorbent assay (ELISA). T-cell suppression ability was evaluated by Transwell. Moreover, the immunoregulatory ability of HL-, NHL- and normal adult bone marrow-derived MSCs was detected by mixed lymphocyte culture assay. Results HL-, NHLand normal adult bone marrow-derived MSCs were similar in morphology and immunophenotype, and they did not express HLA-DR and co-stirnulatory molecules (CD40, CD80 and CD86). All HL-,NHL- and normal adult bone marrow-derived MSCs could significantly suppress T lymphocytes'proliferation in a dose-dependent manner and such an effect could be reversed by anti-TGF-β1 antibody.MSCs differentiated into various mesenchymal lineages did not alter their immunosuppressive effects on T-cell proliferation. Conclusion The similar immunomodulatory effects were found among HL-,NHL- and normal adult bone marrow-derived MSCs, which was not changed by differentiation.
