1.Research on Molecular Biological Characteristics of Proto-oncogene pim-2.

The purpose of this paper is to present the research on the molecular biological characteristics of proto-oncogene pim-2 and to analyze the related mechanism. Proto-oncogene pim-2 was studied and analyzed by the bioinformatics method and technology. With an online server, the chromosomal localization of pim-2 gene was analyzed, and the exon, open reading frame, CpG island and miRNAs complementary fragments and the like were predicted. With bioinformatics software, the physicochemical property of transcription protein of proto-oncogene pim-2 and various modification sites of protein sequence, such as ubiquitination and glycosylation, were predicted, the antigenic index was calculated, and the spatial structural was modeled. The research findings showed that the proto-oncogene pim-2 comprised six exons, the CDS (coding sequence) transcribed a section of peptide chain including 311 amino acids, a gene promoter has a CpG island, and the 3'UTR region contains an miRNA gene. The molecular weight of the Pim-2 protein was 34,188. 47, the isoelectric point was 5.78, the instability index was 45.87, and the extinction coefficient was 279nm. A plurality of covalent modification sites, two ubiquitination sites, four glycosylation sites, an SUMO sumoylation site, a nitrosation site, two palmitoylation sites and sixteen regions with higher antigenic index were distributed in the protein sequence. This research showed that the related regions and modification sites distributed on the sequence of proto-oncogene pim-2 were closely related to the carcinogenic effect thereof.

2.Investigation of Pharmaceutical Care for Pediatric Severe Infection Treatment by Clinical Pharmacists

OBJECTIVE:To investigate the role of clinical pharmacists in pediatric severe infection treatment,and to provide reference for rational drug use in clinic. METHODS:Based on related typical cases,the breakthrough points of clinical pharmacists providing pharmaceutical care in pediatric severe infection treatment plan were introduced. RESULTS:Clinical pharmacists formu-lated multi-drug resistance pathogenic antibacterial plan according to the results of pathogenic microbiological examination,use pharmacokinetics/pharmacodynamics,excluded interference to pediatric anti-infection treatment from ADR,and guideline and eco-nomical factors of patients. CONCLUSIONS:Clinical pharmacists adopt professional knowledge of pharmacy to provide pharmaceu-tical care in pediatric severe infection treatment and play professional assisted role so as to guarantee safe,effective and economical treatment.
3.Diagnosis and treatment for gastrointestinal stromal sarcomas

Objective To explore the clinical behavior,pathological and immunohistochemical characteristics and diagnosis, treatment methods of gastrointestinal stromal sarcomas (GISS). Methods Retrospective analysis was perfomed over 36 GISS patients based on clinical data and pathological data. Results GISS occurred more in the stomach and small intestine, than colon and rectum. Immunohistochemical staining showed CD117, CD34 expression,the positive rates were 91.7% and 86. 1%. Conclusion GISS is a rare malignant tumor, GISS diagnosis depends on the histologic and immunohistochemical test. CD34 and CD117 in the positive diagnosis has its special significance. Surgery is the main treatment.

Fracture healing activity of aqteous extracts of rhizome of Drynaria fortunei (Kunze ) J.Sm.,D.sinica Diels ( "Gusuibu"),and its main constitucnt naringin were examined and coufirmedwith an experimental bone-injury model rat femur inferior. Results showed that they werefefective for healing of experimental rat bone-injury.
5.Non-linear rectification of sensor based on immune genetic algorithm.

A non-linear rectification based on immune genetic algorithm (IGA) is proposed in this paper, for the shortcoming of the non-linearity rectification. This algorithm introducing the biologic immune mechanism into the genetic algorithm can restrain the disadvantages that the poor precision, slow convergence speed and early maturity of the genetic algorithm. Computer simulations indicated that the algorithm not only keeps population diversity, but also increases the convergent speed, precision and the stability greatly. The results have shown the correctness and effectiveness of the method.

7.The biological characterization of Pseudomonas aeruginosa phage PaP3

Objective To identify some basic biological properties,optimal multiplicity of infection(MOI),one-step growth kinetics,serum neutralization rate constant of Pseudomonas aeruginosa phage PaP3 were determined, respectively. Methods The host bacteria were infected with PaP3 at six different MOI(0.0001,0.001,0.01,0.1,1 and 10,then lysated adequately, the phage titer of supernatant was determined.For one-step growth experiments,the host bacteria were infected with PaP3 at MOI=10, following centrifugation after 15 min adsorption,the pellet containing (partially) infected cells was resuspended in 3 ml of pre-warmed LB broth and incubated with 160rpm at 37℃.Samples(50?l) were taken at 10 min-intervals and immediately tittered by the double-layer agar plate method. A rabbit was immunized with purified PaP3 to prepare anti-PaP3 serum, reaction constants between antiserum and PaP1, PaP2, PaP3 were determined using cross neutralization test. Results 0.001 MOI-infected host bacteria gave the highest phage offsprings. One-step growth curve was determined according to the results of one-step growth experiments.Reaction constant was determined using neutralization test. Conclusion The optimal MOI of PaP3 was determined to be 0.001. One-step growth curve for PaP3 shows that the latent period is about 20 min, the rise period is 60 min, and the average burst size is about 31pfu/cell. Rate constant of reaction between antiserum and PaP3 is 262.

9.THE USE OF SMALL-SIZED THORACOTOMY IN MINIMALLY INVASIVE THORACIC SURGERY

To assess the advantage, disadvantage and feasibility of small thoracotomy incision for intrathoracic operations. In 124 patients minimal thoracotomy incisions (MT) were made for infrathoracic surgery from July 1999 to June 2001. The operative approaches consisted of posterolateral incisions in 82 cases, axillary incisions in 17 cases, and anterolateral incision in 25 cases. The operations included 46 lobectomies, 8 pneumonectomies, 27 wedge resections, 13 mediastinum tumor resections, 13 bullae ligations with pleurodeses, 2 esophagectomy and esophagogastric anastomosis, 1 total gastrectomy and esophagojejunostomy, 9 explorations, and 5 other operations. There was no mortality or serious complication in this group. The patients had a faster recovery with less postoperative pain and better cosmetic result. Minimal thoracotomy incisions are safe and feasible approaches for intrathoracic surgery and worth recommending.
10.DETECTION OF LYMPHATIC METASTASIS OF LUNG CANCER BY INTRAOPERATIVE ULTRASONOGRAPHY

Objective To evaluate the value of intraoperative ultrasonography (IUS) in detecting the lymph nodes in mediastinum and hilum during lung cancer surgery. Methods Lymph nodes were detected in every part of the mediastinum and hilum by IUS in 33 cases with lung cancer during surgery. The number, size and location of lymph nodes were recorded, and the metastasis to lymph nodes was predicted by the sonographic features and geometric measurement respectively. Mediastinum was explored and lymph nodes resection was performed in each case, and each resected lymph node was sent for pathological examination. Results The sensitivity of IUS in detecting lymph nodes in mediastinum and hilum was 83.6% (163/195) and the positive predictive value was 97.0%(165/168). The result of detecting lymph nodes with IUS was influenced by their location and size. The sensitivity, specificity and accuracy of IUS in diagnosing lymphatic metastasis with respect to geometric measurement were 70.3%, 71.7%, 71.2%, respectively, and were 85.7%, 75.0%, 83.3%, respectively, with respect to sonographic features. Conclusions IUS was an effective method for lung cancer surgery in detecting mediastinum lymph nodes.
