1.Establishment of rat model of epilepsy induced by lithium-repeated low-dose pilocarpine

Objective:To estabhish rat model of epilepsy induced by the Lithium-repeated low-dose pilocarpine.Methods:A totol of 173SD male rats were divided into the normal control group,seizure-induced group and Chloral Hydrate group randomly.Lithium chloride was injected at a dose of 3 mEq/kg intraperitoneally(ip) 19h prior to the administration of pilocarpine.Rats were injected with pilocarpine,10mg/kg ip,every 30min,and this injection was repeated for 2~4 times.We then determined the rat activity,EEG,the percentage of rats exhibiting status epilepticus(SE),the mortality rate,the proportion of rats developing spontaneous seizures,the latency for the first spontaneous motor seizure,and the frequency of the spontaneous recurrent seizures.Results:The lithium-repeated low-dose pilocarpine resulted in SE after 1~4 injections;the average period between seizure and injection was 16min and the success rate was 73%.The mean dose of pilocarpine needed to induce SE was 25.9mg/kg.When SE was interrupted after 90min,mortality rate was below 10%,Spontaneous seizure occurred 2.4~5.7 times a day averagely,and average latency to the first spontaneous seizure was approximately 40days.All seizure incidence was over level Ⅲ and most were level Ⅳ/Ⅴ;EEG was similar to that of human temporal epilepsy;seizure of rats in control group was level 0.Conclusion:Lithium-repeated low-dose pilocarpine induced rats model of epilepsy is characterized by easy building,with high successful rate and lower mortality of the animal.This model has mimic features of human status epilepticus and temporal epilepsy in seizure behavior and electroencephalographic findings.

Objective:To explore the vascular protective effects and mechanism of fasudil in rats after experimental subarachnoid hemorrhage.Methods:Forty-eight rats were divided into sham-operated control group,subarachnoid hemorrhage group and treatment group.Model induced by autologous blood injection into cisterna magna,and rats were killed on day 1,day 3,day 7 and day 14 after subarachnoid hemorrhage.Morphological change of basilar artery was observed by light microscopy,diameter and wall thickness of basilar artery were measured respectively.eNOS expression in basilar artery was detected by immunohistochemical SP method.Results:Vasospasm of basilar artery initiated on day 1,reached its peak on day 3,and mitigated on day 7 after SAH.eNOS expression decreased at each time point in SAH group.Compare with SAH group,diameter and wall thickness of basilar artery were significantly reduced on day 3 and day 7 in treatment group(P

5.Study on the relationship between mechanical ventilation initiation time and prognosis of in-hospital cardiac arrest patients

Objective To investigate the correlation between mechanical ventilation (MV) initiation time during cardiopulmonary resuscitation (CPR) and the prognosis of patients with in-hospital cardiac arrest (IHCA) in emergency . Methods Retrospective analysis of clinical records of patients with IHCA in emergency from January 2011 to April 2016 treated in Zhangye People's Hospital of Hexi University was performed. Patients with restoration of spontaneous circulation (ROSC) and were on MV with aged over 18 years were divided into early treated group (≤ 10 minutes) and later treated group (> 10 minutes) according to the initiation time of MV. Corneal reflex, pupillary reflex, pain-avoidance responses and motor response 24 hours after CPR, neurological function and cure rate of the two groups were analyzed. Results 210 patients were selected into our study including 130 males and 80 females (mean age: 60.24±13.17 years). There were no significant differences in gender, age, type of heart rate and etiological factor of cardiac arrest (CA) between the MV early stage group (124 cases) and the MV late stage group (86 cases). The restoration of corneal reflex, pupillary reflex, pain-avoidance responses, motor response and achievement ratio of CPR in early group were higher than those of later group (respectively, 59.68% vs. 31.40%, 59.68% vs. 31.40%, 54.84% vs. 24.42%, 54.84% vs. 24.42%, 70.16% vs. 51.16%, all P < 0.01); The D-dimer levels in the early group patients were significantly lower than those in the later group (μg/L: 478.39±57.21 vs. 510.05±62.83, P < 0.01). However, no statistical difference was observed between the two groups with respect to pH (7.24±0.72 vs.7.13±0.67, P > 0.05); The average hospitalized day of the early group was significantly shorter than that of the later group (days: 24.15±3.04 vs. 30.28±4.17, P < 0.01); Besides, the early group showed a higher survival rate at discharge and had more cases with neurologic level of grade 1-2 than those of the later group (Respectively, 41.94% vs. 26.74%, P < 0.05; 44.35% vs. 15.12%, P < 0.01). Conclusion Initiation MV on IHCA patients in the early stage of CPR (≤ 10 minutes) could help improve the hypoxic condition and prognosis of neurological function, and increase the achievement ratio of CPR.
6.Cluster ensemble algorithm based on dual neural gas applied to cancer gene expression profiles.

The microarray technology used in biological and medical research provides a new idea for the diagnosis and treatment of cancer. To find different types of cancer and to classify the cancer samples accurately, we propose a new cluster ensemble framework Dual Neural Gas Cluster Ensemble (DNGCE), which is based on neural gas algorithm, to discover the underlying structure of noisy cancer gene expression profiles. This framework DNGCE applies the neural gas algorithm to perform clustering not only on the sample dimension, but also on the attribute dimension. It also adopts the normalized cut algorithm to partition off the consensus matrix constructed from multiple clustering solutions. We obtained the final accurate results. Experiments on cancer gene expression profiles illustrated that the proposed approach could achieve good performance, as it outperforms the single clustering algorithms and most of the existing approaches in the process of clustering gene expression profiles.

7.Clinical research of treating Alzheimer's disease by Qingkailing injection and donepezil hydrochloric

Objective To observe the clinical treatment effects of Qingkailing injection on Alzheimer's disease.Methods 84 patients with AD were randomly divided into a control group and a treatment group (n=42 cases,per group). The patients in both groups orally took 5 mg of donepezil hydrochloride every night. The Qingkailing injection of 60 ml was administrated by intravenous drip in addition to oral uptake of donepezil hydrochloride to the treatment group. The total treatment period of both groups was 20 weeks. The MMSE, ADL score, HS-CRP, IL-6, MDA and ox-LDL content and the clinical treatment results and differences in the two groups before and after the treatment were observed.Results After treatment, the serum HS-CRP (4.18 ± 0.38 mg/Lvs. 5.78 ± 0.53 mg/L,t=15.900), IL-6 (34.28 ± 4.33?g/L vs.44.75 ± 4.99?g/L,t=10.270), MDA (6.04 ± 0.98?mol/L vs.7.43 ± 0.69?mol/L,t=7.516), ox-LDL (62.87 ± 7.65?g/L vs.102.56 ± 9.81?g/L, t=26.677) content of the treatment group were significantly decreased than the control group (P<0.01). The total effective rate between the treatment group and the control group had significance difference (83.3%vs. 61.9%;χ2=4.850,P=0.028). The MMSE score in both groups after the treatment were higher than those in the same group(t value were 8.036, 7.475, respectively,P<0.01), ADL scores in both groups after treatment were lower than those before treatment(t value were 7.818, 5.960, respectively,P<0.01), but no significance difference between the two groups(t value were 1.633, 1.710,respectively,P value were 0.106, 0.091, respectively).Conclusion The treatment effect of Qingkailing injection on Alzheimer's disease was sound.
8.Preliminary study of adjuvant therapy with autologous platelet-rich gel for refractory wound

Objective To investigate the effects of autologous platelet-rich plasma in the treatment of refractory wound.Methods Fifteen cases with refactory wound ( 1 1 males and 4 females) involving six cases with diabetic ulcer,five with post-surgical ulcer wounds and four with pressure sore were enrolled into the study.The autologous platelet-rich gel was applied to the wounds after debridement and dressing change was done every week.The results of wound bacterial culture were regularly detected and the growth of granulation was observed.Skin graft or flap graft surgery was performed to close the wound when the wound condition was permitted.Results Of all,three patients had spontaneous healing after the change of dressing,four got healing after flap graft and eight attained healing after skin graft.There was no recurrence or sinus after treatment.Conclusion Autologous platelet-rich gel accelerates wound healing,improves wound condition,and raises success rate of surgical closure of the wound.
9.Treatment of infections in patients with primary immunodeficiency disease

Primary immunodeficiency disease (PID) is a heterogeneous group of congenital disorders of the immune system.The impaired immune response is caused by defect of immune active cells and moleculars,which results in increased susceptibility to pathogens.Recurrent infection is the predominant presenting complaint for patients with PID.Early diagnosis,immunoglobulin replacement therapy,antimicrobial treatment,haematopoietic stem cell transplantation and gene therapy,genetic conselling and prenatal diagnosis will help to prevent and control infections in patients with PID.In addition,we should strengthen the education of paediatricians,improve awareness about PID and collaboration across different centers to advance the diagnosis and management of PID in China and save the life of patients at the maximum limit.
10.Research development of triple-negative breast cancer chemotherapy

Triple-negative breast cancer ( TNBC ) is a subtype of breast cancer,and it is characterized by an aggressive clinical course with a poor prognosis.Treatment for TNBC has attracted much attention in recent years,however,there is no standard treatment for TNBC in clinical setting.The pCR rates of neoadjuvant chemotherapy in TNBC ranges from 12% to 48% in the current published data,and it is higher than that in other types of breast cancer,however,the fluctuating range of the TNBC ’s pCR is large in literature.Although the administration of adjuvant chemotherapy for early TNBC is controversial,the regimen without anthracyclines is reported to be suitable for early TNBC patients.Standard cytotoxic agents including taxanes and anthracyclines are still the main choices for TNBC salvage treatment,and the combination with gemcitabine or capecitabine may improve the overall survival.Poly (ADP-ribose) polymerase (PARP) is a new molecular target for TNBC in ongoing studies.Further research on the target-inhibitors such as BSI-201and Olaparib will provide more effective choices to clinical treatment.
