Objective Our aim was to observe the effects of prenatal nicotine exposure on development of cerebral cortex and hippocampus in the offsprings of pregnant mice and expressions of NCAM proteins. Methods We established prenatally nicotine exposed(PNE) animal models.We investigated the offspring's structural changes of cerebral cortex and hippocampus with HE staining.Using immunohistochemistry and Western blotting we observed the changes of expressions of NCAM proteins in the neonatal mice. Results Cerebral cortex and hippocampus as well as hippocampal cell layers of the PNE offsprings were thinner than that of the control group.The expressions of PSA-NCAM in cerebral cortex and hippocampus of the PNE offsprings were higher than the control group while the expressions of NCAM-180,140 and 120 weaker.Conclusion Prenatally nicotine exposure can prevent normal morphogenesis of cerebral cortex and hippocampus of mice and can change the expressions of NCAM proteins in cerebral cortex and hippocampus of neonatal mice.
2.An Experimental Study on the Biocompability of Porcelain Compounded With Nanostructured Material

Objective: Our aim was to evaluate the biocompability of porcelain compounded with nanostructured material. Methods:The proliferation of the NIH3T3 cells were investigated using a tetrazolium-based colorimetric assay(MTT assay).Maceration extract of four tested materials were added into culture plate .The samples were tested with ultraviolet spectrophotometer in 490 wavelength at 1,3,5,7 days after the addition of extract. Morphology of these cells was also observed with phase-contrast microscope. Results: There was no obvious difference among three kinds of prosthetic materials in the optical absorbance and the cells growing in the extract showed normal morphology. Conclusion: The toxicity graduation of the porcelain compounded with nanostructured material is 0. This material has a good biocompability and can be used clinically.

4.Study of obstruction of proximal deferent duct by high-frequency color Doppler ultrasound

Objective To study the diagnostic value of high-frequency color Doppler ultrasound (CDU) on the obstruction of proximal deferent duct. Methods One hundred and twenty-three patients with scrotal diseases (aged 23-67 years old) were examined by CDU with 5-10 MHz transducer. Among the cases, infertility was in 88, vasoligation in 8, and epididymal nodes in 27. The sonographic appearances of the obstruction of the deferent duct were analysed, compared with that of normal deferent duct. Results Of the 123 cases, the obstruction of the deferent duct was in 72, included absence of deferent duct in 6, epididymal malformation in 9, epididymitis in 37, epididymal neoplasms in 5, and dilatation of testic rete and testic efferent duct in 3. Twenty-two patients underwent surgery, confirmed the sonographic appearances of the obstruction of proximal deferent duct. Conclusions Obstruction of proximal deferent duct could be assessed by high-frequency CDU. It is helpful for distinguishing the causes of the obstruction of proximal deferent duct and choosing their therapy scheme.

Objective To investigate the effects of local application of NGF combined with Insulin on wound healing with Ⅱ degree deep scald and the expression of HIF-1α and VEGF in diabetic rats. Methods 60 male Wistar rats were used in the study by intraperitoneal injection of streptozotocin (STZ). Deep partial thickness scalding was engendered on the back of the rats after one month. Then these rats were random divided into group B (scalding of the rats with diabetes, n = 15), C (insulin control, n = 15), D ( NGF control, n = 15) and E ( NGF and insulin supplementation, n = 15). And group A ( nomal control,n = 15) was created by normal rats of partial thickness scalding. The wound area, wound healing rate, and grey of HIF-1α and VEGF immunohistochemistry staining were determined on the 3rd, 7th, 15th, and 21st day post scalding days (PSDs). Results Compared to those in groups A, B, C and D, the wound healing rate in group E increased significantly since the 7th PSD [(25.33 ± 2. 32 ) %, P < 0.05]. The wound healing rate in group C and D increased significantly compared with group B [( 16. 68 ± 1.95 ) %, ( 18.29±1. 70)%, P <0.05]. Compared to group A, the group D increased significantly since the 15th PSD [(54. 84 ±3.60)%, P <0. 05]. The expression of HIF-1α and VEGF in group E began to increase slower than other groups after the 7th day ( P <0. 05 ). Conclusion Local application of NGF combined with Insulin could be beneficial to the regeneration of capillary vessel in the burn wounds of the rats with diabetes,as well as to accelerate wound healing by redress anoxia and decrease the expression of HIF-1α and VEGF.
7.Clustering analysis of anti-nuclear antibodies in systemic lupus erythematosus

Objective To examine clusters of anti-nuclear antibodies (ANA) and their associations with clinical features in patients with systemic lupus erythematosus (SLE).Methods It was a retrospective study.113 SLE patients were reviewed from March 2010 to May 2012 in Department of Rheumatology,Jinhua Central Hospital.ANA and specific autoantibodies to 15 kinds of nuclear antigens were tested by indirect immunofluorescence assay (IIF) and line immunoassay (LIA) respectively.Hierarchical clustering method was performed to analyze specific clusters of ANA profiles in SLE.Chi-square tests were used to investigate relationship between antibody clusters and clinical features of SLE.Results The positive rate of LIA for ANA was 97.3%,consistent with IIF method,and the total accordance rate of the both methods was 98.2%.Thirteen kinds of antigen-specific antibodies were detected in SLE patients by LIA.Clustering analysis for these antibodies showed three specific clusters in SLE,Nuc/His/dsDNA cluster (C1),low-Ro/low-La cluster (C2),and Ro/Sm/RNP cluster (C3),accounting for 36.3%,24.8%,38.9% of the total cases respectively.There were significant difference of AST levels among three clusters [(32.62 ± 21.92)U/L,(25.56 ± 16.63) U/L,(50.41 ± 60.86) U/L respectively for C1,C2 and C3].High incidences of chronic cutaneous lupus,abnormal renal indicators and inflammatory synovitis were found in all three clusters.Besides,there were significant differences among three clusters for the incidences of chronic cutaneous lupus (39.0%,39.3%,63.6% respectively for C1,C2,C3) and leukopenia/lymphopenia (56.1%,25.0%,56.8% respectively for C1,C2,C3) (P < 0.05).Patients in Ro/Sm/RNP cluster showed higher incidences of lupus nephritis (43.2%/26.8% or 39.3%); patients in low-Ro/low-La cluster showed low risk of hypertension (7.1%/19.5% or 22.7%) ; patients in Nuc/His/dsDNA cluster showed high incidences of thrombocytopenia (41.5%/21.4% or 25.0%) and high risk of lung or upper respiratory tract infection (46.3%/28.6% or 29.5%),but low incidence of neurologic symptoms (0%/ 3.6% % or 11.4%).Conclusion Three characterized ANA clusters are identified in SLE patients in this pilot study.Different clusters are associated with certain clinical features and complications ofSLE.However,the correlations found in this study need to be investigated further in larger populations.
8.Clinical observation on treatment for postoperative metastasis of gastric cancer by Jianpi Huoxue Jiedu prescription

OBJECTIVE: To study the effect of Jianpi Huoxue Jiedu (JPHXJD) prescription on prevention of the postoperative metastasis of gastric cancer and to observe the changes of immunity and whole blood viscosity after treatment. METHODS: JPHXJD prescription and chemotherapy were administered to the treated group for 12 successive months, while only chemotherapy was given to the control group. RESULTS: The effective rate of anti-metastasis in the treated group was obviously better than that in the control group. In the aspects of enhancing Karnofsky scores, reducing whole blood viscosity and improving immunity after treatment, the treated group was better than the control group (P<0.05). CONCLUSION: JPHXJD prescription combined with chemotherapy shows apparent effect on anti-metastasis. It can improve the quality of life of the patients, lower the whole blood viscosity and improve the immunity function.
9.Role of protein tyrosine phosphatase SHP-2 in group I metabotropic glutamate receptors-induced allodynia

Aim To investigate whether the pain modi-fication by group I metabotropic glutamate receptors (mGluRs)required the involvement of Src homology-2 domain-containing phosphatase-2 (SHP-2 ).Methods Co-immunoprecipitation was performed to examine the possible interaction between SHP-2 and group I mGluRs in spinal cord dorsal horn of mice.By measur-ing the paw withdrawal thresholds,the effects of SHP-2 inhibitor NSC-87877 or its catalytically inactive SHP-2 (C459S ) mutant on allodynia induced by group I mGluRs agonist DHPG (50 nmol)were observed.Re-sults Anti-mGluR5 antibody was able to co-immuno-precipitate SHP-2 from spinal dorsal horn of mice, while no SHP-2 was precipitated by anti-mGluR1 anti-body.Inactivation of SHP-2 by NSC-87877 (6 nmol) or SHP-2 (C459S ) effectively attenuated allodynia caused by DHPG.Conclusion SHP-2 can physically interact with mGluR5.The activation of SHP-2 may be necessary for group I mGluRs to process the nocicep-tive information.
10.Analysis of clinical use of oral drugs in elderly geriatric inpatients

Objective To analyze the inappropriate use of oral drugs in elderly patients admitted in internal medicine, and to guide clinical rational drugs use in elderly patients. Methods One hundred inpatients′ data over 65 years were collected in the department of geriatrics. The potentially inappropriate use of oral drug were analyzed according to the Beers criteria published in 2012 and drugs instructions. Results In 100 patients, (5.81 ± 2.78) kinds of oral drugs had been taken per inpatient. One patient was involved out-of-label usage. There were potentially inappropriate uses of oral drugs in 21 patients. Inappropriate use of oral drugs in three patients were not related to diagnosis or disease, and in 4 patients were related to morbid state, 14 patients used drugs that need to be used with caution in older adults. Conclusions There are off-label uses and potentially inappropriate medications in clinical practice, so clinical doctors should examine and assess the uses of drugs. Clinical doctors should strengthen the understanding and application of the comprehensive geriatric assessment.
