1.Investigation of T lymphocyte subsets in fluid of the patients by flow cytometry

Objective To evaluate the method of flow cytometric analysis of T lymphocyte subsets in bodyfluid of the pa-tients. Methods In ovder to get T lymphocytes by 3-colour staining method utilizing SSC/CD3Percp antibody,47 bronchoalveolar lavage fluid and 50 were tested by flow cytometery. Results 1)The percentage of Th is (67.98±7.41) % ,Ts(27.36±6.41) % and Th/Ts(2.60±0.64)in BALF with lung cancer,and the percentage of Th is (37.61±4.8)% ,Ts(67.97±4.20)% ,and Th/Ts(0. 47±0.10)in BALF with lung cancer respectively, and the Th cells in BALF with lung cancer were lower than those in the benign pulmonary disorder(P<0.05), and the Th/Ts ratio was significantly lower while the Ts cells were significantly higher in BALF of lung cancer(P<0.01), comparied with the benign pulmonary disorder; 2)The percentage of Th is (35.76±7.71) % and Ts(64.31 ±7.37) % and Th/Ts(0. 57 ± 0. 18) in BALF with lung cancer, and the percentage of Th is(49.27 ± 12.88) %, Ts(44.54 ± 9. 56) %,and Th/Ts is (1.13 ± 0.29) in BALF with benign pulmonary disorder. The Th cells in BALF with lung cancer was lower than those in the benign pulmonary disorder(P<0.05) ,the Th/Ts ratio were significantly lower and the Ts cells were significantly higher in BALF of lung cancer(P<0.01) comparied with the benign pulmonary disorder;3)The percentage of This(35.76±7. 71) %,Tsis(64.31 ± 7.37) % and Th/Ts are(0.57 ± 0. 18) in BALF with lung cancer, and the percentage of Th is (49.27 ± 12. 88) %, Ts is (44.54 ± 9.56) %, and Th/Ts is(1.13 ± 0.29) in BALF with benign pulmonary disorder. Respectively, the Th cells in BALF with lung cancer were lower than those in the benign pulmonary disorder(P<0.05), the Th/Ts ratio were significantly low-er and the Ts cells were significantly higher in BALF of lung cancer(P<0.01)comparied with the benign pulmonary disorder. Con-clusion The immunity function in the patients with lung cancer was significantly decreased. Flow cytometric approach is valuable in analyzing T lymphocyte subsets in BALF.

3.Study on the Influence Factors for the Preparation of Reverse Microemulsions and the Optimization of Its Formula

OBJECTIVE:To study the effect of oil phase,emulsifier and drugs on reverse microemulsion.METHODS:Pseudo-ternary phase diagram was adopted to study the effect of Span-80/Tween-80(emulsifier),thymopentin(model drug), long-chain glyceride,medium-chain glyceride,non-glyceride(oil phase).on the preparation of reverse microemulsions.Optimal formula of reverse microemulsions was selected.RESULTS:The largest water-in-oil(W/O)area was obtained in reverse microemulsion which was prepared using medium-chain glyceride as oil phase.Final formula was confirmed and contained distilled water/ Span-80/Tween-80/caprylic/capric triglyceride(2∶3∶6∶9).In addition,thymopentin was incorporated into the aqueous phase.CONCLUSIONS:The preparation of reverse microemulsions will be influenced by emulsifier,constitution of oil phase and drug.The influence factors for the actual preparation of reverse microemulsions should be optimized.
4.Pathogenic risk factors correlation study of developmental dysplasia hip in Shigatse of Tibet

Objective To perform the epidemiological analysis of the developmental dysplasia hip(DDH) in Shigatse of Tibet in order to explore the correlation between the onset of DDH with some risk factors such as altitude and take targeted intervention measures to reduce the disability rate of DDH.Methods A total of 5900 children aged 0 to 14 year-old from different altitudes in Shigatse of Tibet were surveyed randomly from June 2013 to June 2014.SPSS 18.0 statistical software was used to analyze the correlation of the onset of DDH with residence altitude,production and living ways,binding method of the lower limbs,delivery situation and genetic factor.Results One hundred and ninety-one DDH cases were screened out,and the average incidence of DDH in Shigatse of Tibet was about 32.4‰,and unilateral was higher than bilateral,fight side(33 cases)∶left side(68 cases) =1.00∶ 2.06.The result of Pearson correlation analysis of the onset of DDH with residence altitude showed that there was significant correlation (r=0.820,P=0.046).The result of Spearman correlation analysis of the onset of DDH with production and living ways showed that there was significant correlation(r=0.530,P =0.048).The result of x2 test analysis of the onset of DDH with binding method of the double lower limbs and breech delivery showed that there was statistical significance (P=0.0017,0.0082).Conclusions The DDH incidence of children living in high altitude areas in Tibet has certain regional characteristics indicating that it has clear correlation with some environmental risk factors such as altitude,methods of the lower limbs binding,production and living ways and breech delivery,especially the bundled package of lower limbs is the main environmental risk factor of DDH.
5.Investigation of early appendectomy in appendicular abscess

Objective To evaluate the feasibility and safety of early appendectomy in appendicular abscess. Methods Fifty patients with appendicular abscess who received early appendectomy in our hospital from January 2009 to June 2012 were considered as the abscess group,50 cases patients received acute appendicitis surgery were regarded as the control group. The postoperative hospital stay,cost of hospi-talization,operative time,the amount of bleeding in operation and complications were retrospectively analyzed. Results The operative time of abscess group was (68±23) min,and the control group was (49±14) min (P<0. 05). There were significant differences in operating time between two groups. But there was no significant difference in intraoperatve blood soss,length of stay,the average hospitalization charge and surgical complications (P>0. 05). All patients were cured. Conclusion Early treatment of appendiceal abscess is safe and alternate to conventional conservative treatment followed by interval appendectomy. It is not only to avoid the secondary hospitalization and treatment fail-ure,but also save the cost of hospitalization and minimize the medical expenses.
6.Effect of thrombopoietin on chemical hypoxia-induced apoptosis of PC12 cells

[ ABSTRACT] AIM:To study the effect of thrombopoietin ( TPO) on chemical hypoxia-induced apoptosis of the Rattus norvegicus adrenal pheochromocytoma (PC12) cells.METHODS:The cultured PC12 cells were randomly divided into normal control group, cobalt chloride ( CoCl2 ) group, CoCl2 +TPO group and TPO group.The cell viability was mea-sured by MTT assay.The effect of TPO on CoCl2-induced cell apoptosis was analyzed by flow cytometry with Annexin V/PI double staining.The intracellular reactive oxygen species ( ROS) were detected by fluorescence microscopy, and the chan-ges of the mitochondrial membrane potential ( MMT) were determined by flow cytometry and fluorescence microscopy.RE-SULTS:Chemical oxygen agent CoCl2 significantly inhibited the growth of PC12 cells (P<0.01).The apoptotic rate in CoCl2 group was obviously higher than that in control group ( P<0.05) , while the apoptotic rate in CoCl2 +TPO group was obviously lower than that in CoCl2 group (P<0.05).TPO decreased the production of ROS, and inhibited the decrease in MMP induced by CoCl2(P<0.01).CONCLUSION: TPO has a protective effect against CoCl2-induced apoptosis of PC12 cells by decreasing the production of ROS and inhibiting the decrease in MMP.
7.The comprehensive prevention strategy for occurrence and risk factors of haploidentical hematopoi-etic stem cell transplantation associated oral mucositisunder

Objective To investigate the comprehensive prevention strategy in the prevention of oral mucositis (OM) during allogeneic hematopoietic stem cell transplantation (HSCT). Methods A total of 721 patients who received HSCT from January 2010 to December 2011 at the Institute of Hematology, Peking University were enrolled. All the patients received the comprehensive prevention strategy of OM. Results A total of 315 patients (43.7 percents) suffered from OM during HSCT, the median time from HSCT to OM occurrence was 5 days. There were 173 (54.9 percents), 91(28.9 percents), 37(11.7 percents), and 14(4.4 percents) patients suffered from gradeⅠ,Ⅱ,Ⅲ, and IV OM, respectively. Tongue OM was most common. A total of 111 patients suffered from multiple OM (≥2 locations). Multiplicity showed that duration of agranulocytosis≥15 days was the only independent risk factor of OM in HSCT [ hazard ratio (HR) value 1.5, P<0.05 ]. Neutrophil engraftment time≥12 days was significantly associated with prolonged OM (≥8 days) (HR=2.1, P<0.05). Patient age ≥20 was independent risk factor for occurrance of grade Ⅲ-Ⅳ OM (HR=2.7, P<0.05). Conclusions Comprehensive prevention strategy can overcome the risk factors of OM and prevent the occurrence of OM during HSCT.
8.Mechanism Study on Nano-Realgar Intervention of TWEAK-NF-κB Signal Pathway through Downregulation of Chemokines in Kidney Expression among MRL/lpr Mice

This study was aimed to explore the molecular mechanism of nano-realgar in treatment of systematic lupus erythematosus (SLE) lupus nephritis (LN). Intragastric administration of equal volume of high-, middle-, low-dose nano-realgar suspension, and normal saline (NS) were given to MRL/lpr mice, respectively. The observations were made on levels of ANA, ds-DNA antibody, IgG and IgM in blood serum, as well as TWEAK, NF-κB, MCP-1 mRNA and protein expression levels in renal tissues. The results showed that compared with the NS group, levels of ANA, ds-DNA antibody, IgG and IgM were obviously reduced (P < 0.05); the levels of TWEAK, NF-κB and MCP-1 mRNA were obviously reduced (P < 0.05); the protein expression levels of TWEAK, NF-κB and MCP-1 mRNA were obviously reduced (P < 0.05) in the high-, middle-, low-dose nano-realgar group. It was concluded that nano-realgar intervened the TWEAK-NF-κB signal pathway through downregulating MCP-1 expression among MRL/lpr mice, in order to reduce the levels of ANA, ds-DNA antibody, IgG and IgM for relieving autoimmune damages in the treatment of SLE (LN).
9.Determination of Quinolones in Soils Using Solid Phase Extraction and High Performance Liquid Chromatography-Fluorimetric Detection

An analytical method for the simultaneous determination of four quinolones in soil was developed. Soil samples were extracted by a mixture of 50% magnesium nitrate and 10% ammonia(96∶ 4, V/V)with an ultrasonic-assisted extraction, then purified and concentrated by HLB cartridge, and eluted with acetonitrile and 0.067 mol/L phosphoric acid(5∶ 1, V/V). Using acetonitrile and 0.067 mol/L phosphoric acid(pH=2.5) as the mobile phase, these analytes were quantificated by HPLC(fluorimetric detector) at excitation and emission wavelength of 280 nm and 450 nm respectively. The detective limits for four quinolones in soil were from 0.58 to 0.03 g/kg. The recoveries were 60.4% to 99.3% for soil samples. The method was successfully applied to determine the quinolones in soil samples from vegetable fields. Four quinolone compounds were detected to a different extent with total amounts of quinolones ranged from 27.84 to 129.26 μg/kg.
10.Clinical significance of serum high sensitive C-reactive protein in patients undergone prostate biopsy

Objective To assess the clinical significance of serum high sensitive C-reactive protein (hs-CRP) in patients undergone prostate biopsy.Methods A total of 273 consecutive patients were enrolled,aged 44-95 years (mean,69 years).All the patients underwent prostate biopsy.The pathological findings showed 96 cases with prostate cancer (PCa) and 177 cases with benign prostate hyperplasia (BPH).The difference of hs-CRP level between patients with PCa and those with BPH was analyzed.The positive prostate biopsy rate was compared between the patients with high hs-CRP level and those with normal hs-CRP level.Logistic regression was used to evaluate the effect of factors such as hs-CRP,tPSA,PSA density,prostate volume and age on prostate biopsy.Results The medians (interquartile range) of hs-CRP were 3.22 mg/L (1.22-9.84 mg/L) in patients with PCa and 1.24 mg/L (0.55-2.76 mg/L) in those with BPH,respectively,with significant difference(P<0.05).The positive prostate biopsy rate in patients with high hs-CRP (> 3 mg/L)was 55% (51/92),higher than that in those with normal hs-CRP (≤ 3 mg/L).The odds ratio of hs-CRP was larger than that of all other factors analyzed including tPSA,prostate volume and age according to the Logistic regression analysis.Conclusions Elevated serum hs-CRP level is associated with increased positive prostate biopsy.Serum hs-CRP acts as an independent factor increasing the positive prostate biopsy rate in patients undergone prostate biopsy.
