China Pharmacy 2016;27(12):1618-1620
OBJECTIVE:To observe the effect of metformin on related indicators of obese patients with type 2 diabetes. METHODS:136 obese patients with type 2 diabetes were randomly divided into control group and observation group. All patients were given conventional examination, conventional education about diabetes, diet control and stressed physical exercise. Based on it,control group was given 80 mg Metformin tablet(Ⅱ),once a day,and monitored blood glucose levels, the dosage was adjusted based on fasting blood glucose levels to ensure blood glucose in 3.9-6.1 mmol/L;observation group was given 0.5 g Metformin en-teric-coated tablet, twice a day. The 2 groups were compared after 12 weeks treatment. The glycated hemoglobin (HbA1c),body mass index (BMI),resistin,insulin-like growth factor-1(IGF-1),inflammatory cytokines [interleukin-1β(IL-1β),interleukin-6 (IL-6),tumor necrosis factor-α(TNF-α),high-sensitivity C-reactive protein(hs-CRP)] levels in 2 groups before and after tveat-ment were observed, and the incidence of adverse reactions in 2 groups was recorded. RESULTS:After treatment,resistin,IGF-1, IL-6 and hs-CRP levels in 2 groups were significantly lower than before, and observation group was lower than control group;the differences were statistically significant(P<0.05);BMI,IL-1β and TNF-α levels in obser vation group were significantly higher than before and control group, the differences were statistically significant(P<0.05);HbA1c level in 2 groups was significiantly lower than before, the difference was statistically significant(P<0.05),but there was no significant difference between 2 groups(P>0.05);BMI level in observation group was lower than before and control group, and there was no significant difference in the inci-dence of adverse reactions between 2 groups(P>0.05). CONCLUSIONS:Metformin can effectively improve insulin resistance in obese patients with type 2 diabetes and inhibit the inflammatory reaction.

Objective To discuss the effect of polysaccharides from Tegillarca granosa as an adjuvant on pcDNA3.1?Sj26GST vaccine against Schistosoma japonicum infection. Methods Sixty SPF BALB/c mice were divided into four groups randomly(15 mice each group),including a control group,a polysaccharides group,a vaccine group, and a vaccine plus poly?saccharides group. In the 0,2th and 4th week of the experiment, the mice in the above four groups were immunized for 3 times with 100μl PBS,100μg polysaccharides from Tegillarca granosa,100μg Sj26GST vaccine, and 100μg Sj26GST vaccine plus equivalent polysaccharides, respectively. Two weeks after the last immunization, all the mice were infected with 40 ± 1 Schistosoma japonicum cercariae through the skin of the abdomen. After the infection for 6 weeks, all the mice were sacrificed, and their serums, livers and the adult worm of Schistosoma japonicum in them were collected, the specific sera IgG antibodies were detected by ELISA,and the worm reduction rates and egg reduction rates in the liver were calculated. Results Six weeks after the infection, the IgG antibody levels of the mice in the vaccine group and the vaccine plus polysaccharides group were 18.26 ± 0.42 mg/ml and 20.21 ± 0.89 mg/ml respectively, the difference between them were statistically significant, and both of the IgG levels of the above groups were significantly higher than those in the control(both P<0.05). The worm reduction rate and egg reduction rate in the vaccine group were 28.60%and 35.84%,respectively,and the rates in the vaccine plus polysac?charides group were 38.04% and 49.74%,respectively,the differences between the worm reduction rates and egg reduction rates were both statistically significant(both P<0.05). Meanwhile, the two rates in the two above groups were all significantly higher than those in the control group(all P<0.01). Conclusion Polysaccharides from Tegillarca granosa using as an adju?vant can increase the protection effect of pcDNA3.1?Sj26GST vaccine against Schistosoma japonicum infection.

Objective To investigate the effect of different subtypes of fractionated doses on multidrug resistance in ovarian cancer cells. Methods The human ovarian cancer cell lines SKOV3 and its drug-resistant subtype SKVCR were divided into four groups i.e., sham-irradiated, single dose (10 Gy),fractionated dose (2 Gy × 5 ) and multi-fractionated dose (1 Gy × 2 × 5).Cell sensitivity to vincristine(VCR),etoposide ( VP-16),pirarubicin (THP) and cisplatin (DDP) was measured by MTT assay.Western blot was applied to detect the expression of P-gp after irradiation. Results The doubling time of SKVCR was about 1.8-fold of that of SKOV3 cells.P-gp was expressed in SKVCR but not in SKOV3.IC50 values of SKVCR were higher than those of SKOV3.To SKOV3 cells, single dose irradiation decreased cell sensitivity to THP and DDP and fractionated irradiation decreased cell sensitivity to VCR,THP and VP-16.Multi-fractionated irradiation decreased cell sensitivity to VP-16.In SKVCR cells, all these irradiation treatments increased cell sensitivity to VCR and VP-16 but not to DDP.In addition, single and fractionated irradiation decreased P-gp expression in SKVCR cells. Conclusions Single, fractionated and multi-fractionated radiation induced chemotherapy resistance in SKOV3 cells, while reversed drug resistance to VCR and VP-16 in SKVCR cells.

Objective To investigate the effects of repeated computed tomography (CT) scan on hematological and immunological index in mice, and to explore the related mechanism. Methods A total of 60 BALB/c mice were randomly divided into six groups as control,once,twice,4,6 and 8 times of CT scans, and administered with whole-body scan by 64-slice spiral CT with the pelvis scan mode.Thermoluminescence dosimeter was used to detect the absorbed doses of CT scan, while DNA damage (γ-H2AX),apoptosis,autophagy,cell cycle and percentages of CD4/CD8 in peripheral lymphocytes and bone marrow cells were analyzed by flow cytometry.γ-H2AX was detected 1 h after the last CT scan, and the other indexes were detected 24 h after the last CT scan. Results γ-H2AX increased after more than twice CT scans in peripheral lymphocytes and after than 4 times of CT scan in bone marrow cells (P < 0.05).Apoptosis increased in peripheral blood lymphocytes and decreased in bone marrow cells after more than 4 times of CT scan (P < 0.05),suggesting that lymphocytes were more sensitive while different cell subpopulation in bone marrow showed different sensitivity and response phase. No obvious changes of autophagy level were observed in peripheral blood lymphocytes, while autophagy increased in bone marrow cells after single CT scan (F =81.10,P < 0.05).8 times of CT scan significantly increased the percentages of cells at G0/G1 phase and G2/M phase(x2 =11.29,28.57,P <0.05).The percentage of CD4 + T,CD8 + T cells decreased after CT scan, but no effects on the ratio of CD4 +/CD8 +.Conclusions Repeated CT scans could induce injury effects on hematopoietic system and immune system, and then the apoptosis.Apoptosis, autophagy and cell cycle arrest might be initiated to maintain genome stability but there is no influence on immune balance.
