2.Crystal structure of SMU.2055 protein from Streptococcus mutans and its small molecule inhibitors design and selection.

OBJECTIVEThe aim of this study is to analyze the three-dimensional crystal structure of SMU.2055 protein, a putative acetyltransferase from the major caries pathogen Streptococcus mutans (S. mutans). The design and selection of the structure-based small molecule inhibitors are also studied.
METHODSThe three-dimensional crystal structure of SMU.2055 protein was obtained by structural genomics research methods of gene cloning and expression, protein purification with Ni²⁺-chelating affinity chromatography, crystal screening, and X-ray diffraction data collection. An inhibitor virtual model matching with its target protein structure was set up using computer-aided drug design methods, virtual screening and fine docking, and Libdock and Autodock procedures.
RESULTSThe crystal of SMU.2055 protein was obtained, and its three-dimensional crystal structure was analyzed. This crystal was diffracted to a resolution of 0.23 nm. It belongs to orthorhombic space group C222(1), with unit cell parameters of a = 9.20 nm, b = 9.46 nm, and c = 19.39 nm. The asymmetric unit contained four molecules, with a solvent content of 56.7%. Moreover, five small molecule compounds, whose structure matched with that of the target protein in high degree, were designed and selected.
CONCLUSIONProtein crystallography research of S. mutans SMU.2055 helps to understand the structures and functions of proteins from S. mutans at the atomic level. These five compounds may be considered as effective inhibitors to SMU.2055. The virtual model of small molecule inhibitors we built will lay a foundation to the anticaries research based on the crystal structure of proteins.

3.Study on the relationship between pediatric nurses' emotional intelligence and job performance

Objective To explore the relationship between emotional intelligence and job performance by investigating the level of them in pediatric nurses,and to provide a basis for improving job performance and the implementation of“quality care.Methods 335 pediatric nurses were investigated by using emotional intelligence scale and job performance scale.And the result of the investigation was analyzed statistically.Results The average score of the emotional intelligence of the pediatric nurses were (3.63±0.52),the highest score was interpersonal latitude (3.87± 0.69),the lowest score was management of emotions latitude (3.25±0.90); titles,the form of post and degree of nurses had a certain influence on the emotional intelligence.The score of job performance was(3.65±0.75).By Pearson correlation analysis,the emotional intelligence score and job performance score were obtained a significant positive correlation,self-motivation latitude and job performance were the most related,the management of emotional latitude job performance was the lowest.Conclusions Nursing managers should take corresponding measures to improve nurses' emotional intelligence and job performance levels,and promote the implementation of quality care effectively.
4.Study on the quality of work life status and influencing factors of the contract nurses

Objective To investigate the level and influencing factors of community nurses' quality of work life,and supply reference for improvement of community nursing management quality.Methods Totally 193 community nurses from 11 community health service center in Changchun were investigated with Chinese Version of Quality of Nurse Work Life Scale (QNWL).Results The total score of QWL was (165.00±22.19).The scores of work family balance,workload,work environment,social environment were (25.18±5.65),(39.30±5.07),(82.79±11.88) and (17.73±3.56).Multiple linear regression analysis showed that work like degree were influencing factors of community nurse' quality of work life,professional titles followed.Conclusions Community nurses' quality of work life need to be further improved.Nursing educators and managers should take measures to improve community nurse' quality of work life aimed at the influencing factors.
5.Effect of ptxA and ptxB genes of phosphotransferase system on growth of Streptococcus mutans.

OBJECTIVEThis study aims to evaluate the effect of ptxA and ptxB genes, which are important genes in the L-ascorbate phosphotransferase system (PTS) of Streptococcus mutans (S. mutans).
METHODSThe ptxA-, ptxB-, and ptxAB-double deficient mutant as well as ptxAB-complemented strain were constructed. Quantitative real-time polymerase chain reaction analysis was performed to evaluate the expression of the target genes of wild-type S. mutans when L-ascorbate was used as the sole carbohydrate source. The OD₆₀₀ values of the wild type, deficient, and complemented strains were continuously monitored, and their growth curves were constructed to compare growth capacity.
RESULTSPolymerase chain reaction and sequencing analyses suggested that deficient and complemented strains were successfully constructed. The expression levelsof ptxA and ptxB significantly increased (P < 0.01) when L-ascorbate was used as the sole carbohydrate source. The growth capacity of the deficient mutants decreased compared with that of the wild-type strain. However, the wild-type phenotype could be restored in the complemented strain.
CONCLUSIONptxA and ptxB genes are associated with L-ascorbate metabolism of S. mutans. The construction of deficient strains and complemented strain lay a foundation for further mechanism study on L-ascorbate metabolism in S. mutans.

6.Study on Methylation of DBC2 in Gastric Cancer

Objective To detect the expression of DBC2 and the methylation status of its gene promoter region in gastric cancer,so as to analyze their correlation. Methods Gastric cancer tissues and adjacent normal tissues were collected from 92 cases as research subjects. The DBC2 mRNA expression and the methylation status of DBC2 gene promoter region in gastric cancer were detected by using RT?PCR and MS?PCR respectively. In addition,the correlation of DBC2 gene methylation status with clinicopathological parameters was analyzed. Results In comparison with adjacent normal tissues,gastric cancer tissues displayed a low expression level of DBC2mRNA and a high methylation level of gene promoter region. Besides, the methylation level was significantly correlated with tumor invasion depth,differentiation and TNM staging(P<0.05). With deep invasion,poor differentiation or higher TNM staging,methylation level of gene promoter region was more significant in gastric cancer tissues. Conclusion High methylation of DBC2 gene promoter region may be a significant contributing factor leading to low DBC2 expression.
7.Investigation of the status of self-medication behaviors in children and analysis of its influencing factors

Objective To study the status of the self-medication behavior in children,and analyze the problems existing in medication of children by their parents,in order to provide references for the nurses to give medication guide to the parents of the children.Methods On the principle of convenience sampiing,120 hospitalized children's parents from a grade-Ⅲ A level hospitals in Jilin Province were investigated with a self-designed questionnaire.Results 79.3% of the parents did not behaved standardly in children's self-medication,75.0% of the parents' behaviors were not standardized in the management of the children's drugs; there was significant difference in the level of the children's self-medication among the parents with varied academic background or the age and the children's age.Conclusions The processes of self-medication in children have potential health risks,the nurses should strengthen health education,raise the parents' cognition of the risk of self-medication for children,and thus to help parents establish scientific and rational self-medication behaviors.
8.Local injection of ligustrazine influences the expression of osteoprotegerin in the maintenance phase in rats with orthodontic tooth movement

BACKGROUND:Ligustrazine has been shown to restore the function of the femoral headviathe revascularization, increased blood flow, theabsorption ofnecroticbone, and bone regeneration. OBJECTIVE:To study the effects of ligustrazine on remodeling of periodontal tissues and the expression of osteoprotegerin in the maintenance phase in rats with orthodontic tooth movement. METHODS:Thirty-two healthy male Wistar rats were included and equaly randomized into four groups. Maxilary left first molar mesialization was performed through traction of 50 g force for 21 days to establish the rat model of tooth movement. 5, 10, 15 mg/L ligustrazine (50 μL) were localy injected into the first molar periosteum in model rats on the day before removing the orthodontic forcing device. Same volume of saline was injected in the control group. The injection was administered every other day. At 1 and 4 weeks after injection, the distance of tooth movement, the recurrence distances and percentage were determined and calculated. The pathological changes in periodontal tissues were observed by immunohistochemistry and hematoxylin-eosin staining. The width ofthe parodontium and number of osteoblasts were observed under an optical microscope. RESULTS AND CONCLUSION:The recurrence distance inthecontrol group was increased compared withtheexperimental group, while the number of osteoblasts and osteoprotegerin immunoreactivity were decreased. Good width of the parodontium and smal recurrence trend were found in 10mg/L ligustrazine group. These findings indicate that ligustrazine promotes the proliferation of osteoblasts and enhances the expression of osteoprotegerin, which is beneficial to the retention of teeth after orthodontic surgery.
9.Detection of the Platelet Activation Markers CD62p and PAC-1 in Patients with Hypertension and Coronary Heart Disease

Objective To investigate the clinical significance of platelet activation markers CD62p and PAC-1 in patients with hypertension and coronary heart diease.Methods To select patients who were in the Second Affiliated Hospital of Dalian Medical University between January 2012 and September 2012,42 patients with hypertension,46 patients with coronary heart disease.Flow cytometry was used to test the positive percentage of the platelet surface activation markers CD62p and PAC-1 .To observe the difference among hypertension group,coronary heart disease group and health control group;hyper-tension and coronary heart disease group.Results The positive percentage of platelet activation markers CD62p and PAC-1 in hypertension (36.36±9.62,7.18±8.20)%,coronary heart disease group (42.74±14.60,8.81±12.53)% showed sta-tistically significant differences (t=4.150~5.853,P<0.01)compared with healthy control group (26.82±9.13 ,1.09± 1.05)%.The positive percentage of CD62p in coronary heart disease group (42.74±14.60)% was also higher than that of hypertension (36.36±9.62)%,and the difference between them was of statistical significance (t=2.444,P<0.05).Con-clusion The hypertensive patients and coronary heart disease were in pre-thrombotic state,the activation of platelet increase significantly.Molecular markers should be measured in hypertensive patients and coronary heart disease for prevention and treatment of thrombotic disease complication.
10.CT differentiation between vertebral multiple myeloma and osteolytic metastasis

Objective To explore the imaging differences of vertebral multiple myeloma(VMM)and vertebral osteolytic metastasis on CT. Methods Review the imaging signs of 32 patients with VMM and 52 patients with vertebral osteolytic metastasis,then record and finally statistical analysis was carried out.Results Compare to 1 67 vertebras involved in 52 cases of vertebral metastasis,the patents with VMM had 220 vertebras involved.The incidence of the multiple small circular type of bone destruction in VMM was 42.66%(93/218),which was higher than that in vertebral metastasis 0.00% (0/165)(χ2 =92.963,P =0.000).The incidence of the irregular shape type of bone destruction in VMM was 23.39% (5 1/218),while it was higher in patients with vertebral metastasis 45.45%(75/165)(χ2 =20.704,P=0.000).It was also found that the incidence of the fragmentary type of bone destruction and the involvement of the unilateral pedicle in VMM were lower than that in patients with vertebral metastasis 8.7% (19/218 )& 27.27% (45/165)(χ2 =23.238,P =0.000), 6.82% (1 5/220)& 1 7.96% (30/1 67)(χ2 =1 1.477,P =0.001).The incidence of the crest protrusion type of bone destruction in patients with VMM was 16.06% (35/218),however it was 9.10% (15/165)(χ2 =4.013,P =0.045)in vertebral osteolytic metastasis. Conclusion The imaging features of VMM and vertebral osteolytic metastasis had certain characteristic.And they can be differentiated from each other,combining with clinical traits.
