Objective Curcumin and MS-275,an inhibitor of histone deacetylase (HDAC),are promising anti-tumor agents. The aim of present study was to investigate the mechanisms of apoptosis induced by combined use of MS-275 in low dosage and curcumin in prostate cancer cell line DU145. Methods MTT assay was used to evaluate the lethal effect on DU145 cells by solitary use of MS-275 and or combined with curcumin. The changes in cell life cycle was detected by flow cytometry. The expressions of the survival signaling pathways were determined by Western blotting. Results Solitary application of MS-275 or curcumin may inhibit the growth of DU145 cells in a time and dose-dependent manner. The combination of MS-275 (1?mol/L) and curcumin (20?mol/L) exhibited obvious cytotoxic effect on cell viability, which was only 45.9% 48h after the combined treatment. Cell cycle assay showed that the combination of MS-275 and curcumin resulted in obvious appearance of sub-G1 phase in DU145 cells, implying that the cell apoptosis had been induced. The results of Western blotting showed that after treatment of MS-275 or curcumin singly, the phosphorylation level of Akt and ERK kinase declined slightly, however, when MS-275 and curcumin were used together, there appeared a prominent inhibitory effect on Akt and ERK kinase, indicated by a sharp decline of their phosphorylation level, and at the same time, the level of cleaved PARP,a hallmark of apoptosis, was increased in DU145 cells. Conclusion Combined use of MS-275 and curcumin may exert a synergistic cytotoxic effect on the viability of DU145 cells, and exhibit an inhibitory activity on Akt and ERK kinases to induce apoptosis.

Objective To screen and verify the proteins interacting with phosphorylation cluster of DNA dependent protein kinase catalytic subunit((DNA-PKcs) by yeast two-hybrid assay. Methods To know the proteins interacting with DNA-PKcs phosphorylation cluster, yeast two-hybrid assay was applied to screen the cDNA library of human hepatic tissue with a previously constructed plasmid pGBKT7-DPC.The positive clones were further identified by PCR,rotary validation and sequence analysis. Then the eukaryotic expression vectors of the bait protein and screened positive clone proteins were constructed and transfected into human embryonic kidney 293T cells to detect whether the proteins could been expressed correctly. At last, the bait protein and screened positive clone proteins were co-transfected into 293T cells and protein interaction was detected with Co-Immunoprecipitation (Co-IP) assay. Results After two rounds of screening using the yeast two-hybrid assay,12 candidate clones were obtained. Then 7 clones with different insert fragments were identified by PCR,and 3 positive proteins interacted with DNA-PKcs phosphorylation cluster were further verified by rotary validation.Sequencing analysis demonstrated that these 3 proteins were MBNL1,SIK2 and YY1AP1,respectively.Accordingly,the eukaryotic expression vectors of bait protein and 3 positive clone proteins were constructed successfully and expressed correctly in 293T ceils. Finally, the Co-IP assay confirmed that these 3 positive clone proteins could interact with DNA-PKcs phosphorylation cluster. Conclusions Proteins interacting with DNA-PKcs phosphorylation cluster are successfully screened and identified.

Background To provide clinical evidence for ablative application by comparison of the analgesic effect following different thalamotomy in rats. Methods Thirty rats were randomly assigned into sham and 4 thalamotomies groups: central medial thalamic nucleus ( CM), parafascicular thalamic nucleus ( PF), ventral posterolateral thalamic nucleus (VPL), and CM +Cigulum (cg). Two μL 10% phenol dissolved in glycerin were used for stereotactic thalamotomy. The thermal pain thresholds before and after procedures were evaluated with the tail stimulate test. The formalin test was carried out in an open field apparatus where the animal formalin-induced responses (licking duration, flexing duration, and flinching frequency of the injected paw) were recorded for 60 min. Results Changes of pain thresholds in all ablative groups were significantly higher than that in the sham group, especially it was higher in VPL group. Differences of the factor thalamotomy were found to be due to the shorter licking in the ablative groups than that in the sham group (P <0.01 ), whereas flexing duration and flinching frequency were only slightly affected by thalamotomy. Moreover, licking duration was lower in VPL group than in CM and CM + cg groups ( P <0.05), whereas nociceptive responses did not differ between the CM and CM+cg groups (P >0.05).Conclusions In acute period, CM, PF, VPL, CM + cg neurolysis all showed to elevate the thermal pain threshold and to reduce the pain-induced behavioral responses related to supraspinal neural circuits (licking of the injected paw). Among them, the damage of VPL might be the most active one. CM + cg damage did not get better antinociceptive effect than single CM ablation.

OBJECTIVETo test whether Porphyromonas gingivalis (P. gingivalis) could produce bacterial signal molecule, bis-(3'-5')-cyclic dimeric adenosine monophosphate (c-di-AMP) and lay the foundation for explorations of its roles in life metabolism and periodontitis immunity of P. gingivalis.
METHODSP. gingivalis standard strain ATCC33277 was used as the experimental strain to extract nucleic acids from the bacteria. Then, c-di-AMP was detected using high performance liquid chromatography coupled with mass spectrometry (HPLC-MS/MS). Subsequently, HPLC was used to validate the sample further.
RESULTSBased on the signal/noise (S/N) for 3 : 1, the limit of determination of HPLC-MS/MS for peak time of c-di-AMP standard substances was 7.49 min and nucleic acid extractions from P. gingivalis was 8.82 min (S/N > 3). Further confirmation of HPLC showed that nucleic acid extractions from both P. gingivalis and c-di-AMP standard substances pre- sented goal absorbent peaks at 15.7 min, with the same ultraviolet absorbent spectrum.
CONCLUSIONThe nucleic acid extrac- tions from P. gingivalis contained c-di-AMP, which shows that P. gingivalis could produce c-di-AMP.
