1.Technetium [99Tc] methylenediphosphonate inhibits osteoclast formation from PBMCs in patients with rheumatoid arthritis

Objective To observe the influence of technetium [99Tc] methylenedipho-honate (99Tc-MDP) on osteoclastogenesis induced by receptor activator of NF-κB ligand (RANKL) and macrophage-colony stimulating factor (M-CSF)in peripheral blood mononuclear cells in patients with rheumatoid arthritis, and to study the mechanism of 99Tc-MDP in osteoclast differentiation. Methods The monocytes/macrophages were isolated from peripheral blood in patients with rheumatoid arthri-tis, incubated in RPMI-1640 with receptor activator of NF-κB ligand (RANKL, 25 μg/L), macro-phage-colony stimulating factor (M-CSF, 25μg/L ) and different concentrations of 99Tc-MDP (5, 10, 20,and 50 mg/L) for 4,12, and 20 days. Tartrate resistant acid phosphatase staining was used to observe the formation of osteoclasts. Results After 12 or 16 days culture of peripheral blood mononuclear cells, plenty of large nultinuclear cells could be found on the coverslips. 99Tc-MDP markedly inhibited those changes and the inhibitory effects were increased as the concentration of 99Tc-MDP increased (P<0.05). Conclusion 99Tc-MDP probably has some protective effect on rheumatoid arthritis by inhibiting osteoclast formation.
2.Moderating effect of emotional expressivity on relationship between test anxiety and working memory capacity

Objective:To investigate the moderating role of emotional expressivity in the relationship between test anxiety and working memory capacity (WMC).Methods:Totally 332 college students were selected.The Test Anxiety Inventory (TAI),Emotional Expressivity Inventory (EEI) and Arithmetic Operation-Word Memory Span Task (OSPAN) were used to measure the severity of test anxiety,working memory capacity,and degree of emotional expressivity,respectively.Results:TAI scores were negatively correlated with WMC scores (r =-0.36,P <0.01).Emotional expressivity had a moderating effect between scores of test anxiety and WMC (β =0.19,P <0.01).Test anxiety score negatively predicted WMC score in students with low emotional expressivity (β =-0.74,P < 0.01),whereas it couldn't predict WMC in those with high emotional expressivity (β =-0.07,P >0.05).Conclusion:It suggests that higher emotional expressivity could buffer the negative effect of test anxiety on working memory capacity.
3.Review of ARDs of Ready-made Chinese Drugs in the First OTC List

OBJECTIVE: To review the ARDs of ready-made Chinese drugs included in OTC list .METHODS: 160 cases of ARDs induced by ready-made Chinese drugs,reported in Chinese journals over 40 years(1960~2000),were analysed .RESULTS: 34 kinds of drugs were involved in inducing ARDs, of which, Huoxiang Zhengqi liquid was the most common one( 19 cases), essense of Fengyou and pseudo-ginseng tablet were the next(16 cases and 13 cases respectively).The ARDs in duced by above-mentioned 3 drugs accounted for 41.4% of total ARDs.Hypersensitive reaction was the most common ARD (67.2%) . 115 of 116 cases were cured, one died.CONCLUSION: Ready-made Chinese drugs can induce ARDs, so the consumers should pay attention to it, however,they need not feel frieghtened,so long as the drugs are used rationally, the incidence of ARDs is very low and the prognosis is good.

5.Metabolomics characteristics in a rat model of myocardial infarction based on bibiometrics analyses

BACKGROUND:Metabonomics has been proved to analyze and observe the pathological process of rat myocardial infarction and the underlying mechanism. OBJECTIVE:To further analyze the metabolomic pathways of bioinformatics in rat models of myocardial infarction. METHODS:The experimental studies about rat myocardial infarction were retrieved from CNKI, WanFang, CqVip, PubMed and Embase databases. The metabolic products described in the literatures were col ected and summarized. Signaling pathways were analyzed using KEGG database molecular function annotation, the enzymes, translocators and their properties were analyzed by HMDB database. Metabolites pathway were visualized with MetPA. RESULTS AND CONSLUSION:A total of 26 metabolic products were identified in the included literatures and mainly participated in 29 metabolic pathways. Through topology analysis, 5 of the 10 metabolic pathways were selected and regarded as the metabolic pathways of myocardial infarction in rats, including aminoacyl-tRNA biosynthesis;glycine, serine and threonine metabolism;valine, leucine and isoleucine biosynthesis;biosynthesis of unsaturated fatty acids;phenylalanine, tyrosine and tryptophan biosynthesis. In conclusion, the bioinformatics analysis of metabolites in rats with myocardial infarction show that myocardial infarction is related to the metabolism and metabolic pathways of carbohydrates, proteins, fat and RNA.
6.Imaging Analysis of Lymphoma of Anterior Mediastinum

Objective To investigate the X-ray and CT features of anterior mediastinal lymphoma.Methods 11 cases with lymphoma of anterior mediastinal confirmed by pathology were collected,the X-ray and CT features were reviewed.Results The tumors in all cases appeared as anterior mediastinal tumor,among them,the tumors in six cases grew at anterior mediastinum bilaterally and in five cases at one-side.There were soft tissue nodi around tumor in eleven cases,superior vena cava were involved in 6,neighborhood blood vessel embed by tumors in 5,pleural effusion could be seen in 3,combined with hydropericardium in one.Conclusion The anterior mediastinal lymphomas have certain imaging characteristics,most of them can be distinguish from other anterior mediastinal malignant tumors if one analysing their imaging features carefuly.
7.The expression of livin of kidney tissues in rats with acute kidney injury induced by endotoxin

Objective To investigate the expression of livin of kidney tissues in rats with acute kidney injury (AKI) induced by endotoxin ,and the role of livin in the cell apoptosis of AKI .Methods The rat models with AKI were induced by endotoxin .The de-gree of kidney injury was observed by hematoxylin and eosin stain ,measuring the levels of the serum creatinine and urea nitrogen . The expression of livin and caspase-3 in kidney tissue at different time points was analyzed by immunohistochemistry assay ,and the relationship between the expression of livin and caspase-3 and kidney injury was analyzed .Results Compared to the control group , the rats injected endotoxin had the performance of AKI ,with obviously pathomorphological damage in the kidney tissues ,signifi-cantly increasing in the serum creatinine and urea nitrogen (P<0 .01) .The livin and caspase-3 of kidney tissues in rats with AKI caused by endotoxin were positive expression (P<0 .01) .With the lapse of time ,the trend of increasing of caspase-3 showed gradu-ally slow after the highest expression of livin .Conclusion The livin involved in the pathogenesis of endotoxin-induced AKI ,it might relieve the kidney injury and protect renal function by inhibiting casepase-3 important apoptotic effector protein .
8.Analysis of correlation between hypokalemia and sudden death in young and middle-aged people

Objective To explore the correlation between hypokalemia and sudden death in young and middle-aged people. Methods One hundred and twenty-nine young and middle-aged patients with sudden death during treatment were selected as observation group. Then 100 cases of healthy volunteers were randomly selected as control group. The incidence of cardiovascular disease, incidence of hypokalemia and intake of potassium were compared between 2 groups. Results The incidences of hypertension, arrhythmia, myocardial infarction, heart failure and hypokalemia in observation group were significantly higher than those in control group: 17.05% (22/129) vs. 5.00%(5/100), 13.18% (17/129) vs. 2.00% (2/100), 26.36% (34/129) vs. 9.00% (9/100), 9.30% (12/129) vs. 1.00% (1/100) and 55.04% (71/129) vs. 12.00% (12/100), and there were statistical differences (P<0.01). The ratios of higher, common, lower intake of potassium in observation group were 1.55%(2/129), 27.91% (36/129) and 70.54%(91/129), in control group were 15.00% (15/100), 58.00% (58/100) and 27.00% (27/100), and there was statistical difference (P<0.01). Logistic regression analysis result showed that hypertension, arrhythmia, myocardial infarction, heart failure, hypokalemia and lower intake of potassium were the risk factor for sudden death (P<0.01). The incidences of hypertension, arrhythmia, myocardial infarction and heart failure in hypokalemia patients were significantly higher than those in normokalemia patients: 28.92% (24/83) vs. 2.05% (3/146), 19.28% (16/83) vs. 2.05% (3/146), 44.58%(37/83) vs. 4.11% (6/146) and 13.25% (11/83) vs. 1.37% (2/146), and there were statistical differences (P<0.01). The incidence of hypokalemia in people with lower intake of potassium was significantly higher than that in people with higher and common intake of potassium: 56.78% (67/118) vs. 2/17 and 14.89%(14/94), and there was statistical difference (P<0.01). Conclusions There is a significant correlation between hypokalemia and sudden death in young and middle-aged people. Preventive measures of sudden death should be made according to serum potassium level in clinic. People should pay attention to the uptake of potassium in daily life.
9.Gene Cloning, Construction and Expression of Single-Chain Fv (scFv) Against the Membrane Protein of Schisotosoma japonicum

Objective To construct single chain antibody specific to membrane protein of Schistosoma japonicum by gonetic engineering technique. Methods The V\-H (heavy-chain variable region) and V\-L (light-chain variable region) genes were amplified by PCR from the genomic DNA of NP11-4 cell line, and sequenced by Sanger's method. The ScFv was constructed in pTHA90 vector using V\-H and V\-L genes, then expressed by IPTG. Results The V\-H and V\-L genes were obtained through PCR. The DNA sequences showed that V\-H and V\-L were new variable region genes of antibody. They were registered by GenBank. A ScFv gene with (Gly4Ser) 3 intralinker in the pTHA90 vector was successfully constructed. The ScFv was expressed as thioredoxin-fused proteins about 36\^2 kDa. Conclusion A specific ScFv against the membrane protein of Schistosoma japonicum was constructed and expressed.
