Objective:To investigate the expressions of Th 1-Th2-Th17 cytokines in the coxsackievirus B 3-induced mice chronic viral myocarditis(VMC).Methods:BALB/c mice were intraperitoneally(i.p) infected with increased CVB3 for establishing chronic VMC models.Control mice were treated with phosphate-buffered saline(PBS)i.p.Cardiac tissues were obtained 8 weeks after CVB3 in-jection,myocardial histopathologic changes were observed by HE and Masson staining .Th1-Th2-Th17 cytokines in plasma were detected by protein array technology , and their cardiac mRNA expressions were measured by RT-PCR.Results: Compared with the control group,levels of IL-2,IL-5,IL-10,IL-13,IL-17,IL-6,IL-22,IL-21 and TGF-βobviously increased in chronic VMC group (P all<0.05). Conclusion:The imbalance of Th1-Th2-Th17 cytokines may play an important roles in the pathogenesis of chronic VMC .

Objective To observe the effect of ginsenoside Rb 1 and Re on cardiomyocyte apoptosis and expression of Bcl-2, Bax, Bad and Fas gene proteins after acute ischemia and reperfusion in rats and elucidate the possible mechanisms. Methods The ischemia-reperfusion heart model was made by ligating the left anterior descending branch of coronary artery in Wistar rats. The apoptotic cardiomyocytes were confirmed with transmission electron microscopy and counted with in situ nick labeling (TUNEL) method and light microscopy. The expression of Bcl-2?Bax?Bad and Fas gene proteins were studied by immunohistochemical staining. Mean optical density (OD) value of the gene proteins expression were quantitatively examined by image analysis system. Results A.The apoptotic cardiomyocytes were not observed in the sham-operation group. The number of the apoptotic cells were 134.45?45.61/field in the ischemia- reperfusion group, 51.65?13.71/field in the ginsenoside Rb 1-treated group and 90.66? 19.22/field in the ginsenoside Re-treated group, respectively. The differences were significant among the three groups (P0.05), but Bax?Bad and Fas gene expression were decreased significantly in the ginsenoside Rb 1 and Re-treated group as compared with the ischemia-reperfusion group (P

Objective To investigate the influence of PPARγ excitomotor RSG and ATRA on gastric cancer SGC7901 cell line proliferation in vitro and its potential mechanism study.Methods Human gastric cancer SGC7901 cell line was cultured in vitro.Experiment samples were divided to blank group,10μmol/L ATRA group, 12.5μmol/L RSG group, 25μmol/L RSG group, 10μmol/L ATRA + 25μmol/L RSG group.Proliferation inhibitory effect was determined by MTI assay.Flow cytometry was used to detect cell cycle, H.E stain was used to observed micrography alteration.Expression of PPARγ protein in gastric cancer cells were measured by immunohistochemistry.PPARγ mRNA in gastric cancer cells were measured by RT-PCR.Results ATAR at concentration 10μmol/L, RSG at 12.5 μmol/L and RSG at 25 μmol/L could inhibit the proliferation of SGC7901 cells in a dose-and time-dependent, and when both agents were combined for 72h, growth inhibition ratio was (29.73 ± 0.69) %.Flow cytometry analysis revealed a cell cycle arrest at G1 and S phase, and when both agents combined, S% was (12.87 ± 0.35 )%, cell micrography tended to be normal when both agents combined.Up-regulation of PPARγ protein and PPARγ mRNA expressions were also observed, those effects were enhanced when both agents combined, and grey scale ratio was 0.646.Conclusion The ATRA and RSG could significantly induced growth inhibition of human gastric cancer SGC7901 cell, which may be associated with cell cycle arrest and inducing differentiation, activation of PPARγ protein and PPARγ mRNA expression.Synergistic effect could be caused by the combined use of the two agents.

Objective To discuss the X-ray and CT manifestations of keratocystic odontogenic tumour(KCOT)in mandible,to im-prove the diagnostic accuracy of the disease.Methods The X-ray and CT features of KCOT(n=25)in mandible,which were proved by surgery and pathology,were retrospectively analyzed.Results The KCOT,including solitary tumor(n=23)and multiple tumor (n=2),mainly displayed unilocular or multilocular in shape with distinct sclerosing margin.Disruption of the adjacent cortex was de-tected in 7 cases,growth along the long axis of mandible in 18 cases,compression and displacement of the adjacent teeth in 21 cases, resorption of bevel of roots in 2 cases.Conclusion Most lesions of KCOT in mandible have characteristic manifestation in X-ray and CT findings,which is helpful for diagnosis and differential diagnosis.

Objective To discuss the X-ray and CT findings of pulmonary air leak in neonates and to improve the early diagnostic ability.Methods The supine anteroposterior chest films of 33 neonates with pulmonary air leak were retrospectively analyzed.A-mong them,spiral CT scanning was performed in 5 cases.Radiographic follow-up was made in 30 cases.Results Pulmonary intersti-tial emphysema was found in 2 cases,pneumomediastinum in 4 cases,simple pneumothorax in 14 cases.Pulmonary interstitial em-physema combined with pneumomediasfinum was detected in 2 cases,with pneumothorax in 4 cases.Pneumomediastinum combined with pneumothorax was displayed in 3 cases.Pulmonary interstitial emphysema combined with pneumomediastinum and pneumotho-rax was found in 4 cases.Among them,medial pneumothorax was shown in 26 side,lateral pneumothorax in 9 side.Conclusion X-ray radiography is the main method for the diagnosis of pulmonary air leak in neonates,and CT can further define the location,range and extent of the disease.

BACKGROUND: The studies found that cardiomyocyte apoptosis is related to ischemia-reperfusion injury directly. The clinical and experimental studies proved that ginseng and losartan could improve myocardial iscbemia and prevent the ischemia-reperfusion injury.But the comparative study of their effects on cardiomyocyte injury induced by ischemia and reperfusion has not been reported.OBJECTIVE: To compare the effects of ginseng and losartan on cardiomyocyte Bcl-2 gene expression after ischemia and reperfusion in vivo.DESIGN: Randomized controlled experiment.SETTING: Department of Cardiovascular Internal Medicine, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology.MATERIALS: The experiment was carried out in the Laboratory of Cardiovascular Internal Medicine of Tongji Hospital of Tongji Medical College Affiliated to Huazhong University of Science and Technology from November 2002 to April 2003. Totally 40 healthy adult Wistar rats, weighting 200-250 g, of either gender, were selected and divided into 4 groups:sham operated group, ischemia-reperfusion group, ginseng treated group and losartan treated group with 10 rats in each group.METHODS: Rats were modeled in ischemia-reperfusion group, ginseng treated group and los arran treated group but not in sham operation group.20 mg/kg (1 mL in volume) losartan was given by stomach. The first administration was 2 hours prior to operation. Subsequently, the second and third administrations were given in immediate and 24 hours after operation,respectively; 1 mL of radix ginseng rubra (1 g/mL) was given by stomach.The first administration was 2 hours before operation. Subsequently, the second and third administrations were given in just and 24 hours after operation, respectively. The rats in ischemia-reperfusion and sham-operated group were given the normal saline with the same volume and at the same time. Immunohistochemistry and in situ hybridization histochemistry were used to measure mRNA and protein of Bcl-2 gene expression that were compared with those in the control group.MAIN OUTCOME MEASURES: The expression level of Bcl-2 mRNA and Bcl-2 protein in ginseng treated group and losartan treated group.RESULTS: Data of totally 40 rats was entered the final analysis. ① Content of Bcl-2 mRNA and protein were not significantly different both in ginseng group and control group (P ＞ 0.05). ② Contents of Bcl-2 mRNA and Bcl-2 protein were higher in ginseng treated group than those in the ischemia-reperfusion group (P ＜ 0.05 or 0.01).CONCLUSION: Expressions of Bcl-2 mRNA and Bcl-2 protein in ginseng treated group are higher than those in losartan treated group, which suggests that ginseng has not same effect with losartan in inhibiting cardiomyocyte apoptosis and in preventing and curing cardiomyocyte injury induced by ischemia-reperfusion.

Objective To investigate the effect of hot compress with traditional Chinese herbs with wine and vinegar on lumbago after cesarean section. Methods Ninety patients with lumbago after cesarean section were divided into the control group and the observation group equally according to their admission number. Both groups were cured by the injection at the pressure pain point, the observation group by hot compress of traditional Chinese herbs of Reyanbao with wine and vinegar. The treatment effect between both groups were observed. Results The effective rates of the experiment group and the control group were 93.3%and 68.9%, respectively. The difference was statistically significant (χ2=12.433, P<0.05). Conclusions Based on injection on the pressure pain point, lumbago after cesarean section can be effectively relieved with the extra treatment by hot compress with traditional Chinese herb of Reyanbao with wine and vinegar. The treatment is simple and safe and worthy of being applied clinically.

Objective To investigate the effect of CD151 gene therapy on improving myocardial function in swines with myocardial infarction. Methods CD151, antisense CD151 and green fluorescent protein (GFP) were constructed into the recombinant adeno-associated virus (rAAV). Swines were divided into 4 groups: rAAV-GFP group (6 swines), rAAV-CD151 group (6 swines), rAAV-antiCD151 group (6 swines) and control group (6 swines). The swines were performed with coronary artery ligation and intramuscularly injection with rAAV. Eight weeks after vector administration, western blot was used to detect gene expression of CD151. 13N-labeled NH3 positron emission tomography (PET) was used to evaluate myocardial perfusion. Echocardiography was used to assess myocardial function. Results Compared with the control group and the rAAV-GFP group, the rAAV-CD151 group showed higher CD151 protein expression. Compared with the rAAV-GFP group, the defect size of myocardium was decreased[( 11.3±2.4)% vs. (21.1±2.6)%, t= -5.67,P<0.01] and left ventricular ejection fraction (EF), left ventricular fractional shortening (FS), the ratio of anterior lateral wall thickening (△ALWT) and ratio of interventricular septum thickening (△IVST) were significantly improved in rAAV-CD151 group 8 weeks after vector administration [(65.7±4.6)% vs. (54.7±5.3)%, (36.0±2.9)% vs. (27.6±3.1)%,(55.4± 4.9)% vs. (36.8±7.8)%, (35.2±6.0)% vs. (26.7±4.4)%, t=3.98, 3.35, 3.34, 9.27, all P< 0.05]. The level of diastolic ALWT and diastolic IVST was also increased in rAAV CD151 group compared with rAAV-GFP group ( P<0.05).Compared with rAAV-CD151 group, parameters of myocardial function in rAAV-antisense CD151 group were not improved (P<0.05). Conclusions rAAV-CD151 can effectively transfeet the myocardium, increase the expression ofCD151 protein, promote the blood perfusion of myocardium and improve the ventricular function after myocardial infarction.

Objective To investigate the changes in myocardial proteomics in the late phase of limb ischemic preconditioning (LIP) in rats.Methods Twelve pathogen-free adult male Sprague-Dawley rats,aged 8-9 weeks,weighing 260-280 g,were randomly assigned into LIP group (n=6) and control group (group C,n=6) using a random number table.Limb ischemia was preceded by 3 cycles of 5-min ischemia which was induced by ligation of the root of the right hindlimb with a rubber band followed by 5-min reperfusion in group LIP.At 24 h after LIP,the tissues were obtained from the left ventricle,and the isobaric tags for relative and absolute quantification technique and liquid chromatography-mass spectrometry were applied to detect the differences in protein expression profiles between the two groups (the difference in expression between the two groups＞ 1.2 times and P＜0.05).The identified differentially expressed proteins were analyzed using the bioinformatics,and some were further verified by Western blot.Results A total of 55 proteins were identified to be differentially expressed,and among the 55 proteins,the expression of 35 proteins was up-regulated,and the expression of 20 proteins was down-regulated.Bioinformatics analysis showed that most of the 55 proteins were organelles,cell membrane or macromolecular compounds,were involved in the process such as metabolism,biological regulation,stress response and signal transduction,and showed functions such as the binding affinity to molecules,catalytic activity,anti-oxidant activity,and modulation of the activity of enzyme.The results verified by Western blot were consistent with those shown by using the isobaric tags for relative and absolute quantification analysis.Conclusion The late phase of LIP can induce changes in the expression of the 55 proteins involving regulation of energy metabolism,anti-oxidant action,regulation of gene expression,and protein folding and degradation in the myocardium,which may be the mechanism of myocardial protection in rats.

Protective effect and mechanism of electroacupuncture (EA) on acute reperfusion ventricular arrhthmia was investigated. Ventricular arrhythmia was induced by occlusion of the proximal left anterior descend (LAD) branch of coronary artery for 5 min and followed with 15 min reperfusion. EA on acupoint "Neiguan", "Jianshi" was performed at 30 min before ligation and continued another 5 min during ischemia. Isoprenaline (20, 30 and 50 microg/kg) or atropine (1 mg/ kg) was intravenously injected at 5 min before ischemia. The results showed that EA significantly decreased the incidence of ischemia/reperfusion (I/R) induced ventricular tachycardia (VT), ventricular fibrillation (VF) and mortality as compared to I/R group. Atropine partially suppressed the EA's effect of antiarrhythmia; Isoprenaline increased the incidence and severity of reperfusion arrhythmia, which was inhibited by EA, but this inhibition of EA was blocked with increasing dose of isoprenaline. The results indicated that EA treatment could prevent the occurrence of reperfusion ventricular arrhythmia in rats with myocardial ischemia, and its mechanism might be related to the regulation of EA on the beta-adrenoceptors and M-cholinergic receptor activation in myocardium.
