Objective To observe the effects of serum containing the medicine of Tujian Mixture on phospho-Akt and phospho-PTEN of glomerular mesangial cell (MC) cultured in high concentrations of glucose. Method The glomerular mesangial cells from SD rat were divided into six groups:the normal control group, the group of MC cultured in high concentrations of glucose, and four other groups cultured in high concentrations of glucose + different concentrations of rat’s serum containing Tujian Mixture. After 72 hours, the content level of phospho-Akt (Thr308) and phospho-PTEN (Ser380) in MC was detected by using sandwich ELISA. Objective To observe the therapeutic effect of segmental scleral buckling and vitrectomy with/without lensectomy on the retinopathy of prematurity (ROP) stage 4a,4b and 5.Methods One hundred and thirty-four ROP infants (181 eyes) diagnosed as stage 4a,4b and 5,and performed with segmental scleral buckling or vitreous with/without lensectomy were retrospectively analyzed.The operated 4a-,4b-and 5-stage eyes were 40,51 and 90 eyes.The operational method depended on the location and severity of fibrovascular membrane.Of 181 eyes,segmental scleral buckling was referred for 37 eyes which include 23 eyes with 4a stage and 14 eyes with 4b stage;vitrectomy was referred for 50 eyes which include 14 eyes with 4a stage,29 eyes with 4b stage and 7 eyes with 5 stage;vitrectomy with lensectomy was referred for 94 eyes which include 3 eyes with 4a stage,8 eyes with 4b stage and 83 eyes with 5 stage.The effect was classified as success,improved and failure.Failure includes lost eye.Follow-up for 4a,4b and 5 stage patients are 34,31 and 29 months respectively.Results Segmental scleral buckling was referred for 37 eyes,success in 23 eyes (62.16%),improved in 11 eyes (29.73 %),failure in 3 eyes (8.11 %).Vitrectomy was referred for 50 eyes,and success in 20 eyes (40.00%),improved in 22 eyes (44.00%),and failure in 8 eyes (16.00%).In the total of 94 eyes underwent vitrectomy with lensectomy,20 eyes was success (21.28%),improved in 17 eyes (18.08%),failure in 57 eyes (60.64%).In 40 stage 4a eyes,33 successes (82.50%),6 improved (15.00%) and 1 failure (2.50%).In 51 stage4b eyes,11 successes (21.57%),30 improved (58.82%) and 10 failures (19.61%).For 90 stage 5 eyes,14 successes (17.50%),19 improved (23.75%) and 57 failures (71.25%).The therapeutic effect of segmental scleral buckling for stage 4a was better than that for stage 5 (x2 =6.707,P=0.035).The difference of therapeutic effect of vitrectomy for different stage was significant (x2=21.010,P =0.000);stage 4a was the best;stage 4b was the second,stage 5 was the worst.The therapeutic effect of vitrectomy with lensectomy for stage 5 was worse than that for stage 4a and 4b (x2=16.066,P=0.003).Conclusion The surgery patterns of ROP was determined based on the disease severity,the surgery effects of stage 4a and 4b were better than stage 5,which had nothing to do with the surgical procedures.
3.Correlation between body fat distribution and cardiovascular risk factors in some patients with type 2 diabetes mellitus in Xinjiang region

Objective To investigate the relation between the characteristics of body fat distribution and the cardiovascular risk factors in some patients with type 2 diabetes mellitus (T2DM)in Xinjiang region.Methods The percentage of body fat (PBF)and visceral fat area(VA)were determined in 155 cases of T2DM by using the Inbody 720 body composition analyzer.At the same time triglycerides(TG),total cholesterol(TC),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),blood pressure(BP), glycosylated hemoglobin HbA1c,body weight(W),body height,waist circumference ratio(WC)and hip circumference(HC)were also determined.The body mass index(BMI)and the waist to hip ratio was calculated.Results In both men and women,BMI in the subjects with visceral fat obesity(VFO)was higher than that with subcutaneous fat obesity(SFO).In males,TG in VFO was signif-icantly higher than that in SFO.PBF,TC and LDL-C in the female T2DM patients were significantly higher than those in the male T2DM patients.VA and PBF in the minority male patients were significantly higher than those in the Han nationality male patients (P <0.05).After correcting the age,course of disease and BMI,WHR in the female patients was positively correlated with DBP and PBF was positively correlated with LDL-C.In the male patients,there were correlations between BMI with SBP,between VA with DBP,and between VA with PBF and HDL-C;in the female patients,there were correlations between BMI with SBP and be-tween WHR with DBP.Conclusion The body fat distribution in the patients with T2DM in Xinjiang region is dominated by VFO. The cardiovascular risk factors are not only associated with the increase of body fat content,but more closely associated with VFO, moreover the differences in genders and nationalities exist.
4.Effect of perindopril on the expression of scavenger receptor A gene in diabetic nephropathy

Objective To explore the effect of ACE-inhibitor perindopril on the expression of scavenger receptor A (SR-A) gene in the kidney of diabetic rats.Methods Diabetes were induced in male Sprague-Dawley rats by peritoneal injection with streptozotocin (60mg/kg).The rats were then random di vided into normal control group, diabetes group and ACEI treatment group [4mg/(kg·d) for 24 weeks].Blood glucose concentration and 24h urinary albumin excretion were determined.The renal morphological change was observed.Immunohistochemistry was used to analyze CD68 positive macrophages,and the Mrna of SR-A in renal tissue was detected by quantitative real-time PCR.Results Compared with normal control group,blood glucose concentration,24h urinary albumin excretion and the number of CD68 positive macrophages were significantly increased [(5.3 ± 0.6) mmol/L vs (26.7 ± 3.3) mmol/L;(2.7 ± 1.3) mg/24h vs (26.7 ± 1.8)mg/24h;(0.77 ±0.24)/gcs vs (2.55 ±0.46)/gcs;(6.13 ±0.50)/HPF vs (11.9 ±2.12)/HPF;P <0.05],and the expression of SR-A Mrna were significantly up-regulated in diabetes group [ (5.6 ± 1.2 vs 1.5 ±0.2),P <0.05].After intervention with ACE-inhibitor,the up-regulations of the above mentioned parameters,except blood glucose concentration,were all significantly inhibited [ (3.6 ±1.4)mg/24h;(1.03±0.37)/gcs;(8.28±1.19)/HPF;3.4±0.7;P <0.05].Conclusion ACE-inhibitor might have renoprotective effects of diabetic nephropathy,it probably was associated with inhibiting the expression of SR-A gene.
5.Hepatitis B vaccine enhanced the treating effect of CIK cells on hepatitis B virus transgenic mice

Objective To observe whether hepatitis B vaccine enhance the treating effect of cyto-kine induced kill(CIK) cells on hepatitis B virus transgenic(HBV-Tg) mice. Methods The HBV-Tg mice were treated with CIK cells by peritoneal injection and hepatitis B vaccine by hypodermic injection. The HBV DNA level were tested by real-time PCR,T lymphocyte subgroup were detected by flow cytometry and the pathological diversify of hepatic tissue were observed by HE staining. Results The HBV DNA loading in peripheral blood of HBV-Tg mice decreased after CIK cells were treated and CD3~+ , CD4~+ and CD8~+ cells increased which were enhanced after CIK cells combined with hepatitis B vaccine. Conclusion Hepa-titis B vaccine enhanced the treating effect of CIK on HBV-Tg mice which may be implemented by increased the blood level of CD3~+, CD4~+ and CD8~+ cells, especially CD8~+ cells level.
6.Study on transdermal absorption of piperine in Erxiekang plaster.

OBJECTIVETo research the absorbed character of piperine in Erxiekang plaster, and piperine by transdermal absorption was determined.
METHODThe percutaneous absorption of piperine in vitro at different times was conducted by Franz osmosis and diffusion apparatus as well as high-performance liquid chromatography.
RESULTIt showed that the piperine through skins of mice gradually increased within the experiment time. After 52 h, the penetration of piperine was 78.51%, and remained basically unchanged.
CONCLUSIONThe method is reliable, and can be used for Erxiekang plaster of determination of transdermal absorption.

Objective To investigate the effect of unfractionated heparin (UFH) on the expression of heme oxygenase-1 (HO-1) in intestinal mucosa of mice with sepsis.Methods Thirty-six male C57BL/6J mice were randomly divided into sham group,cecal ligation and puncture (CLP) group and UHF group,n =12 in each group.Model of intestinal injury in sepsis was induced by CLP.In sham group,the mice were exposed without ligation of cecum.In UFH group,the mice were treated intravenously with 8 U of UFH via the tail vein half an hour before the operation and 12 hours after the surgery respectively.Six mice in each group were randomly chosen at 4 hours and 24 hours after operation to collect inferior vena venous blood samples and terminalileum tissues.The serum levels of interleukins (IL-1 β,IL-6),and tumor necrosis factor-α (TNF-α) were determined by enzyme linked immunosorbent assay (ELISA).The serum level of D-lactate was determined by colorimetry.Pathological changes of ileum tissue and Chiu score were observed after hematoxylin eosin (HE) staining.The HO-1 expression was detected immunohistochemically.Results In sham group,no significant changes in the serum levels of IL-1 β,IL-6,TNF-α and D-lactate were observed.Twenty-four hours after the operation,the structure of intestinal mucosa was basically normal without obvious pathology change and no HO-1 positive cells were found.The serum levels of IL-1 β,IL-6,TNF-α,and D-lactate in CLP group were gradually increased,and they were significantly increased as compared with sham group [IL-1 β (ng/L):40.87±2.88 vs.22.60±2.05 at 4 hours,113.73±3.96 vs.22.07±2.74 at 24 hours;IL-6 (ng/L):63.89±3.26 vs.44.89±3.38 at 4 hours,176.56±5.45 vs.45.76±4.02 at 24 hours;TNF-α (ng/L):194.62± 14.13 vs.152.05±6.22 at 4 hours,599.62± 10.20 vs.155.90± 14.18 at 24 hours;D-lactate (mmol/L):0.24± 0.02 vs.0.19 ± 0.01 at 4 hours,0.33 ± 0.04 vs.0.20 ± 0.02 at 24 hours,all P < 0.05].Twenty-four hours after the operation,edema and inflammation in ileal mucosa,intestinal villi structural damage were observed,the Chiu score was significantly higher than those in the sham group [4.5 (3.0-5.0) vs.0 (0-1.0),P < 0.05],and a small amount of HO-1 positive cells were localized in the intestinal mucosa.Compared with CLP group,the serum levels of IL-1 β,IL-6,TNF-α,and D-lactate of UFH group were significantly decreased [IL-1 β (ng/L):31.53 ± 2.90 vs.40.87 ± 2.88 at 4 hours,61.13 ± 2.80 vs.113.73 ± 3.96 at 24 hours;IL-6 (ng/L):51.16 ± 5.68 vs.63.89 ± 3.26 at 4 hours,81.16 ± 4.54 vs.176.56 ± 5.45 at 24 hours;TNF-α (ng/L):171.76± 5.60 vs.194.62± 14.13 at 4 hours,328.48 ± 10.79 vs.599.62± 10.20 at 24 hours;D-lactate (mmol/L):0.21 ±0.01 vs.0.24±0.02 at 4 hours,0.24±0.02 vs.0.33±0.04 at 24 hours,all P < 0.05].Twenty-four hours after the operation,intestinal injury was ameliorated,the Chiu score was significantly lower [1.5 (1.0-5.0) vs.4.5 (3.0-5.0),P < 0.05],and HO-1 positive cells in the intestinal mucosa was remarkably increased.Conclusion UFH can enhance the expression of HO-1 in intestinal mucosa,reduce the release of inflammatory factors,ameliorate the intestinal inflammatory response,and thus play a protective role in intestinal tissue in mice with sepsis.

Objective:
To analyze the mental health status of the Chinese Medical Team (CMT) members and its influencing factors.
Methods:
A total of 1 015 members of the CMT were recruited and surveyed by using a self-administered questionnaire and 12-item General Health Questionnaire (GHQ-12) from May 10 to June 10, 2018. Multivariable logistic regression models were used to identify risk factors for mental health problems.
Results:

OBJECTIVEConstructing a plasmid containing the shRNA of luciferase to suppress the expression of luciferase in BHK-21 cell.
METHODSA 334 bp human U6 snRNA promoter was amplified from human genomic DNA by PCR and ligated to a 21 bp reverse repeated motif of luciferase target sequence with 9 bp spacer and AAV plasmid pSNAV. The recombinant pSNAV/U6/Luc plasmid cotransfected with pMAMneoLuc or transfected luciferase cell line to detect the effect of luciferase expression separately.
RESULTSpSNAV/U6/luc suppresses the luciferase expression from pMAMneoLuc by 50% and luciferase cell line by 70%.
CONCLUSIONSThe results showed that the short hairpin RNA of luciferase can efficiently suppress its expression in BHK-21.
