International Journal of Laboratory Medicine 2015;(3):347-349
Objective To analyze the polymorphism of thymidylate synthase(TS)in colorectal cancer patients, further more,to provide guidance for personalized therapy of colorectal cancer. Methods PCR direct sequencing was used to detect the polymor-phism of TS in 252 patients with colorectal cancer. Results TS genotypes of 252 patients with colorectal cancer were detected total-ly,including 137 male and 115 female.3RG/3RC accounted for the largest proportion in both male and female(36.50% and 36.52%respectively).In female,2RC/3RC and 3RG/3RG both accounted for the second largest proportion(both 18.25%).While in fe-male,3RG/3RC accounted for the second largest proportion(26.09%).If patients were divided according to age groups,in youth patients(n=28),3RG/3RC accounted for the largest proportion(42.86%),and the second was 2RC/3RG(21.43%).In the middle aged patients(n=84),3RG/3RC(45.24%)and 2RC/3RC(16.67%)were the major genotypes. For old patients(n=115),the ma-jor genotypes were 3RG/3RC(36.52%)and 2RC/3RG(16.52%).Conclusion The polymorphism of TS are mainly 3RG/3RC in colorectal cancer patients.

Objective To evaluate the effect of crisis intervention on prevention of patients with mental disorder who had suicide ideas or attempt to commit resuicide. Methods 280 patients with mental disorder, holding suicidal ideas or attempting to suicide were randomly divided into the intervention group and control group with 140 cases in each group. The diagnosis of all patients was accorded with the CCMD-3 criteria. The cases of the control group only accepted routine treatments and cares in the hospital, and drug maintenance therapy after discharge. The cases of the intervention group accepted a comprehensive treatment of crisis intervention except the routine treatments. All cases were evaluated when they were admitted and 6 and 12 months after discharged. Results After discharged 6 months and 12 months, the social supports and treatments compliance of the intervention group were better than that of the control group(P<0.01),while suicidal ideas and the numbers of suicides were less than the control group significantly. Conclusion The crisis intervention can improve patients' social supporting and treatment compliance, eliminate their suicide ideas, and prevent the patients committing resuicide significantly.

Objective To investigate the effect of apatinib on the proliferation, apoptosis and migration of pancreatic cancer cell line AsPC-1 in vitro. Methods Pancreatic cancer AsPC-1 cells were treated by apatinib in different concentrations.Cell proliferation and apoptosis were measured by CCK-8 and flow cytometry, and the effect of apatinib on cell migration ability was observed by wound healing assay. Results In control and 10,20,30,40 and 50umol/L apatinib treatment group, the inhibitory rates of AsPC-1 cells were 0,(1.45 ±0.68)%,(16.92±0.70)%,(23.84±0.84)%,(34.35±1.55)% and (37.33± 0.81) %,respectively.Cell proliferation was obviously inhibited by apatinib as the concentration increased, and the differences were statistically significant (P < 0.05).In control and 20,40 umol/L apatinib treatment group, the apoptotic rates were (9.44 ± 0.18) %,(16.62 ± 0.19) % and (25.42 ± 0.41) %,respectively.Number of apoptotic cells was obviously increased by apatinib as the concentration increased, and the differences were statistically significant (P < 0.05).In control and 20,40 umol/L apatinib treatment group, the migration ability was (29.5 ± 0.7) %,(17.4 ± 0.9) % and (6.6 ± 0.5) %,which was greatly decreased as the concentration increased, and the differences were statistically significant (P < 0.05).Conclusions Apatinib can effectively inhibit the proliferation and migration of pancreatic cancer AsPC-1 cells and induce apoptosis.

Background and purpose: Pancreatic cancer is often diagnosed at advanced stage, therefore, chemotherapy remains the cornstone of treatment for advanced pancreatic cancer. However, no standard regimen has been established as second-line therapy for advanced pancreatic cancer. The purpose of the study was to evaluate the efifcacy and safety of albumin-bound paclitaxel plus S-1 for the treatment of advanced pancreatic cancer patients in second-line setting after the failure of gemcitabine treatment. Methods:Clinical outcomes of 19 patients with advanced pancreatic cancer were analyzed. These patients received albumin-bound paclitaxel plus S-1 as second-line therapy after the failure of gemcitabine treatment. Albumin-bound paclitaxel was administered at a dose of 125 mg/m2 over 30 minutes on day 1 and 8 of a 21-day cycle. From d1-14, all patients received oral S-1 40 mg/m2, twice daily. Results:All patients were available for evaluation. Of the 19 patients, 1 case got complete response (CR), 4 cases had partial response (PR) and 9 cases had stable disease (SD). The objective response rate (ORR) was 26.3%, the disease control rate (DCR) was 73.7%and the median progression free survival (PFS) was 5.2 months. The main toxicities include hematological toxicity, myodynia, gastrointestinal reactions, sensory neuropathy, fatigue and alopecia. Conclusion:The combination of albumin-bound paclitaxel and S-1 is effective and tolerated in the treatment of advanced pancreatic cancer patients who resistant to gemcitabine.

Wearing transfemoral prosthesis is the only way to complete daily physical activity for amputees. Motion pattern recognition is important for the control of prosthesis, especially in the recognizing swing phase and stance phase. In this paper, it is reported that surface electromyography (sEMG) signal is used in swing and stance phase recognition. sEMG signal of related muscles was sampled by Infiniti of a Canadian company. The sEMG signal was then filtered by weighted filtering window and analyzed by height permitted window. The starting time of stance phase and swing phase is determined through analyzing special muscles. The sEMG signal of rectus femoris was used in stance phase recognition and sEMG signal of tibialis anterior is used in swing phase recognition. In a certain tolerating range, the double windows theory, including weighted filtering window and height permitted window, can reach a high accuracy rate. Through experiments, the real walking consciousness of the people was reflected by sEMG signal of related muscles. Using related muscles to recognize swing and stance phase is reachable. The theory used in this paper is useful for analyzing sEMG signal and actual prosthesis control.

Objective To investigate the metabolism change of intestinal flora due to chronic hypoxia in infants. Methods Ten infants with tetralogy of fallot were considered as the chronic hypoxia group,10 healthy infants were regarded as the control group. The urine concen-tration of hippurate in the morning with fasting was detected by 1 H nuclear magnetic resonance. Results The concentration of hippurate was decreased in hypoxia group compared with the control group,(47. 15 ± 32. 88) mg/L vs (346. 698 ± 13. 555) mg/L,with significant differ-ence,P=0. 002. Conclusion Chronic hypoxia alters metabolism of intestinal flora in infants.

AIM:To observe the protective effect of Swertia Punicea on chronic liver injury induced by CCl 4. METHODS: To measure levels of serum ALT、AST and the total protein, and to observe changes of the liver index、the spleen index as well as the liver pathological change. RESULTS: Swetia Punicea obviously lowered the levels of serum ALT and AST, and increased the content of the total protein. The drug decreased the liver index, but it had no obvious effect on the spleen index. It also improved the liver fibrosis. CONCLUSION: Swertia Punicea can protect liver and decrease enzymatic activity, and can retard or ease up the formation of the liver fibrosis.
