1.Consideration on non-clinical safety evaluation of traditional Chinese medicine

Safety,efficacy and quality control are the three essential elements for drugs,including traditional Chinese medicine(TCM). Currently,rapid development of life sciences,medicine and phar?macy provides new prospects for TCM. Considering the long history of TCM application,modern phar? macology and toxicology, global standards and expectations on the safety of TCM, how to evaluate the safety of TCM scientifically has become a key to the research and development of TCM. This review aims to discuss current technical requirements of non-clinical safety study of TCM as well as the challenges.
2.Influence of central aortic and pulse pressure in coronary Artery Disease

Objective To investigate the influence of central aortic pressure and pulse pressure in coronary Artery. Methods The research involed 173 patients who were suspected coronary heart disease (CHD) in the Fifth People′s Hospital of Chengdu city. The patients were divided into non-coronary disease and coronary disease groups. Patients were divided into one- vessel, two- vessels and three-vessels in coronary disease groups. The data were analyzed by using spass 17.0 software. Results SBP-C,DBP-C and PP-C had statistical significance compared with non-coronary disease (P < 0.05).SBP-C and PP-C had significant difference between single-branch and multiple-branch group(P < 0.05).Logistic multivariate analysis indicated that SBP-C [P = 0.019,OR = 1.207, 95%CI(1.032,1.412)]and PP-C [P = 0.023, OR = 1.166,95%CI (1.021,1.332)] had significant correlation. Conclusion Compared with non-coronary, SBP-C and PP-C were higher, while DBP-C was lower. The higher SBP-C and PP-C are,the worse coronary artery is. SBP-C and PP-C are predispose risk factors for multiple-vessels coronary disease.
3.Recognition of walking stance phase and swing phase based on moving window.

Wearing transfemoral prosthesis is the only way to complete daily physical activity for amputees. Motion pattern recognition is important for the control of prosthesis, especially in the recognizing swing phase and stance phase. In this paper, it is reported that surface electromyography (sEMG) signal is used in swing and stance phase recognition. sEMG signal of related muscles was sampled by Infiniti of a Canadian company. The sEMG signal was then filtered by weighted filtering window and analyzed by height permitted window. The starting time of stance phase and swing phase is determined through analyzing special muscles. The sEMG signal of rectus femoris was used in stance phase recognition and sEMG signal of tibialis anterior is used in swing phase recognition. In a certain tolerating range, the double windows theory, including weighted filtering window and height permitted window, can reach a high accuracy rate. Through experiments, the real walking consciousness of the people was reflected by sEMG signal of related muscles. Using related muscles to recognize swing and stance phase is reachable. The theory used in this paper is useful for analyzing sEMG signal and actual prosthesis control.

4.Comparative Study on Pharmacokinetics of Cephradine in Diabetic and Normal Rats

Objective To investigate effects of diabetes mellitus (DM) on pharmacokinetics (PK) of cephradine (CED).Methods DM model was induced by iv.alloxan 60 mg·kg-1.A reversed phase HPLC internal standard method was developed for measurement of CED plasma concentration.After blood collection,rats were sacrificed to collect kidneys for calculating kidney index(KW/BW).DM and normal control (CTL) rats were randomly assigned to receive iv.or ig.CED at a dose of 180 or 90 mg·kg-1.The 3p97 program was used to calculate PK parameters.Results The developed HPLC method was validated to have high specificity,precision,recovery and good storage stability,and met requirements for PK study of CED.The CED in rats of both DM and CTL groups showed the iv.two-compartment PK and ig.one-compartment PK and followed the first-order kinetics.Following iv.dosing,a remarkably decreased t1/2β and MRT,increased CLt were evident in DM group as compared with CTL group (P < 0.05).After ig dosing,a significant decrease in t1/2k and t a remarkable increase in CLt and Cm=were observed for DM group as compared with CTL group (P < 0.05).The DM rats showed a trend of decreased t1/2ka vs CTL rats.There was no significant difference in the oral bioavailability between the two groups (P > 0.05).KW and KW/BW in DM group were increased remarkably compared with CTL group (P < 0.05).Conclusion The DM vs CTL results in faster absorption and elimination of CED in rats,but does not have significantly affect in oral bioavailability.The compensatory hypertrophy and hyperfunction of early-stage diabetic kidneys may constitute one of causes of quick elimination of CED in rats with DM.
5.Early effect of tonsillotomy on children′s snoring with tonsil hypertrophy

Objective To explore short-term effect of tonsillotomy on children′s obstructive sleep apnea hypopnea syndrome (OSAHS) with tonsil hypertrophy by Ellman double-frequency radio-frequency machine .Methods Totally 30 cases children′s OS-AHS with tonsil hypertrophy were had tonsillotomy by Ellman double-frequency radio-frequency machine .They were compared with 30 cases children′s OSAHS with tonsil hypertrophy by traditional tonsillectomy .Short-term effect of tonsillotomy were ana-lyzed by wound bleeding ,operation time ,postoperative pain ,difficulty eating ,PSG testing ,immune function ,growth and develop-ment .Results Postoperative symptom in 1 weeks and 6 months ,compared with the traditional tonsillectomy ,30 cases of OSAHS patients were found that there were less bleeding ,shorter operative time ,mild postoperative pain ,no difficulty in feeding ,improving sleep-disordered breathing ,no decline in immune function ,good growth and development .Conclusion Short-term effect of tonsillot-omy on children′s OSAHS with tonsil hypertrophy is good by Ellman double-frequency radio-frequency machine ,it is a minimally invasive ,safe and effective surgical method children OSA HS .
6.Evidence for Central Neural Regulation of Pacing Strategies

Objective Based on the model of exercise induced central fatigue,we attempt to testify that the whole power output in time-counted sporting events was regulated by the central nervous system(CNS).Methods Eighteen national level rowers participated in the experiment.Each participant completed two successive 6 minutes maximal rowing exercise with an interval of 15 minutes.The power output(PO),heart rate(HR),blood lactate(BL),RPE and EMG were measured.Results It was demonstrated that the pacing strategies adopted by the rowers in the 2 successive 6-minute rowing tests were remarkably similar,and the rowers'PO and iEMG activities in the two tests were highly correlated,especially in the last minute of exercise.Conclusion It is concluded that the hypothesis of pacing strategy is controlled by CNS.
7.Median effective target plasma concentration of propofol inhibiting response to laryngeal mask airway insertion when combined with dexmedetomidine

Objective To determine the median effective target plasma concentration (EC50) of propofol inhibiting the response to laryngeal mask airway (LMA) insertion when combined with dexmedetomidine.Methods ASA Ⅰ or Ⅱ patients of both sexes,aged 20-60 yr,with body mass index 20-25 kg/m2,scheduled for surgeries under general anesthesia,were studied.EC50 of propofol was determined by modified Dixon' s up-and-down sequential experiment.After dexmedetomidine 1.0 μg/kg was infused over 10 min,propofol was infused by targetcontrolled infusion.The initial target plasma concentration of propofol was set at 3.0 μg/ml.LMA was inserted when the target effect-site concentration of propofol and target plasma concentration of propofol reached the balance and BIS value was 50-60.Each time the target concentration increased/decreased by 0.2 μg/ml according to the occurrence of the response to LMA insertion.The response to LMA insertion was defined as the occurrence of coughing,body movement,laryngospasm or systemic voluntary movement.EC50 and 95 % confidence interval (CI)of propofol for inhibition of the response to LMA insertion were calculated.Results The EC50 of propofol required for inhibition of the response to LMA insertion was 2.351 (95% Cl 1.737-2.600) μg/ml when combined with dexmedetomidine 1.0 μg/kg.Conclusion The EC50 of propofol inhibiting the response to LMA insertion is 2.351 μg/ml when combined with dexmedetomidine.
8.Relationship between sternum protection and bone marrow suppression in postoperative radiotherapy for esophageal carcinoma

Objective To evaluate the relationship between sternum protection and bone marrow suppression in postoperative radiotherapy for esophageal carcinoma. Methods Total of 98 postoperative patients with thoracic esophageal carcinoma were randomly divided into experimental group (52 cases) and control group (46 cases). All patients were given intensity-modulated radiotherapy (IMRT), with the dose of 50-50.4 Gy. The patients in experimental group were irradiated by 6 fields (4-fields in front, 2-fields behind) which were crossed to avoid direct exposure to the sternum. The patients in control group were irradiated by 5 fields (3-fields in front, 2-fields behind) with front-middle of the field passing through the sternum. Concurrently all patients received 2 cycles of cisplatin chemotherapy. Results Dmean, V20 and V30 of the sternum in the experimental group were (20.21 ±3.60) Gy, (40.78 ±7.19) % and (33.78 ±9.44) %, which were lower than those in the control group [(30.91±5.21) Gy, (81.01±4.81) %, (51.60±6.84) %], respectively (P<0.05). However, the volume and dose distribution of lung, spinal cord and heart were similar between the two groups (P> 0.05). Both the incidence rates of bone marrow suppression at 14th day and 35th day after radiotherapy were significantly higher in the control group (52.2%, 73.9%) than those in the experimental group (28.8 %, 50.0 %) (P< 0.05), and the incidence rate of bone marrow suppression at 7th day after radiotherapy was similar between the two groups. Conclusion Protecting and sketching for sternum in postoperative radiotherapy for esophageal carcinoma can reduce the incidence of bone marrow suppression effectively, which would not increase the radiation dose in the lung, heart and spinal cord.
9.Binocular rivalry estimated objectively under face awareness and fusiform correlation analysis

Objective To study binocular rivalry (BR)objectively and the correlation between fusiform face area (FFA)and visual cortex.Methods Six subjects participated in this study,with one eye presented a normal face expres-sion picture flickered at 8.57 Hz,while the other presented a fearful face flickered at 12 Hz or 15 Hz,respectively.Electro-encephalogram(EEG)was recorded during this process.Steady state visual evoked potential(SSVEP)evoked by two flick-ering rates was analyzed by time-frequency analysis of short time fourier transformation(STFT).The time index of BR was estimated and the correlation coefficient between FFA and visual cortex compared.Results The total average time was (411.6 ±73.8)ms for the left eye and (547.6 ±126.7)ms for the right eye.The switch rate of the two groups was not different,but the left FFA was more sensitive than the right FFA in process of the fearful face.Neither side of FFA had any frequency preference to the flickered fearful face.Conclusion SSVEP can be used as a frequency tag of BR or as a tool to evaluate visual sensation under BR objectively.SSVEP combined with BR can be used in research of neural mechanisms of visual awareness.
10.Application of temperature sensitive yeast models with definite target in the screening of potential human Pin1 inhibitors.

This study is to explore new lead compounds by inhibition of Pin1 for anticancer therapy using temperature sensitive mutants. As Pin1 is conserved from yeast to human, we established a high-throughput screening method for Pin1 inhibitors, which employed yeast assay. This method led to the identification of one potent hits, 8-11. In vitro, 8-11 inhibited purified Pin1 enzyme activity with IC50 of (10.40 +/- 1.68) micromol x L(-1), induced G1 phase arrest and apoptosis, showed inhibitory effects on a series of cancer cell proliferation, reduced Cyclin D1 expression, was defined as reciprocally matched for protein-ligand complex in virtual docking analysis and reduced cell migration ability. In vivo, we could observe reduction of tumor volume after treatment with 8-11 in xenograft mice compared with vehicle DMSO treatment. Altogether, these results provide for the first time the involvement of 8-11 in the anticancer activity against Pin1.
