1.Clinical Observations on Acupuncture Point Injection of Mouse Nerve Growth Factor for the Treatment of Sciatic Nerve Injury

ObjectiveTo investigate the clinical efficacy of points injection of mouse nerve growth factor in treating sciatic nerve injury.MethodAcupuncture point injection of injectable mouse nerve growth factorwas performed in 60 patients with sciatic nerve injury. Points Huantiao(GB30)and Zusanli(ST36)were selected. Treatment was given once daily, for a total of 30 times. Sensory and motor functions were assessed (MS and SS) and electromyography (EMG) was performed before and after treatment to compare motor nerve conduction velocities (MCV) and sensory nerve conduction velocities (SCV) in the injured nerves and EMG changes in the musclescontrolled by the injured nerves.ResultAfter treatment, nerve function was restored to more than S3M3, with an efficacy rate of 81.7%, in 49 patients. Neuro-electrophysiological study showed that regenerative potentials occurred, accounting for 71.7%,in43patients. Denervated potentials were fewer after treatment than before; there was a significant difference (P<0.05). The mean MCV value increased after treatment compared with before; there was a significant difference (P<0.05). ConclusionAcupuncture point injection of mouse nerve growth factor can markedly improve sensory and motor functions in the regions controlled by the injured sciatic nerve. It provides an effective way to promote the repair of sciatic nerves and the reconstruction of limb function after the nerves are injured.
2.Detection and analysis of stress urinary incontinence patients after non -urethral suspension surgery in tension-free vaginal tape-obturator technique

Objective To explore the influence of non -urethral suspension surgery in tension -free vaginal tape -obturator technique on patients with stress urinary incontinence.Methods 80 patients with stress urinary incontinence were researched,they were divided randomly into observation group (40 cases) and control group (40 cases).The control group was treated by conventional drug therapy +functional exercise,while the observation group was treated by tension -free vaginal tape -obturator technique on the basis of the control group.The urethral dynamics before and after two months treatment were compared,and the clinical therapeutic effect and recurrence rate were compared.Results After treatment,the voiding time of the observation group was longer than that of the control group[(25.78 ±4.15)s vs.(21.42 ±3.41)s,t =3.465,P <0.05],the mean urinary flow rate was lower than the control group[(6.41 ±1.14)mL/s vs.(9.84 ±1.79)mL/s,t =10.222,P <0.05].The functional urethral length, maximum urethral closure pressure of the observation group were higher than those of the control group[(38.94 ± 8.41)mm vs.(33.47 ±5.38)mm,(36.94 ±9.21)cmH2 O vs.(31.73 ±7.19)cmH2 O,t =3.465,2.820,all P <0.05].The total effective rate of the observation group was higher than that of the control group(72.5% vs.70.0%,χ2 =11.113,P <0.05).After six months of follow -up,the recurrence rate of the observation group was lower than that of the control group(0.0% vs.10.0%,χ2 =4.210,P <0.05 ).Conclusion In patients with stress urinary incontinence,tension -free vaginal tape -obturator technique can significantly improve the urethral dynamics,promote recovery of illness,it is worthy of application and promotion.
3.Analysis of curative effect of Metronidazole Suppositories, conjugated estrogen, estrogen ointment and psychological intervention on senile vaginitis

Objective To study and analyze the efficacy of Metronidazole Suppositories, conjugated estrogen, estrogen ointment and psychological intervention in the treatment of senile vaginitis. Methods 100 cases of senile vaginitis patients in our hospital from February 2015 to August 2016 were selected as the research object. They were randomly divided into the control group and the experimental group, each group had 50 cases. The control group was treated with the treatment of patients in the experimental group of Metronidazole Suppositories, Metronidazole Suppositories, conjugated estrogen ointment in the treatment of psychological intervention on the mental status of patients and patients, strengthen communication and exchanges, increase confidence in the treatment and the treatment compliance of patients. The therapeutic effects of the experimental group and the control group were compared and analyzed. Result After the corresponding treatment, 3 patients in the experimental group were invalid. In the control group, 10 patients were ineffective. The effective rate of the experimental group was 96.0%, which was significantly higher than that of the control group (80.0%), with statistical difference (P<0.05). The incidence of adverse reactions in the experimental group and the control group was 2% and 4% respectively, and there was no significant difference. Conclusion Metronidazole Suppositories, combined estrogen ointment and psychological intervention in the treatment of senile vaginitis clinical effect is good, high treatment efficiency, low incidence rate of adverse reaction, has clinical significance.
4.Comparison of effectiveness and safety between Twisted File technique and ProTaper Universal rotary full sequence based on micro-computed tomography

Objective:To evaluate the efficacy and security of two type of rotary nickel titanium system (Twisted File and ProTaper Universal)for root canal preparation based on micro-computed tomography (micro-CT).Methods:Twenty extracted molars (including 62 canals)were divided into two experimen-tal groups and were respectively instrumented using Twisted File rotary nickel titanium system (TF)and ProTaper Universal rotary nickel titanium system (PU)to #25/0.08 following recommended protocol. Time for root canal instrumentation (accumulation of time for every single file)was recorded.The 0-3 mm root surface from apex was observed under an optical stereomicroscope at 25 ×magnification.The presence of crack line was noted.The root canals were scanned with micro-CT before and after root canal preparation.Three-dimensional shape images of canals were reconstructed,calculated and evaluated. The amount of canal central transportation of the two groups was calculated and compared.Results:The shorter preparation time [(0.53 ±0.1 4)min]was observed in TF group,while the preparation time of PU group was (2.06 ±0.39)min (P<0.05).In mid-root level,TF group shaping resulted in less ca-nal center transportation than PU group [(0.070 ±0.056)mm vs.(0.097 ±0.084)mm,P<0.05]. No instrument separation was observed in both the groups.Cracks were not found in both the groups ei-ther based in micro-CT images or observation under an optical stereomicroscope at 25 × magnification. Conclusion:Compared with ProTaper Universal,Twisted File took less time in root canal preparation and exhibited better shaping ability,and less canal transportation.
5.Construction of scFv-psm-ETA recombinant immunotoxin and its antitumor effect on human prostate cancer cells in vitro and in vivo

Objective To evaluate the construction of scFv-psm-ETA immunotoxin and to study its antitumor effect on human prostate cancer cells. Methods The genes encoding the heavy and light chain Fv regions of the monoclonal murine antibody Ed-5,which recognizes prostate specific membrane antigen on the surface of human prostate cell,were cloned by PCR technique and were used to generate single-chain immunotoxins containing Pseudomonas exotoxin A(ETA).The light and heavy chains were connected by a flexible linker to form a single-chain antigen-binding protein,which was in turn fused to truncated forms of ETA.The resulting plasmid,pSW200-psm,was transformed intoE.coli CC118 and the recombinant immunotoxins,scFv-psm-ETA,were expressed and purified to near homogeneity.The cell killing activity and in vivo antitumor activity of scFv-psm-ETA were tested using human prostate cancer cell line LNCaP. Results scFv-psm-ETA was shown to be cytotoxic specifically to 80% of LNCaP cells,which expressed the PSM on their surface at a dose up to 100 ng/ml.In athymic nude mice, administration of the scFv-psm-ETA inhibited the growth of human prostate cancer cells LNCaP.The size of the tumors was 153 mm3 and 272 mm3,respectively, in treatment group and control group (P
6.Humeral head replacement for treatment of complex proximal humerus fractures

Objective To discuss the indication, surgical technique and curative effect of humeral head replacement in treatment of complex proximal humerus fracture in old patients. Methods From 2000 to 2006, 23 patients with complex proximal humerus fractures were treated with humeral head replacement and fixated with bone cement in old patients. Rehabilitation training was performed after operation.Results Of all, 19 patients were followed up for average 1.8 years, which showed no infection, nerve injury or periprosthetic fracture except for one patient with semiluxation. NEER score system was used to evaluate the function joint prosthesis. Mean NEER score was 82.1 points, including 80-89 points ( satisfactory) in 17 patients and 70-79 points (unsatisfactory) in 2, with satisfaction rate of 89%. No patient needed revision. Conclusion With strict indication, active reconstruction of rotating cuff and stable structure of the joint, planned postoperative rehabilitation training, the humeral head replacement is a good method for treating complex proximal humerus fractures in the old patients.
7.Effects of NaokangⅡon cognitive function and neurogenesis in rats with cerebral ischemia WANG

Objective ToobservetheeffectsofNaokangⅡ,atraditionalChinesemedicineon spatial learning and memory abilities and hippocampal neurogenesis in rats with cerebral ischemia. Methods Atotalof54SDratsweredividedinto6groupsaccordingtothedigitalrandommethod:a control,a sham operation,a model,a low-dose,a medium-dose,and a high-dose NaokangⅡ group (the low-,medium-,and high-dose groups were 5,10,and 20 times of adults (n=9 in each group). A rat model was induced by ligate the bilateral common carotid arteries permanently. The bilateral common carotid arteries were only isolated but not ligated in rats of the sham operation group. The rats in the control group were not given any treatment. After modeling,the rats in the treatment groups were treated with NaokangⅡ. The rats in the control,sham operation,and model groups were given equal volume of isotonic saline (2 ml). Morris water maze test and space search experiment were used to evaluate the learning and memory abilities in rats 9 weeks after modeling. Immunofluorescence staining and immunohistochemical methods were used to observe proliferation and differentiation of neural stem cells (NSCs). Western blot was used to detect the expression of autophagy related protein Beclin-1 and LC-3Ⅱ. Results (1 )The escape latencies of the model group from 1 to 5 days were 100 ± 13,98 ± 13,82 ± 14,72 ± 15,and 46 ± 7 s,respectively. They were significantly longer than those of the sham operation group and the control group (all P<0. 05). The search distances were 1239 ± 127,1011 ± 122,959 ± 123,553 ± 66,and 407 ± 29 cm,respectively. They were significantly increased as compared with those of the sham operation group and control group (all P<0. 01). Both the escape latencies and search distances of the rats in the Naokang Ⅱ low-,medium-,and high-dose groups were significantly shorter than those of the model group (P<0. 05). The escape latencies on the fifth day were 20 ± 6,19 ± 8,and 15 ± 6 s,respectively,and the search distances were 234 ± 38, 297 ± 23,and 99 ± 22 cm,respectively. Nine weeks after modeling,the quadrant search time on the original platform in the rats of the model group was 30 ± 9 s,it was significantly shorter than that of the sham operation group (P<0. 01). The quadrant search time on the original platform of the Naokang Ⅱ low-, medium-,and high-dose groups were 34 ± 6,38 ± 8,and 40 ± 10 s,respectively. They were significantly longer than those of the model group (P <0. 01). (2)Nine weeks after modeling,the results of immunofluorescence and immunohistochemistry showed that the number of BrdU-positive cells and the Nestin positive cells in hippocampal dentate gyrus of the model group were higher than those of the control group and the sham operation group (P<0. 05). The number of BrdU-positive cells and Nestin positive cells of Naokang Ⅱ each dose group were significantly higher than those of the model group (P<0. 05). The expression of LC-3Ⅱand Beclin-1 in the model group was significantly higher than those in the control and sham operation groups (P<0. 01). The expression of LC-3Ⅱ in the medium- and high-dose groups was 143 ± 9 and 129 ± 9 respectively,and the expression of Beclin-1 was 139 ± 10 and 124 ± 7 respectively, andtheyweresignificantlylowerthanthemodelgroup(P<0.01).Conclusion NaokangⅡcanimprove the cognitive function in rats with cerebral ischemia,promote neurogenesis in the dentate gyrus,and inhibit autophagic cell death.
8.Antitussive, Expectorant and Antiasthmatic Effects of Tankeqing Capsule

Results of experiments such as ammonia steaming test in mice and citric acid test in guinea pigs, phenol red secretion test in mice and capillary expectorant test in rats, in-vivo and in-vitro antiasthmatic tests in guinea pigs proved that Tankeqing capsule had good antitussive, expectorant and antiasthmatic effects and a significant time-effect relationship was showed. The antitussive effect and expectorant effect arrived to the peak in 1-6h after oral administration, and the antiashmatic effect in about 1h.

Objective To analyse the relationship of blood deficiency syndrome’s principal components of chronic fatigue syndrome (CFS) and mental state and quality of life. Methods 23 cases of CFS blood deficiency syndrome were collected to pick up principal components, and combined with mental state (SCL-90) and quality of life (SF-36). Then, the relationship was analyzed. Results There was significant relationship between CFS blood syndrome principal component 1 and GH, VT and BP in SF-36. And there was significant relationship between CFS blood syndrome principal component 2 and patients’ somatization, obsessive compulsive, interpersonal sensitive, depression, anxiety, hostility and paranoid in SCL-90, and between principal component 1 and somatization. Conclusion There were two principal components in CFS blood deficiency syndrome, one influenced patients’ quality of life, the other influenced their mental state.
