1.Effect of curcumin on sterol regulatory report system in HEK-293

Aim To investigate the lipid-lowering mechanism of Chinese Medicine curcumin.A sterol regulatory report system of LDLR gene expression in HEK-293 cell line was established.Method A green fluorescent protein(GFP) gene was constructed downstream of the sterol regulatory element-1(SRE-1) and the constructed plasmid was introduced into HEK-293 cell line.This SRE-GFP transfected 293 cells were treated with different concentration of curcumin for 2 days.The expression of GFP in the cells was measured by a fluorescence microscope and Flow Cytometry.Results The expression of GFP in SRE-GFP transfected 293 cells was obviously induced by curcumin.Conclusion Curcumin may positively affect the expression of LDLR gene by its influence on SRE-1.
2.Clinical effects of dimple-plasty under needle guidance

Objective To explore the methods and clinical effects of dimple plasty with syringe needle guiding.Methods From February 2008 to May 2012,36 dimple plasties in 21 patients were performed with using a syringe needle guiding the absorbable suture,including 15 cases in bilateral side and 6 cases in unilateral side.There were 4 cases in unilaterally right side and 2 cases in unilaterally left side.Two patients with inborn dimple in unilateral side received the dimple plasty in the other side for facial symmetry.Results 34 dimples in 21 patients were satisfied with the results.Two dimples were re-operated after three months because of disappearance.There were scleromas in 2 cases,which was disappeared by hot compress in three months.There were no hemotoma and infection in 21 patients.Conclusions The procedure of dimple plasty under syringe neddle guiding is safe,reusable and effective.It is a minimally invasive surgery and the patients are satisfied with the dynamic results after operation.
3.The effects of orthodontic force on the expression of IL-6 and the alveolar bone remodeling in rats

Objective; To investigate the expression and distribution of IL-6 in periodontal tissues and the change of the alveolar bone of rats during orthodontic tooth movement, and to study the effects of orthodontic force on the periodontal tissue remodeling. Methods: SO gram orthodontic force was loaded on the left first maxillary molars of 25 rats in experimental group. Immunohistochem-istry and histomorphometric analysis were performed to measure the expression of IL-6 and the loss of alveolar bone at 0, 1, 3, 5, 7 " and 10 days after the application of orthodontic force. Results; The expression of IL-6 was observed to reach maximum level on day 3 and to decline thereafter in experimental group. No obvious alveolar bone loss was detected in the mesial side of the first molars. Conclusion; Although orthodontic force can evoke the local inflammatory response of periodontal tissue and the expression of pro-inflammation cytokines such as IL-6, it can not cause severe periodontal destruction and alveolar bone loss.
4.HLA-A, -B, and -DRB1 gene polymorphism in the Miao ethnic group of Hunan, China.

OBJECTIVE:
To investigate HLA-A, -B, and -DRB1 gene polymorphism in the Miao ethnic group in Hunan, China.
METHODS:
PCR-sequence specific oligonucleotide probes (SSO) reverse flow chip method was used to type HLA-A, -B, and -DRB1 genes of 154 unrelated healthy Miao ethnic individuals in Hunan. The allele and haplotype frequencies were calculated and compared with other populations in China.
RESULTS:
A total of 10 HLA-A, 25 HLA-B, and 13 HLA-DRB1 alleles were observed in the population. The higher frequency alleles included A 11(38.96%), A 02(27.27%), A 24(18.83%), B 40(60)(17.53%), B 46(13.96%), B 15(75)(11.69%), DRB1 09(15.26%), DRB1 12(15.26%), DRB1 15(15.26%), and DRB1 04(12.66%). The most frequent haplotypes were A11-B60(9.60%), A2-B46(9.27%), A11-B75(9.22%), B75-DR12(6.31%), A11-DR12(9.62%), A11-DR4(9.07%), A11-DR15(6.69%), A11-B75-DR12(5.43%), A11-B60-DR4(4.24%), and A2-B46-DR9(3.71%). Compared with other populations in China, HLA gene polymorphism of Hunan Miao population was close to that of southern population.
CONCLUSION
HLA loci are highly polymorphic in the Miao population of Hunan, and their distribution has the character of South China population.

5.Preparation and clinical application of the monoclonal antibodies against the human O6 -methylguanine-DNA methyltransferase

Objectives:To establish hybridomas secreting monoclonal antibodies(McAbs) against O6-methylguanine-DNA methyltransferase(MGMT) and to observe the relationship between MGMT expression and clinical responses to ACNU and BCNU in human brain tumors.Methods:The hybridomas were established by cell fusion.MGMT expression in 60 glioma specimens was detected by means of immunohistochemical assay.Results: Seven hybridomas secreting McAbs against MGMT were obtained.Thirty tumor specimens had no detectable or low level of MGMT expression(Mer-), while 30 specimens had high level of MGMT expression(Mer+). The Mer- patients showed more sensitive to ACNU and BCNU than the Mer+ patients.Conclusions: The high specific hybridomas secreting monoclonal antibodies(McAbs) against MGMT were established.The preliminary study indicated that MGMT negative tumors were sensitive to ACNU and BCNU, whereas MGMT positive ones were more resistant to nitrosourea drugs.
6.Evaluation of sphincter damage degree with anal endosonography for children with fecal incontinence.

OBJECTIVETo evaluate the application of anal endosonography in the morphology of internal anal sphincter (IAS), external anal sphincter (EAS) and puborectalis (PR) in order to provide necessary information for analysis of the etiology of fecal incontinence and formulation of the therapeutic schedule.
METHODSFrom December 2010 to November 2012, 18 children of anorectal malformation (n=14) or Hirschsprung's disease(n=4) with fecal incontinence received anal endosonography. The morphology of IAS, EAS and PR was observed. The damage of anal sphincter was classified according to Starck criteria. Anorectal mamometry and anal clinical score were measured simultaneously. Spearman analysis was performed to examine the correlation of anal sphincter damage with anorectal mamometric score and anal clinical score.
RESULTSAccording to Starck criteria, anal sphincter damage was small in 11 children, moderate in 6, and severe in 1. PR damage was found in 4 cases. Starck score was positively correlated with manometric score(P<0.05), while not correlated with anal clinical score(P>0.05).
CONCLUSIONSAnal endosonography can clearly display the morphology of IAS, EAS and PR, and their integrity and damage degree. It is a very valuable technique to evaluate the anal sphincter of the children with fecal incontinence, which however can not reflect the function of anal sphincter and anal continence thoroughly.

7.Construction and expression of recombinant adeno-associated virus expressing brain-derived neurotrophic factor.

A fusion gene called Ig-BDNF, in which brain-derived neurotrophic factor cDNA fused to the 3' end of signal peptide of Ig coding sequence, was constructed by PCR, digested and subcloned into shuttle plasmid pSNAV to obtain a recombinant plasmid pSNAV-Ig-BDNF. Then the plasmid encoding fusion protein was transfected into 293 cell lines and the stably transfected clones were selected with neomycin. AAV1 containing Ig-BDNF fusion gene vectors were obtained by super-infection by Herpes virus. The resultant adeno-associated virus vectors AAV-Ig-BDNF were confirmed by PCR, Western blotting and a sandwich enzyme-linked immunosorbent assay (ELISA) after infection of 293 cell lines. The results indicated that AAV-Ig-BDNF contained the target gene, and infected cells and produced the fusion protein into the supernatant. The content of BDNF in medium per 5x104 cells over a 24 h incubation period reached 1000 pg/mL. With the help of non-replicative adenovirus during AAV-Ig-BDNF infection, the expression of BDNF increased 7-8 fold, and the enhancement of BDNF gene expression was observed in a concentration-dependent manner. These results suggested that a functional AAV-Ig-BDNF was successfully constructed and it offers basis for further study for gene therapy of neural degeneration diseases.

8.Research progress on nursing quality evaluation indicators of precontrol of venous thromboembolism

Venous thromboembolism is a common surgical complication. It is highly regarded because of its tendency to cause fatal pulmonary embolism, so its nursing quality is of great significance to the outcome of patients. This paper summarizes the structural, process and outcome indicators of nursing quality evaluation in the precontrol of venous thromboembolism, aiming to provide reference for the formulation of nursing quality evaluation indicators in precontrol of venous thromboembolism suitable for our national conditions.
9.Effects of orbital septum fat redistribution on multiple malformation of upper eyelid after blepharoplasty

Objective The multiple malformation of upper eyelid,including the excessive width of fold line,sunken and/or multiple folded upper eyelid are the common complications after blepharoplasty.The aim of this study was to investigate the clinical effect of orbital septum fat redistribution on correcting these deformities.Methods From September 2015 to September 2017,38 patients with multiple malformations of upper eyelid were treated.The incision of upper eyelid and the excessive skin was designed and resected.After completely relieving the scar adhesion zones,we set the lateral orbital septum fat free and transposed it to the inner side of orbital septum or superior border of tarsus with suturing fixation.Then we routinely completed the double eyelid operation.Results Thirty-eight patients with multiple malformations of upper eyelid were treated successfully.The patients were followed up for 6 months to one year,and the results were totally satisfactory.The operation was not performed again.Conclusions The orbital septum fat redistribution can successfully correct the multi ple malformation of upper eyelid.It should become a regular procedure in blepharoplasty.
10.The significance of PCT and SAA detection in early childhood bacterial infection in preschool children

Objective To investigate the clinical significance of procalcitonin(PCT)and serum amyloid A (SAA)in early bacterial infection in preschool children.Methods 67 children with bacterial infection(bacte-rial infection group),62 children with viral infection(viral infection group)and 60 healthy children(healthy control group)were enrolled in this study,latex enhanced turbidimetric immunoassay was used to detect SAA and turbidimetric immunoassay was used to detect the level of PCT,the levels of SAA and PCT and the posi-tive rate were compared among all groups,and the sensitivity,specificity and positive predictive value,negative predictive value and Youden index of SAA and PCT levels.Results The levels of SAA and PCT in the bacte-rial infection group before treatment were significantly higher than those in the viral infection group and the healthy control group(P<0.05),7 days after treatment,the levels of SAA and PCT decreased significantly (P<0.05);the level of PCT in viral infection group was not significantly different from that in healthy control group(P>0.05),but the positive rate of SAA was significantly different from that of the healthy control group(P<0.05).The sensitivity,specificity,positive predictive value and negative predictive value of PCT for early bacterial infection in preschool children were 92.5%,93.5%,93.9%,92.1%,respectively,of the SAA values were 97.0%,59.7%,72.2% and 94.9%,respectively,there were significant differences between the two groups in specificity and positive predictive value(P<0.05).Conclusion Detection of SAA and PCT lev-els was helpful for early diagnosis,differential diagnosis and prognosis of bacterial infection in preschool chil-dren.
