1.Suitable vehicle for gene transfection into human adipose derived adult stem cells: pEGFP-N1, Ad5-EGFP and rAAV-2/1-EGFP

AIM: To observe the enhanced green fluorescent protein (EGFP) gene expression and cytotoxicity in human adipose derived adult stem cells (hADSCs) by pEGFP-N1, Ad5-EGFP and rAAV-2/1-EGFP, and investigate the suitable gene-transferred vehicle. METHODS: The experiment was conducted at Chinese National Human Genome Center and the Third Hospital of Peking University from January to July 2006. ①After the patients and their relative were informed consent, the subcutaneous adipose tissue was obtained from the patients undergoing routine total hip joint replacement in Department of Orthopaedics, Third Hospital of Peking University. pEGFP-N1 was provided by Clotech Company, Ad5-EGFP and rAAV-2/1-EGFP by Vector Gene Technology Company. ②hADSCs were cultured in vitro after isolated from the adipose tissue after dissected and digested with type I collagenase. ③hADSCs of passage 3 were infected with pEGFP-N1, Ad5-EGFP and rAAV-2/1-EGFP and the EGFP expression and the cell toxicity were observed. ④Twenty-four hours after being transfected, 5?104 cells were reseeded in a 24-well plate and the solution was changed three times every week. The growth curves of each group were drawn. Normal non-transfected cells served as control. The influence of different transfection ways on the growth of hADSCs was observed. RESULTS: ①Comparison of transfected efficiency with different ways: pEGFP-N1 transfection showed a higher cytotoxicity and lower efficiency of 10.5%; Ad5-EGFP could efficiently transfect hADSCs (multiplicity of infection=5?102, 82.5%); when MOI was 0.05); however, the growth capability of hADSCs was decreased significantly in the pEGFP-N1 transfection group compared with the control, and the differences were significant at day 3-10 after transfection (P

OBJECTIVE To study the prevention strategy of community-acquired pneumonia(CAP) for retired old cadres of PLA.METHODS We adopted the first and second prevention strategy of disease to deduce the frequency for CAP.RESULTS The old cadres were divided into common prevention group and important prevention group by evaluating their body condition.The common prevention group was taken the first prevention strategy:healthy education,inhabit surroundings improvement and proper exercises.The second prevention was carried out to personnel who have chronic disease of respiratory organs which included eliminating the risk factors and making efforts to find, diagnose and treat infection as early as possible.CONCLUSIONS The first and second prevention is an effective working mode which improves the level of CAP prevention.
3.Experiment Studies of the Eye Movement of Chinese Children with Reading Disorder when Reading Chinese Article

4.Studies of the Executive Function Profiles in Children with Attention Deficit Hyperactivity Disorder

Objectives: To study the executive function (EF) profiles in children with ADHD. Methods: 1.We used some neuropsychological tests,including Stroop test, visual and auditory Go/No-Go tasks, digits reciting inverse, delayed spacial position memory span test, tower of London (TOL), tail making test (TMT),to evaluate the capacity of response inhibition, phonological working memory, visual-spacial working memory, planning and set-shifting of subjects. Results: 1.The ADHD children spent more time to accomplish color naming when the word color was inconsistent with the word meaning than normal control (NC) in Stroop test(p﹤0.05). 2.The ADHD children made more errors than NC in both visual and auditory Go/No-Go test(p﹤0.01). 3.The scores of digits reciting inverse was lower in ADHD than NC(p﹤0.01). 4.The representation of ADHD was poorer than NC in delayed spacial position memory span test(p﹤0.01). 5. In TOL test, the ADHD children spent more time in all of two-move, four-move and five-move tasks than NC, and broke rules more frequently than NC(p﹤0.05). 6.In TMT, the ADHD children spent more time and made more errors on part B than NC(p﹤0.05), and there was no significant difference on part A (p﹥0.05). Conclusions: The obvious impaired executive functions are found in children with ADHD, involving poor response inhibition,impaired working memory, dysfunction of planning and set-shifting.
5.The design of bionic left ventricular auxiliary pump.

This paper reports a novel design of bionic left ventricular auxiliary pump, and the characteristic is that elastic diaphragm of pump driven by hydraulic, having smooth, reliable blood supply, can prevent blood clots, can use the flow sensor, pressure sensor detection showing the blood pressure and blood volume at the inlet and outlet of the pump. The pump can go with heart rate synchronization or asynchronous auxiliary by the R wave of human body's ECG. The design goal is realization of bionic throb. Through the animal experiment, the blood pressure waveforms are close to expectations, stable flow can stroke according to the set value, which prove that the pump can meet the requirement for heart disease patients for bionic left ventricular assistant.

6.DIGITAL ANATOMY OF THE DEEP CIRCUMFLEX ILIAC ARTERY OSTEOCUTANEOUS PERFORATOR FLAP

Objective To determine the anatomical basis of an algorithm to safely elevate the deep circumflex iliac artery osteocutaneous perforator(DCIAP) flap. Methods 1.Six unfixed corpses underwent whole body gelatine/lead oxide injection.Specimens were dissected by layers.Angiography and photography were used to document the precise course,size,location,and type of individual perforators in the lateral lumbar region.The surface areas of cutaneous territories and perforator zones were measured and calculated with Photoshop and Scion Image.2.One specimen also underwent whole body carboxymethylcellulose/lead oxide injection,CT scan and 3D-Reconstrution. Results An average of 1.6 DCIA perforators with a diameter of 0.7mm was present in 92% of specimens.Perforators were located 5~10 cm posterior to the anterior superior iliac spine,12~35mm above the crest,with a perforator zone of 31 cm~2.The DCIA reliably perfused the medial aspect of the iliac crest.Conclusion The DCIA reliably perfused the medial aspect of the iliac crest and lateral lumbar region.It offers a large quantity of bone on a pedicle of large diameter.The mobility of the skin component allows better tissue positioning during complex reconstructions.
7.Mononuclear cells were induced into endothelia progenitor cells by VEGF165 and bFGF

Objective To determine the biological traits and optimal condition for the induction and differentiation of endothelial progenitor cells from peripheral blood in healthy adults. Methods Mononuelear cells were isolated from peripheral blood of healthy adults by Ficoll-density eentrifugation. The isolated ceils were cultured in 1640 medium supplemented with VECF165 and bFGF. The EPC specific surface mark CD34 and KDR were assessed by fluorescence activated cell sorter(FACS)analysis: EPC were characterized as adherent cells double positive for DiL-acLDL uptake and lectin binding by direct fluorescent staining under a hser scanning confocal microscope. EPC migration were assayed by MTr assay. Result The number and migration ability of EPC were increased by VEGFl65 and bFGF. Conclusion Endothelial progenitors cells can be derived from mononuclear cells of peripheral blood at specific conditions.
8.Effect of alprostadil on early proinflammatory cytokines and its therapeutic effect in patients with severe acute pancreatitis

Objective To investigate the effects of alprostadil on expression of proinflammatory cytokines in patients with severe acute pancreatitis (SAP) and evaluate the clinical efficacy.Methods Seventy-three SAP patients were collected from January 2014 to May 2016,and then were randomly divided into control group (n=37) and experimental group (n=36).On the basis of routine treatment,the experimental group patient was given alprostadil at a dose of 15μg/d.The expression of C-reactive protein (CRP),white blood cell (WBC) count,amylase,alanine aminotransferase (ALT),aspartate aminotransferase (AST),creatinine,serum proinflammatory cytokine tumor necrosis factor alpha (TNF-α),interleukin-1 beta (IL-1[β),interleukin-6 (IL-6) were detected in serum on the 1st,3rd and 7th day.Results The biochemical indexes and expression ofproinflammatory cytokines were significantly increased in the two groups on the 1st day,and decreased gradually,with a significant difference between the time points (P<0.05),but the between-group difference was not significant (P>0.05).These indexes were decreased significantly with the passage of time and there were significant differences between the two groups at the 3rd and 7th day (P<0.05).Conclusion Alprostadil can effectively reduce the severity of early inflammatory reaction in SAP patients,and has important significance for improving the prognosis.
9.The profile of cognitive response to facial expressions in 6- to 12-month-old infants

Objective To explore the profile of cognitive response to five facial expressions in 6-to 12-month-old infants. Method One hundred and fifty-five 6-to 12-month-old healthy infants were recruited to view five kinds of normative facial emotional expression pictures which presented on computer screen. The visual attention, self emotional and behavioral responses of each infant toward the facial expressions were recorded by a web camera and then were coded and analyzed from the videotaped observations. Results The visual preference to expressions was different in different groups. 6-to 8-month-old infants looked back ( such as 1.50 ± 0.73,1.39 ±0.81,1.67 ±0.87 ) more to all expressions than 9-to 12-month-old infants( 1.10 ± 0.53,1.04 ± 0.51,1.12 ±0. 61 ). 6-to 8-month-old infants exhibited no obvious difference(P> 0.05 ) while 9-to 12-month-old infants exhibited significant difference in self emotional responses (include positive and negative) to different expressions (P <0. 05 ). Infants appeared some positive behaviors including looking back at mother,finger-pointing and phonating to expressions during the observation. Conclusions Infants over 6-month-age could discriminate different facial expressions,recognize and comprehend happy expression earlier. 9-to 12-month-old infants began to understand the negative emotional significance of negative facial expressions gradually, but their comprehension to negative emotional expressions was rudimentary.
10.Beta-1,3-glucomannanase assisted lipid extraction from Rhodosporidium toruloides.

To evaluate the effectiveness of enzymatic assisted extraction (EAE) of lipid from the oleaginous yeast Rhodosporidium toruloides in the presence of beta-1,3-glucomannanase at a larger scale, we investigated the effects of enzymatic treatment and extraction conditions on lipid extraction yields at 10-L scale by using the broth of R. toruloides Y4 as the feed and ethyl acetate as the solvent. When it was treated for 0.5 h, the lipid extraction yield reached 71.1%, indicating that the enzymatic treatment process reached similar efficiency to that obtained at 10-mL scale. The inhibitory effect of emulsification was greatly reduced by repeated extraction. After extracted for three times, yields of lipid extraction, solvent recovery and total material recovery reached 92.9%, 87.0% and 94.2% respectively. As it can use the lipid production slurry with good extraction efficiency, EAE technology is promising for industrial production of microbial lipids.
