Objective:To study the therapeutic effect of warming needle plus tuina for degenerative knee osteoarthritis.Method:Randomize 120 cases into a treatment group and a control group,60 cases in each group by the visiting sequence,and then apply warming needle plus tuina for the cases in the treatment group and articular cavity injection of Sodium Hyaluronate for the cases in the control group.Results:The total effective rates in the treatment group and control group were 93.3%and 83.3%respectively,showing a significant difference (P＜0.05).Conclusion:Warming needle plus tuina works well on degenerative knee osteoarthritis.

Objective To investigate the effects of serum from burned rats on the gene expression in cultured rat mesenchymal stem cells (MSCs). Methods Bone marrow was extracted from sacrificed Wistar rats, and MSCs were then incubated for 24h in F-12 medium in the presence of normal rat serum (group N) or serum obtained from burned rats 3 days after injury (group B). Total RNA was extracted from both groups.The mRNA was isolated.An Oligo microarray containing 5705 genes was used to compare the differences of gene expression between two groups. Results There were four genes which expressed differently in two groups.In comparison with group N, the expression of steroid sensitive gene 1 was decreased, but that of fibroblast growth factor-4, dihydroxyacetonephosphate acyltransferase and a EST, which was moderately similar to Bmp2-inducible kinase, were increased in group B. Conclusion Serum from burned rats was able to change the gene expression of MSCs, and it might play the key role in wound repair.

Objective To investigate the effects of serum from rats with burn injury on the differentiation of bone marrow mesenchymal stem cells(MSCs). Methods Bone marrow was extracted from sacrificed Wistar rats and MSCs were then incubated in F-12 medium in the presence of fetal calf serum (FCS) (group F),normal rat serum (group N),or serum harvested from rats with burn injury(group B). Cells of the fifth passage were harvested to investigate the expression of CK and FⅧ by immunohistochemistry and flow cytometry. Results With immunohistochemical staining,both CK and FⅧ were positive in MSCs of group B and negative in the cells of the other two groups. The flow cytometry analysis showed that the positive rates of CK and FⅧ staining MSCs of group B were higher than that of the other two groups ( P 0.05). Conclusions The serum obtained from rats with burn injury could induce differentiation of MSCs to both epithelial cells and vascular endothelial cells,implying that serum after barm injury might promote the MSCs to participate in the process of wound repair.

Objective To investigate the effect of bFGF and MSCs autografting on expression of PCNA and Ⅷ-R Ag of porcine skin defect wound. Methods Six male minipigs were used to produce 48 full-thickness skin wounds, each wound area about 2.54 cm~ 2 on both sides of the back (8 wounds per pig). These 48 wounds were randomly treated with bFGF, MSCs, bFGF+MSCs, and normal saline (12 wounds for each treatment). The tissue from the wounds was harvested on day 3, 7, 14 after surgery, then used for HE staining and immunohistochemical staining for proliferating cell nuclear antigen (PCNA) and factor Ⅷ related antigen (Ⅷ-R Ag). Results Granulation tissue was presented in all groups on day 3 and 7 after wound, most abundant in the wounds treated with bFGF+MSCs, as well as blood capillary formation. PCNA expression increased after skin injury, reached the peak on day 7, most in the granulation tissue, and lasted to day 14, then began to decrease. On day 3, 7, 14 after skin injury, PCNA expression was the most highest in the wounds treated with bFGF+MSCs. Objective To investigate the changes of the cell cycle regulators ATM, Chk2 and p53 and cell cycle arrest in HeLa cells after cisplatin therapy. Methods The proliferation-inhibiting rates of HeLa cells induced by eisplatin of different concentrations were measured by MTT assays. The mRNA and protein expressions of ATM, Chk2 and p53 of HeLa cells with and withont cisplatin were detected by RT-PCR and Western blot, respectively. The cell cycle analysis was conducted by flow cytometric analysis. Results Cisplatin inhibited the proliferation of HeLa cells in a dose- and time-dependent manner. The mRNA and protein expressions of ATM, Chk2 and p53 were increased in HeLa cells treated with cisplatin. The cell cycle was arrested in G2/M phase in HeLa cells treated with cisplatin. Conclusion Activation of ATM, Chk2 and p53 might be critical in determining whether cells survive or undergo apoptesis. Targeting ATM, Chk2 and p53 pathway might he a promising strategy for reversing chemoresistance to clsplatin in cervical cancer.

Objective To investigate the expression of heat shock protein (HSP) 60 and analyze the significance in cervical carcinoma. Methods The cervical tissue samples of 57 cases of cervical cancer and 57 normal cervical tissues were collected. The expression of HSP60 were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). The protein expressions of HSP60 were determined by Western blot analysis. To observe the relationship between HSP60 expression and the occurrence and development of cervical cancer. Results HSP60 mRNA expression in normal cervical tissue was significantly lesser than that in cervical cancer tissue (t=2.65, P0.05). AIM: To investigate gene expression of bax, bcl-2 and p53 in fetal skin at different gestational ages and children skin in order to explore their potentially biological significance. METHODS: Apoptosis in skin specimens was determined by terminal deoxynucleotidy transferase mediated dUTP-biotin nick-end labeling technique (TUNEL). Gene expressions of bax, bcl-2 and p53 in skin at different developmental stages was examined with reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: Along with fetal growth and development, the incidence rate of apoptosis was increased progressively in skin. In skin from early gestational fetus, bcl-2 was strongly expressed. Objective To study the effects of tetrandrine on apoptosis of HeLa cervical cancer cells qualitatively and quantitatively. Methods We measured tetrandrine-induced inhibition of HeLa cell proliferation at different concentrations and time points by MTT assay. The rate of Hela cell apoptosis induced by tetrandrine was detected by flow cytometer and confocal laser scanning microscope (CLSM). Results Tetrandrine inhibited the proliferation of HeLa cells in dosage- and time-dependent manners. Flow cytometry showed that the apoptosis rate was (51.8±0.97)% at the concentration of 15μmol/L, which was significantly higher than that in the control group (24.3±1.23)% (P<0.05). The cells treated with tetrandrine showed typical apoptotic morphology under CLSM. Conclusion Tetrandrine can inhibit proliferation and induce apoptosis of HeLa cervical cancer cells.

Purpose To describe clinicopathological features, diagnosis and differential diagnosis of Castleman′s disease. Methods Retrospective analyses of the clinical data, clinicopathology and immunohistochemistry were conducted in ten cases of Castleman dis-ease and reviewed of literature. Results There were 8 cases of unicenrtic Castleman′s disease and 2 cases of multicentric Castleman′s disease. Pathologically, there were 6 cases of hayline vascular types, one case of plasmatcyic type and 3 cases of mixed type in all Castleman′s disease. Immunohistochemically, all cases were negative for BCL-6 and CD10, and Ki-67 expression was less than or e-qual to 30%. There were 4 cases with complete follow-up data, including one case of intermediate type, 3 cases of hyaline vascular type which were healed by surgical resection without recurrence. Conclusions Castleman′s disease is a rare and lymphoproliferative disorders with unknown cause, it is not easy to diagnose before the operation. Whether immunohistochemical features reflect abnormal immune function or play unknown role in the pathogenesis of Castleman′s disease is also demanded further study.

Objective To investigate whether the proliferation of HepG2 ceils is influenced by interfering BC047440 gene with small hairpin RNA (shRNA) expression plasmid. Methods According to the sequence of BC047440 gene, 2 pairs of BC047440 gene-specific shRNA (shRNA1 and shRNA2) were designed and synthesized. After primer annealing, they were inserted into plasmid pGenesil-1 to construct the shRNA expression plasmids. The recombinant plasmids were transfected into HepG2 cells. The expression of BC047440 gene was detected by quantitative fluorescent PCR, the proliferation of HepG2 cells by MTT assay and the changes of cell cycle by flow cytometry. Results Two shRNA expression plasmids were constructed successfully and were confirmed by restriction enzyme digestion and sequencing. Quantitative fluorescent PCR analysis showed that shRNA1 and shRNA2 could specifically inhibit the expression of BC047440 gene in HepG2 cells, with the inhibition rate of 80.22% and 58.63%, respectively. The shRNA effectively inhibited the proliferation of HepG2 cells, and arrested the cell cycle in S phase. Conclusions The shRNA significantly inhibits the expression of BC047440 gene and the proliferation of HepG2 cells. The expression of BC047440 may be correlated with the proliferation of HepG2 cells.
