1.Determination of the Concentration of Zaleplon in Human Plasma by HPLC

OBJECTIVE:To build up a method for determining the concentration of zaleplon in human plasma by HPLC. METHODS:The Kromasil-C 8 column was used and mobile phase consisted of methanol-water(65∶35);the flow rate was1.0ml/min,fluorescence excitation wave-length was233nm;fluorescence emission wave-length was460nm.RESULTS:The calibration curve revealed linearity in the concentration range of0.42~73.92ng/ml(r=0.9999)with a regressive equation of Y=1.870?147.The average recovery was99.47%.The with-day and between-day RSDs were4.9%and4.73%respec?tively.CONCLUSION:The method is accurate,reliable and highly sensitive so it can be used in study of pharmacokinetics of zaleplon.
2.Determination of the Content of Ciprofloxacin Lactate in Eye Drops by Dual-wavelength Spectrophotometry

OBJECTIVE:Dual-wavelength spectrophotometry was developed for the determination of ciprofloxacin lactate in its eye drops.METHODS:Detection was performed at wavelength 237nm and 279nm with 0.1mol/L HCl as diluent.RESU_LTS:The assay was linear for ciprofloxacin lactate in the concentration range of 3~9?g/ml,r=0.9 998;The mean recovery was 101.6%,RSD was 2.16%.CONCLUSION:This method is simple,quick,accurate and suitable for the determination of ciprofloxacin lactate in its eye drops.
3.APS on Inducing the Effect of Differentiating Cord Blood Monocyte into the Dendritic Cells Induced by Astragalus Polysaccharides In Vitro and its Immunological Characteristic.

Objectives To elucidate the effect of APS replacing cytokine on inducing the cord blood monocyte in vitro into the dendritic cells (DCs) and its cellar immunological characteristic. Methods The cord blood monocytes were isolated and obtained by lymphocyte isolation. three groups were divided: ②Cultured in the RPMI-1640 culture with GM-CSF/IL-4/TNF-?,as the positive control group, with APS in concentration (100mg/L) as the experimental group,and without GM-CSF/IL-4/ TNF-?and APS,as the negative control group, respectively. The morphotype of DCs was identified by inverted optical microscope or scanning electron microscope. The phenotype of cultured 12 days DCs (CD1a, CD80, CD86, and CD83) was identified by flowcytometry. Results Cultured for 72 hours , the morphous of cell of the experimental group grew clustering and began to change from round to irregularity, appearing rough cell face and barb pustute. The longer cell cultured, the more obvious the dendritic structure is. The experimental group cell cultured for 12 days had the most typical dendritic structure. the negative control group cell had no dendritic structure and became the macrophage when cultured for 12 days. The experimental group cell cultured for 10 days showed typical dendritic morphotype by SEM. The experiment group cell and the positive group cell cultured for 12 days significantly expressed the high level phenotype of DCs((CD1a, CD80, CD86, and CD83))by flowcytometry. Conclusions APS and cytokine both could induce the cord blood monocyte to direofive differentiate into functional DC.
4.Effects of different levels of hemodilution during extracorporeal circulation on lactic acid and postoperative outcomes in children with congenital heart disease

Objective To study the effects of different levels of hemodilution during extracorporeal circulation(ECC) on the concentration of lactic acid and postoperative outcome in children with congenital heart disease.Methods Sixty children with congenital heart diseases undergoing open heart surgery with ECC were randomly divided into three groups: groupⅠ(n=20) underwent mild hemodilution [Hb(80—90)g/L],groupⅡ(n=20) with medium hemodilution [Hb(70—79)g/L],and groupⅢ(n=20) with deep hemodilution [Hb(60—69)g/L].The concentration of lactic acid and PaO_2 in groupⅡ and group Ⅲ were compared with groupⅠ before ECC,during ECC,after ECC and 24h after the operation.The rate of homologous transfusion during ECC and the postoperative dosage of furosemide were also compared among the three groups.Results The concentrations of lactic acid and PaO2 in the three groups were all within normal range before ECC,during ECC,after ECC and 24h after operation.The homologous transfusion rate during ECC were lower in groupⅡ(30%)and groupⅢ(15%) compared to that of groupⅠ(50%)(P
5.Application of Acupoint Massage Plus Body Massage for Intervention of Premature Pathological Jaundice

Objective To observe the intervention effect of premature pathological jaundice treated with conventional blue light phototherapy combined with acupoint massage plus body massage. Methods Ninety premature infants with pathological jaundice were randomized into control group 1, control group 2 and observation group, 30 cases in each group. The control group 1 was given blue light phototherapy plus oral use of vitamin B2, the control group 2 was given body massage based on the treatment for the control group 1, and observation group was given acupoint massage based on the treatment for the control group 2, the treatment for the three groups lasting for 7 continuous days. Before and after treatment, the levels of transcutaneous bilirubin (TCB), serum total bilirubin (TB), and serum direct bilirubin (DB) were observed, and then the therapeutic effects on jaundice in the three groups were compared. Results After treatment, the overall therapeutic effect and the daily decline of TCB, TB and DB in the observation group were superior to those in the two control groups, the difference being significant (P < 0.05 or P < 0.01). In the control group 2, the post-treatment average decline of TB and the decline of TCB on treatment day 4-7 were superior to those in the control group 1, the difference being significant(P<0.05). Conclusion Acupoint massage plus body massage is effective and safe for the treatment of pathological jaundice in premature infants, and the therapy being simple and practical.
6.Expression of β-catenin and Oct-4 in colonal cancer and the relationship with metastasis after operation

Objective To investigate the expression of β-catenin and Oct-4 in colonal carcinoma and explore the relationship with recurrence and metastasis after operation. MethodsImmunohistochemical analysis was used to evaluate the expression of β-catenin and Oct-4.The correlation of β-catenin and Oct-4 expression with tumor cell differentiation,T stage,N stage and metastasis was analyzed.The gene expression of Oct-4 was examined by RT-PCR in 20 frozen tumor tissues and normal tissues adjacent to tumor.Results Thirty-five patients had metastasis. The positive rates of β-catenin and Oct-4 expression were significantly higher in metastasis group than in the non-metastasis group (65.71% vs 31.11%,51.43 %vs 13.33 %,x2 =9.843,P =0.002,x2 =13.605,P =0.001).Expression of β-catenin and Oct-4 was not associated with differentiation,T stage or N stage.The positive expression rate of Oct-4 in colonal carcinoma tissues was significantly higher than that in normal tissues.Metastatic rates in patients with positive expression of β-catenin and Oct-4 was higher than that in negative expression.The survival analysis showed that time of metastasis was significantly different in two groups of patients (P <0.05).Conclusion The expression of β-catenin and Oct-4 in tumor tissues is related to metastasis of colonal cancer after surgery and might be used to predict metastasis of colonal cancer after operation.
7.The value of quantitatively fetal brainstem-vermis and brainstem-tentorium angles evaluation by three-dimensional ultrasonography

Objective To evaluate the role of fetal brainstem-vermis (BV) and brainstem-tentorium (BT) angles in differential diagnosis of fetal cereballar vermis anomalies by three-dimensional ultrasonography. Methods A total of 335 normally developed fetuses at 18-34 gestational weeks were examined prospectively in Prenatal Ultrasound Center of Suzhou Municipal Hospital from July 2013 to July 2014 by three-dimensional ultrasonography scanning. The BV angles and BT angles were measured at the mid-sagittal sections of fetal heads which were obtained by three-dimensional ultrasonography. Sixteen fetuses with abnormal cerebellar vermis were selected. The BV angles and BT angles were measured and compared with the measurements of normal group. Results In normal fetuses, the mean of BV angles was (3.3±1.7)° , ranging from1.0 to 10.0°;the mean BT angle was (31.5±6.9)° , and ranging from 16.6 to 57.8° . In 16 cases of abnormal cerebellar vermis, 4 cases of Dandy-Walker Malformation (DWM) showed significantly upward rotated cerebellar vermises, the BV angles increased significantly (88.9±18.1)° , the cerebellar tentoriums were superior displaced, the BT angles increased remarkably (89.0±12.8) ° ;6 cases of Dandy-Walker variant (DWV) showed upward rotated cerebellar vermises, with increased BV angles (23.7±5.2) ° , and slightly increased BT angles (54.5±12.0)°;6 cases of Blake′s pouch cyst (BPC) showed upward rotated cerebellar vermises were, mildly increased BV angles (16.7±1.8)° , and slightly increased BT angles (50.3±8.2)° . The comparison results showed:BV angles increased with increasing severity of the condition in BPC, DWV and DWM;BT angles increased significantly in DWM. Conclusions The measurements of BV angles and BT angles are valuable in the differential diagnosis of fetal cereballar vermis anomalies. From 19 gestational weeks, a BV angle>60° is strongly suggestive of a Dandy-Walker malformation, while a measurement>10.0° and<20° favors the diagnosis of Blake′s pouch cyst.

Objective:To observe the effect of Astragalus polysaccharides(APS) on inducing the cord blood monocytes into mature dendritic cells(DCs) in vitro and to investigate their morphous,cellar immunological characteristics,and contribution to T cell proliferation.Methods:①The cord blood monocytes were isolated by lymphocyte isolating solution under axenic condition,and three groups were divided.②Cells cultured with APS in concentration of 100 mg/L as the experiment group, that with the cytokines of GM-CSF/IL-4/TNF-? as the positive control, and another without either GM-CSF/IL-4/TNF-? or APS as the negative control, respectively. The morphology of DCs was identified by inverted optical microscope or scanning electron microscope. The phenotype of 12 days cultures of DCs(CD1a, CD80, CD86 and CD83) were identified by flowcytometry. The DCs preparations from the experiment group were treated with mitomycin for 45 min to remove their proliferative activity as incentive cells in the mixed cultures with allogenic peripheral blood mononuclear cells from healthy volunteers as responders. T cell proliferation induced with the DCs preparations was detected by MTT chromatometry.Results:After cultured for 72 hours, the cell of both the experiment group and the positive control grew dusteringly and began to change from round to irregularin shapes. The longer the cells were cultured, the more obvious the dendritic structure is. The cells of experiment group and the positive control group when cultured for 12 days had the most typical dendritic structure. The negative control group cells had no dendritic structure and became macrophages when cultured for 12 days. The experiment group cells cultured for 10 days showed typical dendritic morphology by SEM. The experiment group cells and the positive group cells cultured for 12 days significantly expressed high level of the phenotypes of DCs(CD1a, CD80, CD86 and CD83) by flowcytometry.And the difference exhibitied statistical significance when compared with the negative control group(P0.05).The mixed lymphocyte reaction showed that the DCs induced by APS trigerred proliferation of allogenic T cells obviously.Conclusion:Both APS and cytokine could induce the cord blood monocytes to differentiate into functional DCs committedly. DCs reduced by APS stimualate proliferation of the allogenic T cells obviously.
9.The comparison of Wechsler intelligence test outcomes between children with Asperger syndrome and children with attention deficit hyperactivity disorder

Objective To compare the intelligence quotient (IQ) between children with Asperger syndrome (AS) and children with attention deficit hyperactivity disorder (ADHD) in order to provide reliable evidence for differential diagnosis.Methods The intelligence of 141 children with AS,154 children with ADHD and 102 normal control (NC) children aged 6-13 years old examined by the China-Wechsler Intelligence Scale for Children was analyzed,and the diagnoses of AS and ADHD was made according to the Fourth edition of the Diagnostic and Statistical Manual of Mental Disorder.Results The proportions of children at low level of IQ (borderline and mental retardation range) in children with AS and with ADHD group were both significantly higher than those of NC group.The proportions of children at top level of IQ (superior and very superior range) in ADHD group were significantly lower than those of AS and NC groups.In the AS group,higher proportions of verbal IQ (VIQ) at top level and performance IQ (PIQ) at low level were found.The average VIQ,PIQ and full IQ (FIQ) of the AS,ADHD and NC groups were (102.55 ± 17.27,91.31 ± 16.01,97.10 ± 15.88),(95.39 ±13.49,94.16 ± 12.92,94.40 ± 12.78) and (104.06 ± 13.66,103.05 ±11.80,103.91 ± 12.92),respectively.The VIQ,PIQ and FIQ in ADHD group were lower than those in NC group significantly(P < 0.01);compared with NC group,the PIQ and FIQ in AS group were significantly lower (P < 0.01),while the VIQ was not significantly different(P >0.05).The AS and ADHD group scored both lower than the NC groupin the subtests ofcomprehension, coding, picture completion andpicture arrangement (P < 0.01).The difference values between VIQ and PIQ in the AS,ADHD and TD group were(11.23 ± 17.29),(1.23 ± 13.10) and (1.00 ± 10.91),respectively,among which only the value in AS group was statistically significant (P < 0.01) but not in ADHD and NC group(P >0.05).The proportion of children having a VIQ-PIQ difference value more than 15 in AS group was 54.60% (77/141 cases),which was significantly higher than that in ADHD [22.07% (34/154 cases)] and NC groups [13.72% (14/102 cases)] (P < 0.01),while the latter 2 value had no statistical difference (P > 0.05).The differences among scores on the three Kaufman factors were significant in AS group (P < 0.01) but not in ADHD and NC groups(P > 0.05).Conclusions Compared with ADHD and normal control children,there are some unique intelligence profiles in children with AS,which show VIQ and PIQ separation,imbalance intelligence structure as well as strengths and weaknesses coexisting.Furthermore,children with ADHD and AS children share common features in some subtests scores.
10.Comparison of the efficacy and adverse effects of docetaxe or oxaliplatin combined with capecitabine for late-staged gastric cancer in aged patients

Objective To compare the short-term efficacy and adverse effects of docetaxe or oxaliplatin combined with capecitabine in the treatment of late-staged gastric cancer in aged patients. Methods Eighty-two aged patients with late-staged gastric cancer were randomly divided into two groups,of which 38 patients were treated group) ,and 44 patients were treated with oxaliplatin (100 mg/m2 ivgtt on 1st day) and eapecitabine (2000 mg/1 cycle). Results There is no failure of follow-up. In the docetaxe group,the effective rate was 52.63% (20/38) and 54.55 % (24/44) for the docetaxe and oxaliplatin group,respectively (P>0.05). The median progression-free survival(PFS) in the docetaxe group (6.1 months) was similar to that in the oxaliplatin group (6.3 months) (P>0.05). Gastrointestinal response,myelosuppression and neurotoxicity (Ⅰ or Ⅱ level) were the most common ad-verse effects observed in both groups (P>0.05). No chemotherapy-related death was observed. Conclusions The short-term efficacy of decetaxe or oxaliplatin combined with capecitabine in the treatment of late-staged gastric cancer in aged patients is similar,and the adverse effects are all within tolerance limits.
