OBJECTIVE:To investigate the effects of different concentration of methanol on 5 anthraquinone constituents in Semen Cassiae.METHODS:Semen Cassiae was extracted with different concentration of methanol(95%,80% and 30%).The contents of aloe-emodin,rhein,emodin,chrysophanol and physcione in Semen Cassiae were determined by HPLC.RESULTS:The contents of aloe-emodin,rhein,emodin,chrysophanol and physcione were 0.001 0 ～0.018 1 mg?g-1,0.010 4～0.063 7 mg?g-1,0.020 9～0.253 5 mg?g-1,0.003 1～0.915 7 mg?g-1 and 0.005 1～0.126 3 mg?g-1,respectively,The average recoveries were 96.34%～97.16% and the RSDs of all were less than 3%(n=6).CONCLUSION:There were great differences in the contents of the 5 anthraquinone constituents in Semen Cassiae extracted with different concentration of methanol,and a best extraction efficacy was achieved for Semen Cassiae extracted with 95% methanol while the worst extraction efficacy was noted if extracted with 30% methanol.

OBJECTIVE:To investigate the effect of rat liver and intestine on the metabolism of Baicalein.METHODS: The microsomal drug-metabolizing enzyme incubation method was employed,and the contents of bacicalein and its glucuronidation metabolite at different incubation time were determiend by HPLC. The effects of various microsomes on the glucuronidation rates of Baicalein were studied.RESULTS:Different microsomal drug-metabolizing enzymes contributed to the metabolism of Baicalein in time-and concentration-dependent manner,with Jejunum microsome the strongest while the colon microsome the weakest in efficacy.CONCLUSION:Baicalein had a strong metabolism in liver and intestine especially in the intestine.

OBJECTIVE:To explore the use of antibiotics in the department of respiratory diseases and pediatric outpatient department after the introduction of initiative intervention stragety on the rational use.METHODS:A field investigation was carried out on the international RDU index of antibiotics in the department of respiratory diseases and pediatric outpatient department by self-control test.Initiative intervention on physicians' prescribing practices was carried out at four stages:befter intervention,following first intervention,second intervention,and third intervention,respectively.Data statistics was analyzed by SPSS11.0 software.RESULTS:The percentages of patients treated with antibiotics and injcetable antibiotics in the department of respiratory diseases dropped from 61.6% and 19.2% before intervention to 30.0% and 14.0% after third intervention(P

Objective:To discuss the influence and curative effect of intensive lipid-lowering with atorvastatin on blood fat and ser-um inflammatory factors levels in the patients with acute cerebral infarction. Methods:Totally 94 cases of patients with acute cerebral infarction were divided into the intensive group(n=47) and the ordinary group (n=47). The patients in the two groups were given the basic medical treatment, such as reducing intracranial pressure and dehydration, controlling blood pressure and blood sugar, anti-platelet aggregation, neural protection and etc. The patients in the ordinary group were orally given 20mg atorvastatin calcium tablets, once a day, while the patients in the intensive group were additionally given 40mg atorvastatin calcium tablets, once a day, and the treatment course was 8 weeks. The changes of blood fat index and serum inflammatory factors of hs-CRP, TNF-αand IL-10 in the two groups before and after the medical treatment were detected, and the clinical curative effect was compared as well. Results:After the 8-week medical treatment, TC, TG and LDL-C levels in the two groups were declined at different degree, while HDL-C levels were in-creased at different degree (P<0. 05 or P<0. 01), and the changes in the intensive group was more notable than those in the ordinary group (P<0. 05). After the treatment, the serum hs-CRP and TNF-αlevels in the two groups were declined at different degree, while serum IL-10 levels were increased at different degree (P<0. 05 or P<0. 01), and the changes in the intensive group was more signifi-cant than those in the ordinary group (P<0. 05). Meanwhile, the total clinical efficiency in the intensive group (95. 74%) was much higher than that in the ordinary group (80. 85%, P<0. 05). Respectively 2 and 4 cases of untoward effect were appeared in the ordi-nary group and the intensive group without statistical difference between the two groups(P>0. 05). Conclusion:Intensive lipid-lower-ing with atorvastatin has significant curative effect with favorable security on acute cerebral infarction, which can obviously improve the degree of neural function defect, and the mechanism may related with reducing blood fat, serum hs-CRP and TNF-αlevels, increasing serum IL-10 levels and inhibiting topical inflammatory reactions.

OBJECTIVE: To investigate the effects of serum containing Scutellaria Barbata extract (ESB) on apoptosis rate and mitochondrial transmembrane potential (MTP) of liver cancer cell line H22 from mice in vitro. METHODS: H22 cells were cultured in vitro and divided into 5 groups: blank control group, low-dose ESB group, medium-dose ESB group, high-dose ESB group and fluorouracil (5-Fu) group. Methyl thiazolyl tetrazolium assay was utilized to determine the proliferation rates of H22 cells. Cellular morphology was observed under a transmission electron microscope (EM). The rhodamine 123 was used as a fluorescence probe to label the H22 cells, and the fluorescence intensities were observed with a laser scanning confocal microscope. The fluorescence intensity of H22 cells indicated the MTP of H22 cells. RESULTS: The inhibition of serum containing ESB on the proliferation of H22 cells in vitro was observed in a time-dependent manner. The typical morphological changes of apoptosis were observed after incubation with ESB-containing serum in high dose for 48h. The apoptosis rates of blank control group, 5-Fu group, low-dose ESB group, medium-dose ESB group and high-dose ESB group were (0.51+/-0.32)%, (11.26+/-2.97)%, (1.07+/-0.46)%, (3.15+/-1.12)%, (7.83+/-2.25)% respectively. ESB could reduce the MTP of H22 cells from mice as compared with the untreated group. The MTPs of the blank control group, 5-Fu group, and low-, medium- and high-dose ESB groups were (245.45+/-67.37), (127.42+/-41.35), (213.68+/-65.52), (186.34+/-56.37) and (142.65+/-39.44) respectively, which were negatively correlated with the apoptosis rates. CONCLUSION: ESB-containing serum effectively induces apoptosis, which may be related to the decrease of MTP in H22 cells.

Objective:To observe the effect of Erfukang oral liquid on microcirculation disturbance in mice with acute blood stasis caused by adrenaline. Methods:The mice were randomly divided into the blank group,Naodesheng group(1.35 g·kg-1),Erfukang oral liquid group respectively at high(30 ml·kg-1),medium(15 ml·kg-1) and low(7.5 ml·kg-1) dose,and a microcirculation apparatus was used to observe the number of capillary opening in the normal mice at one hour after the administration. The microcircu-lation disturbance of auricle was induced by the injection of epinephrine into caudal vein. The number of capillary opening and blood flow in 2 min in the mice were observed. Results:Compared with the blank group,Ergukang oral liquid at high,medium and low dose showed effect on the number of opening capillaries without statistical significance (P>0.05);compared with the model group, Erfu-kang oral liquid at high and medium dose could significantly improve the microcirculation induced by epinephrine (P<0.05 or P<0.01),and significantly confront the decreased number of capillary network opening induced by adrenaline(P<0.05). Conclusion:Erfukang oral liquid can improve microcirculation disturbance.

Objective:To evaluate the value of B-Raf proto-oncogene, serine/threonine kinase (BRAF) V600E mutation detection in the differentiating malignant from benign with Bethesda system for reporting thyroid cytopathology (BSRTC) categories Ⅰ and Ⅲ nodules. Methods:From January 2019 to December 2020, a total of 264 nodules from 263 patients (79 males, 184 females; median age 46 years) were retrospectively enrolled and all patients underwent BRAF V600E mutation detection, fine-needle aspiration cytology (FNAC) and thyroid nodulectomy in the Affiliated Drum Tower Hospital of Nanjing University Medical School. Thirteen nodules of 12 patients had repeat aspirate samples and 51 nodules were examined with multiple genes assay in formalin fixed paraffin embedded tissues. Taken the postoperative histopathological results as the gold standard, the diagnostic efficiency of BRAF V600E mutation was analyzed by comparing the results of multiple genes assay and BRAF V600E mutation detection of repeated puncture samples. Results:Of 264 nodules, 230 were malignant (papillary thyroid cancer (PTC)) and 34 were benign, with BSRTC categories Ⅰ (nondiagnostic) and Ⅲ (follicular lesion) nodules of 58 and 206. The sensitivities of BRAF V600E mutation detection in BSRTC categories Ⅰ and Ⅲ nodules were 77.1%(37/48) and 78.0%(142/182), the specificities were 9/10 and 91.7%(22/24), the positive predictive values were 97.4%(37/38) and 98.6%(142/144), the negative predictive values were 45.0%(9/20) and 35.5%(22/62), and the accuracy rates were 79.3%(46/58) and 79.6%(164/206). The diagnostic concordance of BRAF V600E mutation detection was 90.2%(46/51) in the preoperative and postoperative samples of 51 nodules with preoperative BRAF V600E wild type but postoperative pathology confirmed as PTC and was 11/13 in repeat puncture samples. Conclusion:BRAF V600E mutation detection is an effective diagnostic method for BSRTC categories Ⅰ and Ⅲ nodules.
