1.ENZYME-LINKED LMMUNOSORBENT ASSAY (ELISA) FOR SOYBEAN RHIZOBIA STRAIN IDENTIFICATION

The direct ELISA was used to detect cells from pure cultures and nodules of Bradyrhizobium japonicum USDA110 and Rhizobium fredii RTt50. The optimum dilution of enzyme-linked conjugates, HRP-Ab110 and HRP-Ab50, was 1:3200 and 1:800 respectively. The optimum dilution of antibodies, Ab110 and Ab50, was 1:3200 and 1:800 separately. The optimum concentrations of antigens, USDA110 and RTt50, were both 6?10~7 cells/ml. Slow and fast-growing soybean rhizobiz can be detected and differentiated specifically by direct ELISA. Among a few strains of these two groups of soybean rhizobia, cross-reaction occurred. This was eliminated by absorption, so that specific strain could be identified by ELISA. The minimal concentration of antigen for detected was 2?10~5 cells/ml. It was found that nodules preserved by drying over silica gel or freezing were equally good, without loss in sensitivity of ELISA. ELISA was used to study the competition of USDA110 and RTt50 with indigenous rhizobia in soil pot experiment. Nodule occupacy of USDA110 ranged from 75-87.5% in different growing season of soybean and RTt50 ranged from 25-45%. The result showed that ELISA was more sensitive than agglutination.
2.Biomechanical properties of a novel automatic anti-rotation posterior atlantoaxial internal fixation system: a finite element analysis

BACKGROUND: Atlantoaxial posterior approach is currently the main surgical treatment for atlantoaxial instability and/or dislocation, but the shape of rod in normal screw-rod device system is cylindrical. To obtain satisfactory reduction of atlantoaxis, the rod will be pre-bent obviously before fixation; however, the cylindrical rod will be rotated when tighten the nuts. Extra devices will be required to adjust and maintain the direction of the rod, thereafter, the surgical field will be blocked by the device, and spinal injury will occur once the devices are not held tightly. While the novel automatic anti-rotation rod has the function of anti-rotation during nut-tightening process, and also holds all the advantages of normal rod. Further research should be performed for the differences in biomechanical characteristics between two methods.OBJECTIVE: To investigate the biomechanical properties of the novel automatic anti-rotation rod for internal fixation system of atlantoaxial posterior approach based on three-dimensional finite-analysis model of upper cervical spine.METHODS: The three-dimensional finite element model of upper cervical spine with internal rod fixation system was developed. The biomechanical characteristics of the internal fixation system were analyzed. RESULTS AND CONCLUSION: The traditional and novel three-dimensional finite element model with realistic and geometric similarity contained 198330 elements, 964747 nodes and 246788 elements, 996069 nodes,correspondingly. There was no obvious stress concentration in both two systems, stress was concentrated mainly in the screw-bone and screw-rod interfaces. The stress values of the novel system were higher than those of the traditional system, but the maximum Von Misses Stress of two systems was lower than the yield (795-827 MPa ) and ultimate (860-896 MPa) strength of titanium alloys. These results show that the design of the novel automatic anti-rotation rod-screw fixation system has matched the biomechanical requirements for new internal fixation instruments, and is one safe, effective and practical device for atlantoaxial posterior procedure showing promising application prospect.
3.Clinical significance of the changes of coagulation and fibrolysis parameters in malignant lymphomas

Objective To explore the clinical significance of the coagulation and fibrolysis parameters changes for the knowledge of complicated thrombosis after chemotherapy in malignant lymphomas. Methods Morning fasting anti-coagulation blood samples were taken to detect plasma prothrombin time (PT), activated partial thromboplastin time (APTT), fibrinogen (FIB) and thrombin time (TT) with automatic coagulation analyzer in 71 hospitalized malignant lymphomas and 20 normal controls. The plasma D-dimer levels of the two groups were detected with immunoturbidimetry. Results The levels of plasma APTT, Fib and D-dimer in 71 malignant lynphomas were (30.44±1.43) s, (3.28±0.20) g/L, (297.05±56.59) μg/L respectively, which were significantly higher than those in normal controls at the levels of (23.72±0.76) s, (2.57±0.22) g/L, (94.50±26.07) μg/L respectively (P <0.05). The coagulation and fibrolysis parameters were of no statistic differences between the normal controls and lymphomas of stage Ⅰ and Ⅱ (P >0.05). The APTT and Fib levels in lymphomas of stage Ⅲ and Ⅳ were higher than those in normal controls and lymphomas of stage Ⅰ and Ⅱ (P <0.05). There are 7 malignant lymphomas complicated venous thrombosis . Of the 7 cases, the FIB and D-dimer levels were higher than those of stage Ⅰ and Ⅱ, furthermore the D-dimer levels were higher than those of stage Ⅲ and Ⅳ. Conclusion The abnormalities of coagulation and fibrolysis parameters occurred in malignant lymphomas with requirement of periodic monitoring. After chemotherapy, the lymphoma patients had a high incidence of venous thrombosis, which need early prevention for prolongation of the survival.
4.Preparation and in Vitro Release of Poly(lactide-co-glycolide) Microspheres Loaded with Ropivacaine

OBJECTIVE: To optimize the preparation technology of poly(lactide-co-glycolide) (PLGA) microspheres loaded with ropivacaine (ROP-PLGA-MS) and study their powder particle characteristics and in vitro release characteristics. METHODS: ROP-PLGA-MS was prepared with PLGA as carriers using the water-in-oil-in-water(W/O/W) emulsion solvent evaporation method. The micromeritic characteristics of ROP-PLGA-MS,such as the particle size,loading and entrapment efficiency were taken as indexes for evaluation,and the preparation technology was optimized by orthogonal design. Its dissolution characteristics in vitro were studied. RESULTS: The prepared microspheres were spherical with smooth surfaces,with an average particle size of (2.525?0.047) ?m and over 80% falling in the range of 1.8~5.0 ?m. The encapsulation efficiency was (58.05?1.169)% and the drug loading efficiency was (6.067 ?0.312)%. The in vitro release curves of ropivacaine microspheres could be fitted with Higuchi equation. The accumulated release percentage of which was 82 % in 192 h,and t1/2=60.16 h. CONCLUSION: ROP-PLGA-MS has obvious sustained release.
5.Protective Effect of Swertia Punicea on Experimental Hepatic Injury

Objective: to observe the protective effect of Swertia Punicea on experimental hepatic injury induced by CCl4 and BCG/LPS respectively. Methods: Mouse models of hepatic injury were induced by CCl4 and BCG/LPS. Serum levels of ALT and AST, MDA content and the pathological changes of hepatic tissue were observed. Results: Swertia Punicea obviously inhibited the rising levels of serum ALT and AST, improved the histological features and relieved hepatic edema induced by CCl4 in mice. However, it has no obvious action on the rising levels of ALT and AST induced by BCG/LPS. It also reduced MDA content and decreased the hepatic index and the splenic index. Conclusion: Swertia Punicea has antioxidation and a protective action on the chemical and immunological hepatic injury, especially on the chemical hepatic injury.
6.CT study of dependability in cerebral hemodynamic changes of carotid artery stenosis and the degree of carotid stenosis

Objective To investigate cerebral hemodynamic changes in patients with unilateral stenosis of carotid artery by using CTP and CTA,and to analyze the dependability of the degree of artery stenosis and cerebral hemodynamic changes.Methods Fifty patients with unilateral stenosis of carotid artery underwent toshiba aquilion one CTP and CTA.The patients were divided into mod-erate stenosis group,severe stenosis group and occlusion group according to the NASCET criteria.The rCBF,rCBV,rMTT and rTTP were measured ,and compared between stenosis and contralateral hemispheres ,and the dependability were analyzed between the de-gree of unilateral carotid artery stenosis and relative perfusion parameters by using ranking-test.Results (1)There were 20 cases of unilateral carotid artery occlusion,13 severe stenosis,1 7 moderate stenosis.(2 )No statistical significance on rCBF and rCBV was found among the three stenotic groups (P>0.05).There were statistical significances on rMTT and rTTP among the three stenotic groups(P<0.05).(3)There were statistically no correlations between the values of rCBV,rCBF and the degree of artery stenosis.There were statistically positive correlations between the values of rMTT,rTTP and the degree of artery stenosis with the pearson correlations 0.512 and 0.708. Conclusion rMTT and rTTP are more sensitive parameters for evaluation of cerebral hemodynamics and dependability of carotid ar-tery stenosis on the CTP and CTA.These have an important value to guide the clinical treatment.
7.Research progress of gallbladder neuroendocrine tumor

Neuroendocrine tumors of the gallbladder(GB-NET) are rare, and it lacks early clinical manifestations and has no specific tumor markers, it is difficult to distinguish GB-NET from gallbladder adenocarcinoma. The diagnosis of GB-NET is based on histopathology of the tumor and the assessment of proliferation fraction, which makes it difficult to achieve early diagnosis. GB-NET has a high degree of malignancy, 32.39% of patients have liver metastases at diagnosis, and 51.60% of patients have lymph node metastases, the median survival time is 9 to 10 months.There are currently no specific guidelines or consensus for the treatment of GB-NET. The treatment strategies are choosen mainly by the principles of gallbladder adenocarcinoma. We reviews the clinical and basic researches of GB-NET and case reports from China and across the world, as well as the data from SEER database, and we discuss the research progress on the classification, clinicopathological features, diagnosis, treatment advances and the prognosis.
8.Effects of intravenous infusion of lidocaine on colorectal resection

Objective To investigate the effects of intravenous infusion of lidocaine on colorectal resection.Methods Sixty patients undergoing colorectsl resection were randomly divided into group L [receiving intravenous infusion of 1%lidocaine,bolus injection of 1.5 mg/kg lldocaine at induction of anesthesia,followed by a continuous infusion of 1.5 mg/(kg·h)intraoperatively and for 12 h postoperatively]and group C(an equal volume of saline).Postoperative pain scores(VAS score)and morphine consumption were measured.Times of flatus and first defecation were recorded.Results The end expiration sevotturane concentration maintaining hemedynamics stable in group L was lower than that in group C(P<0.05).The dose of sulfentsnyl was(17.5±3.8)μg in group L,(25.6±4.5)μg in group C,there was significant difference between two groups(P<0.05).The times of flatus first,defecation and morphine conaumption at 24 and 48 h after operation in group L were lower than those in group C[(23±6)h vs(28±7)h,(31±7)h vs(43±9)h,(32±6)mg vs(46±8)nag and(58±7)mg vs(71±10)mg,P<0.05].There was no significant difference at resting VAS score,but there was significant difference at movement and cough VAS score between twp groups(P<0.05).Conclusion Perioperative administration of low doses of intravenous infusion of lidocaine reduces intraoperative anesthetic requirements and has a clinically relevant beneficial effect on postoperative recovery after colorectal resection.
9.Variation of serum ferrin, folic acid and vitamin B_(12) levels in patients with acute promyelocytic leukemia

Objective:To observe the dynamic variation of serum ferritin (SF), folic acid, and vitamin B_(12) levels in patients with acute promyelocytic leukemia (APL) at different disease stages. Methods:Serum SF, folic acid and vitamin B_(12) levels were successively tested in thirty-six patients with primary APL every 1 to 3 months by using chemiluminescence analysis. Five different disease stages were selected as dynamic observation time points: first diagnosed, first complete remission (CR1), six months after CR1, relapsed stage,and CR1 for three years. Results:There were 75.0%(27/36)of patients with abnormal high levels of SF, 77.8% (28/36)of patients with abnormal low levels of folic acid, and 100%(36/36)of patients with increased vitamin B_(12) levels in first diagnosed stage. The number of patients with abnormal variations of SF, folic acid and vitamin B_(12) level was decreased in CR1 stage compared with those in first diagnosed stage (SF: P<0.05;folic acid and vitamin B_(12): P<0.01). The serum SF, folic acid and vitamin B_(12) levels tended to recover step by step with chemotherapy. At six months after CR1 the three parameters of most patients recovered to normal levels. APL was relapsed in 4 patients after 1-year CR. Both SF and vitamin B_(12) levels were increased and the folic acid level was decreased compared with those before replase, but the difference had no significance (P>0.05). The serum SF, folic acid and vitamin B_(12) levels were in normal ranges in the patients who had 3-year CR. Conclusion:The serum SF, folic acid and vitamin B_(12) levels had dynamic variation in APL course. Increase in serum SF and vitamin B_(12) as well as decrease in folic acid are related with the active degree of APL and its tumor load.
10.Application of the xenogenic acellular dermal matrix membrane application used in the postoperative tissue shortage repair.

OBJECTIVE:
To observe the short-term and long-term curative effect of the xenogenic acellular dermal matrix membrane (or joint muscle flap transfer) application used in the 82 cases postoperative tissue shortage repair that after the head neck carcinoma resection.
METHOD:
To held the 82 cases head neck carcinoma postoperative mucosa shortage repaired after resection by the xenogenic acellular dermal matrix membrane (or joint muscle flap transfer), 65 cases mucosa shortage wound be directly covered by the repair membrane and the other 17 cases mucosa shortage wound be repaired by the tranfered muscle tissue flap with the repair membrane covered; 53 cases underwent additional postoperative radiotherapy between 2-4 weeks and follow-up in 1, 3, 6, 12, 18, 24, 30, 36, 48, 60 months and observed the operation site repair process through the electronic laryngoscope, observed the patients respiration, swallow, phonation function.
RESULT:
Seventy-seven cases patients operation incision reached I phase healing standard, another 5 cases patients operation incision reached II phase healing standard because of the wound infection and fully-recovered through the local wound drainage,dressing process. All the patients tracheal cannula,the stomach tube be extubated successfully and without the local cicatricial constriction occurred. Seventy-eight cases follow up period reached 1 year including 53 cases who underwent postoperative radiotherapy, 49 cases follow up period reached 3 years including 32 cases who underwent postoperative radiotherapy, 14 cases follow up period reached 5 years including 12 cases who underwent postoperative radiotherapy. The patients with static local lesions discovered no reaction such as exclusion, allergy.
CONCLUSION
The application of xenogenic acellular dermal matrix membrane (or joint muscle flap transfer used in in the postoperative tissue shortage repair that after the head neck carcinoma resection have several advantage such as comparatively easily implementation, operation safety edge enough,well preserved organ function, comparatively low incidence about the laryngeal stenosis, the short-term and long-term repair effect are all exact.
