1.Dose-effect research using nanopatch to deliver siRNA in the inhibition of HPV gene expression.

Hela is the cell line of adenocarcinoma of the uterine cervix, and human papillomavirus (HPV) 18 shows positive. We delivered siRNA with target specifically to HPV18 E7 mRNA into nude mice Hela tumor xenografts by nanopatch to inhibit the HPV gene expression, and further to study the superiority, the best action time and concentration of siRNA of using nanopatch to transfer siRNA in vivo. We designed siRNA that target specifically to HPV18 E7 mRNA (siE7) and checked the effect of siE7 in vitro. Tumor xenografts were transfected with siE7 and GenEscort III by nanopatch. Expression of HPV18 E7 mRNA and protein were detected 0 hours, 24 hours, 48 hours, 72 hours after transfection with PT-PCR and Western blot, and the best action time was analyzed using nanopatch to thansfect siRNA in vivo. We transfected GenEscort III and siE7 of Different concentration into tumor xenografts respectively by nanopatch and intraperitoneal injection. Expression of HPV18 E7 mRNA and protein was detected 72 hours after transfection by PT-PCR and Western blot, to analyze the best action concentration of siRNA and the superiority of using nanopatch to thansfect siRNA in vivo. The results proved that SiE7was efficient to inhibit expression of HPV18 E7 mRNA and to advance Hela apoptosis in vitro. SiE7 transfected by nanopatch into xenografts could inhibit effectively expression of HPV18 E7 mRNA and protein. The best action time and concentration of siRNA of using nanopatch to thansfect siRNA in vivo are 72 hour post-transfection and 2 micromol/L siE7. To compare intraperitoneal injection in delivering siRNA in vivo, the effect of nanopatch is very predominant. It can be well concluded that Nanopatch can effectively transfer siRNA in vivo, which can effectively inhibit the HPV gene expression.

2.Application value of the transesophageal echocardiography in perioperative of percutaneous left atrial appendage closure with Watchman

Objective To assess the feasibility and accuracy of transesophageal echocardiography (TEE) in screening patients,intraoperative guidance of occluder releasing and efficacy assessment in the patients of percutaneous left atrial appendage (LAA) occlusion with Watchman.Methods Ultrasonic instrument Philips iE33(proble model X7-3t) were used to measure the maximum diameter and the depth of LAA ostium in enrolled patients by TEE,and intraoperative guided puncture of interatial septum,positioning Watchman transmission system and instruction closure release under TEE,immediate evaluate the postoperative therapeutic effect and record the complications by TEE.At 45 days after implantation,patients should repeat TEE to assess the efficacy.Results Eighteen patients underwent device implantation.Seventeen patients implant successfully,except one patient can't implant device beacuse of the LAA morphologe was not suitable.There was one patient showing thrombus formation on the surface of device,one patient of LAA and the device axial angulation was 90 °.At 45 days after implantaion,16 patients were fullowed up,there were no major stroke at all.Conclusions TEE has important application value in screening of preoperative patients,intraoperative guidance of occluder releasing and efficacy assessment in the patients of percutaneous left atrial appendage occlusion with Watchman.
3.Survey on status quo of psychological pressure of female infertility patients in Chongqing

Objective To investigate the status quo of psychological pressure in female infertility patients so as to provide the basis for carrying out the psychological counseling aiming at these patients .Methods The random sampling was adopted and the questionnaire survey was conducted on 1 760 female infertility patients .Results The patients with psychological pressure accounted for 84 .43% of the investigated patients .The cases number of psychological pressure in the patients with in‐vitro‐fertilization and embryo transfer(IVF‐ET) treatment was significantly more than that in the patients with pregnancy‐guiding and artificial insemina‐tion (P<0 .01);the cases number of psychological pressure in the patients with higher educational degree was more than that in the patients with lower educational degree (P<0 .01);the cases number of psychological pressure in technicians ,teachers ,staffs ,cadres and civil servants was more than that in the workers ,farmers ,merchants and the people with the other occupations (P<0 .01);the cases number of psychological pressure in the patients with 2-5 year infertility was more than that in the patients with the other infertility years (P<0 .01);the cases number of psychological pressure in the patients with visiting doctors for twice or over was more than that in the patients with visiting doctors for the first time (P< 0 .01) .Conclusion The female infertility patients in Chongqing have large psychological pressure .The doctor should actively communicate with patients ,answer the patient doubt ,ease the patient emotion ,mobilize the patients′family and society to care the patients .
4.Corrosion performance of medical titanium alloys in three different physiological electrolytes

BACKGROUND: Titanium alloy implant has been widely used in the clinic. But there are few reports addressing on corrosion performance of medical titanium alloy dental implant, in particular in different physiological electrolytes. OBJECTIVE: To investigate the corrosion behaviors of medical titanium alloys in physiological saline, simulated saliva, and simulated body fluid. DESIGN, TIME AND SETTING: A randomized grouping, controlled observation was performed at the School of Stomatology, Lanzhou University and State Key Laboratory of Solid Lubdcation, Lanzhou Institute of Chemical Physics, Chinese Academy of Sciences between November 2008 and March 2009. MATERIALS: Medical titanium alloys (Ti-6Al-4V) were cut into 10 mm×10 min×1 mm sheets and were randomly divided into 3 groups, with 10 sheets in each group. METHODS: Corrosion performance of titanium alloys in physiological saline, simulated saliva, and simulated body fluid was investigated using electrochemical measurements. Following electrochemical corrosion, surface morphologies and wettability of samples were also observed using scanning electron microscope and CA-A type contact angle tester. MAIN OUTCOME MEASURES: Tafel curve, surface morphology, and contact angle of medical titanium alloys in three physiological electrolytes after 4 days of electrochemical corrosion. RESULTS: The corrosion of titanium alloys was most severe in simulated saliva, followed by simulated body fluid, and mildest in physiological saline. Scanning electron microscope results demonstrated that medical titanium alloy surface exhibited many holes after corrosion by physiological saline, showed rough surface with a slightly changed number of holes after corrosion by simulated body fluid, and presented with an increased number of holes, with aperture greatly increased in some holes after corrosion by simulated saliva. The contact angles of titanium alloys were all reduced after corrosion of three electrolytes. CONCLUSION: The corrosion of titanium alloy was most serious in simulated saliva. Thus, corresponding protection measures should be given in clinical application.
5.Targeted and thrombolytic effect of urokinase-loaded immunoliposomes on acute pulmonary embolism in rabbits

AIM: To prepare urokinase-loaded immunoliposomes, with anti-D-dimer mouse monoclonal antibody that can selectively target to thrombotic site and test the thrombolytic effect in the rabbit model of acute pulmonary embolism (PE) in early phase. METHODS: 40 New Zealand white rabbits were randomly divided into five groups: TBS group (TBS buffer, negative control group), urokinase (UK) group (150 000 IU/kg UK, positive control group), Lip group (Lip-UK, containing 50 000 IU/kg UK), Ab group (Ab-Lip-UK, containing 50 000 IU/kg UK) and 2 Ab group (2 Ab-Lip-UK, containing 50 000 IU/kg UK but double DDmAb of Ab group). PE model was established. Five minutes later, five solutions (TBS, UK, Lip-UK, Ab-Lip-UK, 2 Ab-Lip-UK) were transfused through femoral vein respectively. Right ventricular systolic pressure (RVSP) and right ventricular diastolic pressure (RVDP) were estimated within one hour. The pathological changes of lung, heart, liver and kidney were all observed. RESULTS: RVSP in TBS group had no significant changes within one hour after administration. However, RVSP had respective notable decline at 30 min, 40 min, 30 min, 20 min in UK, Lip, Ab and 2 Ab group, respectively. The means of residual emboli were as follows: 4.0±0 in TBS group, 2.4±0.9 in UK group, 3.1±0.6 in Lip group, 2.4±0.9 in Ab group and 1.9±0.6 in 2 Ab group. The lungs in each group showed scattering local congestion, effusion and swelling on the surface. Obvious hemorrhage of heart, kidney and liver was found with HE staining in UK group, no pathologic change was observed in other groups. CONCLUSION: 2 Ab-Lip-UK with early thrombolytic effect and security may be an ideal thrombolytic agent for treating pulmonary embolism.
6.Application of the Animal Energy Metabolism Monitoring System (AEMMS) in Acute Alcoholic Liver Injury with the Treatment of Acupuncture

This study aimed to explore the value of animal energy metabolism monitoring system (AEMMS) to evaluate the energy metabolism of acute alcohol liver injury in rat with the treatment of acupuncture.Tirty rats were randomly divided into the control group,the model group and the EA group.The model group and the EA group were gavaged with 50% ethanol for the establishment of the animal model of acute alcoholic liver injury.EA was given by needling Zusanli (ST-36) 30 min after intragastic administration every day in the EA group.The treatment of acupuncture continued 3 days.In addition,the RER,heat,feeding and drinking were continuously recorded for 96 h in each group at the same time by AEMMS.As a result,the RER,feeding and drinking indices significantly decreased in rats with ALI with an increase in the heat index.There was an up-going tendency of the RER,feeding and drinking indices,and a declined tendency of the heat index in the model rats after EA treatment.It was concluded that AEMMS presented a practical value in the study of the energy metabolism of rats with ALI with the treatment of acupuncture.
7.Effect of nicotine on alkaline phosphatase activity of human dental papilla mesenchymal cells in vitro

Objective: To study the effect of nicotine on alkaline phosphatase (ALP) activity of human dental papilla mesenchymal cells in vitro. Methods: Human dental papilla mesenchymal cells were cultured in the presence of nicotine at various concentrations, ALP level of the cells was meseased at different time with an enzyme dynamical method and was expressed as A value at 410 um. Results: After 3day-exposure of the cells to nicotine(g/L) at 0, 0.05, 0.1, 0.2 and 0.4 the A values(per 10 5cells) were 0.64 ? 0.15, 0.61?0.03, 0.47 ? 0.05, 0.44 ? 0.06 and 0.39? 0.03; after 5-day-exposure those were 0.55 ? 0.12, 0.42?0.16, 0.29? 0.10, 0.19 ? 0. 4 and 0.10?0.02; after 7 day exposure those were 0.40? 0.12, 0.31 ? 0.10, 0.26? 0.06, 0.13? 0.02 and 0.09? 0.02, respectively. Conclusion: Nicotine inhibited the alkaline activity of human dental papilla mesenchymal cells in a dose and time dependant manner. Nicotine may play a role on the differentiation of the cells.
8.Application of micro transesophageal echocardiography (micro-TEE) in neonat and infant cardiac surgery

Objective To evaluate the value of phased array probe of transesophageal echocardiography miniature (micro-TEE) in the neonatal and infant cardiac surgery intraoperation.Methods Micro-TEE probe was used in children with weight ≤5 kg undergoing cardiac surgery to detect the atrial and ventricular cavity size,the left and right ventricular outflow tract diameter were detected by transthoracic echocardiography(TTE) 3-7 d after operation,and compared with the measured value immediately after the TEE.Results Before and after intubation,airway index not statistically significant (P > 0.05).Preoperative TEE and TTE inspection results were consistent with intraoperative diagnosis.Postoperative TEE measuring intracardiac structural data (each room,chamber size and left and right ventricular outflow tract diameter) and postoperative TTE measurement value representing the difference was not statistically significant (P > 0.05).Conclusions Micro TEE intraoperative guardianship of low birth weight infants is safe and feasible,and can supplement and improve the preoperative diagnosis,and can accurately and timely evaluate postoperative curative effect.
9.The express and research of chemerin and CRP in 45 patients with early-onset preeclampsia

Objective The purpose of this study was to compare chemerin and CRP levels between women with and without ear‐ly‐onset preeclampsia based on maternal body mass index (BMI) .Methods 45 patients with early onset preeclampsia (gestation week is less than 34 weeks) ,30 normal pregnancy women were enrolled in the study .The serums levels of chemerin and CRP were measured in these pregnancy women .Results The chemerin [(379 .18 ± 52 .09)ng/L ,(481 .27 ± 63 .12)ng/L] and CRP [(7 .59 ± 1 .41)mg/L ,(11 .74 ± 1 .61)mg/L] levels in MPE and SPE were significantly higher than those [(316 .08 ± 51 .90)ng/L ,(3 .47 ± 1 .27)mg/L]in normal pregnancy women(P<0 .05) .While the chemerin(424 .64 ± 46 .61)ng/L and CRP (8 .32 ± 3 .82)mg/L levels increased with maternal BMI in patients overweight and obese women levels compared with normal weight women [(331 .66 ± 51 .77)ng/L ,(5 .93 ± 2 .95)mg/L] ,the difference are significant(P<0 .05) .The correlation analysis showed that the chemerin were positively correlated with BMI ,systolic blood pressure ,diastolic blood pressure ,triglycerides ,and CRP were positively correlated with BMI .Conclusion The expression of chemerin and CRP were associated with severity of preeclampsia and BMI .
